Effect of Spermine Treatment on Growth, Yield, and Quality of Strawberry under Low-Temperature Condition (이상 저온 조건에서 Spermine 처리가 딸기의 생육과 수량 및 품질에 미치는 영향)

  • The present study was carried out to investigate the effects of spermine treatment on the growth, yield and quality in strawberry under low-temperature condition, and thereby develop a chemical method to minimize damages by low temperature in greenhouse cultivation. Spermine treatment significantly improved the growth of strawberry in terms of leaf number, leaf area, leaf length, leaf diameter, plant height and plant weight. The highest effect was observed in the 250 uM spermine treatment and the effect tended to be maintained during the entire growth period of 90 days. Fresh weight and dry weight were significantly different depending on the concentrations of spermine. Strawberry plants treated with 250 uM spermine showed higher fresh weight and dry weight compared to untreated control plants during the growth period. Fruit weight, fruit length and fruit diameter were relatively higher when treated with $100{\mu}M$ spermine compared to other treatments. The fruit yield was the highest with 14 fruits per plant at $250{\mu}M$ spermine treatment and the coloration of the fruit was the best at this treatment with the Hunter a and b values of 46.56 and 28.75, respectively. The hardness of strawberry fruit tended to increase higher than 2N at $250{\mu}M$ and $500{\mu}M$ 250 uM spermine treatment. The sugar content of strawberries treated with $250{\mu}M$ spermine was $9.5^{\circ}$ Bx which was $1.6^{\circ}$ Bx higher compared to that in untreated control. However, spermine treatment did not affect the acidity of fruit and it remained 0.68-0.76% regardless of treatment concentrations. These results suggest that spermine treatment has a positive effect on the growth and productivity of strawberry fruit under abnormal low-temperature condition. The positive effect was the highest at $250{\mu}M$ spermine treatment and gradually decreased in the order of $100{\mu}M$, $500{\mu}M$, and untreated control.

  • ZnO nanostructures have a lot of interest for decades due to its varied applications such as light-emitting devices, power generators, solar cells, and sensing devices etc. To get the high performance of these devices, the factors of nanostructure geometry, spacing, and alignment are important. So, Patterning of vertically- aligned ZnO nanowires are currently attractive. However, many of ZnO nanowire or nanorod fabrication methods are needs high temperature, such vapor phase transport process, metal-organic chemical vapor deposition (MOCVD), metal-organic vapor phase epitaxy, thermal evaporation, pulse laser deposition and thermal chemical vapor deposition. While hydrothermal process has great advantages-low temperature (less than $100^{\circ}C$), simple steps, short time consuming, without catalyst, and relatively ease to control than as mentioned various methods. In this work, we investigate the dependence of ZnO nanowire alignment and morphology on si substrate using of nanosphere template with various precursor concentration and components via hydrothermal process. The brief experimental scheme is as follow. First synthesized ZnO seed solution was spun coated on to cleaned Si substrate, and then annealed $350^{\circ}C$ for 1h in the furnace. Second, 200nm sized close-packed nanospheres were formed on the seed layer-coated substrate by using of gas-liquid-solid interfacial self-assembly method and drying in vaccum desicator for about a day to enhance the adhesion between seed layer and nanospheres. After that, zinc oxide nanowires were synthesized using a low temperature hydrothermal method based on alkali solution. The specimens were immersed upside down in the autoclave bath to prevent some precipitates which formed and covered on the surface. The hydrothermal conditions such as growth temperature, growth time, solution concentration, and additives are variously performed to optimize the morphologies of nanowire. To characterize the crystal structure of seed layer and nanowires, morphology, and optical properties, X-ray diffraction (XRD), field emission scanning electron microscopy (FE-SEM), Raman spectroscopy, and photoluminescence (PL) studies were investigated.

