구기자 추출액을 첨가한 양갱의 품질특성 및 묘사적 관능평가 (Quality Characteristics and Descriptive Analysis of Yanggaeng added with Lycii Fructus Extract)

  • The purpose of this study was to investigate the quality characteristics and descriptive analysis of Yanggaeng prepared with Lycii fructus extract (LD). LD were added in ratios (w/w) of 0 (C), 1.5 (LY1), 3.0 (LY2), 4.5 (LY3), and 6% (LY4), and then proximate compositions, physicochemical properties, and sensory evaluations of the Yanggaeng were measured LY1~LY4 samples showed higher contents of crude lipid, crude protein and crude ash as well as $^{\circ}Brix$ compared to control (p<0.001). pH and lightness (L) of samples decreased as the LD increased. With regard to the texture of Yanggaeng samples, the scores of hardness, adhesiveness, springness, and cohesiveness was significantly increased by the Addition of LD (p<0.05, p<0.01). For the descriptive analysis, ten panelist generated and evaluated 29 sensory attributes for the Yanggaeng, and there were significant differences among the samples for all 26 sensory attributes. For the descriptive data, principal component analysis (PCA) was performed to summarize the sensory characteristics of the Yanggaeng. The results of PCA showed that the positive attributes, e.g. savoury, were closely in relationship with LY2 and LY3. Form the findings, this study suggests that 3~4.5% addition of LD was effective for preparation of Yanggaeng in the aspects of the consumer acceptability.

구기자 추출물이 피부 미백 및 주름에 미치는 영향 (Effects of Lycii Fructus Extracts(LFE) on Skin whitening and Elasticity using Melanoma cells)

  • Objective : Lycii Fructus Extracts(LFE) can do Anti-hypertension activity, Antidepressant, Anti-diabetic activity. This study was designed to investigate effects of LFE on skin whitening and elasticity using melanoma cells. Methods : In this experiment, effect of LFE on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. Results : More than $250{\mu}g/ml$ of LFE treated group showed lowered proliferation rates significantly compared to non-treated group. More than $125{\mu}g/ml$ of LFE treated groups were lower levels of melanin synthesis respectively. LFE showed inhibitory effect on tyrosinase activities in vitro. And, LFE suppressed tyrosinase activities in B16F10 cells significantly. Finally, LFE suppressed elastase type I and IV activities in dose-dependent manner in vitro. And LFE also slightly suppressed elastase activities in vivo. Conclusion : These results suggest that LFE can inhibit melanin synthesis through ihhibitory action on tyrosinase activity and inhibt elastase activity, and also suggest that these results can be used for the study on maintaining skin elasticity or whitening.

수확시기에 따른 구기자 품종의 성분변화 (Changes in Composition of Gugija (Lycii Fructus) Species According to Harvest Time)

  • 시험장에서 육성한 품종인 명안, 불로, 청대, 장명, 청운과 육성중인 청양6호, 청양7호 및 청양재래의 가용성 무질소물, 조단백질, 조지질, 회분, 총 폴리페놀, 엑스, 베타인의 함량을 8, 9, 10 및 11월의 수확시기별로 조사한 결과, 가용성 무질소물 및 엑스는 육성중인 청양6호가 가장 높은 함량을 보였고, 조단백질, 총폴리페놀 및 베타인의 함량에서는 육성중인 청양 7호가 가장 높은 함량을 보였으며, 조지질은 청대에서, 조회분은 청운에서 가장 높았다. 건과비율은 육성중인 청양6호에서 가장 높았고, 건과비율이 높을수록 가용성 무질소물의 함량은 증가 (r = 0.70)하는 반면 조단백질 (r = -0.83) 및 조회분 (r = -0.71)등은 감소하였다. 일반성분 중에는 서로 일정한 상관을 보였는데, 가용성 무질소물의 함량이 증가하면 엑스의 함량 (r = 0.81)은 높아지는 반면, 단백질 (r = -0.88), 지질 (r = -0.82) 및 회분 함량 (r = -0.81)은 감소하고, 총폴리페놀 함량이 증가하면 베타인 함량이 증가 (r = 0.90)하는 경향이었다. 품종특성과 성분을 분석한 결과 육성중인 청양 6호는 가용성 무질소물 및 엑스분이 높게 관찰되었을 뿐만 아니라, 건과비율이 높아 타 품종에 비해 쉽게 건조될 수 있어 건조비용을 줄일 수 있는 것으로 예상되었고, 청양 7호는 조단백질, 총폴리페놀 및 베타인 함량이 높아 기능성 품종으로서의 가치를 보여주었다.

