The youngkaechulgamtang (Y) composed of four herb drugs, including Hoelen (H). Cinnamomi Ramulus (C). Atractylodis Rhizoma Alba (A) and Glycyrrhizae Radix (G). In oriental medicine literatures, Youngkaechulgamtang is described to be effective in headache, inflammation, uremia, gastritis, diarrhea and hypertension. To estimate the clinical effectiveness of Youngkaechulgamtang, several pharmacological experiments were carried out. The results are summerized as follows; On acetaminophen-induced hepatotoxicity, C+A, Y-G, Y-H, MIX and Y showed the significant elevation of glutathione-S-transferase. But, C+A, Y-G, Y-H, MIX and Y showed the significant suppression of serum aminotransferases. On ANIT-induced cholestasis, U (Ursodesoxycholic acid 50 mg/kg)+$Y_l$ (760 mg/kg) showed the significant increase of bile juice volume. $Y_l,\;Y_2$ (1520 mg/kg), U, $U+Y_l$ showed the remarkable increase of cholic acid. U and $U+Y_l$ showed the significant decrease of total bilirubin. From these results, it is suggest that Y shows liver protective effect against various hepatic injury. Especially, Youngkaechulgamtang was more effective than mixture of 4 ingredients in the elevation of glutathione-S-transferase in acetaminophen-induced hepatotoxicity.
High fructose intake induces hyperglycemia and hypertension. However, the mechanism by which fructose induces metabolic syndrome is largely unknown. We hypothesized that high fructose intake induces activation of the renin-angiotensin system (RAS), resulting in hypertension and metabolic syndrome. We provided 11-week-old Sprague-Dawley rats with drinking water, with or without 20% fructose, for two weeks. We measured serum renin, angiotensin II (Ang II), and aldosterone (Aldo) using ELISA kits. The expression of RAS genes was determined by quantitative reverse transcription polymerase chain reaction. High fructose intake increased body weight and water retention, regardless of food intake or urine volume. After two weeks, fructose intake induced glucose intolerance and hypertension. High fructose intake increased serum renin, Ang II, triglyceride, and cholesterol levels, but not Aldo levels. High fructose intake increased the expression of angiotensinogen in the liver; angiotensin-converting enzyme in the lungs; and renin, angiotensin II type 1a receptor (AT1aR), and angiotensin II type 1b receptor (AT1bR) in the kidneys. However, expression of AT1aR and AT1bR in the adrenal glands did not increase in rats given fructose. Taken together, these results indicate that high fructose intake induces activation of RAS, resulting in hypertension and metabolic syndrome.
In this study, we investigated whether PLGA microspheres in combination with hyaluronic acid (HA) gel have appropriate properties as a bulking agent for urologic injection therapies and whether the implantation of PLGA microspheres and HA gel induces angiogenesis in the newly formed tissues. In order to investigate whether this bulking agent is injectable, this material was injected through 24-gauge needles into the subcutaneous dorsum of the mouse. The bulking agent was easily injected without needle obstruction. Histological analyses of the hybrid tissues at 2 weeks showed that host cells at the surrounding tissues migrated into the spaces between the implanted PLGA microspheres and formed tissue-like structures. An inflammatory response to the implants was mild at 2 weeks and diminished at 8 weeks. Importantly, extensive ingrowth of blood vessels was observed in the hybrid tissues formed by the injection of PLGA microspheres and HA, whereas blood vessels rarely formed in the hybrid tissues formed by the injection of PLGA microspheres only. The implant volume was conserved for almost the entire implantation period. Histological analyses of the distant organs of the bulking agent-implanted animals, such as the lungs, liver, heart, brain, kidney, and spleen, showed no evidence of the injected microsphere migration. These results show that PLGA microspheres in combination with HA possess the appropriate characteristics for a bulking agent for urologic injection therapies and induce extensive blood vessel formation in the hybrid tissues.
This study was attempted to investigate the pharmacokinetics of cyclosporine (10mg/kg, oral) in rabbits with $CCI_4$ and bile duct ligation-induced hepatic disorder. The area under the curve (AUC) of blood cyclosporine concentration versus time was significantly increased ($CCI_4$-induced hepatic disorder. Elimination rate constant (Kel) was significantly decreased (p<0.05, p<0.01) in rabbits with $CCI_4$ and bile duct ligation-induced hepatic disorder. Volume of distribution (Vdss) and total body clearance (CLtot) were significantly decreased (p<0.01) in rabbits with $CCI_4$-induced hepatic disorder. But Vdss was significantly increased (p4-induced hepatic disorder were 874ng/ml and 2.71 hr, respectively. Cmax and Tmax values in rabbits with bile duct ligation were 105ng/ml and 2.834 hr, respectively. From results of this experiment. It is desirable to do therapeutic drug monitoring of cyclosporine for effective treatment when the cyclosporine is administered to patients with liver disorder m clinical practice.