Efficacy of live NNV immersion vaccination in three grouper species (Seven band grouper Hyporthodus septemfasciatus, Red spotted grouper Epinephelus akaara, Long tooth grouper Epinephelus bruneus) (바리과 어류 3종(능성어 Hyporthodus septemfasciatus, 붉바리 Epinephelus akaara, 자바리 Epinephelus bruneus)에 대한 신경괴사증바이러스(Nervous Necrosis Virus, NNV)의 생백신 처리 효과)

  • In this study, we investigated safety and efficacy of a low temperature immunization protocol with NNV in red spotted grouper, Epinephelus akaara and long tooth grouper, Epinephelus bruneus and seven band grouper, Hyporthodus septemfasciatus. Further, growth rate between immunized and naïve fish was evaluated during the experiment to check side effect of immunization. Three grouper species were immunized by immersion method with live NNV at 105.0 TCID50/mL at 16.5℃ for 30 min and reared for 120 days at natural sea water temperature. To evaluate growth rate, total length and wet weight was measured 7 times after immunization. Immunized three grouper species were challenged by intramuscular inoculation with NNV at 104.2 TCID50/100 µL/fish. Immunization at low temperature with live NNV did not show any clinical symptoms of infection, mortality and inhibition of growth. After challenge, cumulative mortality of naïve seven band grouper, red spotted grouper, long tooth grouper were 45, 10, 20 %, respectively. However no mortality was observed at immunized groupers. Thus, it was demonstrated that immunization at low temperature with live NNV are able to protect three different species of groupers without inhibition of growth.

  • Graphene has drawn enormous attention owing to its outstanding properties, such as high charge mobility, excellent transparence and mechanical property. Synthesis of Graphene by chemical vapor deposition (CVD) is an attractive way to produce large-scale Graphene on various substrates. However the fatal limitation of CVD process is high temperature requirement(around $1,000^{\circ}C$), at which many substrates such as Al substrate cannot endure. Therefore, we propose plasma enhanced CVD (PECVD) and decrease the temperature to $400^{\circ}C$. Fig. 1 shows the typical structure of RF-PECVD instrument. The quality of Graphene is affected by several variables. Such as plasma power, distance between substrate and electronic coil, flow rate of source gas and growth time. In this study, we investigate the influence of these factors on Graphene synthesis in vacuum condition. And the results were checked by Raman spectra and conductivity measurement.

A Study on the Low Temperature Growth of SiC Film with a 1,3-DSB Precursor (단일전구체(1,3-DSB)에 의한 저온 SiC박막 성장에 관한 연구)

  • Silicon carbide thin film was deposited in APCVD and LPCVD system with 1,3-DSB precursor 1,3-DSB is the single precursor to deposit SiC on Si at low temperature. SiC film was deposited at $850^{\circ}C$ lower than ordinary temperature ($1000~1200^{\circ}C$) in CVD process. SiC thin film glowed to high oriented (111) plane in APCVD system. In LPCVD system, SiC film groved to preferred (220) plane at same temperature. This discrepancy between preferred planes can be described by the difference of deposition mechanism. Amorphous phase and crystal defect were observed in APCVD system with the main growth mechanism of mass transport limited region. But in LPCVD system, we got the SIC film of uniform, faceted structure and high quality.

Impacts of Temperature, Salinity and Irradiance on the Growth of Ten Harmful Algal Bloom-forming Microalgae Isolated in Korean Coastal Waters (한국연안에서 분리한 적조형성 미세조류 10종의 성장에 미치는 온도, 염분, 광도의 영향)