  • Objective : This study was conducted to observe the anti-hyperlipidemic and anti-inflammatory effects, as well as the metabolic benefits, of pear extract and herbal drug mixture (Lycii Fructus, Coicis Semen, Alimatis Rhizoma, and Astragali Radix) on rats fed with a high fat diet. Methods : The animals used were male rats and the control group was fed a high fat diet only. The experimental groups were divided into four. Exp I group was fed a high fat diet with a mixture of pear extract and 3% Lycii Fructus; Exp II group was fed a high fat diet with a mixture of pear extract and 3% Coicis Semen; Exp III group was fed a high fat diet with a mixture of pear extract and 3% Alimatis Rhizoma; and Exp IV group was fed a high fat diet with a mixture of pear extract and 3% Astragali Radix for 4 weeks. Results : The body weight gain increased in all groups, but attenuated gradually in the experimental groups compared to the control group. The food intakes were significantly lower in Exp I and Exp III groups than the control group. The concentrations of serum total cholesterol (TC), HDL-cholesterol, LDL-cholesterol, and triglyceride (TG) were significantly higher in Exp II than in the control group, and lower in Exp III group than in the control group. Also the concentration of serum free fatty acid was significantly lower in the Exp III group than in the control group. In inflammatory activities, the Exp II group was significantly lower than the control group. Conclusion : The results indicated that Exp III group (administered a mixture of pear extract and Alimatis Rhizoma) most efficiently reduced fat accumulation and body weight, while the Exp II group (administered the mixture of pear extract and Coicis Semen) had the highest elevated lipid metabolism and immune activity.

구기자 분말이 첨가된 유과의 품질특성 및 항산화효과 (Quality Characteristics and Antioxidative Effect of Yukwa Prepared with Lycii fructus Powder)

  • 유과의 저장성 향상과 기능성 효과가 있는 소재를 개발하기 위하여 구기자 분말(Lycii fructus powder)을 첨가한 유과를 제조하여 $50^{\circ}C$에서 40일 동안 저장하면서 유과의 저장과정 중 항산화효과와 품질특성을 조사한 결과는 다음과 같다. 구기자 첨가 유과 중 구기자 5% 첨가 유과의 밀도와 부피가 가장 높게 나타났고 대조군에 비해 구기자 첨가 유과의 퍼짐성과 수분함량이 더 많이 증가하였다. 색도에서 구기자 첨가량이 많을수록 L값은 낮게, a값과 b값은 높게 나타났으며 조직감은 구기자 함량이 증가할수록 높게 나타났다. 구기자 유과의 관능평가에서는 유과의 색, 맛, 풍미, 조직감에서 구기자 5% 첨가 유과가 가장 높은 점수를 보였으며 대조군이 전반적인 기호도에서는 가장 낮은 점수를 나타냈다. 저장 기간 중 구기자 유과의 산가를 살펴보면 저장기간이 경과함에 따라 모든 실험군에서 유의적으로 증가하였고 대조군보다 구기자 첨가군에서 훨씬 낮았으며 구기자 5%첨가 유과에서 가장 낮은 산가를 나타냈다. 과산화물가는 저장 20일까지는 약간씩 증가하다가 그 후 30일까지는 급격히 증가하다가 감소하는 경향을 보였으며 과산화물가 역시 구기자 5% 첨가 유과에서 가장 낮은 과산화물가를 나타냈다. TBA가는 대조군에 비해 구기자 첨가군이 낮은 값을 나타내 지방의 산화를 지연시켰음을 알 수 있었으며, 항산화 활성이 뛰어난 것을 확인할 수 있었다. 이와 같은 결과로부터 구기자 분말을 유과에 첨가 제조 시 저장성 향상과 기능성 효과를 갖는 소재로써의 가능성이 확인되었다.

