Life History of the Siberian Stone Loach Orthrias toni (Pisces: Balitoridae) in the Buk Stream, Goseong, Korea (한국자생종 종개 Orthrias toni(Pisces: Balitoridae)의 생활사)

  • Life history of the Siberian stone loach, Orthrias toni, were investigated using samples collected from Buk Stream, Goseong-gun, Gangwon-do, Korea. Growth was rapid between April and October when water temperature exceeded $10^{\circ}C$, but growth almost stopped from November-March when water temperature was below $10^{\circ}C$. Age groups for O. toni estimated by frequency distribution of total length in spawning season (May) indicated the 60~81 mm is 1-year old, the 82~99 mm group is 2-years old, the 100~119 mm is 3-years old, and the 120~140 mm is more than 4-years old. Total length range by sex was similar, and sex ratio (♂/♀) was 0.73. Spawning season according to the gonadosomatic index (GSI) was May with water temperature $14{\sim}17^{\circ}C$, and almost all fish were mature after one year old. Fecundity increased rapidly with age with an average of $4,460{\pm}3,302$ ($mean{\pm}SD$). Size of mature eggs was $0.87{\pm}0.03mm$ in diameter irrespective of age.

  • This study was conducted to investigate food and meal preference of workers (435 male and 212 female) in the Chonnam Yeosu industrial area. The results of the survey were analyzed by principal components analysis. The results were obtained as follows: 19.3% of the subjects were twenties, 28.3% were thirties, 28.8% were forties and 23.7% were fifties. Females liked rice rolled in dried laver, rice cakes, janchi-noodle and breads, on the other hand males liked thick beef soup, loach soup, an eel stew and soju. The young liked instant noodles, fried chicken, sweet and sour pork, pork cutlet, pizza, hamburger, ham, sausage and fruit beverage, on the other hand the old liked a fish pot stew, loach soup, eel stew, fish boiled in soy with spices, panfried fish, sea slug, ascidian, bunder, green laver and boiled burdock-lotus root in soy. Females who are young liked hamburger and sweet and sour pork whereas males who are young liked instant noodles, pork cutlet, ham, sausage and fruit beverage. Aged females liked fish boiled in soy with spices, panfried fish, bunder, green laver, boiled burdock-lotus root in soy whereas aged males liked a fish pot stew, loach soup, eel stew, sea mussel and oysters. Boiled rice was located within the middle irrespective of age and sex, but noodles, wheat flour meal, fast foods and fruits were situated at the young female side. Aged males liked soup and pot stew. Young males liked meats and eggs whereas fish and shellfish and kimch were located at the aged people side. Aged females liked sea weeds and most people disliked vegetables but females liked some vegetables irrespective of age. Processed foods, salted foods, and alcohol were generally disliked foods by subjects but males liked those foods.   • This study was carried out to investigate the appearance time of the second polar body for producing Gynogenesis or Triploid which could be obtained by arresting the second polar body by cold shock, and then blastoderm was used to measure fertility that revealed the nature of oogenesis, the effects of water temperature on fertility, hatchability, abnormality, viability and growth rate, and the water temperature and the breeding methods to prevent early death of larvae in Korean loach(Misgurnus anguillicaudatus) ; the results obtained in this study were summarized as follows. The second polar body was observed ont he surface of plasma disc close to micropyle within 10~40 min after fertilization at 29$^{\circ}C$. Artificial inseminatin had to be done immediately after the egg spawning because the spermatozoa of loach their mobility within 2 minutes when they were exposed to water. The amount of time needed to reach at blastoderm stage was 12 hours if fertilized eggs were incubated at 16$^{\circ}C$, 8 hours at 19$^{\circ}C$, 6 hours at 21$^{\circ}C$, 5 hours at 23$^{\circ}C$, 4 hours at 26$^{\circ}C$ and 3 hours 30 min at 29$^{\circ}C$ showing the shorter time for development of eggs at higher temperature. Fertilization rates in water temperatures of 19$^{\circ}C$, 21$^{\circ}C$, 23$^{\circ}C$, and 26$^{\circ}C$ were higher than those of water temperatures, 16$^{\circ}C$ and 29$^{\circ}C$. Water temperatures at 19$^{\circ}C$, 21$^{\circ}C$, and 23$^{\circ}C$ showed higher hatching rates that those of 16$^{\circ}C$, 26$^{\circ}C$, and 29$^{\circ}C$, while abnormal rates in 16$^{\circ}C$, 19$^{\circ}C$, 21$^{\circ}C$ and 23$^{\circ}C$ were lower than that of 26$^{\circ}C$ and 29$^{\circ}C$. Water temperatures at 16$^{\circ}C$, 19$^{\circ}C$, 21$^{\circ}C$, 23$^{\circ}C$ and 26$^{\circ}C$ respectively, were more different than 29$^{\circ}C$ in survival rates. The embryos were hatched at 72 hours after fertilization in 16$^{\circ}C$ water temperature, 48 hours in 19$^{\circ}C$, 40 hours in 21$^{\circ}C$, 32 hours in 23$^{\circ}C$, 25 hours in 26$^{\circ}C$, and 16 hours in 29$^{\circ}C$. Within three days after hatched out, the larvage grew 3mm in total length, the yolk granules were entirely consumed and the head and the trunk became thicker. Within 45 days after hatched out, the larva grew 25mm at 29$^{\circ}C$, 21mm at 26$^{\circ}C$, 16mm at 23$^{\circ}C$, 15mm at 21$^{\circ}C$, 12mm at 16$^{\circ}C$ in a 30 litreglass aquarium.

