Study on Multiscale Analysis on Drought Characteristics

  • One of the hazard of nature is a drought. Its impact varies from region to region and it is difficult for people to understand and define due to differences in hydrometeorological and social economic aspects across much of the country. In the most general sense, drought originates from a deficiency of precipitation over an extended period of time, usually month, season or more, resulting in a water shortage for some activity, group, or environmental sector. Palmer Drought Severity Index (PDSI) is well known and has been used to study aridity changes in modern and past climates. The PDSI index is estimated over US using USHCN historical data.(e.g. precipitation, temperature, latitude and soil moisture). In this study, low frequency drought variability associated with climate variability such as El-Nino and ENSO is mainly investigated. With respect to the multi-scale analysis, wavelet transform analysis is applied to the PDSI index in order to extract the low frequency band corresponding to 2-8 years. Finally, low frequency patterns associated with drought by comparing global wavelet power, with significance test are explored.

백두옹엑스 분획물의 소염진통작용 (The Anti-inflammatory and Analgesic Actions of the fractions from Pulsatilla koreana Root Extract)

  • From our previous report, the water extract of Pulsatilla koreana root was found to have potent anti-inflammatory and analgesic actions in intravenous administration in animals. Among chloroform, ethyl acetate, butanol and water fractions which were obtained through successive fractionation of the extract, only the water fraction was found to have the antiinflammatory and analgesic actions. The fraction did not affect normal body temperature at the effective doses in mice and showed low acute toxicity of which $LD_{50}$ was less than 500 mg/kg i.v. in mice. It is interesting that its anti-inflammatory action might be attributed in part to inhibition of cyclooxygenase-1 and -2.

Modified Cultivation Methods Improve Shelf-life and Quality of Soybean Sprouts, Effects of Treatment with Oak Charcoal and Citrus sunki Seed Extract

  • The effects of cultivation methods (cultivation of curled-shaped type, M-1; conventional cultivation, M-2; growing after treatment with a growth regulator, M-3; cultivation by the combination of M-1 and treatment with oak charcoal, M-4) on the quality characteristics of soybean sprouts were studied by the measurement of growth characteristics. This study also investigated the changes in shelf-life stability of the new soybean sprouts (NSB) using M-4, which was cultivated with oak charcoal and treated with antimicrobial Citrus sunki seed extract. Among the soybean sprouts grown for six days at the high temperature and humidity environment (90$\pm$5% RH, 25$\pm$1$^{\circ}C$), M-1 revealed no significant difference in terms of quality, such as the harvest yield, the rot rate and the growth characteristics when compared with M-2. M-3 showed no significant difference in growth characteristics, of hardness, and sensory evaluation scores when compared with the soybean sprouts grown by conventional methods. NSB had a low number of total microorganisms and had a better appearance after five days of storage than did the control group (M-2). These findings demonstrate that chemical-free and clean soybean sprouts can be grown by combining oak charcoal and antimicrobial Citrus sunki seed extract, thereby meeting the consumer demand for safe, chemical free sprouts.

건조 황칠나무 부위별 추출물을 이용한 견직물의 염색성과 기능성 (Dyeability and Functionality of Dried Dendropanax morbiferus Extracts)

  • This study aims to examine the possibility of using dried Dondropanax morbiferus extract as a functional dye. The leaves and branches of were extracted with distilled water and 30% ethanol, and the dyeability and functionality of silk fabrics were examined according to the color characteristics of the extract and dyeing conditions. As a result of analyzing the ultraviolet and visible light absorption spectrum of the extract, it was possible to confirm the peak of flavonoid belonging to polyphenol, and the peak of riboflavin expressing yellow color was confirmed. Adsorption equilibrium was observed at 4% dyeing concentration and 60 minutes of dyeing time, and as the temperature increased, dyeing amount increased without color change of Y-series. Aluminum mordanting also increased the yellow color. The color fastness of washing and UV irradiation was low, but the color fastness of rubbing was evaluated as relatively good. The silk fabric dyed with the distilled water extract of the leaves showed a 99.9% bacteriostatic reduction against Staphylococcus aureus and Klebsiella pneumoniae, showing excellent antibacterial properties.

