항산화제와 Growth Factor 첨가배양이 소 체외수정란의 체외발육에 미치는 효과

  • 도살장에서 구입한 소의 난소로부터 회수한 미성숙 난포란을 체외에서 성숙, 수정시킨 후, 2~8세포기 수정란을 CR$_1$aa 체외배양액에 일정량의 aesculetin, taurine을 첨가하여 체외배양을 실시한 두 처리구간의 항산화 효과를 비교 검토하고, aesculetin과 taurine에 growth factor(EGF, PDGF)를 첨가배양하여 체외수정란의 체외발육에 항산화제와 growth factor의 상호작용 효과를 검토하였다. 대조구, aesculetin(1$\mu\textrm{g}$/$m\ell$) 및 taurine(2.5mM)을 첨가하여 체외수정란을 체외배양시킨 결과 상실배이상 발육된 체외발육율은 각각 46.5%, 64.3% 및 60.5%로서 대조구보다 aesculetin과 taurine 첨가구가 유의하에 높은 성적을 얻어, aesculetin과 taurine이 체외수정란의 체외발육에 높은 효과를 나타냈다. 대조구, aesculetin (1$\mu\textrm{g}$/$m\ell$) + PDGF (1ng/$m\ell$) 첨가구, aesculetin (l$\mu\textrm{g}$/$m\ell$) + EGF (10ng/$m\ell$)첨가구, taurine (2.5mM) +PDGF (1ng/$m\ell$) 첨가구 및 taurine (2.5mM)+EGF (10ng/$m\ell$) 첨가구에서 배반포기 발육율은 각각 46.0%, 70.0%, 58.0%, 56.0 및 66.0%로써 처리구가 무첨가구에 비해 유의하게 높은 성적을 얻어(P<0.05), 항산화제와 growth factor의 첨가 배양이 체외수정란의 체외발육에 상승효과가 있음이 입증되었다. 배양액에 천연추출 aesculetin과 상품화된 aesculetin을 첨가하여 체외배양을 실시한 결과 배반포기 발육율은 대조구, 천연추출 aesculetin 및 상품화된 aesculetin 첨가구에서 각각 38.6%, 54.6% 및 53.5%로써 첨가구가 무첨가구보다 높은 성적을 얻어, aesculetin의 항산화 효과가 입증되었다(P<0.05). 그러나 모든 처리구에서 배반포까지 발육된 체외수정란의 세포수에는 커다란 차이가 인정되지 않았다.

Effect of Synthetic CaM and NFAT Oligodeoxynucleotide on MPP+-Stimulated Mesencephalic Neurons

  • Background: Ca2+ signaling plays a vital role in neuronal signaling and altered Ca2+ homeostasis in Parkinson's disease (PD). Overexpression of αSYN significantly promote the Ca2+-Calmodulin (CaM) activity and subsequent nuclear translocation of nuclear factor of activated T cells (NFAT) transcription factor in dopaminergic neurons of midbrain. However, the exact role of Ca2+-CaM and NFAT in PD pathology is yet to be elucidated. Methods: We designed the CaM-NFAT-oligodeoxynucleotide (ODN), a synthetic short DNA containing complementary sequence for NFAT transcription factor and CaM mRNA. Then, the effect of CaM-NFAT-ODN on 1-methyl-4-phenylpyridinium (MPP+)-mediated neurotoxicity was investigated in mimic PD model in vitro. Results: First, the expression of αSYN and CaM was strongly increased in substantia nigra (SN) of PD and the expression of tyrosine hydroxylase (TH) was strongly increased in control SN. Additionally, the expression of apoptosis marker proteins was strongly increased in SN of PD. Transfection of CaM-NFAT-ODN repressed CaM and pNFAT, the target genes of this ODN in rat embryo primary mesencephalic neurons. It also reduced ERK phosphorylation, a downstream target of these genes. These results demonstrated that CaM-NFAT-ODN operated successfully in rat embryo primary mesencephalic neurons. Transfection of CaM-NFAT-ODN repressed TH reduction, αSYN accumulation, and apoptosis by MPP+-induced neurotoxicity response through Ca2+ signaling and mitogen-activated protein kinases (MAPK) signaling. Conclusion: Synthetic CaM-NFAT-ODN has substantial therapeutic feasibility for the treatment of neurodegenerative diseases.

Effects of Flos Lonicerae Japonicae Water Extract on Cytokine Production in RAW 264.7 Mouse Macrophages (금은화(金銀花)물추출물이 마우스 대식세포의 사이토카인 생성에 미치는 영향)