Intraspecies Androgenesis in Mud Loach (Misgurnus mizolepis): I. Optimization of the Egg Inactivation and Haploid Androgenesis Using Transgene Marker (미꾸라지(Misgurnus mizolepis)의 웅성발생성 처녀생식: I. 형질전환 유전자 표지를 이용한 웅성발생성 반수체 유도의 최적화)

  • Induced androgenesis, a form of artificial parthenogenesis is an important tool for the generation and use of genetically isogenic or clonal fish strain. An optimized protocol for the genetic inactivation of mud loach (Misgurnus mizolepis) oocytes (i.e. production of androgenetic haploid) was developed using UV-irradiation. Various dose levels of UV significantly affected the fertilizing capacity of the eggs, hatchability of embryos and incidence of haploidy. Based on the extensive examinations of treatment conditions on embryo viability and haploid incidence, the optimum dose level of UV irradiation was turned out to be $10,800ergs/mm^2$ with 56.9% of hatching success and 94.6% of haploidy. The overall yield of putative androgen under optimized treatment condition was more than 50% out of total eggs inseminated. The success of androgenetic reproduction of haploid genome was verified by flow cytometry and PCR amplification of transgene that is exclusive to either one of parental sexes. However, a small portion $(8\sim11%)$ of presumed androgenetic haploid larvae was proven to contain residual DNA fragment(s) from maternal parent.

The Habitat, Age and Spawning Characteristics of the Korean Eastern Spined Loach, Iksookimia yongdokensis (Pisces: Cobitidae) in the Chuksancheon (stream), Korea (한국고유종 동방종개 Iksookimia yongdokensis(Pisces: Cobitidae)의 서식지와 연령, 산란기 특징)

  • The habitat, age and spawning characteristics of the Korean eastern spined loach, Iksookimia yongdokensis, were investigated to obtain baseline data for its ecological characteristics in the Chuksancheon (stream), Yeongdeok-gun, Gyeongsangbuk-do, Korea. The difference between the habitats of juveniles (0+) and 1~4 year old loaches was that juveniles mainly inhabited sandy bottoms (substratum particle size $0.6{\pm}1.44\;cm$) with very slow velocities of water current ($2.4{\pm}2.09\;cm/sec$) and low depths ($29.2{\pm}7.57\;cm$), whereas 1~4 year old loaches inhabited pebble and cobble bottoms (substratum particle size 5.5~6.1 cm) with relatively fast velocities of water current (3.8~4.6 cm/sec) and deep depths (50~70 cm). The age groups for I. yongdokensis (female) estimated by the frequency distribution of total length in the spawning season (July) indicated that the 35~64 mm is 1-year old, the 65~94 mm group is 2-year old, the 95~119 mm is 3-year old, and the 120~145 mm is more than 4-year old. The sex ratio (♂/♀) was 0.69, and females were 10~30 mm larger than males. The lamina circularis at the base of the pectoral fins in males, a secondary sexual characteristic, formed at the 13th month after hatching. The spawning season according to the gonadosomatic index (GSI) was June to July with water temperatures of $20{\sim}25^{\circ}C$. The average number of eggs was $2,292{\pm}618$, with a diameter of $1.46{\pm}0.02\;mm$ for mature eggs.

