A Super-Absorbent Polymer Combination Promotes Bacterial Aggressiveness Uncoupled from the Epiphytic Population

  • Plant leaf surface is an important niche for diverse epiphytic microbes, including bacteria and fungi. Plant leaf surface plays a critical frontline defense against pathogen infections. The objective of our study was to evaluate the effectiveness of a starch-based super-absorbent polymer(SAP) combination, which enhances water potential and nutrient availability to plant leaves. We evaluated the effect of SAP on the maintenance of bacterial populations. In order to monitor bacterial populations in situ, a SAP mixture containing Pseudomonas syringae pv. tabaci that expressed recombinant green fluorescent protein(GFPuv) was spray-challenged onto whole leaves of Nicotiana benthamiana. The SAP combination treatment enhanced bacterial robustness, as indicated by disease severity and incidence. Unexpectedly, bacterial numbers were not significantly different between leaves treated with the SAP combination and those treated with water alone. Furthermore, young leaves treated with the SAP combination had more severe symptoms and a greater number of bacterial spots caused by primary and secondary infections compared to young leaves treated with the water control. In contrast, bacterial cell numbers did not statistically differ between the two groups, which indicated that measurement of viable GFP-based bacterial spots may provide a more sensitive methodology for assessing virulence of bacterial pathogens than methods that require dilution plating following maceration of bacterial-inoculated leaf tissue. Our study suggests that the SAP combination successfully increased bacterial aggressiveness, which could either be used to promote the ability of biological agents to control weedy plants or increase the robustness of saprophytic epiphytes against competition from potentially harmful microbes.

  • Pulmonary thromboembolism (PTE) increases the pressure of the right ventricle and leads to symptoms and signs, such as dyspnea and hypoxia. If PTE causes hemodynamic instability, thrombolytic therapy should be considered. A mechanical thrombectomy is an alternative treatment to thrombolytic therapy and should be considered when thrombolytic therapy is contraindicated. Various devices are used in mechanical maceration and catheter-directed thrombolysis, but there is no standard mechanical device for PTE as yet. We report here on 2 clinical experiences of mechanical thrombectomy using the Arrow-Trerotola percutaneous thrombolytic device to remove residual clots after systemic thrombolysis in patients with massive PTE.

Anti-Proliferative Activity and Apoptosis Induction of an Ethanolic Extract of Boesenbergia pandurata (Roxb.) Schlecht. against HeLa and Vero Cell Lines

  • Rhizomes of Boesenbergia pandurata (Roxb.) Schlecht have been reported to contain active compounds with anticancer properties. This research was carried out to examine anti-proliferative and apoptotic induction against HeLa and Vero cells-line. Dried powder of B. pandurata rhizomes was extracted by a maceration method using 90% ethanol. Cytotoxic assays to determine $IC_{50}$ and anti-proliferative effects were carried out by MTT methods. Observation of apoptosis was achieved with double staining using acridine orange and ethidium bromide. The results showed that ethanolic extract of B. pandurata was more cytotoxic against HeLa cells ($IC_{50}$ of $60{\mu}g/mL$) than Vero cells ($IC_{50}$ of $125{\mu}g/mL$). The extract had higher anti-proliferative activity as well as apoptotic induction in HeLa than Vero cells. Therefore, it was concluded that the ethanolic extract of B. pandurata had anti-proliferative as well as apoptosis induction activity dependent on the cell type.

Antioxidant and Antimicrobial Activities of Various Solvent Fractions of Fine Ginseng Root

  • This study was carried out to investigate the changes of yield, total phenolics, saponin content and composition, antimicrobial, and antioxidant activities of various fractions of fine ginseng root (Panax ginseng C.A. Mayer) by maceration method in the order of increasing polarity (hexane, chloroform, ethyl acetate, butanol, and water). Butanol fraction showed the highest total saponin content compare to other fractions. Hexane fraction could harvest significantly high ginsenoside Rg2, Rg1, and Rf (p<0.05). And the contents of ginsenoside Rh1, Rg3, and Rg1 showed relatively higher in the fraction of ethyl acetate than other fractions. The system of hexane-chloroform-ethyl aceate-butanol showed relatively high content of ginsenoside Re, Rd, Rc, Rb3, and Rb1. However, the last fraction of water still remained lots of Rb2 content. The fraction of water was the highest phenolics. The 1,1-diphenyl-2-picryhydrazil, superoxide, and hydroxyl radical scavenging activity of water fraction was higher than the other fractions. In antimicrobial activity, the fraction of hexane showed relatively high antimicrobial activity against Pseudomonas aeruginosa, Salmonella typhimurium, Staphylococcus aureus, Bacillus cereus, and Escherichia coli. And the fractions of the chloroform and ethyl acetate showed higher antimicrobial activities than the other samples in against P. aeruginosa and S. typhimurium.

