Acridine Orange Stained Micronucleus Assay in Human B and T-lymphocytes after Low Dose ${\gamma}-irradiation$ (아크리딘 오렌지 형광염색법을 이용한 저선량 감마선 유도 말초혈액 B와 T-림프구 미소핵 분석)

  • Firstly, we compared the two staining techniques, Giemsa and Acridine orange, to determine micronuclei on samples of cultures of five healthy human peripheral blood lymphocytes after ${\gamma}-irradiation\;(^{137}Cs)$ in dose ranges of 0 to 800cGy. It was found that the Acridine orange staining method gives more reliable results than the usual Giemsa staining method in micronucleus tests. Moreover, the frequency of micronuclei in cytokinesis-blocked human B-lymphocytes was studied after in vitro irradiation in dose ranges of 0 to 50cGy. After setting and separating the B-lymphocytes, the frequency of radiation-induced micronuclei were observed as the end-point markers for the low-dose radiation dosimetry after staining with Giemsa and Acridine orange dyes. The micronuclei frequency in B-lymphocytes was significantly elevated from 10 to 30cGy ${\gamma}-irradiation$. The determination of micronuclei in B-lymphocytes after staining with Acridine orange was higher than that of Giemsa. The frequency of micronuclei in B-lymphocytes was observed to be at least two times higher than those of T-lymphocytes Giemsa in dose increasing. Therefore, the determination of low-dose radiation-induced micronuclei in B-lymphocytes after staining with Acridine orange is likely to have the greatest potential in the estimation of low dose radiation exposure.

Influence of the Low Dose ${\gamma}$-ray Radiation on the Old Seed Germination and Growth of Chinese Cabbage (묵은 배추종자의 발아와 생육에 미치는 저선량 ${\gamma}$선 효과)

  • To investigate the stimulating effects of the low- dose radiation, the seeds of chinese cabbages (Brassica campestris L. cv. Hanyoreum and cv. Tropic emperor) were irradiated at the dose of 0.5 Gy~20 Gy with the ${\gamma}$-ray radition. In case of ‘Hanyoreum’cultivar, there were no significant promoting effects on the germination rate except the 0.5 Gy irradiated group. The increase in seedling height and fresh weight in 8 Gy and 12 Gy irradiated groups was 20% and 40%, respectively, which suggested the stimulating effect of the low-dose radiation on the early growth. In case of‘Tropic emperor’cultivar, the germination rate of 1 Gy irradiated group increased approximately 10% and the seedling height and fresh weight of 1 Gy and 12 Gy irradiated group in the greenhouse 20% on the average, and plant height and fresh weight of 0.5 Gy and 4 Gy irradiated group in the field increased 10% and 20% on the average, respectively.

  • Acetylsalicylic acid (ASA) has been issued recently in contaminated water environments because of potential impacts on ecosystem and public health. This study was aimed at investigating the possibility of ASA degradation using gamma ray irradiation. In addition, the use of sodium persulfate, hydrogen peroxide, ferrous sulfate were tested in order to examine a synergistic effect with gamma ray. The absorbed dose was ranged from 0.2 to 10 kGy and the concentration of oxidants were from 0.1 to 10 mM in this study. The concentration of ASA was gradually decreased corresponding to the increase of the absorbed dose. When soudium persulfate was simultaneously applied, most of the parent compound was completely degraded even at a low dose of 0.8 kGy. The removal efficiency of total organic carbon was 90% even at the highest dose of 10 kGy without sodium persulfate. However, the efficiency was dramatically enhanced up to 98% at the same dose by adding 10 mM of oxidants. It was suggested that hydroxyl radical ($OH{\cdot}$) and sulfate radical ($SO{_4}^-{\cdot}$) were formed in the system and made roles in degrading ASA at the same time.