  • Egg development and early life history of the Korean spine loach, Cobitis tetralineata, were investigated in the present study. Adult fish were sampled using spoon nets at the Seomjin River in Jeongsan-ri, Bokheong-myeon, Sunchang-gun, Jeollanbuk-do, Korea, July 2013. Eggs were obtained after injecting Ovarprim into females. Eggs were then artificially fertilized using the Dry method in the laboratory. Mature eggs were slightly adhesive and transparent with grey coloring, and measured $1.04{\pm}0.03mm$ ($mean{\pm}SD$) in diameter. Hatching of the embryo occurred approximately 56 hours after fertilization at $25^{\circ}C$ of water temperature, and the average size of newly hatched larvae was about $4.6{\pm}0.11mm$ in total length. At fifth day after hatching, the larval full length reached $6.8{\pm}0.28mm$ on average and their yolk sac had been completely absorbed. At 17th day after hatching, larva entered the juvenile stage and reached $9.8{\pm}0.50mm$ in total length. At 100th day after hatching, the formation of Gambetta's zone of four line was complete and juveniles were similar in general appearance to adults, and they averaged $34.7{\pm}5.77mm$ in total length.

  • In the present study, egg development and early life history of Korean spined loach, $Iksookimia$ $koreensis$, were observed. Adult fish were sampled using spoon nets in Okgye-ri, Hoengseong-gun, Gangwon-do, Korea in July 2010. Eggs were obtained after injecting females with Ovarprim. Eggs were then artificially fertilized in the laboratory using the dry method. Mature eggs were slightly adhesive and transparent with a light yellowish color, and measured $1.40{\pm}0.04mm$ (mean${\pm}$SD) in diameter. Hatching of the embryo occurred approximately 50 h after fertilization in the water at $23^{\circ}C$, and newly hatched larvae were averaged $4.7{\pm}0.21mm$ in total length. 5 days after hatching, the averaged total length of larvae was $7.1{\pm}0.25mm$ and their yolk sacs had been completely absorbed. 17 days after hatching, fish started to enter the juvenile stage and reached $12.2{\pm}1.10mm$ in total length. 80 days after hatching, the band patterns and external form of juvenile fish were similar to those of adults, and they averaged $31.0{\pm}3.98mm$ in total length.

  • Effects of intraperitonial injections of human chorionic gonadotropin (HCG) on ovulation and spawning of the loach, Misgurnus mizolepts, were investigated. Matured females spawned successfully by a single dose of 6 IU (HCG) per gram body weight. Spawning usually occured 13 to 25 hours after hormone injection. Most of the eggs were fertilized and hatched normally. Fertilization rate, hatching percentage and Pseudo-gonadosomatic index were not correlated with increasing HCG doses. For the study of multiple spawning of this species, fatness, hepatosomatic and gonadosomatic indices of artificially spawned females were checked for 42 days after first spawning. According to this result, females could spawn again 40 days after first spawning.

  • Intraspecific diploid androgenesis was achieved in mud loach (Misgurnus mizolepis) by the inhibition of the first mitotic division using combined thermal treatment. A combined thermal treatment (heat shock at $40.5\;^{\circ}C$ for 120 sec followed by cold treatment at $1\;^{\circ}C$ for 45 min) applied to the 1st metaphase of cell division (28 min post insemination at $25\;^{\circ}C$) successfully recovered viable androgenetic diploidy. Mean hatching success of the androgenetic diploid group was 29.6%, and the average yield out of total eggs taken was about 7% assessed at 1 week of age. However, relatively large variations in the yield of diploid androgenesis were observed among different egg batches used as cytoplasmic donors. Successful diploidization was confirmed by flow cytometric analysis, and parthenogenic reproduction in a paternal exclusive manner was verified with transgene dosage. Significant mortality was found in most androgenetic groups especially from hatch to 1 month of age, although such mortality was stabilized later.

  • A 123.2 mm TL female of Iksookimia longicorpa with a laminar circularis at the base of its pectoral fin was collected in the Hoeya River, Korea, on May 5, 2008. The specimen had full-grown ovaries like those of normal females. Its pectoral fins were larger than those of normal females, but was smaller than those of males. The laminar circularis of the pectoral fin base was more similar to normal males in its shape but was less developed. The ovaries in the individual had about 3,148 mature eggs, with egg sizes approximately $1.26{\pm}0.11mm$.