아보카도 씨와 씨 껍질의 항산화 효과 (Antioxidant Effects of Avocado Seeds and Seed Husks as a Potential Natural Preservative)

  • As the consumption of avocado fruits and avocado oils is steadily increasing, the amount of avocado seeds which are thrown away as by-products is also inceasing. Thus, the possibility of use of avocado seeds as natural preservatives was studied focused on the antioxidant effect. The extraction of avocado seeds and seed husks with 100% ethanol by maceration showed highest antioxidant activities and lowest IC50 values compared to 80% ethanol extract. Furthermore, 100% ethanol extract of avocado seeds and seed husks included significantly higher amount of polyphenols than 80% extract. However, total flavonoid content of 100% avocado seed extract was not signigicantly different from 80% seed extract, whereas that of 100% avocado seed husk extract was significantly higher than 80% seed husk extract. In case of acid values, heating of oil alone for 120 and 180 min significantly increased the acid values, whereas the treatment of oil with seed and seed husk extract signficantly decreased the acid values. These results suggest that antioxidant effects of avocado seeds and seed husks protected the oil against heat-induced acidification. Thus, avocado seeds and seed husks have a potential to be developed as an natural antioxidant and natural preservative which could be used commercially.

Effect of Curcuma xanthorrhiza Gel on Methicillin-Resistant Staphylococcus aureus-Infected Second-Degree Burn Wound in Rats

  • Methicillin-resistant Staphylococcus aureus (MRSA) infection often complicates burn wounds. Mupirocin is the antibiotic of choice for superficial MRSA infection, and its resistance is on the rise due to its frequent and widespread use. This study aimed to develop and evaluate Curcuma xanthorriza extract (CXE)-containing gel as a topical agent against MRSA-infected second-degree burn wound in rats. CXE was obtained using maceration with 96% ethanol. Xanthorrhizol level, antibacterial, and antioxidant activity were evaluated using a standardized method. In vivo, the wound's healing and bacterial load were evaluated every three days, whereas the histopathology of the wound was examined on day 12 of treatment. One-Way ANOVA and Kruskal-Wallis test were used to analyze the data. In this study, 27.0% and 7.10% of the obtained CXE were xanthorrhizol and curcumin, respectively. Additionally, an IC50 of 64.27 ppm was shown in antioxidant activity measurement, and MIC against MRSA was 5 mg/ml. Treatment with CXE-containing gels showed a significant reduction in bacterial load and proliferation of connective tissue in a dose-dependent manner. In conclusion, CXE-containing gel showed a greater reduction of bacterial load and more advanced wound healing phase than mupirocin.

Fiber Quality of Seven Mangrove Wood Species

  • As an archipelagic country, Indonesia is surrounded by large and small islands. Many mangrove plant species are found along the coasts surrounding large and small islands. Besides their conservation value, mangrove plants provide various benefits, including the use of their leaves, fruit, bark, and wood as raw materials in pharmaceutical and other industries. Additionally, mangrove wood is a potential raw material for pulp and paper production. It is essential to study the fiber dimensions of the wood to identify the appropriate characteristics of raw material for pulp and paper. Therefore, in this study, we comprehensively analyzed the differences in the wood fiber dimensions of seven mangrove species from the Indramayu Regency, West Java Province, namely Avicennia alba, Bruguiera gymnorhiza, Bruguiera cylindrica, Hibiscus tiliaceus, Sonneratia ovata, Sonneratia caseolaris, and Excoecaria agallocha. For this analysis, maceration followed the Forest Product Laboratory guidelines, and preparation followed the Sass method. The fiber length, diameter, and lumen diameter were measured using a light microscope. Based on the values of the length and dimension derivatives, the fibers of these mangrove wood species were grouped into quality classes II or III for use as raw materials for the pulp and paper industry. The wood fibers of H. tiliaceus, A. alba, S. caseolaris, and E. agallocha met the quality class criterion II, whereas wood species fibers of B. gymnorhiza, B. cylindrica, and S. ovata met the quality class criterion III.

