Effect of the Cultivated Fruit body of Phellinus linteus on Low Density Lipoprotein Oxidation (배양상황버섯의 저밀도지질단백질 (LDL)산화에 미치는 영향)

  • Phellinus linteus (Hymenocaetaceae) has been used for the treatment of gastric cancer, noninsulin dependant diabetes, diarrhea, and menstrual irregularity. The antioxidative effect of cultivated fruit body of Phellinus linteus on low density lipoprotein oxidation was investigated. The MeOH and water extracts were examined by TEARS assay on human plasma LDL in vitro system. The MeOH ex. showed antioxidative activity, and then was fractionated into Precipitates, Water Insoluble fr., Water fro and ether fro The results showed that the lipophilic fractions, Water Insoluble fr. and Ether fr. of cu 1-tivated fruit body of Phellinus linteus, inhibited the oxidative modification of LDL.

Inhibition effects of flavonoid on Oxidation of Human Low Density Lipoprotein

  • This study was designed to investigate the antioxidative activity of a substance isolated from. The antioxidative activity of procyanidine was higher than that of dl-tocopherol and BHA on low-density lipoprotein (LDL) oxidation by thiobarbituric acid reactive substance (TBARS). Procyanidine inhibited the copper-mediated oxidation of human LDL in a dose dependent manner with almost complete inhibition at $60{\mu}g/mL$. Procyanidine at a concentration of $80{\mu}g/mL$ in hibited oxidation of LDL induced by J774. LDL oxidized by copper-mediated or cell-induced oxidation was degraded at a much greater rate than native LDL. These results suggested the importance of further research to procyanidine in the investigation of atherosclerosis and free radical-induced injury.

  • Sulfur containing compounds in garlic have all be used as one of the traditional folk medicine as well as food source. The present study was performed to investigate the antioxidative compounds of 1-methyl-1-cysteine, dimethyl trisulfide and 2-vinyl-4H-1,3-dithiin. The antioxidative activity of sulfur containing compounds on human LDL was investigated by monitoring a thiobarbituric acid substances (TBARS). Sulfur containing compounds inhibited on oxidation of LDL mediated by $CuSO_4$ and macrophages in dose dependent manner with almost completely inhibition at $80{\mu}g/ml$. Antioxidant activities of sulfur containing compounds on LDL oxidation were 2-vinyl-4H-1,3-dithiin, 1-methyl-1-cysteine, and dimethyl trisulfide in order. Inhibitory effects of sulfur containing compounds on oxidation of LDL mediated by $CuSO_4$ and macrophages were degraded at much greater rate than native LDL, the LDL oxidation process was arrested as shown by the lower conjugated dienes formation at the concentration of $60{\mu}g/ml$. Sulfur containing compounds in garlic revealed at high antioxidative activity at low physiological concentration for human LDL oxidation in vitro specially, it was indicated that the antioxidative activity of 3-viny l-4H-1,2-dithiin was higher than that of the other sulfur containing compounds.

Low-density Lipoprotein Improves Motility and Plasma Membrane Integrity of Cryopreserved Canine Epididymal Spermatozoa

  • Cryopreservation of caudal epididymal spermatozoa is an effective technique to conserve genetic potentials of superior dogs when it is not possible to collect ejaculated spermatozoa. Although hen egg yolk is commonly supplemented into the semen extender, active substances within the egg yolk which protect sperm against cryoinjury remain to be discovered. Among its compositions, low-density lipoprotein (LDL) has been reported to have a cryoprotective property for sperm cryopreservation. However, the effects of LDL on dog epididymal spermatozoa during cryopreservation have not yet been investigated. This study aimed to investigate the effects of LDL on epididymal spermatozoa quality following cryopreservation and thawing. After routine castration of 12 dogs, caudal epididymides from individuals were separated from the testes and cut into a few pieces in a Tris-buffer. Spermatozoa recovered from each sample were examined at once for sperm quality and divided into six groups of extender: no LDL, 20% egg yolk, 4%, 8%, 16%, and 24% LDL, before cryopreservation. The sperm aliquots were then equilibrated and conventionally frozen. After thawing, sperm motility, morphology, plasma membrane integrity, and acrosome integrity were evaluated. The results revealed that 4% LDL and 20% egg yolk yielded significantly higher sperm motility (57.69% and 52.69%, respectively, p<0.05) than other LDLs. In addition, 4% LDL yielded the significantly highest plasma membrane integrity (70.54%, p<0.05). In conclusion, the supplementation of 4% LDL in Tris-glucose extender could be applied for cryopreservation of canine epididymal spermatozoa.

