Protective Effect of Herbal Mixture Including Lycii Fructus on Hepatotoxicity Induced by Thioacetamide in Mice (구기자 복합물 약침액이 간기능 개선에 미치는 영향)

  • Objectives : This study investigated the hepatoprotective effect of herbal mixture including Lycii fructus (HML) in thioacetamide (TAA)-induced hepatotoxicity in mice. Methods : To confirm the liver protective effect, induced by TAA for 3 days injection at 100 mg/kg mice, HML were treated for 8 weeks at 300 mg/kg/day, 1000 mg/kg/day. Positive control was treated silymarin 50 mg/kg/day after TAA injection. The changes of mortality rate, clinical signs, organ weight, relative liver, blood chemistry and histopathological findings were analyzed after experiment. Results : Body weight gain was observed in all groups, but TAA treated group at 4th week and all treated groups decreased weight compared to the untreated group. As a result of organ weight measurement, organ weight gain due to hepatic injury was observed statistically significantly in TAA-treated group and TAA+Silymarin treated group, and the herbal mixture-treated group showed a tendency to decrease compared to the TAA treated group. Blood biochemistry showed that total cholesterol and very low density lipoprotein cholesterol decreased statistically in TAA+low-dose and high dose herbal mixture treated group compared to the TAA-treated group. Histopathological examination showed that liver abnormalities were not observed in untreated group, liver fibrosis was observed in liver injury with TAA treated and herbal mixture treated group. And, TAA+high dose herbal mixture group showed relaxation tendency on liver calcification compared to the TAA treated group. Conclusions : According to the above results, HML provided hepatoprotective effects on the hepatic injury by reduction of inflammatory responses.

Effect of Elsholtzia splendens Extracts on the Blood Lipid Profile and Hepatotoxicity of the Mice

  • Effects of extracts obtained from the flowers of Elsholtzia splendens on the serum lipid profile and hepatotoxicity in mice were investigated. Female ICR mice were given E. splendens ethanolic extract (ESEs) orally at a dose of 10 or 50 mg/kg BW for 50 days. Significant dose-dependent decreases in triglyceride and low-density lipoprotein (LDL)-cholesterol of serum were observed. In addition, ESEs prolonged the lag-time of LDL oxidation in vitro. In the serum of ICE mice given ESEs orally at 10 and 50 mg/kg BW, the serum levels of aspartate aminotransferase (AST) and lactic dehydrogenase (LDH) increased significantly, while total protein, albumin, creatinine, alanine aminotransferase (ALT), and total bilirubin did not change. Therefore, ESEs may be beneficial to human health, although it has some hepatotoxicity.

Risperidone as a Janus in Mood Disorder (기분장애에서 risperidone의 양면성)

  • To examine the double-faced thymoleptic(antidepressant and antimanic) effects of risperidone in mood disorders, this article reviews the psychotropic-induced mania, thymoleptic effects of antipsychotics, therapeutic effects of risperidone and risperidone(RIS)-induced mania(RIM) in mood disorders, risk factors of RIM, possible neurochemical mechanism of these thymoleptic effects, pathophysiological and clinical significance of thymoleptic effects, and suggestive clinical guideline of RIS in mood disorders. RIS appeared effective for bipolar disorder at a lower dose than that recommended for schizophrenia, especially in the cases of maintenance of mood stabilizers, and gradual titration from low doses. Manic induction/exacerbation can occur by chance during RIS treatment in mood disorders, schizoaffective disorders, and schizophrenias. The possible risk factors for RIM are refractory mood disorder, especially in bipolar I disorder with poor initial response ; refractory schizoaffective disorders, especially in bipolar type with poor initial response ; refractory chronic schizophrenias, especially with initial responses ; psychotic features ; higher initial doses ; rapid titration ; combined therapy with antidepressants in refractory depression ; and RIS monotherapy in mania/hypomania. RIS is a drug that preferentially block 5-HT2 receptors. The effects of low dose are due mainly to the blockade of 5-HT2 receptors. There are more gradual increase in D2 blockade with increasing dose and this D2 blocking properties become apparent at higher doses. This may be related to a modulation of dopaminergic transmission by 5-HT2 antagonism at lower doses with the direct action of RIS on DA receptors coming into play at higher dose. The serotonergic antagonistic effect may be important for its effects on depressive symptoms. This, together with adequate blo-ckade of D2 receptors, may not necessarily lead to destabilization of mood disorder, but rather to more therapeutic effects. Therefore, this dose-receptor affinity relationship with both antidepressant and antimanic effects according to treatment duration can explain a continuum of antidepressant effect, antimanic effect, behavioral stimulation, and manic/hypomanic induction/exacerbation. It was the recognition of a useful psychiatric side effects by a thoughtful observer with fertile minds that led to their ultimate utilization as psychotropic drugs, i.e., phenothiazine, MAOI, TCA, and lithium. And, in vivo pharmacological challenge by novel psychotropics, as a neurochemical probe, with more specific actions is a useful tool to select pharmacologically homogeneous subgroup of the same phenotypical(clinical) condition, to further study the unknown underlying pathogenesis of various mental illnesses. Finally, RIS may be a useful alternative or adjunctive drug for patients with mood disorders without psychotic features or refractory to treatment with standard antipsychotic drugs. The more conservative doses(tirated slowly from 1-3 mg/d) of RIS, and maintenance of mood stabilizer in the cases with risk factors of RIM are recommended in mood disorder.

