Reduced Autophagy in 5-Fluorouracil Resistant Colon Cancer Cells

  • We investigated the role of autophagy in SNUC5/5-FUR, 5-fluorouracil (5-FU) resistant SNUC5 colon cancer cells. SNUC5/5-FUR cells exhibited low level of autophagy, as determined by light microscopy, confocal microscopy, and flow cytometry following acridine orange staining, and the decreased level of GFP-LC3 puncta. In addition, expression of critical autophagic proteins such as Atg5, Beclin-1 and LC3-II and autophagic flux was diminished in SNUC5/5-FUR cells. Whereas production of reactive oxygen species (ROS) was significantly elevated in SNUC5/5-FUR cells, treatment with the ROS inhibitor N-acetyl cysteine further reduced the level of autophagy. Taken together, these results indicate that decreased autophagy is linked to 5-FU resistance in SNUC5 colon cancer cells.

Comparative Study of Autophagy in Oxaliplatin-Sensitive and Resistant SNU-C5 Colon Cancer Cells

  • Few studies have evaluated the role of autophagy in the development of oxaliplatin (OXT) resistance in colon cancer cells. In this study, we compared the role of autophagy between SNU-C5 colon cancer cells and OXT-resistant SNU-C5 (SNU-C5/OXTR) cells. At the same concentration of OXT, the cytotoxicity of OXT or apoptosis was significantly reduced in SNU-C5/OXTR cells compared with that in SNU-C5 cells. Compared with SNU-C5 cells, SNU-C5/OXTR cells exhibited low levels of autophagy. The expression level of important autophagy proteins, such as autophagy-related protein 5 (Atg5), beclin-1, Atg7, microtubule-associated proteins 1A/1B light chain 3B I (LC3-I), and LC3-II, was significantly lower in SNU-C5/OXTR cells than that in SNU-C5 cells. The expression level of the autophagy-essential protein p62 was also lower in SNU-C5/OXTR cells than in SNU-C5 cells. In SNU-C5/OXTR cells, the production of intracellular reactive oxygen species (ROS) was significantly higher than that in SNU-C5 cells, and treatment with the ROS scavenger N-acetylcysteine restored the reduced autophagy levels. Furthermore, the expression of antioxidant-related nuclear factor erythroid 2-related factor 2 transcription factor, heme oxygenase-1, and Cu/Zn superoxide dismutase were also significantly increased in SNU-C5/OXTR cells. These findings suggest that autophagy is significantly reduced in SNU-C5/OXTR cells compared with SNU-C5 cells, which may be related to the production of ROS in OXT-resistant cells.

Transient intubation for surfactant administration in the treatment of respiratory distress syndrome in extremely premature infants

  • Purpose: To investigate the effectiveness of transient intubation for surfactant administration and extubated to nasal continuous positive pressure (INSURE) for treatment of respiratory distress syndrome (RDS) and to identify the factors associated with INSURE failure in extremely premature infants. Methods: Eighty-four infants with gestational age less than 28 weeks treated with surfactant administration for RDS for 8 years were included. Perinatal and neonatal characteristics were retrospectively reviewed, and major pulmonary outcomes such as duration of mechanical ventilation (MV) and bronchopulmonary dysplasia (BPD) plus death at 36-week postmenstrual age (PMA) were compared between INSURE (n=48) and prolonged MV groups (n=36). The factors associated with INSURE failure were determined. Results: Duration of MV and the occurrence of BPD at 36-week PMA were significantly lower in INSURE group than in prolonged MV group (P<0.05), but BPD plus death at 36-week PMA was not significantly different between the 2 groups. In a multivariate analysis, a reduced duration of MV was only significantly associated with INSURE (P=0.001). During the study period, duration of MV significantly decreased over time with an increasing rate of INSURE application (P<0.05), and BPD plus death at 36-week PMA also tended to decrease over time. A low arterial-alveolar oxygen tension ratio (a/APO2 ratio) was a significant predictor for INSURE failure (P=0.001). Conclusion: INSURE was the noninvasive ventilation strategy in the treatment of RDS to reduce MV duration in extremely premature infants with gestational age less than 28 weeks.

