파킨슨병 관련 실험 연구에서 육계의 신경 보호효과에 대한 고찰 (A Review of the Neuroprotective Effects of Cinnamon in Experimental Studies on Parkinson's Disease)

  • Objective: The purpose of this study was to investigate the effect of cinnamon on the treatment of Parkinson's disease (PD) and to introduce its use in Korea. Method: We searched the experimental studies in electronic databases (PubMed, CNKI, Wanfang, CiNii, J-STAGE, Science ON, and OASIS) using the key search terms "cinnamic acid", "cinnamon", "cinnamomum", "Parkinson's disease", "Parkinson disease", "Parkinsonism", and "dopamine". This study only involved experimental studies (in vivo and in vitro) that adopted cinnamon as a single administration and measured indicators relating to Parkinson's disease, including parkin, tyrosine hydroxylase (TH), and dopamine. Results: A Total of 11 literature studies were selected, and they all showed that treatment with cinnamon has a neuroprotective effect. Cinnamon activated neuroprotective factors and restored neurotransmitters and it reduced the rate of oxidative stress and inflammation in neurons. As a result, cell viability was upregulated, while cell apoptosis and neurodegeneration were downregulated. Five in vivo studies, through behavioral tests, also confirmed that cinnamon recovers locomotor function in PD models. Conclusion: We identified that cinnamon is an effective neural protector and improves motor performance in behavioral testing in the experimental PD studies.

Coping with large litters: management effects on welfare and nursing capacity of the sow

  • A number of management issues can be used as drivers for change in order to improve animal welfare and nursing capacity of the hyperprolific sow. Group housing of sows during gestation is a recommended practice from the perspective of animal welfare. Related health issues include reproductive health and the locomotor system. It appears that management of pregnant sows in groups is challenging for a producer and considerable skill is required. We explored the benefits and challenges of group housing, including feeding issues. Increasing litter size requires additional attention to the mammary gland and its ability to provide sufficient nursing for the growing litter. We discuss the fundamentals of mammary development and the specific challenges related to the hyperprolific sow. We also address challenges with the farrowing environment. It appears that the old-fashioned farrowing crate is not only outdated in terms of welfare from the public's perspective, but also fails to provide the environment that the sow needs to support her physiology of farrowing, nursing, and maternal behaviour. Studies from our group and others indicate that providing the sow with a loose housing system adequate in space and nesting material, along with reasonable chance for isolation, can be considered as fundamental for successful farrowing of the hyperprolific sow. It has also been shown that management strategies, such as split suckling and cross fostering, are necessary to ensure proper colostrum intake for all piglets born alive in a large litter. We thus conclude that welfare and nursing capacity of the sow can be improved by management. However, current megatrends such as the climate change may change sow management and force the industry to rethink goals of breeding and, for instance, breeding for better resilience may need to be included as goals for the future.

Ginseng extract and ginsenosides improve neurological function and promote antioxidant effects in rats with spinal cord injury: A meta-analysis and systematic review

  • Spinal cord injury (SCI) is defined as damage to the spinal cord that temporarily or permanently changes its function. There is no definite treatment established for neurological complete injury patients. This study investigated the effect of ginseng extract and ginsenosides on neurological recovery and antioxidant efficacies in rat models following SCI and explore the appropriate dosage. Searches were done on PubMed, Embase, and Chinese databases, and animal studies matches the inclusion criteria were selected. Pair-wise meta-analysis and subgroup analysis were performed. Ten studies were included, and the overall methodological qualities were low quality. The result showed ginseng extract and ginsenosides significantly improve neurological function, through the Basso, Beattie, and Bresnahan (BBB) locomotor rating scale (pooled MD = 4.40; 95% CI = 3.92 to 4.88; p < 0.00001), significantly decrease malondialdehyde (MDA) (n = 290; pooled MD = -2.19; 95% CI = -3.16 to 1.22; p < 0.0001) and increase superoxide dismutase (SOD) levels (n = 290; pooled MD = 2.14; 95% CI = 1.45 to 2.83; p < 0.00001). Both low (<25 mg/kg) and high dosage (25 mg/kg) showed significant improvement in the motor function recovery in SCI rats. Collectively, this review suggests ginseng extract and ginsenosides has a protective effect on SCI, with good safety and a clear mechanism of action and may be suitable for future clinical trials and applications.