식물세포배양으로부터 Paclitaxel 회수를 위한 메탄올 추출액의 여과 및 농축 조건 최적화 (Octimization of Conditions of Filtration and Concentration of Methanol Extract for Recovery of Paclitaxel from Plant Cell Culture)

  • 식물세포배양으로부터 paclitaxel을 효율적으로 대량추출하기 위한 추출액의 여과 및 농축 조건을 최적화하였다. 메탄올을 이용한 biomass 추출물을 여과할 경우, biomass 대비 6% (w/w) 여과보조제인 규조토를 첨가할 경우 여과시간 단축에 가장 효과적임을 알 수 있었다. 규조토 6% (w/w) 첨가를 통하여 1회 biomass 추출물 여과 시 4.2%, 2회 추출물 여과 시 30.3%, 3회 추출물 여과 시 22.8%, 4회 추출물 여과 시 19.0%의 여과시간 단축 효과를 각각 얻을 수 있었다. 여과액의 농축은 paclitaxel 순도 및 수율을 고려할 때 rotary evaporator의 bath 온도를 50$^{\circ}C$ 이하로 운전하는 것이 바람직하였으며, 이때 농축액의 온도는 증발잠열 때문에 실제 bath의 온도보다는 상당히 낮음을 알 수 있었다. 또한 여과액의 농축 완료 시점은 paclitaxel 순도, 수율, 농축시간, 액-액 추출에서의 상 분리 시간 등을 고려할 때 농축액의 비중이 0.95에서 종료하는 것이 가장 적당함을 알 수 있었다.

묵은 배추김치의 휘발성 성분 특성 (Characterization of Volatile Compounds in Low-Temperature and Long-Term Fermented Baechu Kimchi)

  • 묵은 배추김치의 휘발성 성분들을 고진공승화법을 이용하여 분리, 농축하였으며, GC-MS로 분석하였다. 총 62가지의 휘발성 성분들이 검출되었으며, 7개의 sulfur-containing compounds, 8개의 terpenes, 5개의 esters, 8개의 acids, 15개의 alcohols, 2개의 nitriles, 2개의 ketones, 11개의 aliphatic hydrocarbons, 4개의 miscellaneous compounds등으로 구성되어 있었다. 이 중 특히, acids인 acetic acid와 butanoic acid가 높은 함량을 나타내었다. 묵은 배추김치에서 향기활성성분을 규명하기 위해 고진공승화법을 이용하여 휘발성 성분들을 분리 및 농축 후 향 추출물 희석분석법으로 flavor dilution(FD) factor($Log_3FD$)를 구하였으며, GC-MS 및 GC-Olfactometry(GC-O)법으로 휘발성 성분들을 동정하였다. 이 결과 묵은 배추김치에서 총 16종의 화합물이 검출되었다. 이중 시큼한 냄새, 군덕내의 특성을 지닌 acetic acid, butanoic acid가 높은 FD factor를 보였으며, acids외에 높은 (FD > 9)를 나타내는 성분들로는 linalool, 2-vinyl-4H-1,3-dithin 등이 있었다.

저온숙성마늘의 라디칼 소거 활성을 통한 산화스트레스 억제 효과 (Oxidative Stress Inhibitory Effects of Low Temperature-Aged Garlic (Allium sativum L.) Extracts through Free Radical Scavenging Activity)