Ethanol Eluted Extract of Rhus verniciflua Stokes Showed both Antioxidant and Cytotoxic Effects on Mouse Thymocytes Depending on the Dose and Time of the Treatment

  • For a long time Rhus verniciflua Stokes (RVS) has traditionally been used as a herbal plant. It is known to contain various biological activities. Previously, a crude ethanol extract from RVS was reported to have antioxidant effects, and antiproliferative activities, on human cancer cell lines. In this report, we prepared a highly purified ethanol extract from RVS, which did not contain the urushiol derivatives, named REEE-1 ($\underline{R}$hus $\underline{e}$thanol $\underline{e}$luted $\underline{e}$xtract-1), to investigate the mechanisms of the scavenging activity of hydroxyl radicals using mouse thymocytes. The results from the deoxyribose, DNA nicking, and glucose/glucose oxidase enzyme assays showed that REEE-1 contained a strong scavenging activity of oxygen free radicals, especially of hydroxyl radicals. However, interestingly, REEE-1 also showed cytotoxicity against the thymocytes, although the effect was variable, depending on the concentrations and times of treatment. The REEE-1-mediated cytotoxicity against thymocytes, which has been used as one of the well-characterized models for apoptosis studies, was verified to be apoptotic. This was proven by the following: the appearance of DNA laddering, increases in DNA fragmentation, low fluorescence intensity in the nuclei after propidium iodide staining, and positive Annexin V staining of the cells. These results suggested that REEE-1 had both antioxidative activity and cytotoxicity against the thymocytes, although the effect of the cytotoxicity was variable, depending on the dose and time of the treatment.

Identification of AMPK activator from twelve pure compounds isolated from Aralia Taibaiensis: implication in antihyperglycemic and hypolipidemic activities

  • The root bark extract of Aralia taibaiensis is used traditionally for the treatment of diabetes mellitus in China. The total saponin extracted from Aralia Taibaiensis (sAT) has effective combined antihyperglycemic and hypolipidemic activities in experimental type 2 diabetic rats. However, the active compounds have not yet been fully investigated. In the present study, we examined effects of twelve triterpenoid saponins on AMP-activated protein kinase (AMPK) activation, and found that compound 28-O-${\beta}$-D-glucopyranosyl ester (AT12) significantly increased phosphorylation of AMPK and Acetyl-CoA carboxylase (ACC). AT12 effectively decreased blood glucose, triglyceride (TG), free fatty acid (FFA) and low density lipoprotein-cholesterol (LDL-C) levels in the rat model of type 2 diabetes mellitus (T2DM). The mechanism by which AT12 activated AMPK was subsequently investigated. Intracellular ATP level and oxygen consumption were significantly reduced by AT12 treatment. The findings suggested AT12 was a novel AMPK activator, and could be useful for the treatment of metabolic diseases.

Enhancement of Arsenic Trioxide ($As_2O_3$)-Mediated Apoptosis Using Berberine in Human Neuroblastoma SH-SY5Y Cells

  • Objective : Arsenic trioxide ($As_2O_3$) has been used as an anticancer agent in traditional Chinese medicine for thousand years and berberine is an isoquinoline alkaloid present that has indicated significant antimicrobial activity. We have examined the combined anticancer effects of $As_2O_3$ and berberine against the human neuroblastoma (HNB) SH-SY5Y cells in vitro, and to elucidate underlying molecular mechanism. Methods : HNB SH-SY5Y cells were treated with $2\;{\mu}M\;As_2O_3$ and $75\;{\mu}g/ml$ berberine, and their survival, cell death mechanism as well as synergistic cytotoxic effects were estimated by using MTT assay, DAPI staining, agarose gel electrophoresis, flow cytometric analysis, and western blot analysis. Results : The combined treatment of two drugs also markedly decreased cell viability. The cytotoxic effects of two drugs were revealed as apoptosis characterized by chromatin condensation, DNA fragmentation, and the loss of mitochondrial membrane potential. The apoptotic cytotoxicity was accompanied by activation of caspase-3 protease as well as decreased the expression of Bcl-2, Bid, and Bcl-x/L. In addition, the cells treated with combination of two drugs also showed significantly increased intracellular reactive oxygen species levels and lipid peroxidation compared to cells $As_2O_3$or berberine only. Conclusion : Combined treatment of $As_2O_3$ with berberine induced activation of apoptotic signaling pathways in HNB SH-SY5Y cells. These results suggest that the possibility of the combined treatment of two chemotherapeutic agents with low concentration improving cytotoxic effect for cancer cells with minimal side effects.