Antioxidant and Free Radical Scavenging Potential of Justicia gendarussa Burm. Leaves in vitro.

  • Antioxidant activity of 70% aqueous ethanolic extract of leaves of Justicia gendarussa (EJ) was evaluated. EJ was prepared by cold maceration method. The antioxidant potency of EJ was investigated employing various established in vitro systems, such as DPPH radical scavenging, nitric oxide (NO) scavenging, ${\beta}-carotene$ linoleic acid module system (${\beta}$ CLAMS), hydroxyl (OH) radical scavenging, anti lipid peroxidation. $IC_{50}$ values were determined in each experiment. Also, ferric ion reduction capacity of extracts in presence and absence of chelating agent (EDTA) and total antioxidant capacity were determined. Preliminary phytochemical investigation was carried out to know the nature of constituents present in the leaves and correlate it with antioxidant activity. Further total phenolic content was determined in EJ. $IC_{50}$ values of EJ were 123.09 ${\pm}$ 3.01, 643.0 ${\pm}$ 61.10, 132.3 ${\pm}$ 6.03, 68.5 ${\pm}$ 11.5 and 68.13 ${\pm}$ 1.38 ${\mu}g/mL$ in DPPH radical scavenging, NO scavenging, ${\beta}$ CLAMS, OH radical scavenging and anti lipid peroxidation activity respectively. In total antioxidant capacity assay, ascorbic acid equivalent value was found to be 205.56 ${\pm}$ 4.69 ${\mu}g/mg$ of extract. Total phenolic content was found to be 43.76 ${\pm}$ 4.27 ${\mu}g$ equivalent of gallic acid per mg of extract. Phytochemical investigation reveals the presence of flavonoids. The results indicate that EJ possess antioxidant activity and flavonoids are responsible for this activity.

잔대종자 발아촉진에 관한 연구 (Studies on the Promoting of Seed Germination of Adenophora triphylla var. Japanica $H_{ARA}$)

  • 본 연구는 잔대 종자를 재료로 하여 생장조절제(生長調節劑), 저온처리(低溫處理) 기간(期間), 발아온도(發芽溫度) 등을 처리하여 발아율 향상 및 발아촉진(發芽促進) 방법을 구명(究明)코자 1995년 수행하였고 결과를 요약하면 다음과 같다. 1. 종자 발아율은 무처리 22%에 비하여 BA $10mg/\iota$에서 53%, $GA_3\;500mg/\iota$ 24시간 침지 처리 하였을 때 94%로 발아율이 양호하였으며 평균 발아일수는 5일로 무처리보다 1.5일 빨랐다. 2. $KNO_3\;1g/\iota$에서 54%의 발아율을 보였으나 KOH 처리 효과를 인정할 수 없었다. 3. 휴면타파를 위한 저온처리($1^{\circ}C$) 기간은 2주 이상에서 86% 이상의 발아율을 보였다. 4. 발아촉진을 위한 최적 온도조건은 $25^{\circ}C$였다.

  • 이태리포푸라 I-214 (Populus euramericana cv. I-214)의 기내배양(器內培養)한 엽육조직(葉肉組織)에서 원형질체(原形質體) 분리(分離)에 미치는 몇가지 요인(要因)에 대(對)해 조사(調査), 검토(檢討)하였다. 기내(器內)에서 배양(培養)된 아(芽)를 다량(多量)으로 증식(增植)하기 위한 배지(培地)는 MS 기본배지(基本培地)에 $0.1mg/{\ell}$의 BAP를 첨가(添加)한 것이 가상 좋은 성적을 나타냈다. 엽(葉) 1g 당(當) $2.4{\times}10^6$개의 가장 높은 원형질체(原形質體) 분리(分離) 빈도를 나타낸 것은 Cellulase R-10 2 %, Macerozyme R-10 0.8 %, Hemicellulase 1.2 %, Driselase 2.0 %, Pectolyase Y-23 0.05 %에 DTT와 MES 완충액을 첨가(添加)한 후 삼투압 안정제로 0.6 M의 Mannitol을 넣고 pH를 5.6으로 조정한 효소용액(酵素溶液)이었다. CPW 용액(溶液)으로 세정(洗淨)한 후 0.6 M의 Sucrose 용액(溶液)에 처리(處理)한 것이 회수율(回收率) 51.8 %로 가장 높게 나타났다.
