Effects of the Particulate Matter2.5 (PM2.5) on Lipoprotein Metabolism, Uptake and Degradation, and Embryo Toxicity

  • Particulate $matter_{2.5}$ ($PM_{2.5}$) is notorious for its strong toxic effects on the cardiovascular, skin, nervous, and reproduction systems. However, the molecular mechanism by which $PM_{2.5}$ aggravates disease progression is poorly understood, especially in a water-soluble state. In the current study, we investigated the putative physiological effects of aqueous $PM_{2.5}$ solution on lipoprotein metabolism. Collected $PM_{2.5}$ from Seoul, Korea was dissolved in water, and the water extract (final 3 and 30 ppm) was treated to human serum lipoproteins, macrophages, and dermal cells. $PM_{2.5}$ extract resulted in degradation and aggregation of high-density lipoprotein (HDL) as well as low-density lipoprotein (LDL); apoA-I in HDL aggregated and apo-B in LDL disappeared. $PM_{2.5}$ treatment (final 30 ppm) also induced cellular uptake of oxidized LDL (oxLDL) into macrophages, especially in the presence of fructose (final 50 mM). Uptake of oxLDL along with production of reactive oxygen species was accelerated by $PM_{2.5}$ solution in a dose-dependent manner. Further, $PM_{2.5}$ solution caused cellular senescence in human dermal fibroblast cells. Microinjection of $PM_{2.5}$ solution into zebrafish embryos induced severe mortality accompanied by impairment of skeletal development. In conclusion, water extract of $PM_{2.5}$ induced oxidative stress as a precursor to cardiovascular toxicity, skin cell senescence, and embryonic toxicity via aggregation and proteolytic degradation of serum lipoproteins.

도지(桃枝) 메탄올 추출물과 용매별 분획물이 활성산소종, 활성질소종 및 저밀도 지단백의 산화에 미치는 효과 (Effects of the Young Branch of Prunus persica Methanol Extracts and Solvent Fractions on ROS, RNS and Oxidized Low-Density Lipoprotein)

  • This study was designed to analyze the effects of methanol extracts and solvent fractions of the young branch of Prunus persica on scavenging activity of ROS, RNS and inhibiting activity of oxidative modification in human LDL induced by copper ion or free radical generator. The scavenging capacities of the fractions on DPPH radical, superoxide radical, nitric oxide and peroxynitrite were exhibited the highest in ethylacetate fraction, followed by butanol fraction. In the copper-induced LDL oxidative modification system, the highest antioxidant activity was revealed in ethylacetate fraction, and butanol fraction exhibited a similar activity. However, solvent fractions of the young branch of Prunus persica showed a relatively low antioxidant activity in the AAPH-mediated LDL oxidation. In addition, ethylacetate and butanol fractions also inhibited the copper-mediated LDL oxidation in the REM assay, which was comparable to that of the positive controls, including EDTA, ascorbic acid and BHT. Futhermore, a content of total phenolics in these two fractions was higher than that of the other fractions. These results indicated that ethylacetate and butanol fractions of the young branch of Prunus persica were useful for the prevention of the free radical- or metal ion-induced oxidative damages.

사람의 저밀도 지방단백질의 산화에 대한 생열귀나무 추출물의 항산화 효과 (Antioxidant Effect of Rosa davurica Pall Extract on Oxidation of Human Low Density Lipoprotein (LDL))

  • 비타민-C를 다량 함유하고 있는 생열귀나무의 자원화를 위한 연구로서 페놀성 화합물의 함량 및 사람의 저밀도지방단백질의 산화에 대한 항산화 활성을 측정하였다. 생열귀나무 추출물의 페놀성 화합물 함량을 측정한 결과, 모든 부위에서 100g당 8.3-10.6g으로 매우 높은 양을 함유하고 있었다. 또한, 생열귀나무 추출물 및 분획물에 대한 항산화 물질의 함량을 측정 한 결과, 생열귀나무 뿌리 메탄올 추출물과 뿌리의 에틸아세테이트 및 부탄을 분획물이 285nm에서 흡수를 나타내는 페놀성 화합물을 다량 함유하고 있음을 확인하였다. TBARS에 의한 LDL산화 억제효과 측정시, 생열귀나무 뿌리의 메탄올 추출물이 가장 좋은 효과를 나타내었다. 생열귀나무 뿌리의 에틸아세테이트 및 부탄을 분회물의 경우, $30\;{\mu}g/mL$ 첨가시 85.3% 및 93.2%까지 LDL 산화를 억제함을 확인하였다. 또한 $CHCl_3$ 분획물, EtOAc 분획물, 그리고 BuOH 분획물은 $30\;{\mu}g/mL$ 분획물 첨가시 $Cu^{2+}$에 의한 LDL 산화를 8시간까지 억제함을 확인하였다. $Cu^{2+}$에 의한 apoB carbonylation과 conjugate diene형성 억제효과 측정시, EtOAc 분획물과 BuOH 분회물이 가장 탁월하게 억제함을 보여주었다. 생열귀나무는 다량의 페놀성 화합물을 함유하고 있으며 항산화 활성이 매우 탁월하므로 새로운 기능성식품 소재로의 활용이 기대되는 약용식물자원이다.