The Evaluation of the Acute Toxicity and Safety of Verbenalin in ICR Mice

  • Background: Verbenalin is an iridoid glucoside, which is among the active components of some medicinal herbs such as Verbena officinalis Linn, and Cornus officinalis Siebold and Zucc. Previous studies have confirmed the antioxidant activity and neuroprotective potential of verbenalin. To confirm the safety of verbenalin, an approximate lethal dose was determined based on a single oral dose toxicity study. Methods: Institute of Cancer Research mice were randomly assigned to three verbenalin exposure groups (250, 500, and 1,000 mg/kg) and a control group (5% methylcellulose solution). There were (5 male and 5 female mice per group). Mortality, clinical signs, and body weight were monitored for 14 days, and necropsies were conducted. Results: No mortalities were observed in the control group or the verbenalin 250 mg/kg group, whereas mortalities were observed in the 500 mg/kg and 1,000 mg/kg verbenalin groups. During the observation period, stool abnormalities such as mucous stools were observed. Clinical signs such as loss of locomotor activity were observed in the 500 mg/kg and 1,000 mg/kg verbenalin groups. During the study period, significant changes in body weight were observed in the 500 mg/kg and 1,000 mg/kg verbenalin groups; however, no gross abnormalities were observed at necropsy. Overall, no toxicity was found in the 250 mg/kg group. Conclusion: The approximate lethal dose of verbenalin was estimated to be 500 mg/kg. For a more accurate assessment of the safety of verbenalin, other types of studies such as repeated-dose toxicity studies should also be conducted.

Longevity, tumor, and physical vitality in rats consuming ginsenoside Rg1

  • Background: Effects of the major ginsenoside Rg1 on mammalian longevity and physical vitality are rarely reported. Purpose: To examine longevity, tumor, and spontaneous locomotor activity in rats consuming Rg1. Methods: A total of 138 Wistar rats were randomized into 2 groups: control (N = 69) and Rg1 (N = 69). Rg1 (0.1 mg/kg per day) were orally supplemented from 6 months of age until natural death. Spontaneous mobility was measured by video-tracking together with body composition (dual energy x-ray absorptiometry) and inflammation markers at 5, 14, 21, and 28 months of age. Results: No significant differences in longevity (control: 706 days; Rg1: 651 days, p = 0.77) and tumor incidence (control: 19%; Rg1: 12%, p = 0.24) were observed between the two groups. Movement distance in the control group declined significantly by ~60% at 21 months of age, together with decreased TNF-α (p = 0.01) and increased IL-10 (p = 0.02). However, the movement distance in the Rg1 group was maintained ~50% above the control groups (p = 0.01) at 21 months of age with greater magnitudes of TNF-α decreases and IL-10 increases. Glucose, insulin, and body composition (bone, muscle and fat percentages) were similar for both groups during the entire observation period. Conclusion: The results of the study suggest a delay age-dependent decline in physical vitality during late life by lifelong Rg1 consumption. This improvement is associated with inflammatory modulation. Significant effects of Rg1 on longevity and tumorigenesis were not observed.

Repeated Neonatal Propofol Administration Induces Sex-Dependent Long-Term Impairments on Spatial and Recognition Memory in Rats

