다시마와 다시마 요구르트의 변비해소 효과 (Effects of Sea Tangle Extract and Sea Tangle Yogurt on Constipation Relief)

  • In an effort to make a functional and stable yogurt, this study investigated the improvement effects of sea tangle extract and sea tangle yogurt on intestinal function. The intestinal improvement effect of the extract was measured by the charcoal meal transit method, employing Balb/C mice. And constipation relief was compared utilizing the loperamide-induced constipation method, employing SD rats. Charcoal meal transit was remarkably increased in the mice receiving sea tangle extract as compared to the controls. The constipation relief effects of the sea tangle and sea tangle yogurt were evaluated by measuring fecal amounts in the rats after adding them to water. The fecal contents increased remarkably in the sea tangle administered rat groups as compared to the control group. In addition, different yogurt samples were used to evaluate the characteristics of the sea tangle yogurt. During storage, pH slightly decreased in the yogurt with sea tangle as well as without. At the same time, acidity slowly increased as the storage duration increased. As time elapsed, the amounts of viable cells increased in both yogurts (with and without sea tangle). In the sensory evaluation, significant differences were shown between the sea tangle yogurt and the control for color, flavor, sweetness, and overall quality. Overall, based on the combined results of the intestinal function effects and sensory evaluation, the 0.25% sea tangle yogurt proved to be superior.

  • Objectives : The aim of this study was to compare the effect of acpuncture and electroacupuncture of low(EA(L)) and high(EA(H)) frequencies at San Yin Jiu($SP_6$) and sham point in rats. We made suppressed state of intestinal motility with loperamide in rat and carried out needle retention acupuncture, low frequency electro-acupuncture and high frequency electro-acupuncture at ($SP_6$) in rat devided into pre-treatment group and post-treatment group. We fed charcoal to them after the treatment and measured the travel rate of charcoal in the gastrointestine track to analyze which treatment affected more in intestinal hypermotility Results : 1. Examining intestinal motility of normal rat treated with electro-acupuncture at $SP_6$, high frequency electro-acupuncture significantly increased intestinal motility. 2. Pre-treated with acupuncture at $SP_6$ significantly suppressed intestinal motility. 3. Post treatment with acupunture and low frequency electro-acupuncture at $SP_6$ showed significant increase of intestinal motility. Conclusions : As folowing study there were 21 groups to find out which treatment showed significant increase of intestinal motility and pre-treated acupuncture, post-treated with acupunture and low frequency electro-acupuncture at $SP_6$ was highly significant with control at the figure than any other groups.

초임계 열처리된 무 복합추출물의 장내세균총 및 장질환 예방 효과 (The Prevention of Gut Microbiome and Intestinal Diseases from Supercritical Heat-treated Radish Complex Extracts)

  • 본 연구는 한국산 초임계 열처리된 무 복합 추출물을 이용하여 장내세균의 생육, 특히 식중독 및 위염, 장염 등 감염성 유해균과 정장효과를 나타내는 유익한 세균군의 생육에 미치는 영향을 알아보고자 하였다. 흰쥐를 3개 시험구 즉 정상대조군, Loperamide 투여군, 초임계 열처리된 무 복합추출물(HRE) 투여군 등으로 나누어 동물실험을 수행하면서 장내 유익한 세균의 생육에는 아무런 영향을 주지 않으면서 유해균의 생육을 저해함으로써 장기능 개선 및 세균성 장질환에 효과적이고 안전한 건강기능식품 조성물로도 유용하게 사용할 수 있다는 결과를 얻었다. 특히 무는 오랫동안 복용 시에도 전혀 부작용을 나타내지 않는 안전한 식품으로 장기간 꾸준히 복용하는 것에 의해 장내 세균총 및 세균성 장질환에 효과를 얻을 수 있다는 결론을 얻을 수 있었다.