  • 본 연구에서는 마늘의 숙성 기간(15일, 30일, 60일)과 온도($60^{\circ}C$, $70^{\circ}C$)를 달리한 저온숙성마늘과 생마늘의 항산화 효과를 비교 분석하였다. DPPH와 ABTS의 라디칼 소거능과 FRAP법에 의한 환원력을 측정한 결과 $250{\mu}g/mL$에서 생마늘 추출물보다 30일 $70^{\circ}C$ 추출물과 60일 $60^{\circ}C$ 추출물의 항산화 활성이 우수하였다. 세포 내 활성산소 생성은 15일 $60^{\circ}C$ 추출물과 30일 $70^{\circ}C$ 추출물에서 높은 억제 효과를 보였으며, xanthine oxidase에 대한 활성 저해 효과 역시 15일 $60^{\circ}C$ 추출물에서 우수하였다. 항산화 효소의 유전자 발현은 LPS를 처리한 군과 생마늘 추출물보다 30일 $70^{\circ}C$ 추출물에서 높은 효과를 보였다. 본 연구 결과를 통해 저온숙성마늘이 생마늘보다 항산화 활성이 우수하다는 것을 확인함으로써 차후 항산화 건강기능식품 소재로서의 활용이 가능할 것으로 판단되나, 저온숙성마늘 추출물의 체내 생리활성 메커니즘 규명을 위해 동물실험 등의 추가적인 연구를 계속적으로 수행할 예정이다.

고체배지 상에서 목재부후성 버섯 균주들의 균사생장 특성과 세포외효소 활성 (Mycelial Growth and Extracellular Enzyme Activities of Wood-decaying Mushroom Strains on Solid Media)

  • 우리나라 산림에서 수집한 목재부후성 버섯 균주들에 대한 배지 및 온도별 균사생장 특성을 고체평판배지 상에서 조사하였다. 대부분의 균주들은 malt extract agar (MEA)나 sabouraud dextrose agar (SDA)보다는 PDA (potato dextrose agar) 배지에서 균사 생장력이 더 높았다. 몇몇 균주들을 제외하고, 균주들의 대부분은 PDA 배지 상에서 배양 시 $25^{\circ}C$에서 가장 잘 생장하였으며, 고온($30^{\circ}C$)보다는 저온($10^{\circ}C$)에서 균사 생장이 저조하였다. 2종류의 서로 다른 세포외효소 활성에 각각 특이적으로 반응할 수 있는 기질이 함유된 고체평판배지 상에서 모든 시험균은 carboxymethyl cellulase 활성과 laccase 활성을 나타냈다.

Application of hybrid material, modified sericite and pine needle extract, for blue-green algae removal in the lake

  • The present study assessed the efficient removal of nutrients and Chlorophyll-a (Chl-a) by using methyl esterified sericite (MES) and pine needle extracts (PNE), a low cost and abundant green hybrid material from nature. For this purpose, the optimal conditions were investigated, such as the pH, temperature, MES and PNE ratio, and MES-PNE dose. In addition, a Microcystis aeruginosa control using MES-PNE was also analyzed with various inhibition models. The removal of the nutrient and Chl-a onto MES-PNE was optimized for over 95% removal as follows: 2-2.5 for the MES-PNE ratio, 7-8 pH and a $22-25^{\circ}C$ temperature. In this respect, approximately 1.52-2.20 g/L of MES-PNE was required to remove each 1 g of dry weight/L of Chl-a. Total phosphorus (TP) has a greater influence on the increase in Chl-a than total nitrogen (TN) according to the correlation between TN, TP and Chl-a. Moreover, the Luong model was the best model for fitting the biodegradation kinetics data from Chl-a on MES-PNE from lake water. The novel hybrid material MES-PNE was very effective at removing TN, TP and Chl-a from the lake and can be applied in the field.

허브간장의 항산화성 및 품질 특성에 미치는 영향 (Effects of Antioxidant and Quality Characteristics of Herb Soy Sauce)

  • Herb soy sauce was prepared by extracting rosemary, lemon balm, spearmint and peppermint at a low temperature in fresh soy sauce. The antioxidant and physicochemical properties of herb soy sauces were examined. Herb soy sauces were prepared by adding 2.5% (w/v) of herbs to fresh soy sauce at 60℃. The total polyphenol, DPPH%, ABTS% increased with extracting time but five minutes extraction with sufficient antioxidant herb soy sauce could be prepared. The antioxidant powers of herb soy sauces were higher in lemon balm extract, and the rosemary extract showed the lowest. Soy sauces added with herbs had lower pH and salinity (p<0.05) and higher sugar content than without herbs. The colorness according to the type of herbs showed significant difference.