  • Flos Lonicerae Japonicae (the flower buds of Lonicera japonica Thunberg) has been used as an antibacterial and antiviral drug in Korean Medicine. The aim of this study is to evaluate the effect of Flos Lonicerae Japonicae water extract (FL) on the production of cytokines in RAW 264.7 mouse macrophages stimulated by lipopolysaccharide (LPS). After 24 h treatment, the production of various cytokines from RAW 264.7 was measured with multiplex cytokine assay using Bio-Plex 200 suspension array system. FL at concentrations of 50, 100, and 200 ㎍/mL significantly inhibited productions of tumor necrosis factor-α, macrophage inflammatory protein (MIP)-1β, and MIP-2 in LPS-stimulated RAW 264.7 cells; FL at concentrations of 100 and 200 ㎍/mL significantly inhibited productions of leukemia inhibitory factor, LIX (CXCL5), and RANTES in LPS-stimulated RAW 264.7 cells; FL at concentrations of 200 ㎍/mL significantly inhibited productions of granulocyte-macrophage colony-stimulating factor and macrophage colony-stimulating factor in LPS-stimulated RAW 264.7 cells; FL at concentrations of 50 and 100 ㎍/mL significantly increased productions of interleukin (IL)-10 in LPS-stimulated RAW 264.7 cells; FL at concentrations of 50, 100, and 200 ㎍/mL significantly increased productions of IL-6 and interferon gamma-induced protein-10 in LPS-stimulated RAW 264.7 cells; FL at concentrations of 100 and 200 ㎍/mL significantly increased productions of monocyte chemoattractant protein-1 in LPS-stimulated RAW 264.7 cells. Taken together, these data mean that FL might modulate productions of cytokines, chemokines, and growth factor in LPS-stimulated macrophages. Further study needs to verify the exact mechanism for modulatory activities of FL with macrophages.

Role of Endothelium-derived Relaxing Factor in Cerebral Autoregulation in vivo (뇌혈류 자가조절에 대한 Endothelium-derived Relaxing Factor의 역할)

  • In anesthetized rats, we examined the possibility that endothelium-derived relaxing factor (EDRF) or nitric oxide (NO) released in response to cholinergic mechanism may contribute to the reflex autoregulation of cerebral blood flow. Suffusion with mock cerebrospinal fluid (CSF), containing acetylcholine (ACh, $10^{-9}{\sim}10^{-6}M$) evoked concentration-dependent vasodilatation of the resting pial artery (mean, $19.3{\pm}1.7{\mu}m$, n=36), which was significantly inhibited not only by $N{\omega}$-nitro-L-arginine (L-NNA, $10^{-5}M$) but also by methylene blue ($10^{-6}M$) and oxyhemoglobin ($10^{-6}M$). The muscarinic receptors in the endothelium of pial artery implicated in the release of EDRF were considered to be $M_1\;and\;M_3$ subtypes. When suffused with mock CSF containing L-arginine it caused a transient vasodilatation, which was strongly inhibited by LY 83583 ($10^{-5}M$), but not by L-NNA ($10^{-5}M$). Additionally, both ACh- and L-arginine-induced vasodilation were significantly inhibited by glibenclamide, a specific ATP-sensitive $K^+$ channel blocker. On the other hand, changes in pial arterial diameter were plotted as a function of changes in systemic arterial blood pressure. The slopes of regression lines for vasodilation and vasoconstriction were not affected by pretreatment with $10^{-5}M$ L-NNA, but significantly reduced by $3{\times}10^{-6}M$ glibenclamide. Thus it is suggested that the reflex vasodilation of rat pial arteries in response to a transient hypotension is not mediated by EDRF (NO).

A Study of Electro-optical Characteristics of Full-HD LCOS Panel Depending on Various Cell Gaps (Full-HD LCOS Panel의 Cell Gap의 변화에 따른 전기광학적 특성에 대한 연구)

  • The electro-optical characteristics of the Liquid Crystal on Silicon (hereinafter "LCOS") micro-display on vertically alignment (VA) mode were studied with 3-dimensional LC code. 5 different cell gaps, such as 1.4 ${\mu}m$, 1.8 ${\mu}m$, 2.1 ${\mu}m$, 2.4 ${\mu}m$ and 2.8 ${\mu}m$, were selected. The reflectance-voltage (R-V) characteristics, distribution of reflected light, reflectance, optical fill factor and contrast ratio were calculated and investigated depending on various cell gaps. Due to the surface anchoring effect, higher cell gap showed higher reflectance. However, considering the optical fill factor and contrast ratio, middle-height 2.1 ${\mu}m$ showed the best electro-optical characteristic. 0.7 inch Full-HD LCOS panels having same geometry and material property were fabricated. The reflected light intensity and contrast ratio were measured and the measured results were well-matched to the calculated results.

Removal and Inactivation of Hepatitis A Virus during Manufacture of a High Purity Antihemophilic Factor VIII Concentrate from Human Plasma

  • A validation study was conducted to evaluate the efficacy and mechanism of the cryo-precipitation, monoclonal anti-FVIIIc antibody (mAb) chromatography, Q-Sepharose chromatography, and lyophilization steps involved in the manufacture of high purity factor VIII (GreenMono) from human plasma, in the removal and/or inactivation of hepatitis A virus (HAV). Samples from the relevant stages of the production process were spiked with HAV and subjected to scale-down processes mimicking the manufacture of the high purity factor VIII concentrate. Samples were collected at each step and immediately titrated using a 50% tissue culture infectious dose (TCID$\_$50/) and then the virus reduction factors were evaluated. HAV was effectively partitioned from factor VⅢ during cryo-precipitation with the log reduction factor of 3.2. The mAb chromatography was the most effective step far removal of HAV with the log reduction factor of $\geq$4.3. HAV infectivity was not detected in the fraction of factor VⅢ, while most of HAV infectivity was recovered in the fractions of flow through and wash during mAb chromatography. Q-Sepharose chromatography showed the lowest efficacy for partitioning HAV with the log reduction factor of 0.7. Lyophilization was an effective step in inactivating HAV with the log reduction factor of 2.3. The cumulative lag reduction factor, $\geq$10.5, achieved for tile entire manufacturing process was several magnitudes greater than the potential HAV load of current plasma pools.