Effect of yoghurt with a Bifidobacteria enhancer and dietary fiber on irritable bowel syndrome

  • This study was carried out to investigate the effects of supplementation with a Bifidobacteria enhanced yogurt (BE0623 yogurt), which includes Bifidobacterium lactis BB12, Lactobacillus acidophilus, Streptococcus thermophilus, and Bifidobacterium lactis, in the management of irritable bowel syndrome (IBS) using animal models and clinical trials. In a rat study, a loperamide-treated group (LOP) showed reduced water content in fecal pellets but showed an increased number of fecal pellets in the distal colon. In addition, the BE0623 yogurt (L-BE0623Y) group had the fewest fecal pellets in the distal colon. Regarding the serum lipid parameters, the LOP group had a high-density lipoprotein (HDL)/total cholesterol ratio that was 43% lower than that of a normal water group (NOR), but the outcome for the L-BE0623Y group was 27% lower than the NOR group. In a human study, 116 adults with IBS were sampled as subjects and fed 300 mL of yogurt per day for an eight week period. There was an IBS improvement in the L-BE0623Y and commercial yogurt (L-CY) groups, though flatulence, stool consistency and frequency of defecation outcomes were also noted. Specifically, the L-BE0623Y treatment group showed significant effects with regard to defecation duration and urgency after the consumption of the yogurt used in this study. These results suggest that the Bifidobacteria-enhanced yogurt has superior effects with regard to relieving loperamide-induced constipation in rats and that regular consumption of L-BE0623Y is effective to improve IBS in humans.

Comparison of laxative effects of fermented soybeans (Cheonggukjang) containing toxins and biogenic amines against loperamide-induced constipation mouse model

  • BACKGROUND/OBJECTIVES: Cheonggukjang is a traditional fermented soybean paste with significant health-promoting effects. On the other hand, there have been insufficient studies on the safety and efficacy of Cheonggukjang, which is produced using traditional methods containing toxins and biogenic amines (BAs). This study compared the laxative effect of Cheonggukjang, containing high or low levels of toxins and BAs (HTBC or LTBC) in a loperamide (Lop)-induced constipation mouse model. MATERIALS/METHODS: To induce constipation, Lop (5 mg/kg) was administered orally to ICR mice twice a day for 4 days, and the dose was increased to 8 mg/kg after a 3-day rest period. Cheonggukjang (500 mg/kg, HTBC, or LTBC respectively) was administered for four weeks before the Lop treatment. RESULTS: The number of stools, fecal weight, water contents, gastrointestinal transit, and histological alterations were recovered significantly in the HTBC or LTBC groups. HTBC and LTBC administration did not induce significant changes in body weight, dietary intake, and behavior. The opioid-receptor downstream signaling pathway in colon tissues was also evaluated. The c-Kit, stem cell kinase, and mitogen-activated protein kinases subfamilies, including extracellular signal-regulated kinase 1/2, c-Jun N-terminal kinases, and p38, were all downregulated in the HTBC or LTBC-administered mice colon compared to the Lop group. CONCLUSION: These results show that Cheonggukjang, containing high levels of toxins and BAs, have a similar laxative effect in a mouse model of Lop-induced constipation.

과채복합추출물의 장내 마이크로바이옴 변화 분석 (Analysis of Intestinal Microbiome Changes in Fruit and Vegetable Complex Extracts)

  • 우리는 Loperamide로 유도한 변비 Sprague-Dawley 랫드 모델에서 프락토올리고당 및 과채류복합 추출물을 2 주간 경구 투여 한 후 랫 분변의 그룹 기간별로 수집 한 후 장내 마크로바이옴 변화 경향을 분석하였다. 프락토올리고당 및 과채복합추출물(FVCE)에 대한 미생물 군집 분석을 16S rDNA 클로닝 및 pyrosequencing을 통해 수행하여 표준화 및 체계화를 위한 기초 데이터를 얻었다. 과채복합추출물(FVCE) 제조 공정은 원핵생물 군집에 대한 미생물 분석을 통해 문 수준에서 미생물 verrucomicrobiota의 약간의 차이가 우세한 것으로 나타났다. 속 수준에서는 prevotella와 muribaculaceae가 종 수준에서 더 많은 차이를 보였다. 이러한 결과는 사용된 미생물 군집이 생산되는 과일 및 채소 복합 추출물(FVCE)의 품질에 영향을 미친다는 것을 시사한다고 할 수 있다. 따라서, 일관된 품질의 과일 및 채소 복합 추출물(FVCE)을 생산하기 위해서는 안정적인 미생물 군집이 유지되어야 한다.