구기자 및 구기엽 추출물의 피부과색소 조절효과 (Moderating Effects of Skin Hyperpigmentation from Lycii fructus and Lycii folium Extracts)

  • 구기자, 건조구기엽, 생구기엽 추출물의 미백효과를 검증하였다. Tyrosinase 저해활성 측정결과 건조구기엽 에탄올추출물 $500{\mu}g/mL$에서 44%의 저해활성을 나타내었으며, melanoma cell (B16F10)에서의 melanin 생합성 저해율 측정결과 건조구기 엽 에탄올 추출물 $50{\mu}g/mL$에서 14%의 저해효과를 나타내었다. 구기자, 건조구기엽, 생구기엽 추출물을 처리한 B16F10군에서는 tyrosinase protein의 발현이 처리하지 않은 군보다 감소하였다. 특히, 건조구기엽 추출물의 경우 tyrosinase의 단백질 발현량을 많이 저해하였다. 결과적으로 건조구기엽 추출물의 미백활성이 가장 우수하였으며, 식품 및 화장품의 기능성 소재로 이용이 가능할 것으로 판단된다.

비알코올성 지방간 세포 모델에 대한 택사, 산사, 구기자, 울금, 단삼, 인진의 효능 비교 (Comparison of the Therapeutic Efficacy of Rhizoma Alismatis, Fructus Crataegi, Fructus Lycii, Radix Curcumae, Radix Salviae Miltiorrhizae, Herba Artemisiae Scopariae on the Experimental Cellular Model of Nonalcoholic Fatty Liver Disease)

  • Objectives : We try to compared the efficacy of six herbal medicines, Rhizoma Alismatis (RA), Fructus Crataegi (FC), Fructus Lycii (FL), Radix Curcumae (RC), Radix Salviae Miltiorrhizae (RSM), and Herba Artemisiae Scopariae (HAS), constituting KHchunggan-tang which was previously proven to be hepatoprotective on non-alcoholic fatty liver disease with combined properties of cellular steatosis, ROS production, and cytoprotection. Methods : HepG2 cells were pretreated with aqueous extracts of the six herb medicines at concentrations of 1, 10, 50 and 100 ${\mu}g/ml$ each, and treated with 0.5 mM palmitate consecutively. After 21 hrs, cell viability was assessed using MTT assay, and the percentage of cells with sub-G1 DNA content was measured using fluorescence-activated cell sorting after propidium iodide staining. Results : The first three extracts, RA, FC, and FL restored cell viability reduced by palmitate in MTT assay, and RA, FC, FL and RC inhibited palmitate-induced apoptosis in sub-G1 analysis. FL showed relatively weak potential only at tested maximal dose, and RA showed the greatest higher efficacy on this experimental cellular model of nonalcoholic fatty liver disease. Conclusions : According to this comparative experiment, Rhizoma Alismatis seems to have the most powerful potential among the six herbs constituting KHchunggan-tang, and consecutive further study seems to be required for more standardized and effective clinical application of KHchunggan-tang for treatment of non-alcoholic fatty liver disease.

사육탕가미방(四六湯加味方)이 streptozotocin으로 유발(誘發)된 백서(白鼠)의 실험적(實驗的) 탕뇨(糖尿)에 미치는 영향 (Effect of Saryuktangkamibang on Streptozotocin-induced Diabetic Rat)

