Protective effects of Jucha-whan on liver injury of guinea pigs induced by $CCl_4$ and d-galactosamine (주차환(舟車丸)이 $CCl_4$ 및 d-galactosamine으로 유발된 흰쥐의 간손상(肝損傷)에 미치는 영향)

  • Objective : In order to investigate the curative effect of Jucha-whan on the protective liver of guinea pigs induced by $CCl_4$ and d-galactosamine, serum transaminase(GOT, GPT), lactic dehydrogenase(LDH), alkaline phosphatase(ALP), glutathione S-transferase(GST), superoxide dismutase(SOD) were used to measure enzyme activities and lipid peroxide level. Method : The subject animals were divided into 5 groups; a control group(untreated), a subject group(administered with 0.9% Saline solution), a sample I (500mg/kg administered), sample II group (1000mg/kg administered), positive control group(administered with 200mg/kg silymarine). Result : The inhibitory effects on the serum GOT, LDH, ALP, SOD and Lipid peroxide level activities in protective liver of mice induced by $CCl_4$ were noted both in the sample I group and sample II. The inhibitory effects on the serum GPT activities in protective liver of guinea pigs induced by $CCl_4$ were noted in sample II group, but it was not noted in the sample I. The inhibitory effects on the GST activities in protective liver of guinea pigs induced by $CCl_4$ were not noted in both sample I and sample group II. The inhibitory effects on the serum GOT, GPT activities in protective liver of guinea pigs induced by d-galactosamine were noted in both sample I and sample II, but it was not recognizable statistically. The inhibitory effects on the serum LDH activities in protective liver of guinea pigs induced by d-galactosamine were noted in sample II, but it was not noted in sample I group. Conclusion : According to the above results, it is considered that Jucha-whan has treatment effects on liver injury in guinea pigs induced by $CCl_4$ and d-galactosamine. So it is required to study about the actions of mutual relation of medicines and patho-mechanism through experiment.

  • The aim and objective of this study are to carry out the liver protective activities against the $CCl_4$ intoxication in mice with some Iranian medicinal plants traditionally used for liver injuries. The methanol extracts of Cichorium intybus, Lactuca scarJoia, Eucalyptus camadulensis were evaluated. With various doses of these plants, liver protective activities were performed after $CCl_4$ administration to mice. The serum aminotransferases activites, liver sizes, and histopatological examinations of liver were studied. At a dose of 50 mg/ kg, all three plants were able to protect liver damages induced by $CCl_4$.

  • The studies on concurrent administrations of herb preparations(Daesiho-tang, Daehwangmogdanpi-tang, Dohaegseunggi-tang, Baenongsangeub-tang and Jeoryeong-tang) with indomethacin and tetracycline were carried out to investigate the antiinflammatory, antimicrobial, liver-protective and antiulcer activities. The results showed that the concurrent administrations of herb preparations with indomethacin and tetracycline increased significantly the antiinflammatory activities and antimicrobial activities respectively, and also improved the liver-protective activities and antiulcer activities against indomethacin or tetracycline-induced liver defect and Shay ulcers in rats, respectively.

  • Alisol A monoacetate, alisol B monoacetate, alisol C monoacetate and alisol B were isolated from Alismatis Rhizoma, which is a herbal drug used frequently in the oriental prescriptions. Potential liver-protective activities of the isolated alisol compounds were evaluated against $CCl_4-induced$ liver damage. The results obtained from liver microsomal enzyme assay, measurement of serum glutamic pyruvic transaminase (EC 2.6.1.2) and serum triglyceride content indicated that alisol A,B and C monoacetates showed significant liver-protective activities against $CCl_4$ poisoning. Alisol B monoacetate exhibited slightly higher activity than that of alisol B.

  • Objectives : This study was done to investigate the protective effects of Samgiinjin-tang on liver injury of rats induced by CCI4 and d-galactosamine. Methods: All animals were divided into .5 groups, those were normal group(untreated), control group(treated with 0.9% Saline solution), sample I group(2,250mg/kg administrated), sample II group(4,500mg/kg administrated), Silymarin 200mg/kg administrated group. Liver injury of rats were induced by CCI4 and d-galactosamine, and then the serum transaminases(ALT&AST) alkaline phosphatase(ALP), lactic dehydrogenase(LDH) for enzyme activities, liver weight, lipid peroxidation and catalase, glutathione S-transferase(GST) for enzyme activities were measured. Results : The inhibitory effects on the serum ALT, AST activities in liver injury of rats induced by CCI4 were noted in both sample I and sample II group. The inhibitory effects on the serum ALP, LDH activities and the Lipid peroxidation of Mitochondria & Cytosol were noted in only sample II group. The decreased effects on the GST activities of Homogenate & Cytosol were inhibited in both sample I and sample II groups. The decreased effects on the GST activities of Mitochondria & Microsome were inhibited in sample II group. The inhibitory effects of the serum ALT, AST, LDH activities in liver injury of rats induced by d-galactosamine were noted in both sample I and sample II groups. In serum AST activities, sample II group. Conclusions : Samgiinjin-tang has protective effects against liver injury of rats induced by CCI4 and d-galactosamine. So it is required to study about the actions of mutual relation of medicines and patho-mechanism by experiment.