Analysis of Treatment Efficiency according to Open-water in Constructed Wetland (인공습지 내 개방수역 조성에 따른 처리효율분석)

  • The field scale experiment which is constructed with four sets (0.88 ha for each set) of wetland (0.8 ha) and pond (0.08 ha) systems was performed to examine the effect of plant coverage on the constructed wetland performance and to recommend the optimum development and management of macrophyte communities. After six growing seasons of wetlands, plant coverage was about 100%. And the concentration of DO showed low value (1.0~5.4 mg/L). This is caused by a blighted plant consumed dissolved oxygen with decay in water column. As the result, water column went to be anaerobic conditions and T-N removal rate are 58~67%. Dead vegetation increased nitrogen removal during winter because it is a source of organic carbon which is an essential parameter in denitrification. However, wetland released phosphorus caused by a blighted plant and accumulation, the removal rate of phosphorus might be decreased. To rise of DO concentration, the three open-waters were constructed in cell 3 and 4. Cell 3 has two open-waters (width 10 m, depth 1.8 m) and cell 4 has one open-water (width 20 m, depth 1.8 m). As the result, DO concentration and treatment efficiency of nutrient and BOD were improved. In case that constructed wetland is operated for a long time, physical circulation structure such as open water help continuous circulation of aerobic and anaerobic conditions. Through the constructed open-water, treatment efficiency of phosphorus and nitrogen in wetland could be improved effectively.

The Protective Effects of the Ethyl Acetate Fraction and Flavonoids from Taraxacum coreanum against Oxidative Stress in Neuronal Cells Induced by Hydrogen Peroxide and Amyloid Beta (흰민들레 Ethyl Acetate 분획물 및 Flavonoid 화합물의 Hydrogen Peroxide와 Amyloid Beta에 의한 신경세포의 산화적 스트레스 보호 효과)

  • The protective role against oxidative stress under cellular system using C6 glioma cells was studied using the ethyl acetate (EtOAc) fraction, luteolin (1), and luteolin-7-glucoside (2) of Taraxacum coreanum. C6 glioma cells showed low cell viability and high generation of reactive oxygen species (ROS) by the treatment with generator of hydrogen peroxide ($H_2O_2$) and amyloid beta ($A{\beta}_{25-35}$). However, the treatment of the EtOAc fraction attenuated the cellular oxidative stress, resulting in significant elevation of cell viability. In addition, the production of ROS formation was also decreased by the treatment of the EtOAc fraction. Compounds 1 and 2 were isolated from the EtOAc fraction, and the protective effect was evaluated. Compounds 1 and 2 led to the increase of cell viability and decrease of production of ROS against oxidative stress by $H_2O_2$ and $A{\beta}_{25-35}$. The present study indicated that the EtOAc fraction, compounds 1 and 2 from T. coreanum demonstrated protective effects against oxidative stress, suggesting the preventive role against neurodegenerative diseases.

  • Experiments were carried out to determine the optimum dose for the preservability of each Korean chestnut varieties by means of gamma-radiation. They were associated with control of spots in the edible portion of chestnut due to irradiation at the final stage of storage. With this result the following summary can be made. 1. In consideration of those factors such as rooting, sprouting, rotting and undesirable spots on the flesh, it was shown that the optimum radiation dose was checked to be 25-35 Krad though there are some difference among the varieties of chestnut. 2. Rotting and the spots on the flesh of chestnut were remarkably inhibited by low oxygen atmosphere during irradiation. This condition greatly contributed to inhibit sprouting with the low radiation dose(15 Krad). 3. The nuts irradiated had a marked increase in carbon dioxide evolution immediately after the irradiation. However on the day succeding the treatment, these increased curves gradually normalized as same as that of untreated.

A Study on Comparison of the Quality Changes Powder Yam and Sliced Yams (분말 마와 절편 마의 품질변화 비교에 관한 연구)

  • In this experiment, alcohol and blanching treatments were used for food packaging materials, quality of powder yam and sliced yams (5 mm size) were compared by alcohol concentration and time for measuring change of color, weight, moisture content saccharinity and decomposition. 5% and 10% alcohol were used to alcohol treatment for 5, 10 seconds each. Packaging materials were used OPP($30{\mu}m$), PET($25{\mu}m$), ON($25{\mu}m$) and all materials heat sealable were coated. Quality of powder yam were not changed during storage at low temperature however were changed during storage at room temperature after 5 days. In conclusion, powder yam were needed packaging materials with high water barrier property from moisture, sliced yams were high quality when packaged by ON and stored at low temperature. Therefore, using packaging materials with low oxygen permeability were expected to reduced quality degradation about browning of powder yam and sliced yams.