  • In order to understand growth characteristics of ten major species of microalgae responsible for frequent harmful algal blooms in Korean coastal waters, the growth rates of the isolates were examined in relation with the impacts of water temperature, salinity and irradiance. In addition, their bloom events since 1990 as well as monthly abundance of vegetative cells were analyzed. Heterocapsa triquetra, Eutreptiella gymnastica and Alexandrium tamarense were considered as relatively mid temperature adapted species in that growth rates were comparatively high at low water temperatures of $10{\sim}16^{\circ}C$ and drastically decreased at above $22^{\circ}C$. Prorocentrum micans and Pyramimonas sp. were categorized as relatively high temperature adapted species by showing comparatively better growths at high water temperatures above $25^{\circ}C$. Akashiwo sanguinea, Heterosigma akashiwo, Prorocentrum minimum and Scrippsiella trochoidea were eurythermal species with relative high growth rates in a broad ranges of water temperature, $16{\sim}25^{\circ}C$ were slightly halophobic, showing better growths at low salinities of $10{\sim}30$ psu than at above 35 psu. H. akashiwo, P. minimum and H. triquetra were euryhaline species with remarkable growths in a broad ranges of salinity, 15-40 psu. Frequent algal blooms by these three species at extremely low salinities below 25 psu after rainfall were attributed to their euryhaline and slightly halophobic physiological characteristics. Growth rates of H. akashiwo, P. minimum and Pyraminonas sp. increased with the increase of irradiance within the experimental ranges of $2{\sim}150\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$. However, A. sanguinea, A. tamarense and H. triquetra showed better growths at comparatively low irradiance of $50{\sim}100\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$ and drastic decreases in growth rates above $150\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$ of irradiance. Overall, relatively high temperature adapted species make blooms frequently in high water temperature season with strong natural irradiance, and relatively low temperature adapted species grow better at low water temperature with relatively weak natural irradiance.

Dormancy of Ginseng Seed as Influenced by Temperature and Gibberellic Acid (인삼종자의 휴면기간단축에 미치는 온도 및 지베레린의 영향)

  • This study was conducted to know the effects of temperature and gibberellic acid on the dormancy block-ing of ginseng seeds at different embryo growth stage. Optimum temperature for embryo growth appeared to be I soc at the beginning stage of post ripening (up to dehiscent time), 1$0^{\circ}C$ at middle stage (for 30 days after dehiscence) and 5$^{\circ}C$ at last stage (between 30 and 92 days after dehiscence). And optimum temperature for dehiscence is about 17$^{\circ}C$, also the optimum temperature for dehiscence is higher than that of embryo growth. Germination of ginseng seed with full grown embryo was accelerated at high temperature (25-3$0^{\circ}C$). Germina-tion percentage was 80% at 105th date after dehiscence under 5$^{\circ}C$, 28% at 147th date under 1$0^{\circ}C$, but no germination under the over 15$^{\circ}C$. Gibberellic acid increased the dehiscent rate, whereas the gibberellic acid treatment may not be substituted for effect of low temperature on the germination. Low temperature may be reguired to finish the embryo growth in thickness.

  • One of the major drawbacks associated with the high-level expression of the recombinant proteins in Escherichia coli is the formation of insoluble inclusion bodies in the cytoplasm. Production of recombinant protein at reduced temperature has proven effective in improving the solubility of a number of structurally and functionally unrelated proteins, but a major limitation of using low temperatures for recombinant protein production in E. coli is the reduced rate of synthesis of the heterologous protein caused by the significant reduction of cell growth rate. Here we investigated the effect of co-expression of CspA, a cold-shock protein known to be RNA chaperone at low temperature, on the productivity of recombinant protein at various temperatures by using green fluorescence protein (GFP) as a model recombinant protein. We could observe that the co-expression of CspA enhanced the productivity of GFP at $15^{\circ}C$ by accelerating the growth of E. coli at the temperature. On the other hand, the CspA coexpression didn't affect the cell growth rate as well as the specific GFP production rate at other tested temperatures, $20^{\circ}C$, $25^{\circ}C$, and $37^{\circ}C$.

  • Effects of plant growth regulators on the germination of ginseng (Panax ginseng C.A. Meyer) seeds were investigated. Ginseng seeds germinated more vigorously in the treatments of kinetin and BA, and the promoting effect of kinetic on the germination and the growth of rootlet enhanced in low temperature ($10^{\circ}C$). However, GA did not promote the germination of dehiscent seed. The optimum temperature for germination of dehiscent seed was $10^{\circ}C$ and the range of effective concentration of kinetin for germination was 50 to 100 ppm.

  • BSCCO thin films are fabricated via a co-deposition process at an ultra-low growth rate using ion beam sputtering. The sticking coefficient of Bi element exhibits a characteristic temperature dependence. This temperature dependence of the sticking coefficient was explained consistently on the basis of the evaporation and sublimation processes of Bi$_2$O$_3$.