  • Magnetic shielding effect of cylinderical ahields made of commericial amorphous ribbons has been studied. The shell-arrangement-order of double shell shield has been found to show a striking differencein shielding factor. In low applied field region, a 2605CO-2705M-shield (outer shell: 2605CO, inner shell: 2705M) yields two times higher shielding factor than a 2705M-2605CO-shield (outer shell: 2705M, inner shell: 2605CO). The reasons are as follows: In case of 2605CO-2705M-shield, the outer shell is not easily saturated and effectively shields the applied field. In addition, the inner shell shows high shielding factor in the field shielded by the outer shell. In case of 2705M-2605CO-shield, the outer shell is saturated at very low-field as well as the inner shell shows low shielding factor in the field shielded by the outer shell.

  • Connes [2] showed that if M is an injective .sigma.-finite factor of type II $I_{1}$ and $B_{\phi}$=C1 for some normal faithful state .phi. on M then M is isomorphic to the Araki-wood factor. In [7], Haagerup has succeeded to show that if M is an injective factor of type II $I_{1}$ with separable predual, then $B_{\phi}$=C1 for every normal faithful state on M. Since injective factors of type II $I_{\lambda}$, 0.leq..lambda.<1, were classified [9], this together classifies all injective factors of type III with separable predual. It is not known for non-injective case. In this paper we consider some conditions under which the bicentralizers be trivial.ivial.

A Study of the relevant factors affecting the satisfaction of leisure activities among the dental technicians (일부 치과기공사의 여가만족도 및 관련요인에 대한 연구)

  • This study explored the status of free time activities among the dental technicians who are working at Seoul, Gyonggido, and Gangwondo. On the basis of this research, we aim to present needs and applicable solutions to improve the working conditions and enhance the quality of life through more effective time management in free time activities that lead to their self development and far more enriched life. 500 subjects were randomly chosen and the questionary sheets were provided. Among them, 305 sheets were returned to be analysed through SPSS Win 10.0 software and the results are as follows; First, mean in the satisfaction of leisure activities was 3.44(SD=.73). Mean in sub categories was as follows; psychological factor(M=3.65, SD=.71), social factor(M=3.54, SD=.75), rest factors(M=3.52, SD=.63), physical factor(M=3.43, SD=.88), other factors(M=3.40, SD=.66), circumstantial factor(M=3.33, SD=.63), educational factor(M=3.24, SD=.73). Second, the most relevant sub categories in the satisfaction of leisure activities were social, educational, physical, and other circumstantial factors by sequence. Factors that affect leisure activities satisfaction were career and status of health, which shows statistical significance(p<.001). That is to say, contention of this study is that the general characteristics are closely related with and affect the types and the level of satisfaction in leisure activities of the subjects. Considering these results, the dental businesses and the associations of dental technicians should recognize the needs to improve the time management through more efficient cooperations between dentists and back office dental team, systemized working time, and introduction of incentive system among others. On the basis of this appreciation, the dental business should provide more time and various opportunities with the dental technicians. Dental technicians also need to reconsider their appreciations of leisure activities, that is, leisure activities are not just spending time doing something but they refresh them and enable them to be more active and faithful to home and work.

Characteristics of Groundwater Quality by Elevation in Cheju Island (고도에 따른 제주도 지하수의 수질특성)

  • This study purpose to elucidate the characteristics of water quality by elevation and groundwater samples has been studied with the samples from 150 selected groundwater represented the watershed of groundwater wells in Cheju Island. The evaluation of the characteristics of water quality utilized the physical and chemical property and the statistical analysis. According to the piper diagram, groundwater in the under 50 m region is shown N $a^{+}$$K^{+}$-C $l^{[-10]}$ type, and that groundwater in the 50~100 m region is shown N $a^{+}$$K^{+}$-HC $O_3$$^{[-10]}$ +C $O_3$$^{2-}$ type. and partly N $a^{+}$$K^{+}$-C $l^{[-10]}$ type. In the above 100 m region belongs to N $a^{+}$$K^{+}$-HC $O_3$$^{[-10]}$ +C $O_3$$^{2-}$ type. The result of factor analysis, commonly two factors as TDS(Total Dissolved Solid) and the contaminants extracted in the under 50m region and above 100 m region. Three factors were obtained from the result of the factor analysis in the 50~100 m region. Factor 1, consisting of TRS content. Factor 2, consisting of the contaminant and the dissolution of minerals. and Factor 3, consisting of HC $O_3$$^{[-10]}$ content. content.