The Protective Effects of water Extracts of ZoaGumHwan (ZGH) on the Oxidized LDL-induced Monocyte Adhesion to Human Umbilical Vein Endothelial Cells

  • It is well known that oxidized low-density lipoprotein (oxLDL) is the most characterized humoral factor that plays an important role in the pathogenesis of atherosclerosis. The water extract of the Korean herbal remedy, ZoaGumHwan (ZGH), which is composed of roots of Coptis chinensis Franch and fruits of Evodia officinalis Dode with the ratio of 6 to 1, reduced the in vitro oxidation of low density lipoproteins (LDL). Also, the ZGH extract and berberine, one of the major components of ZGH, significantly prevented oxLDL-induced adhesion of monocytes to human umbilical vein endothelial cells (HUVEC). Furthermore, the ZGH water extract and berberine decreased oxLDL-induced expression of CC chemokine receptor 2 (CCR2), a dominant monocyte chemotaxis receptor, in U937 human monocytic cells. The protective effects of the ZGH water extract and berberine were similar to those of simvastatin, an effective lipid-lowering drug. The results suggest that Korean herbal remedy, ZGH, seems to have protective effect against oxLDL-induced monocyte chemoattractant protein (MCP)-1/CCR2-dependent monocyte recruitment onto endothelial cells.

칠기탕(七氣湯) 약침액(藥鍼液)이 활성산소종, 활성질소종, 지질과산화 및 LDL의 산화에 미치는 영향 (Effect of Chilgitang Extract for Herb-Acupuncture on ROS, RNS, Lipid Peroxidation and LDL oxidation)

  • Objectives : This study was undertaken to determine the in vitro antioxidant activity of the extract of Chilgitang herb-acupuncture solution (CHAS). Methods : The radical scavenging capacity was tested by 2,2-diphenyl-1-picryl-hydrazyl (DPPH), hypoxanthine-xanthine oxidase system, DCFH-DA assay, nitric oxide and peroxynitrite generating system. In addition, antioxidant activity on copper and AAPH mediated human low-density lipoprotein (LDL) oxidation was measured by using TBARS assay and relative electrophoretic mobility assay. The amount of total phenolic compounds was assayed by the Folin-Ciocalteu method. Results : CHAS revealed a potent scavenging activity on DPPH radical(82%), superoxide anions(73%), hydroxyl radical(63%), nitric oxide (99%) and peroxynitrite (99%). Moreover, CHAS showed a strong inhibitory effect (59%) on $FeCl_2$-ascorbic acid induced lipid peroxidation of rat liver homogenate. CHAS also markedly inhibited copper(81%) and AAPH(56%)-mediated LDL oxidation, and effectively suppressed the electrophoretic mobility during exposure of human LDL to copper ions. CHAS (82 mg/g) contained higher concentration of total phenolic compounds than that of water extract (45 mg/g) obtained from Chilgitang. Conclusions : These results indicate that CHAS may protect against ROS- or RNS involved diseases, including cardiovascular diseases.

시호 추출물의 oxLDL 유도 Foam Cell 형성 억제 작용 (Inhibitory Effects of Bupleuri Radix on ox-LDL induced Foam Cell Formation)

  • The oxidative modification of low density lipoprotein(LDL) has been implicated in the development of atherosclerosis. Oxidized LDL(oxLDL) is captured into macrophage and stimulates to form macrophage foam cell. And it can induce an inflammation and smooth muscle proliferation in atherosclerotic plaque. Objective : In this study, we aimed to investigate the effect of Bupleuri radix(SH) on the foam cell formation, a critical initiation stage of atherosclerosis. Methods : To achieve the goal, we examined the effect of SH on LDL oxidation, nitric oxide production in RAW264.7, and the effect of SH on cupuric sulfate-induced cytotoxicity, LDH release, and macrophage activity. Results : SH inhibited the formation of oxidized LDL from native LDL in RAW264.7 cell culture, and decreased the release of LDH from cupric sulfate-stimulated RAW264.7 cell. In other experiments, SH activated RAW264.7 cell, and prolonged the survival time, and inhibited foam cell formation induced by oxLDL in Raw 264.7 cells. Conclusion : These results showed that SH might prevent atherosclerosis by controlling the early stages of foam cell formation.

