Stevia rebaudiana (Asteraceae), a perennial plant, has been used as a low-calorie sweetener and is being developed as a therapeutic agent for diabetes, hypertension, myocardial diseases, and microbial infections. Despite the common use of its leaves and stem, the bioavailability of the components present in S. rebaudiana flowers, when used as ingredients of cosmetics, has not been well investigated. Herein, we investigated the antioxidative and antimelanogenic effects of an aqueous extract of S. rebaudiana flowers (Stevia-F). Total flavonoid and phenolic content in Stevia-F were determined to be $8.64{\pm}0.23mg$ of quercetin equivalents/100 g and $631.5{\pm}2.01mg$ of gallic acid equivalents/100 g, respectively. The $IC_{50}$ values of Stevia-F for reducing power, and 2,2-diphenyl-1-picryl-hydrazyl-hydrate radical, hydrogen peroxide, and nitric oxide scavenging activities were 5541.96, 131.39, 466.34, and $10.44{\mu}g/mL$, respectively. Stevia-F showed inhibitory effects on the tyrosinase ($IC_{50}=134.74{\mu}g/mL$) and ${\alpha}$-glucosidase ($IC_{50}=114.81{\mu}g/mL$) activities. No significant cytotoxicity of Stevia-F was observed in B16F10 cells, treated with up to $100{\mu}g/mL$ of the extract for 24 and 48 h (p > 0.05). Stevia-F ($1-100{\mu}g/mL$) suppressed ${\alpha}$-melanocyte stimulating hormone-induced melanin production in B16F10 cells (p < 0.05) and also inhibited the cellular tyrosinase activity (p < 0.05). Overall, our results show that Stevia-F possesses potential for inhibiting tyrosinase and ${\alpha}$-glucosidase activities and has significant antioxidant capacity. The antimelanogenic potential of Stevia-F should extend the usage of S. rebaudiana flowers in the development of skin-whitening products.
Conventionally, immunotoxins have been produced as a single polypeptide from fused genes of an antibody fragment and a toxin. In this study, we adopted a unique approach of chemical conjugation of a toxin protein and an antibody fragment. The two genes were separately expressed in Escherichia coli and purified to high levels of purity. The two purified proteins were conjugated using a chemical linker. The advantage of this approach is its ability to overcome the problem of low recombinant immunotoxin production observed in some immunotoxins. Another advantage is that various combinations of immunotoxins can be prepared with fewer efforts, because the chemical conjugation of components is relatively simpler than the processes involved in cloning, expression, and purification of multiple immunotoxins. As a proof of concept, the scFv of trastuzumab and the PE24 fragment of Pseudomonas exotoxin A were separately produced using E. coli and then chemically crosslinked. The new immunotoxin was tested on four breast cancer cell lines variably expressing HER2. The chemically crosslinked immunotoxin exhibited cytotoxicity in proportion to the expression level of HER2. In conclusion, the present study revealed an alternative method of generating an immunotoxin that could effectively reduce the viability of HER2-expressing breast cancer cells. These results suggest the effectiveness of this method of immunotoxin crosslinking as a suitable alternative for producing immunotoxins.
Purpose : This study aimed to analyze the antibacterial activity of Liriope platyphylla ethanol extract (LPEE) against Streptococcus mutans and Porphyromonas gingivalis and to validate its potential for the prevention and treatment of dental caries, gingivitis, and periodontal disease. Methods : To verify the antibacterial effect of L. pulsatilla ethanolic extract (LPEE) against S. mutans and P. gingivalis, the disk diffusion method was used to determine the inhibition zones at concentrations of 50, 100, 200, and 300 mg/㎖. To determine the minimum inhibition concentration (MIC), the final dose of LPEE was .2, .4, .8, 1.6, 2.5, and 5.0 mg/㎖, and the minimum bactericidal concentration (MBC) was determined based on the MIC results. To confirm the growth inhibitory effect of LPEE on both pathogens, the absorbance was measured at 600 nm after each incubation for 0, 3, 6, 12, and 24 hr at concentrations of .8, 1.6, 2.5, and 5.0 mg/㎖. Results : The cytotoxicity of LPEE was evaluated and the cell viability was more than 70 % at 400 mg/㎖. Therefore, concentrations of 50, 100, 200, and 300 mg/㎖ were used in this study. The antimicrobial effect against S. mutans was seen at 100 mg/㎖ and grew in a concentration-dependent manner, while P. gingivalis was effective at 50 mg/㎖ with the dose dependency. The MIC was .8 mg/㎖ for both strains, and the MBC was 1.6 mg/㎖ with the same results. The growth inhibitory effect of LPEE on S. mutans and P. gingivalis was observed, even at low concentrations. Conclusion : The antibacterial effect of LPEE was evaluated through the analysis of MIC, MBC, and growth inhibition effect on S. mutans and P. gingivalis, which suggests LPEE might have the possibility of utilization as a preventive and therapeutic composition for oral diseases.

The CRISPR-Cas9 system has significantly advanced regenerative medicine research by enabling genome editing in stem cells. Due to their desirable properties, mesenchymal stem cells (MSCs) have recently emerged as highly promising therapeutic agents, which properties include differentiation ability and cytokine production. While CRISPR-Cas9 technology is applied to develop MSC-based therapeutics, MSCs exhibit inefficient genome editing, and susceptibility to plasmid DNA. In this study, we compared and optimized plasmid DNA and RNP approaches for efficient genome engineering in MSCs. The RNP-mediated approach enabled genome editing with high indel frequency and low cytotoxicity in MSCs. By utilizing Cas9 RNPs, we successfully generated B2M-knockout MSCs, which reduced T-cell differentiation, and improved MSC survival. Furthermore, this approach enhanced the immunomodulatory effect of IFN-r priming. These findings indicate that the RNP-mediated engineering of MSC genomes can achieve high efficiency, and engineered MSCs offer potential as a promising therapeutic strategy.