  • Propofol is an anesthetic agent that gained wide use because of its fast induction of anesthesia and rapid recovery post-anesthesia. However, previous studies have reported immediate neurodegeneration and long-term impairment in spatial learning and memory from repeated neonatal propofol administration in animals. Yet, none of those studies has explored the sex-specific long-term physical changes and behavioral alterations such as social (sociability and social preference), emotional (anxiety), and other cognitive functions (spatial working, recognition, and avoidance memory) after neonatal propofol treatment. Seven-day-old Wistar-Kyoto (WKY) rats underwent repeated daily intraperitoneal injections of propofol or normal saline for 7 days. Starting fourth week of age and onwards, rats were subjected to behavior tests including open-field, elevated-plus-maze, Y-maze, 3-chamber social interaction, novel-object-recognition, passive-avoidance, and rotarod. Rats were sacrificed at 9 weeks and hippocampal protein expressions were analyzed by Western blot. Results revealed long-term body weight gain alterations in the growing rats and sex-specific impairments in spatial (female) and recognition (male) learning and memory paradigms. A markedly decreased expression of hippocampal NMDA receptor GluN1 subunit in female- and increased expression of AMPA GluR1 subunit protein expression in male rats were also found. Other aspects of behaviors such as locomotor activity and coordination, anxiety, sociability, social preference and avoidance learning and memory were not generally affected. These results suggest that neonatal repeated propofol administration disrupts normal growth and some aspects of neurodevelopment in rats in a sex-specific manner.

증가된 글루타민에 의해 초래되는 뇌신경질환의 초파리 모델에 대한 연구 (Expression of Expanded Polyglutamine Disease Proteins in Drosophila (Drosophila Polyglutamine Disease Models))

  • 목 적 : 폴리글루타민 질환은 해당 발현단백질의 연속되는 글루타민 아미노산 서열이 신장되기 때문에 일어나는 질환군이다. 현 연구는 폴리글루타민 질환 형질전환 초파리 모델들이 환자들 에서와 유사한 장애를 나타내는지 확인하기 위해 수행되었다. 방 법 : 폴리글루타민 질환 (SCA3) 형질전환 초파리를 대상으로 벽을 기어오르는 운동 능력을 검사하였다. 또한 유전학적인 방법을 통해서 아폽토시스를 억제하는 bcl-2 유전자와 화학적 샤페론이 뇌신경의 퇴행에 어떤 영향을 미치는지 확인하였으며 향후의 연구를 위해 척수소뇌 운동실조증 타입 2 (SCA2) 질환을 발현하는 형질전환 초파리를 생산하였다. 결 과 : SCA3 형질전환 초파리에서 신장된 폴리글루타민 배열을 지니는 질환성 초파리의 경우 신경계에서 해당 단백질을 발현하였을 경우 전형적인 운동 능력 상실을 나타냈다. 아폽토시스를 억제하는 유전자인 bcl-2를 함께 발현했을 경우, 신장된 단백질의 유독한 영향을 약화시키지 못했지만, 화학적 샤페론인글리세롤의 경우 적어도 눈에서의 유독한 영향은 억제하는 것으로 보인다. 본 연구진에 의해 개발된 SCA2 형질전환 초파리의 경우 유해 단백질의 발현 정도가 낮아서 정확한 분석이 어려웠다. 결 론 : SCA3 형질전환 초파리는 환자들에서 발견되는 운동실조증을 보였다. 글리세롤과 같은 화학적 샤페론이 현재 치료가 전무한 이 종류의 질환군의 치료에 효과적일 것으로 사료된다.

20대 여성의 신발종류에 따른 족저압 영역별 비교 연구 (A comparison study for mask plantar pressure measures to the difference of shoes in 20 female)

  • The purpose of this study was to investigate the test-retest of plantar pressures using the F-Scan system over speeds and plantar regions. 6 healthy female subjects in 20's were recruited for the study. Plantar pressure measurements during locomotor activities can provide information concerning foot function, particularly if the timing and magnitude of the loading profile can be related to the location of specific foot structures such as the metatarsal heads. The Tekscan F-Scan system consists of a flexible, 0.18mm thick sole-shape having 1260 pressure sensors, the sensor insole was trimmed to fit the subjects' right. left shoes - sneakers shoes & dress shoes. It was calibrated by the known weight of the test subject standing on one foot. The Tekscan measurements show the insole pressure distribution as a function of the time. This finding has important implications for the development of plantar pressure test protocols where the function of the forefoot is important. According to the result of analysis it is as follows 1) Center of force trajectory in women's dress shoes display direct movement, compare with center of force trajectory in Sneaker shoes displays a little bit curved slow pronation movement. Sneaker shoes in forefoot part display very quick supination movement, therefore, this shoes effects negative effectiveness for ankle's stability Considering center of force trajectory analyzing the more center of force close straight line, the more movement can be quick movement for locomotion. For foot pressure distribution, center of force trajectory in locomotion is better to curved trajectory with pronation movement. So sneaker shoes style is good shoes considering center of pressure distribution trajectory compare with women's dress shoes. 2) Women's dress shoes increased peak pressure in medial, this is effected by high hill's height. The more increased women's dress shoes's height, the more women's peak pressure will increase, pronation can increase compare with before. Supination movement increase, this focused pressure in lateral, also, supination increased more. If the supination movement increased, foot pressure focused in lateral, therefore, it is appeared force distribution in gait direction. This is bad movement in foot's stability. 3) Women's dress shoes in landing phase displayed a long time, this is when women's dress shoes wear, gait movement is unbalance, so, landing phase displayed a long time. For compensation in gait, swing phase quick movement. 4) Women's dress shoes displayed peak pressure distribution in lateral of rearfoot part, Sneakers shoes displayed peak pressure distribution in medial of forefoot part. Its results has good impact absorption compare with women's dress shoes. In forefoot part, sneakers shoes has good propulsive force compare with women's dress shoes.

