Development of Liquid Culture Media for the Mass Production of Beauveria bassiana Blastospore (흰굳음병균(Beauveria basiana) 단균사의 대량배양생산을 위한 액체배지 개발)

  • To develope a liquid culture media for the mass culture of Beauveria bassiana blastospore, wheat bran and soybean cake were selected as a carbon and nitrogen source, respectively. The composition and final concentration of wheat bran and soybean cake in liquid culture media were determined by the growth of B. bassiana blastospore. The results showed that the growth of B. bassiana blastospore was effective in the rate of 3 wheat bran: 2 soybean cake and in 5% of final concentration of the liquid culture media, suggesting that the liquid culture media using wheat bran and soybean cake are available.

  • The rate of growth of Leptospires were studied by culturing in Korthof's media and Cox's liquid media which were treated at four different temperatures. The effect of thiamine hydrochloride in the culture of leptospires were also studied. The results obtained were summerized as followings: 1. The best growth of Leptospira icterohaemorrhagiae and Leptospira canicola was observed in Korthof's media with the rabbit serum heated for 60 minutes at $56^{\circ}C$. 2. A good degree of growth of Leptospira icterohaemorrhagiae and Leptospira canicola was observed in both Cox's liquid media with rabbit serum heated for 30 minutes at $100^{\circ}C$ and with fresh rabbit serum. 3. The growth of Leptospira was enhanced by adding small amount of thiamine to the both media which were shown to be unsatisfactory for the growth of the organisms. 4. Culture of leptospires were attained by simply heating for 30 minutes at $100^{\circ}C$ both Korthof's media and Cox's liquid media containing rabbit serum obtained without aspetic procedures.

  • Freezing of saturated porous media contained in a rectangular cavity has been studied experimentally. Water and different diameter glass beads consitituted the liquid and porous media. Solidification front shape, the effects of bead diameter and initial liquid temperature was investigated. When the hot wall temperature was below $4^{\circ}C$, the freezing rate was higher at the top than at the bottom due to the density inversion, but with increasing the hot wall temperature the freezing rate at the top was effected by the liquid temperature and was lower than at the bottom. With increasing the bead diameter, the difference of freezing rate between top and bottom was increased and depends on thermal conductivity. When the liquid temperature was low in the beginning, the freezing rate was high, but with increasing the time almost the same with those of high temperature liquid.

Comparison of Cultivation, Mushroom Yield, and Fruiting Body Characteristics of Lentinula edodes Strains according to the Inoculation Method

  • The cultivation in sawdust media, mushroom productivity, and fruiting body characteristics of Lentinula edodes strains NIFoS 2778 and NIFoS 3363 were compared according to the inoculation conditions. The cultivation period was 5% shorter when liquid spawn was used. Fruiting bodies were induced after 113 days of incubation on media inoculated with liquid spawn, and the cultivation period was 119 days on media inoculated with solid spawn. Mushroom productivity of NIFoS 2778 was the highest (661.4 g) when 36 mL of liquid spawn was used. For NIFoS 3363, mushroom production was higher under liquid inoculation conditions when the same amount of liquid and solid spawns were used. The mushroom characteristics of the two strains were not significantly different, except for gill width and stipe diameter.

HIGH-SPEED FLOW PHENOMENA IN COMPRESSIBLE GAS-LIQUID TWO-PHASE MEDIA (압축성 기-액 이상매체중의 고속 유동현상)

  • A high resolution numerical method aimed at solving gas-liquid two-phase flow is proposed and applied to gas-liquid two-phase shock tube problem. The present method employs a finite-difference 4th-order Runge-Kutta method and Roe's flux difference splitting approximation with the MUSCL TVD scheme. By applying the homogeneous equilibrium cavitation model, the present density-based numerical method permits simple treatment of the whole gas-liquid two-phase flow field, including wave propagation and large density changes. The speed of sound for gas-liquid two-phase media is derived on the basis of thermodynamic relations and compared with that by eigenvalues. By this method, a Riemann problem for Euler equations of one dimensional shock tube was computed. Numerical results such as detailed observations of shock and expansion wave propagations through the gas-liquid two-phase media and some data related to computational efficiency are made. Comparisons of predicted results and exact solutions are provided and discussed.