Effects of Opuntia ficus-indica Complex on Lipid Metabolism in Streptozotocin-induced Diabetic Rats (손바닥선인장 복합물이 Streptozotocin으로 유발된 당뇨 쥐의 지질대사에 미치는 효과)

  • This study was conducted to examine the effects of Opuntia ficus-indica complex (OF) on the lipid metabolism, bile acid in feces, alanine aminotransferase (ALT) activity, aspatate aminotransferase (AST) activity, composition of urine and expression of cholesterol related mRNA in streptozotocin (STZ)-induced diabetic rats. Thirty two male Sprague-Dawley rats were randomly divided into non-diabetic control (NC), diabetic control (DC), diabetic OF of 2% (OF-2) and diabetic OF of 5% (OF-5), then each group was fed for 3 weeks. Plasma total cholesterol, non-esterified fatty acids (NEFA), total cholesterol, low density lipoprotein (LDL), very low density lipoprotein (VLDL) were decreased significantly (p<0.05) in OF-5 group compared to DC, but high density lipoprotein (HDL) was not changed. AST and ALT were also reduced and bile acid excretion was improved. Composition of urine in OF-5 was almost same in NC. The expression of cholesterol $7{\alpha}$-hydroxylase (CYP7A1), 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA-R), Low density lipoprotein receptor (LDL-R) mRNA indicated that feeding OF have the effects of cholesterol decreation in plasma by synthesis of bile acid from cholesterol. These results provide experimental evidence about improved lipid metabolism of the OF feeding in the STZ-induced diabetic rats.

Dietary Lipid, Plasma Lipoprotein and Fatty Acid Composition of Young Korean Women (일부 젊은 여성의 지질 섭취와 혈장 지단백 및 지방산 조성에 관한 연구)

  • This study was conducted to investigate dietary lipid intakes and the plasma lipoprotein levels and fatty acid composition of eight healthy young Korean women for 7 days. After an overnight fast, blood samples were taken on the last day of dietary survey. The plasma were fractinated to very low-density lipoprotein(VLDL), low-density lipoprotein(LDL), and high-density lipoprotein(HDL) by ultracentrifugation. From each fraction, the content of triglyceride (TG), phospholipid(PL), cholesterol(CHOL), free fatty acid(FFA) and protein were determined. Fatty acid composition of total plasma lipid was alos analyzed. The subjects consumed 34.7$\pm$2.8g of fat daily. The ingested amounts of EPA and DHA were 0.2 and 0.4g/day, respectively. They also consumed 112.2$\pm$12.9mg of cholesterol per day. The concentrations of VLDL, LDL and HDL in the plasma were 66.5$\pm$6.1, 114.0$\pm$8.8 and 129.4$\pm$5.1mg/dL, respectively, so the percentages of VLDL, LDL, and HDL were 20.7, 44.8 and 34.5%, respectively. The VLDL was composed of 27.5mg/dL of TG, 39.7 of PL, 58.8 of CHOL, 3.1 of FFA, and 22.4 of protein. The HDL had 10.7mg/dL of TG, 23.7 of PL, 18.7 of CHOL, 2.6 of FFA, and 73.7 of protein. In the plasma, linoleic acid was the most abundant fatty acid, followed by palmitic and oleic acid, the percentages of these 3 fatty acids were 30.8, 23.4 and 17.7%, respectively. The result indicated that the plasma levels of lipoprotein in the healthy young Korean women were slightly low and the levels of EPA and DHA were relatively high, compared to the data of other investigatiors. This might be due to the fact that they consumed less fat and cholesterol and higher EPA and DHA.

Effect of Low Density Lipoprotein (LDL) on Motility, Viability, Membrane Integrity and Acrosome Integrity of Frozen-thawed Sperm in Korean Jeju Black Bull (제주흑우 동결정액 제조 시 Low Density Lipoprotein (LDL)의 첨가가 동결 융해 후 정자의 성상에 미치는 영향)

  • This study was designed to determine whether low-density lipoporoteins (LDL) extracted from egg yolk in extender improve the function of Korean Jeju Black Bull semen. The semen was cryopreserved with 5% ethylene glycol (EG) or 7% glycerol (G) extenders containing 10% egg yolk (EY), 4% LDL and 5% EY or 8% LDL. Frozen-thawed sperm were evaluated sperm motility, viability, membrane integrity and acrosome integrity. Post-thawed sperm motility has been significantly higher (p<0.05) in 4% LDL + 5% EY ($69.00%{\pm}4.18$; EG and $63.00%{\pm}9.75$; 7% G) than 8% LDL ($57.00%{\pm}5.70$; EG and $52.00%{\pm}4.47$;G). Treatment of 4% LDL + 5% EY-EG ($66.85%{\pm}5.06$) has been significantly improved sperm viability compared to other treatments except 10% EY - EG. Moreover, in membrane integrity, swollen sperm ratio has been only significantly increased (p<0.05) in 4% LDL + 5% EY - EG ($64.65%{\pm}6.10$) among all treatments. In assess to detect acrosome integrity, especially, AR pattern ratio has been significantly decreased (p<0.05) in 4% LDL + 5% EY - EG among all treatments. In sperm viability as time passes, between 4% LDL + 5% EY and 10% EY, there was no significant difference, but 8% LDL was significantly decreased sperm viability in EG (1 and 2 hrs) and G (30 min, 1, 2, 5 and 12 hrs) extender. However, there were no significant differences among all treatments except 8% LDL-G in sperm membrane integrity. 8% LDL-G has been significantly decreased swollen sperm ratio at 5 hrs after thawed. It is concluded from these results that 4% LDL + 5% EY to the freezing extender showed more positive effect on the frozen-thawed spermatozoa in Korean Jeju Black bull.