저항전분 투여가 Loperamide 유도 변비 쥐의 대장환경과 혈청지질에 미치는 효과 (Effect of Resistant Starch on the Large Bowel Environment and Plasma Lipid in Rats with Loperamide-Induced Constipation)

  • 본 연구는 저항전분이 Loperamide 유도 변비 쥐의 대장내 환경과 혈중지질 농도에 미치는 영향에 대해 실험 조사하였다. 실험동물은 Sprague-Dawley계 수컷 쥐를 사용하였으며 60마리를 정상 셀룰로오스군, 변비 셀룰로오스군, 변비 펙틴군, 변비 RS2군, 변비 고RS2군, 변비 RS3군의 6개 군으로 나누었다. AIN-76 식이를 기본으로 각각 다른 식이 섬유원 즉, 셀룰로오스, 펙틴, RS2, RS3를 첨가하여 에너지 밀도가 동일한 실험식을 변비 유발 쥐에 5주 동안 투여하여 다음과 같은 결과를 얻었다. RS3 투여군과 고농도 RS2 투여군은 맹장 내용물의 비피더스균 수를 정상대조군에 비해 유의적으로 높이는 효과를 나타내었다. 맹장 내용물의 단쇄지방산총량은 셀룰로오스 군에 비해 펙틴과 저항전분군들에서 유의적으로 높았고, 개별 단쇄지방산은 프로피온산의 경우 펙틴군에서 가장 높았으며 부티르산은 CR3군에서 가장 높았다. 저항전분의 투여는 맹장 내용물의 pH를 셀룰로오스군에 비해 유의적으로 낮추었다. RS3 투여는 대장 관내 점막의 두께를 정상대조군에 비해 증가시키는 경향을 보였고, 점액분비 세포의 활성도를 정상대조군과 비슷한 수준으로 나타내었다. 염증지표 $PGE_2$의 수준은 저항전분 보충군들이 정상셀룰로오스군에 비해서는 높았으나 변비 셀룰로오스군이나 펙틴군에 비해서는 유의적으로 낮았다. 뿐만 아니라 저항전분들의 투여는 종류에 관계없이 변비동물에서 혈중 중성지질과 총 콜레스테롤 농도를 낮추고 HDL-콜레스테롤의 비율을 높임으로써 동맥경화지수를 낮추어 지질대사를 개선하는 유익한 효과를 나타내었다. 이상의 결과들에서 저항성 전분들은 장내 비피더스균과 같은 유용균의 증식을 활성화 시키며 단쇄지방산의 생산을 증가시킴으로써 장내 pH를 낮추는 효과를 나타낼 수 있으며, 또한 대장 점막층의 두께와 점액분비 세포수를 증가시키고 염증지표 수준을 낮춤으로서 변비동물의 대장 내의 환경을 건강하게 유지하고 장 기능을 효과적으로 개선시킬 뿐 아니라 혈중지질의 개선효과를 함께 나타낼 것으로 사료된다.

Analgesic and antidiarrhoeal activities of Treama orientalis Linn. in mice

  • Trema orientalis Linn. is commonly grown in many parts of Bangladesh. Its leaves have been used for analgesic and anti-diarrhoeal activity in traditional medicine. This study evaluates the potential analgesic and anti-diarrhoeal activity of methanol and aqueous extracts of leaves in experimental acetic acid induced writhing and castor oil induced diarrhoea in mice. The aqueous extract of leaves showed significant (P < 0.001) analgesic effect in acetic acid induced writhing in mice at a dose of 500 mg/kg body weight In castor oil induced antidiarrhoeal screening both extract increased latent period (P < 0.025) and decrease the number of stool (P < 0.025) at the dose of 500 mg/kg body weight comparable with that of the standard drug loperamide. The results provide a support for the use of this plant in traditional medicine and suggest its further investigation.

Antinociceptive and antidiarrhoeal activities of Sonneratia caseolaris

  • The crude ethanol extract of leaves of Sonneratia caseolaris Linn. (Sonneratiaceae) was screened for its antinociceptive and antidiarrhoeal activities. The extract produced significant writhing inhibition in acetic acid induced writhing in mice at dose of 250 and 500 mg/kg body weight (P<0.01) comparable to the standard drug diclofenac sodium at the dose of 25 mg/kg of body weight. When tested for its antidiarrhoeal effects on castor oil induced diarrhea in mice, it increased mean latent period and decreased the frequency of defecation significantly at the dose of 500 mg/kg body weight (P < 0.05) comparable to the standard drug loperamide at the dose of 50 mg/kg of body weight. The overall results tend to suggest the antinociceptive and antidiarrhoeal activities of the extract.