Anti-Obesity Effect of Pine Cone (Pinus koraiensis) Supercritical Extract in High-Fat Diet-Induced Obese Mice (고지방식이로 유도한 비만 Mice에서 잣송이 초임계 추출물의 항비만 효과)

  • The present study investigated the anti-obesity effect of pine cone (PC, Pinus koraiensis) supercritical extract in high-fat diet (HFD)-induced obese mice. Male C57BL/6J mice were treated with HFD, HFD+catechin, and HFD+PC [two different doses, 20 mg/kg body weight (b.w.) and 100 mg/kg b.w.] in each AIN93G supplement for 8 weeks. Treatment of HFD mice with both low and high doses of PC significantly reduced body weight gain compared to HFD mice. Liver weight of mice was reduced in both the low and high dose PC-supplemented groups (24.19% and 19.83%, respectively). Total adipose tissue weight of mice was reduced in both the low and high dose PC-supplemented groups (45.54% and 62.66%, respectively). Serum total cholesterol, triglyceride, LDL cholesterol, and HDL cholesterol were reduced in the low and high dose PC-supplemented groups, and ratios of HDL cholesterol to LDL cholesterol increased by 94.55% in the high dose PC-supplemented group. Serum leptin was significantly reduced in the low and high dose PC-supplemented groups (28.14% and 62.72%, respectively). These results were supported by genetic expression of protein and enzymes related to lipid metabolism assessed by real-time PCR. There was significant reduction of lipid regulatory transcription factors such as $PPAR-{\gamma}$, C/EBP, and SREBP and lipid enzymes such as fatty acid synthesis and lipoprotein lipase in the low and high dose PC-supplemented groups. However, there was no statistical difference between low and high dose PC treatments. These results suggest that pine cone supercritical extract supplementation is able to regulate serum lipid profiles by reducing total cholesterol, triglyceride, and LDL cholesterol levels, followed by regulation of expression of lipid metabolic factors, resulting in reduction of weight gain in HFD-induced obese mice.

  • The high efficiency moderated-type neutron spectrometer and doseequivalent counter were developed for the measurement of low level environmental neutrons. By using these detectors, the neutron energy spectra and dose equivalent rates due to skyshine effect were measured in the environment surrounding the accelerator facilities and also the altitude variation of cosmic ray neutrons in the aircraft flying over Japan.

The effect of low-dose intravenous bisphosphonate treatment on osteoporosis in children with quadriplegic cerebral palsy

  • Purpose: Quadriplegic children with cerebral palsy are more susceptible to osteoporosis because of various risk factors that interfere with bone metabolism. Pamidronate is effective for pediatric osteoporosis, but there are no guidelines for optimal dosage or duration of treatment in quadriplegic children with osteoporosis. We aimed to evaluate the efficacy of low-dose pamidronate treatment in these patients. Methods: Ten quadriplegic patients on antiepileptic drugs (6 male, 4 female patients; mean age, $10.9{\pm}5.76years$), with osteoporosis and gross motor function classification system level V, were treated with pamidronate (0.5-1.0 mg/kg/day, 2 consecutive days) every 3-4 months in a single institution. The patients received oral supplements of calcium and vitamin D before and during treatment. The lumbar spine bone mineral density (BMD) z score and biochemical markers of bone metabolism were measured regularly during treatment. Results: The main underlying disorder was perinatal hypoxic brain damage (40%, 4 of 10). The mean cumulative dose of pamidronate was $4.49{\pm}2.22mg/kg/yr$, and the mean treatment period was $10.8{\pm}3.32months$. The BMD z score of the lumbar spine showed a significant increase from $-4.22{\pm}1.24$ before treatment to $-2.61{\pm}1.69$ during treatment (P=0.008). Alkaline phosphatase decreased during treatment (P=0.037). Significant adverse drug reactions and new fractures were not reported. Conclusion: Low-dose pamidronate treatment for quadriplegic children with cerebral palsy increased lumbar BMD and reduced the incidence of fracture.

Effects of Low Dose Ionizing Radiation on the Growth and Yield of Soybean Cultivars (저선량의 이온화방사선이 대두품종의 생육과 수량에 미치는 효과)

  • To investigate the hormetic effect of the low dose ${\gamma}-ray$, soybean(Glycine max L.) seeds of three cultivars (Hwangkeumkong, Bansakong, S900 cv.) were irradiated with the dose of 0.5∼20.0 Gy and cultivated in the experimental field. The hormetic effects of the low dose ${\gamma}-ray$ on the growth of field experiment were different from each cultivar, that is, more effective in the S900 cultivar of black seedcoat color than in the Hwangkeumkong and Bangsakong of yellow ones. The germination rate of S900 cultivar increased about 30% in the 0.5Gy irradiated group compared with that of the control. The plant height of the 4.0Gy irradiated group in both Bangsakong and S900 cultivar increased 15% more than that of the control. Grain yield and the number of pod in 1.0 Gy and 4.0 Gy irradiated group of S900 cultivar and that of 2.0 Gy irradiated group of Hwangkeumkong increased 25 and 35% on the average, respectively.