Effect of Low Dose γ Radiation on the Dormancy Breaking and Growth of in vitro Microtubers of Potato (Solanum tuberosum L.) Stored at Low Temperature (저선량 방사선이 저온 저장한 감자 기내 소괴경의 휴면타파와 생육에 미치는 효과)

  • To observe the stimulating effect of low dose ${\gamma}$ radiation on the dormancy breaking and growth, microtubers of two potato cultivars (Solanum tuberosum L. cv. Dejima and cv. Superior) were irradiated at the dose of 0.5-30 Gy. Though it varied with cultivars and storage duration, sprouting rate, plant growth and tuber yield were promoted by 2-8 Gy irradiation in microtuber of 'Dejima' stored at low temperature. On the other hand, in microtuber of 'Superior', sprouting rate was promoted by 2 and 4 Gy irradiation, and the growth and tuber yield by 4 Gy irradiation. These results suggest that low dose of ${\gamma}$ radiation could have stimulating effects on the dormancy breaking of microtuber and potato growth.

  • Germination rate and early growth in crop such as rice,soybean,and perilla were observed after irradiation of $^{\gamma}$-ray (Co-60) in order to determine the effects of low does radiation. The low dose radiation was able to improve the early growth in crops and their agricultural charaters. Germination rate of 2Gy-irradiatied rice seeds was high and also were seeding height and fresh weight of the 0.5 Gy-irradiated. Germination rate and early growth of soybean were high in 4Gy-irradiated group. Perilla gew ot so promisingly after after low dose irradiation,however there slightly increasing effects on germination rate, seeding height and fresh weightat 2Gt-, 1Gy-, and 1Gy irradiated group, respectively.

  • We fabricated a 15 m long position-sensitive plastic scintillation optical fiber (PSOF) detector consisting of a PSOF, two photomultiplier tubes, four fast amplifiers, and a digitizer. A single PSOF was used as a sensing part to estimate the gamma-ray source position, and 137Cs, an uncollimated solid-disk-type radioactive isotope, was used as a gamma-ray emitter. To improve the sensitivity, accuracy, and measurement time of a PSOF detector compared to those of previous studies, the performance of the amplifier was optimized, and the digital signal processing (DSP) was newly designed in this study. Moreover, we could measure very low dose rates of gamma-rays with high sensitivity and accuracy in a very short time using our proposed PSOF detector. The results of this study indicate that it is possible to accurately and quickly locate the position of a very low dose rate gamma-ray source in a wide range of contaminated areas using the proposed position-sensitive PSOF detector.

  • Gamma-rays effectively decomposed TPA and EG, thus removal of $1.0{\times}10^{-3}$ M pollutants was near 65 and 95%, respectively, at an absorbed dose of 10 kGy. However, TOC removal in the radiation treatment was less than 5% due to a low transformation of both TPA and EG to $CO_2$. For TPA, gamma-ray treatment largely reduced biodegradability($BOD_5/COD$) by degrading TPA to non-biodegradable organic acids. This implies that the change of biodegradability should be considered when the radiation treatment is combined with conventional biological techniques. A weight-loss wastewater containing TPA and EG was also purified by gamma-ray treatment. Extraordinarily, biodegradability of the wastewater was increased at a low dose of 1 kGy. Though underlying mechanism was not clearly identified, this result stresses the effect of wastewater composition and absorbed dose on the biodegradability change.

Effects of Low Dose Gamma-irradiation on Alleviation of Salt Stress in Rice (Oryza sativa L.) Seedling (저선량 감마선 조사가 벼 유묘의 염 스트레스 경감에 미치는 영향)

  • To investigate alleviation effects of salt stress by low dose gamma-irradiation on seedling growth of rice (Oryza shrive L), in plants of two rice cultivars (Ilpumbyeo and Gancheokbyeo), seeds were irradiated with the different doses $(0{\sim}32\;Gy)$ of gamma-ray by irradiator ($^{60}Co$, ca.150 TBq of capacity, AECL). The salt stress caused a remarkable decrease of seedling growth, particularly for dry weight. In addition, relative growth rate was decreased over 50% by the salt stress. However, the salt stress-induced inhibition of growth was alleviated by gamma-irradiation. Especially, the relative growth rate of shoot showed the reduction effect over 30% at 4 Gy-irradiated groups as compared with that of control. Also, electrolyte leakage and relative water content were damaged, respectively 4 times and 30% by the salt stress but the damages were alleviated by the gamma-irradiation. These alleviation effects were observed similarly in both the cultivars tested. These results suggest that the low dose gamma-irradiation may have a better protection or the salt stress.