  • Methanol extracts of the roots of Atractylodes japonica (alba) (Compositae) and Gentiana scabra (Gentianaceae) were further fractionated with n-butanol, chloroform, n-hexane and water. Then each fraction was p.o. administered to an animal model of hepatitis caused by $CCl_4$ intoxication. Measurements of sleeping time of mice induced by hexobarbital administration indicated that n-butanol and chloroform fractions of Atractylodes japonica (alba), and chloroform and n-hexane fractions of Gentiana scabra showed significant liver protective activities. Measurements of the activities of serum transaminase, S-GOT and S-GPT appeared to support these results.

  • The protective effect of Yangguksanwha-tang (YST) against hypoxia-reperfusion insult was investigated in PC12 cells. To elucidate the mechanism of the protective effect of YST, cell viability, the changes in activities of superoxide dismutase, glutathione peroxidase, catalase, caspase 3 and the production of malondialdehyde were observed after treating PC12 cells with YST which was metabolized by rat liver homogenate. Pretreatment of YST with liver homogenate appeared to increase its protective effect against hypoxia-reperfusion insult. The result showed that YST had the highest protective effect against hypoxia/reperfusion at the dose of $2\;{\mu}g/ml$ in PC12 cells, probably by recovering the redox enzyme activities and MDA to control level.

Application of Iranian Medicinal Plants to the Treatment of Liver Injury.

  • Matricaria Chammomillal L., Foemiculum Vulgare mill, and Plantago Psylium L. have been screened for their hepato protective activities against liver damge induced by $CCl_4$ intoxication in mice. Hydroalcoholic extractions (2:8) of herbal drugs were concentrated in vacuo and concentrated crude extracts of Matrica Chammomilla L. and Foeniculum Vulgare mill were orally administered at doses of 100 mg/kg, 200 mg/kg, 400 mg/kg, and 800 mg/kg. Plantago Psyllium was given at doses of 50 mg/kg, 100 mg/kg, 200 mg/kg, and 400 mg/kg. Liver protective activities of these herbs were determined after administration of $CCl_4$ Liver size, serum enzyme activities, sleeping time, and histopatology of the liver were examined one hour after administration of $CCl_4$. ALT and AST activities, liver weight and sleeping time decreased in groups that received 400 mg/kg of Matricaria Chammomilla L. or Foeniculum Vulgare. Histological investigation showed significant increase in hepatic cell regeneration and reduction in liver injury. The group that received 100 mg/kg Plantago Psylium showed liver protection but protection was not significant in other doses.

  • Water fractions prepared from Plantaginis Semen and Plantaginis Folium exhibited both significant liver-protective activities against $CCl_4$ intoxication. Aucubin, an iridoid glycoside. isolated from both leaves and seeds of Plantago asiatica(Plantaginaceae) also showed potent liver protective activities against the animal model of hepatitis induced by $CCl_4$ intoxication.

Methods : All animals were divided into 5 groups, those were normal group(untreated), control group(treated with 0.9% Saline solution), sample I group(200mg/kg administrated), sample II group(400mg/kg administrated), Silymarin(200mg/kg administrated) group. Results : The inhibitory effects on the serum ALT activities were noted in both sample I and sample II group. The inhibitory effects on the serum AST activities were noted in only sample II group. The inhibitory effects on the serum ALP activities were noted in both sample I and sample II group. The inhibitory effects on the serum LDH activities were noted in only sample II group. The inhibitory effects on the liver cytosol malondialdehyde were noted in only sample II group. The decresed effects on the liver cytosol catalase activities were inhibited in only sample II group. The decresed effects on the liver cytosol superoxide dismutase activities were inhibited in only sample II group. The decresed effects on the liver cytosol GST activities were inhibited in only sample II group. The decresed effects on the liver cytosol GPX activities were inhibited in only sample II group. The inhibitory effects of the serum ALT activities were noted in both sample I and sample II. The inhibitory effects of the serum AST activities were noted in only sample II group. The inhibitory effects of the serum ALP activities were noted in only sample II group. The inhibitory effects of the serum LDH activities were noted in both sample I and sample II group. Conclusions : Gagaminjakdowha-Tang has protective effects against liver injury in rats induced by $CCl_4$ and d-galactosamine.