  • Objectives : The objective of this study is to observe the effect of Saryuktangkamibang, Saryuktang with Liriopis Tuber, Lycii Fructus, Schizandrae Fructus, and Bombyx Batryticatus, on the experimental diabetes. Methods : In order to induce diabetes experimentally, injected streptozotocin to the vein in the tail of rats and then treated oral administration of Saryuktangkamibang water extracts. In the measurement of the variation levels of glucose, ALP, creatinine, and BUN concentration for each concentration levels for serum (0.2g/100g, 0.4g/l00g, 0.6g/100g), concentration level of glucose significantly decreased in 0.4g/l00g concentration level of Saryuktangkamibang. With this 0.4g/l00g concentration level of Saryuktangkamibang, the following conclusion was derived from the measurement of the serum levels of glucose. ALP, AST, ALT, creatinine, and BUN concentration for the injection of each 4th, 11th, and 18th day. Results : 1. In the measurement for each concentrations, the glucose concentration level in the serum significantly decreased on the 9th day in the Saryuktangkamibang-dosed group. 2. The creatinine and BUN concentration level in the serum significantly decreased on the 9th day in the Saryuktangkamibang-dosed group to which 0.6g/100g injected. 3. In the measurement for each dosage dates, the glucose concentration level in the serum significantly decreased on the 11th day in the Saryuktangkamibang-dosed group to which 0.6g/100g injected. 4. The AST concentration level in the serum significantly decreased on the 11th day in the Saryuktangkamibang-dosed group. Conclusion : Saryuktangkamibang, Saruktang with Liriopis Tuber, Lycii Fructus, Schizandrae Fructus, and Bombyx Batryticatus, has an effect of hypoglycemia, with no hepato-renal dysfunction. For the experimental results above, in that nephrotoxicity significantly decreased on the 0.6g/100g concentration, another research such as study for effect in other ranges and study for effect of dosage term except the concentration range of this study is necessary. Also dosage of appropriate concentration of Saryuktangkamibang is thought to provide valuable aid to cure on diabetes.

진도산(珍島産) 구기자(枸杞子)의 아미노산조성(組成)과 유리당(遊離糖)의 분석(分析) (Quantitative Analysis of Total Aimno Acids and Free Sugars in Lycii fructus)

  • 구기자(枸杞子)의 총(總)아미노산조성(組成) 및 유리(遊離)당을 각각 amino acid autoanalyzer, HPLC로 분석(分析)한 결과(結果)는 다음과 같다. 1. 17종(種)의 총(總)아미노산이 검출(檢出)되었다. 즉 asparatic acid, threonine, serine, glutamic acid, proline, glycine, alanine, cystine, valine, methionine, isoleucine, leucine, tyrosine, phenr-lalanine, hisidtine, lysine, arginine인데 이중 threonine을 가장 많이 함유하였으며 aspartic acid는 가장 적었다. 2. 총(總) essential amino acid량은 총(總)아미노산량의53.93%로 양질(良質)의 아미노산을 함유하였다. 3. Glucose, fructose, saccharose 등 3종(種)의 유리(遊離)당이 검출(檢出)되었는데 이중 fructose를 가장 많이 함유하였다.

에탄올 음용으로 유도된 발기부전 동물모델에 대한 구기자 추출물의 개선효과 (Effects of Lycii fructus Extracts on the Erectile Dysfunction by Chronic Ethanol Consumption in Rats)

  • Erectile dysfunction (ED) is a highly prevalent disorder that affects millions of men worldwide. ED is now considered an early manifestation of atherosclerosis, and consequently, a precursor of systemic vascular disease. Lycii fructus extracts (LFE) were administered for 4 weeks to assess the improving effects on ED. Animals were divided into one normal group and four LFE-treated groups (0, 0.3, 0.6, and 1.2 g/kg). We induced ED in the study animals by oral administration of 20% ethanol instead of water everyday for 4 weeks. This study was designed to investigate the effects of LFE on the mRNA levels of inducible nitric oxide synthase (iNOS) and endothelial NOS (eNOS) expression; NO levels of nitric oxide (NO) and cyclic guanosine monophosphate (cGMP); blood profile; and erectile response of the corpus cavernosum of the rat penis. The libido of the LFE-administered male rats was higher than that of the ethanol control group. The erectile response of the corpus cavernosum was restored after LFE administration, to a level similar to the normal group. In addition, the iNOS in the corpus cavernosum of the male rats administered LFE decreased. In contrast, compared to the control group, LFE-administered male rats showed increased eNOS, NO and cGMP levels in the corpus cavernosum. These results indicate that LFE effectively restored ethanol-induced ED in male rats.