  • The process of flow through porous media is of interest a wide range of engineering fields and areas, and the importance of fluid flow with a change in phase arises from the fact that many industrial processes rely on these phenomena for materials process, energy transfer. Especially, the flow phenomena of cryogenic liquid subjected to evaporation is of interest to investigate how the cryogenic liquid behaves in the porous structure. In this study, thermo physical properties, morphological properties of the glass wool with different bulk densities in terms of its temperature-dependence and permeability behaviors under different applying pressure are discussed. Using the experimentally determined properties, characteristics of two main experimental results are investigated. In addition, simulation results are used to realize the cryogenic liquid's flow in porous media, and are compared with experimental results. By using the experimentally determined properties, more reasonable results can be suggested in dealing with porous media flow.

Evaluation of Absorption and Release, Survival Efficiency and Recovery Rate of Transport Medium according to the CLSI M40-A2 Standard (CLSI M40-A2 기준에 따른 수송배지의 흡수 및 배출, 생존 효율, 회수율 평가)

  • The absorption and release capacities, survival efficiency, and recovery rates of five kinds of transport media were evaluated based on the swab elution method (Quantitative) of CLSI (Clinical and Laboratory Standards Institute) M40-A2. Liquid media showed mostly better results than semi-solid media in the three evaluations. The flocked swabs had better ability to absorption and discharge bacteria than the standard swabs. The liquid medium (S4) had the best survival efficiency. Pneumococcal strains with poor growth had a higher survival efficiency and recovery rate in liquid media (S4, S5). The results of microbial recovery showed that S. pyogenes met all the CLSI standards in all media. S. pneumoniae was inadequate in the semi- solid media (S2, S3) and all the remaining media met the criteria. H. influenzae was unsuitable in semi-solid media (S1, S3) and met the criteria in semi-solid medium (S2) and liquid medium (S4, S5). The viability of the H. influenzae, pneumococcal strain causing respiratory disease, was poor in most media. Overgrowth of P. aeruginosa was observed at room temperature. The combination of liquid medium and flocked swab confirmed the best results in the three evaluation methods.

Optimization of in Vitro Cultivation of Inonotus Obliquus

  • This study was performed to get the basic information concerned to the optimum culture condition of Inonotus obliquus. Several solid media, PDA, MEA and Czapek-Dox, and three liquid media were adopted for the in vitro cultivation. Some main features of the fungal morphological characteristics under cultivation conditions were observed and described. Preliminary results showed that appearance of the mycelial mat, hyphal size and substrate pigmentation differed according to the media. The PDA medium was the most favorable substrate for the growth on solid culture, followed by MEA and Czapek-Dox media. Concerned to the addition of amino acids, 5 amino acids, such as alanine, alginine, isoleucine, leucine and threonine, enhanced to the mycelial growth. Isoleucine was shown the best fungal growth. An important morphological hyphal structure for the fungus, the setae, was found in abundance and diverse its shape and size. In liquid culture, fresh potato broth was the best growth stimulant of the fungus, followed by Malt extract and potato broth. Addition of yeast extract to the liquid media had improved the biomass, but not laccase production.

Role of Crossflow Module Media in Gas-liquid-solid Separation and Biomass Retention in Hybrid Anaerobic Filter (교차흐름식 모듈 충전 hybrid 혐기성여상의 기·액·고 분리능 및 슬러지보유능)

  • Performances and internal behaviors of the upflow hybrid anaerobic filters treating a dairy wastewater were analyzed to identify the functions and roles of the modular crossflow media and sludge bed layer and to discover their interrelationship in the filter. The media could perform independent biological and physical separation role without buildup of sludge bed, while the role of sludge bed was dependent on the function of the media. The filter packed with the crossflow media did not necessarily require the formation of sludge bed when treating a dairy wastewater. Biological contribution of the media was controlled by that of biologically active sludge bed complementing mutually each other. The gas-liquid-solid separation capability of the media was indispensible to ensure the active biological role of sludge bed, since sludge bed buildup without the media had no independently effective biological function. It was believed that the filter in itself could also function as a selector for physical gas-liquid-solid separation resulting in selectively concentrating particles with superior settleability in sludge bed. The sludge bed in the filter played a key role in the physical solids capture from influent as well as biological organics removal.

Liquid culture of entomopathogenic nematodes

  • An in vitro liquid culture media for the cultivation of entomopathogenic nematode Steinernema carpocapsae was developed. Supplementation of whole milk powder with basal liquid culture media showed a remarkable increase in productivity compared to that without whole milk powder and the maximum nematode concentration reached about $1.5{\times}10^5/mL$ within 20 days. Five to twenty gram per liter of liver extract addition revealed highest pathogenicity against 3rd instar of Galleria mellonella which was above about 90% mortality after 48 hr.