Neuroprotective potential of imatinib in global ischemia-reperfusion-induced cerebral injury: possible role of Janus-activated kinase 2/signal transducer and activator of transcription 3 and connexin 43

  • The present study was aimed to explore the neuroprotective role of imatinib in global ischemia-reperfusion-induced cerebral injury along with possible mechanisms. Global ischemia was induced in mice by bilateral carotid artery occlusion for 20 min, which was followed by reperfusion for 24 h by restoring the blood flow to the brain. The extent of cerebral injury was assessed after 24 h of global ischemia by measuring the locomotor activity (actophotometer test), motor coordination (inclined beam walking test), neurological severity score, learning and memory (object recognition test) and cerebral infarction (triphenyl tetrazolium chloride stain). Ischemia-reperfusion injury produced significant cerebral infarction, impaired the behavioral parameters and decreased the expression of connexin 43 and phosphorylated signal transducer and activator of transcription 3 (p-STAT3) in the brain. A single dose administration of imatinib (20 and 40 mg/kg) attenuated ischemia-reperfusion-induced behavioral deficits and the extent of cerebral infarction along with the restoration of connexin 43 and p-STAT3 levels. However, administration of AG490, a selective Janus-activated kinase 2 (JAK2)/STAT3 inhibitor, abolished the neuroprotective actions of imatinib and decreased the expression of connexin 43 and p-STAT3. It is concluded that imatinib has the potential of attenuating global ischemia-reperfusion-induced cerebral injury, which may be possibly attributed to activation of JAK2/STAT3 signaling pathway along with the increase in the expression of connexin 43.

부하유무에 따른 수영운동이 중추신경계 손상 흰쥐의 혈청 BDNF 농도 및 행동변화에 미치는 영향 (The Effect of the Swimming Exercise by Load on Concentration of BDNF in Serum and Behavioral Change of CNS Injury in the Rats)

  • 본 연구의 목적은 중추신경계 손상의 쥐에 운동강도에 따른 신경계 회복을 알아보기 위해 부하수영과 무부하수영이 혈청 BDNF 농도와 행동학적 변화에 어떠한 영향을 미치는가를 확인하고자 하였다. 실험동물로는 Sprague-Dawley 수컷 흰쥐를 사용하였으며, Ketamine을 복강 내에 주사하여 마취한 뒤 제 1-2요추의 척수에 6-OHDA를 $100{\mu}{\ell}$ 주입하여 척수손상을 유발시켰다. 척수 손상으로 유발된 행동학적 변화를 알아보기 위해 BBB와 경사판 검사를 기록하였다. 혈청 BDNF는 수술 후 14일에 혈액을 채취해 흡광도를 측정하였다. 척수 손상 후 무부하군에서는 행동변화가 대조군에 비해 유의하게 증가하였고, 혈청 BDNF 농도는 다른 군에 비해 무부하군에서 증가하였다. 이러한 결과로 보아 저강도 수영운동을 중추신경 손상 후 초기에 적용하면 신경조직의 회복에 영향을 주리라 생각된다.