The phannacokinetics and tissue distribution of DWQ-013, a new quinolone, were examined in rats and mice following a single intravenous and oral administration. DWQ-013 in plasma and urine was determined by both HPLC and microbiological assay. The plasma concentration of the drug declined biexponentially. The terminal half life of the drug was 11.11$\pm$0.14 hour after intravenous dosing. The volume of distribution at terminal elimination phase(Vd$_\beta$) and total clearance of the drug were 1.29$\pm$0.15 l/kg and 0.78$\pm$0.09 l/h/kg. The bioavailability of DWQ-013 after oral administration was 56.0% (HPLC) and 77.2%(bioassay), respectively. Twelve-hour urinary recovery of drug was measured by HPLC and bioassay to 0.035$\pm$0009% and 4.71$\pm$066% after oral dosing, to 0.055$\pm$0.014% and 7.65$\pm$1.53% after intravenous dosing, which may indicate the presence of biologically active metabolites. Binding of the drug to plasma proteins ranged from 97%~99% at various concentrations. The drug was highly distributed in order of liver, kidney and lung after 1.5 hours in mice.
Objectives : To investigate the effects of Cornu cervi parvum pharmacopuncture with regard to the blood picture and antioxidative activity in rats. Methods : Sprague-Dawley rats were divided into 3 groups (n=5 each) and were treated with Cornu cervi parvum pharmacopuncture every other day for 2 weeks. The groups are classified as follows; normal control without treatment (control group), Cornu cervi parvum pharmacopuncture at CV4 (CV4 group), and Cornu cervi parvum pharmacopuncture at BL23 (BL23 group). Thereafter, the blood and liver samples were obtained for blood analysis and superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activity measurement. Results : Cornu cervi parvum pharmacopuncture groups showed higher values of red blood cell count and plasma cell volume compared with those of the control (p<0.05). However, hemoglobin level showed no significant differences among groups. With regard to the blood picture, plasma concentration in total protein and albumin showed no significant differences in pharmacopuncture groups, while higher ratio of albumin/globulin was observed in CV4 group. White blood cell counts and its composition showed no significant differences among groups. Pharmacopuncture groups showed higher values in SOD, CAT, and GSH-Px activities compared with those of control group. Conclusions : Cornu cervi parvum pharmacopuncture alleviates oxidative activities in rats.
Objectives: To investigate the effects of Hominis placenta pharmacopuncture on the blood picture and antioxidative activity in rats. Methods: Sprague-Dawley rats were divided into 3 groups; normal control (n=5), pharmacopuncture at CV12 (CV12 group, n=5), and pharmacopuncture at ST36 (ST36 group, n=5) once every other day for 4 weeks. Blood cell counting was performed and liver superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities were analyzed. Results: Values of red blood cell and plasma cell volume were significantly higher in the ST36 group than the normal control. Values of hematocrit, total protein, and albumin were not significantly different among groups. White blood cell count and the percentage of neutrophils, lymphocytes, and eosinophils were not significantly different among groups. However, monocytes and basophils were significantly increased in the ST36, and CV12 groups, respectively. SOD and CAT in the CV12 and ST36 groups were significantly activated than in the normal control group, while the activity of GSH-Px showed no significant difference among groups. Conclusions: Based on the findings of this study, Hominis placenta pharmacopuncture may have positive impact on antioxidative capacity, thus activate various functions of the body.
The aim of this study was to examine the ameliorating effect of black ginseng on the growth of the HepG2 cell transplanted tumor in BALB/c nude mice. 27 male BALB/c nude mice (all six weeks old) were randomly divided into three groups: the control group, the first treatment group (HepG2300RG, using 300 mg/kg red ginseng), and the second treatment group (HepG2300BG, using 300 mg/kg black ginseng). The HepG2300BG in the HePG2 cells showed increased mean survival time than that of red ginseng group. The size and volume of the tumor in the 300BG group showed significant reduction compared to those of the HepG2300RG group (p<0.05). The body weight and liver weight of the HepG2300RG group was not significantly different with control and HepG2300BG group. The serum levels of ALT and AST in the HepG2300RG and HepG2300BG group were significantly lower than those of the control group. In conclusion, these results suggest that the black ginseng may have possible anti-tumor activities.

A 22-year-old male patient was diagnosed with autosomal dominant polycystic kidney disease (ADPKD). He received conservative treatment with an angiotensin-converting enzyme inhibitor. Two years later, oral therapy, consisting of 60 mg tolvaptan per day, was initiated. Compared with height-adjusted total kidney volume, the rate of kidney growth reduced significantly from 7.33% to 0.66% annually, since commencement of the tolvaptan therapy. The liver enzyme profile and serum sodium level and osmolality were constantly within normal ranges. In Korea, this is the first reported case of a patient with ADPKD who received tolvaptan treatment for more than 1 year. This case demonstrates that long-term tolvaptan treatment appears to be safe, well tolerated, and effective for ADPKD.
Nerium oleander (N. oleander) is a well-known poisonous shrub that is frequently grown in gardens and public areas and contains numerous toxic compounds. The major toxic components are the cardiac glycosides oleandrin and neriin. The aim of our study was to evaluate the toxic effects of an ethanolic N. oleander leaf extract on haematological, cardiac, inflammatory, and serum biochemical parameters, as well as histopathological changes in the heart. N. oleander extract was orally administered for 14 and 30 consecutive days at doses of 100 and 200 mg of dried extract/kg of body weight in 0.5 mL of saline. The results showed significant increases in mean corpuscular volume, white blood cell counts, platelet counts, interleukins (IL-1 and IL-6), tumour necrosis factor alpha, C reactive protein, alanine aminotransferase, lactate dehydrogenase, creatine kinase and creatine kinase MB, especially at high doses. Marked pathological changes were perceived in the heart tissue. Thus, it can be concluded that exposure to N. oleander leaf extract adversely affects the heart and liver.