Enhanced Uptake of Modified Low-Density Lipoprotein by Eicosapentaenoic Acid-Treated THP-1 Macrophages

  • Animal and clinical studies as well as epidemiological data have provided convincing evidence that n-3 polyunsaturated fatty acids can protect against atherosclerosis. However, the effects of the fatty acids on atherogenesis are contradictory. This discrepancy could derive from great susceptibility of the fatty acids to oxidation. We investigated the effect of eicosapentaenoic aced(EPA) on cellular atherogenesis via the scavenger receptor of THP-1 derived macrophages. THP-1 cells were fully differentiated into macrophages by incubating with phorbol 12-myristate 13-acetate for seven days. Atherogenic features of EPA were compared by subsitituting for linoleic acid (LA). Macrophages were also incubated without treatment of the fatty acids as controls. EPA (5-50 nmol/mL) was not cytotoxic and did not measurably induce cellular oxidation compared to bovine serum albumin (BSA) vehicle or identical doses of LA. EPA increased macrophage uptake and degradation of acetylated LDL(AcLDL) up to 14% and 88%, respectively. EPA increased markedly total cellular sterol synthesis and heparin-releasable lipoprotein lipase activity of macrophages, indicating that EPA may enhance accumulation of cellular cholesteryl ester and possibly facilitate formation of foam cells. These results demonstrate that EPA promotes the modified LDL-triggered atherosclerotic process by the modulation of the scavenger receptor and the activation of LPL in macrophages.

Reductive effect of body weight in rats fed a high-fat diet by Sense-line

  • Sense-line (SL), a dietary functional food, is our invention for weight loss. To evaluate such an effect of SL, we analyzed the plasma levels of total cholesterol, low-density lipoprotein (LDL) cholesterol, high-density lipoprotein (HDL) cholesterol, and weight changes after rats were fed on high fat diet with SL for 20 days. Plasma total cholesterol level and LDL-cholesterol level was decreased by 52% and 91.89% in 5% SL treated group and by 13% and 51.45% in 10% SL treated group, respectively. But HDL-cholesterol was not changed. In addition, the weight was significantly lower in SL group than in high-fat diet group (P<0.05). Our findings indicate that SL may contain compounds with actions, which can treat blood circulatory trouble as well as overweight.

The Change of the Important Blood Factors According to the Recovery of Motor Function with Ischemic Stroke Patients (허혈성 뇌졸중 환자의 운동기능회복에 따른 중요 혈액인자들의 변화)

  • Background: This study had been carried out with 18 ischemic stroke patients as its object for about eight months from October, 2006 to May, 2007 in order to observe the recovery of motor function and the change of important blood factors according to the different quantitative exercises. Methods: Subjects were assigned randomly either experimental group (n=19) or the control group (n=19), when the study began the halfway on this study dropout 20 patients, and final subjects remained experimental group's 9 patients and control group's 9 patients. Both groups received thermotherapy and functional electrical stimulation (FES), also taken different quantitative exercise therapy (experimental group 180 minutes, control group 80 minutes). Subjects were assessed for upper and lower extremities motor function Fugl-Meyer Scale; FMS), blood test (white blood count; WBC, low density lipoprotein -cholesterol; LDL-C, high density lipoprotein-cholesterol; HDL-C, Troponin) during pretest, after 2 months, after 3 months. Results: The results of this study were as follows; 1. FMS has no statistically significant difference with intergroup(p>.05). But there was a statistically significant difference with each groups (p<.05). 2. WBC has no statistically significant difference with intergroup (p>.05). But there was a statistically significant difference in control group (p<.05), without experimental group (p>.05). 3. LDL-C has no statistically significant difference with intergroup (p>.05). But there was a statistically significant difference in control group (p<.05), without experimental group (p>.05). 4. HDL-C has no statistically significant difference with intergroup (p<.05). But there was a statistically significant difference with each groups (p>.05). 5. Troponin Ⅰ has no statistically significant difference with intergroup (p>.05). Also there was no statistically significant difference with each groups (p>.05). Conclusion: These findings suggest that different quantitative exercises has no effect on FMS, LDL-C, HDL-C, WBC, Troponin Ⅰ with ischemic stroke patients. But the treatment period that there's less correlation between the recovery of motor function and the different quantitative exercise, also less correlation between the change of important blood factors and the different quantitative exercises with ischemic stroke patients.