곤충자원으로부터 항동맥경화 활성물질 탐색 (Screening of Anti-atherogenic Substances from Insect Resources)

  • Lipoprotein-associated phospholipase $A_2\;(Lp-PLA_2)$ is a potential biomarker of coronary heart disease and plays an important proinflammatory role in the progression of atherosclerosis. Also acyl-CoA: cholesterol acyltransferase (ACAT) and oxidized low-density lipoprotein (LDL) playa key role in atherosclerosis, respectively. And so, the inhibitory activities of the methanol extracts of 42 insect resources were examined on $Lp-PLA_2$, ACAT, and LDL oxidation for screening of anti-atherogenic substances. Among them, the methanol extracts of Eurydema rugosa significantly inhibited all of upper three activities. Several kinds of tested insects having high inhibitory effect with the methanol extracts were extracted with n-hexane, ethyl acetate, and acetone, and their inhibitory activities were tested.

  • Low density lipoprotein (LOL) is the major cholesterol carrier in the blood stream. Also, LDL oxidation has been proposed as an important step in the formation of atherosclerotic lesion. Thus, protection of LDL from oxidation is needed to prevent or to retard the progression of atherosclerosis. In this study, 6-hydroxyeugenol was isolated from the methanolic extract of the Spiraea blumei and the sructure was elucidated by spectroscopic data analysis. (omitted)

송절(松節) 약침액이 자유기와 금속 이온으로 유도된 인체 저밀도 지단백질의 산화 반응에 미치는 효과 (The Effect of Pinus Densiflora Gnarl Extract for Pharmacopuncture on Human LDL Oxidation Induced by Free Radical and Metal Ion)

  • 목적 : 이 연구는 관절 및 심혈관계 질환 치료에 사용되는 송절(松節)(Pinus densiflora Gnarl)을 약침용 시료로 조제하여 본 약물의 항산화 효능을 규명하고자 하였으며 이를 다양한 시스템에서 검토하였다. 방법 : $FeCl_2$-ascorbic acid system에서 흰쥐 간조직의 지질과산화 반응을 관찰하였고, Fenton reaction system에서 자유기에 의한 plasmid DNA 분절을 유도하였다. 또한 deoxyribose assay를 통해 hydroxyl radical 소거능을 관찰하였고, NBT reduction assay로 superoxide radical 소거능을 검토하였다. 또한 human low-density lipoprotein(LDL)의 산화를 유도하기 위해 $CuSO_4$와 AAPH를 사용하였으며 relative electrophoretic mobility (REM) assay로 LDL 산화 억제 효능을 대조 항산화물질과 비교 검토하였다. 결과 : 송절 약침액은 자유기에 의한 간조직의 지질과산화(p < 0.01)및 DNA 분절을 현저하게 억제하였으며, hydroxyl radical, superoxide radical (p < 0.01), nitric oxide 및 peroxynitrite를 강하게 소거하였다. 또한 $CuSO_4$ ($IC_{50}=9.2{\pm}0.2\;{\mu}g/ml$)와 AAPH ($IC_{50}=34.8{\pm}5.1\;{\mu}g/ml$)에 의해 유도된 human LDL의 산화를 억제하였고, REM assay에서도 산화 억제 효능을 재확인할 수 있었다. 결론 : 송절 약침액은 활성산소종 및 활성질소종를 소거하였고, 지질과산화를 억제하였으며, 특히 human LDL의 산화적 손상을 방어하였다. 이에 본 약물은 자유기에 의한 심혈관의 산화적 손상을 효과적으로 보호할 것으로 판단된다.

  • A major protein was isolated from ginseng root (Panax ginseng C.A. Meyer) using a combination of ammonium sulfate fractionation, gel filtration chromatography, ion-exchange FPLC. Electrophoretic and gel permeation chromatographic studies revealed that the major protein, GMP, is composed of two subunits of approximately 28 kDa. In this, investigated the ability of GMP to inhibit the oxidation of low-density lipoprotein (LDL). GMP inhibited $Cu^{2+}$ (5$\mu$M)-promoted oxidation of LDL (125$\mu$g protein/mL) in a dose-dependent mamer (0~5 $\mu$M), with a maximal inhibitor at GMP/copper ratio of 1:10 and an $IC_{50}$ value of 0.2 $\mu$M, as determined by measurement TBARS.
