  • To develop a new natural cosmetic hair care product for scalp treatment, we investigated the effect of enzyme modified (Viscozyme, Pectinex) oriental medical prescriptions on hair growth and melanogenesis. Enzyme modified oriental medical prescriptions showed the antioxidative effect, inhibitory effects on lipoxygenase activity and $5{\alpha}$-reductase activity and melanogenic activity. In human irritation test, they did not show any adverse effect. Based on these results, we suggest that enzyme modified oriental medical prescriptions may potentially be used in a cosmeceutical hair care product for scalp treatment.

  • This study was performed to investigate the change of lipoxygenases and urease activities, trypsin inhibitor, tannin and phytic acid contents during heat treatment of Jinpum soybean. The lipoxygenase-1 and urease possessed their activities after heating at $60^{\circ}C$ for 100 min, but their activities disappeared rapidly at $80^{\circ}C$ and $100^{\circ}C$ for 20 and 10 min. There were no lipoxygenase-2 and -3 activities in Jinpum soybean with and without heating. Trypsin inhibitor was lost 91.9%, 78,1% and 58.6% of the activity after heat treatment at $100^{\circ}C$ for 50 min, at $80^{\circ}C$ for 100 min and at $60^{\circ}C$ for 100 min, respectively. The tannin content was increased by heat treatment. The content of and phytic acid was increase after heating at $60^{\circ}C$ for 100 min, unchanged at $80^{\circ}C$ for 100 min and decreased at $100^{\circ}C$ for 100 min.

  • There have been several reports regarding the effects of Panax ginseng on allergy reactions. However, they are very sporadic and no systemic yet. To study the effects of Panax ginseng on hypersensitivity, either ginseng total saponin (GTS, 200mg/kg, oral, two hours prior to experiments) or ethanol extract (50 and 200 mg/kg, oral, one week) was administered. Various parameters were employed to assess the anti-allergic actions of Panax ginseng 48hr passive cutaneous anaphylaxis (PCA), skin reactions, histamine release from rat peritoneal mast eel Is, and lipoxygenase activity. In 48hr PCA, and in skin reactions induced by chemical mediators (histamine, serotonin) and mediator releaser (compound 48/80), Panax ginseng did not suppress sensitized immune functions, rather showed tendency to increase the histamine-induced vascular permeabi1ity. Panax ginseng did not inhibit lipoxygenase activity either.

  • We investigated the effects of butanol fraction of Asterias amurensis (BFA) on the anti-allergic activity. BFA inhibited both cyclooxygenase-2 dependent prostaglandin $D_2$ and 5-lipoxygenase dependent leukotriene $C_4$ generation in a concentration-dependent manner with $IC_{50}$ values of 174.6 and 22.2 ${\mu}g$/ml, respectively. In addition, BFA also inhibited the degranulation in a dose dependent manner. Furthermore, oral administration of BFA inhibited IgE-mediated passive cutaneous anaphylaxis in mice. These results suggested that BFA may be useful in regulating mast cell-mediated allergic diseases.

  • An investigation was conducted to ascertain the basic information on characteristics of embryonic tissue development among Korean Ginseng, American ginseng, $F_{1}$ hybrids and $BC_{1}F_{1}(F_{1}$ pollen back cross to $BC_{1}=Korean\;Ginseng)seeds$. The specific activities of lipoxygenase (LOX) and the protein assays were made during the above embryonic growth period at 5 or 25 degree C. The results are summarized as follows: The fresh weights(mg) of germinating seeds were significantly different among species. Ginseng seeds were stratified at $5^{\circ}C\;for\;60\;days$ the ratios of embryo/endosperms in $P.g.,\;P.q\;or\;F_{1}\;were\;about\;90{\%}$. The ratio was in $BC_{1}F_{1},\;79.2{\%}$ during the same period. The ratios of embryo/endosperms of seeds of P.g. or P.q. germinated at $5^{\circ}C$ showed rather higher values as compared to that at $25^{\circ}C$ LOX specific activity the highest in the $F_{1}$ and decreased in the order of P.g., P.q., and $BC_{1}F_{1}$. The highest LOX specific activity was shown at 80 days after sowing, followed by 70 days sowing, the least LOX activity was shown at 60 days.

  • Biotin is a water-soluble vitamin used as a skin conditioning agent and promotes the formation of intercellular lipid layers through increased lipid synthesis, which improves the skin's natural barrier function. The anti-inflammatory effects of biotin have been investigated using in vitro assay models, such as MTT assay, measurements of concentrations of nitric oxide(NO), prostaglandin E2(PGE$_2$), and inhibition rate of 5-lipoxygenase(5-LOX). In comparison with biotin, other plant extracts were tested at the same time which were kudzu vine extract, sage extract, paeonia extract, and dipotassium glycyrrhetinate. Nitric oxide is a signal molecule with functions such as neurotransmission, local vascular relaxation, and anti-inflammation in many physiological and pathological processes. NO can cause apoptosis and necrosis of target cells such as keratinocytes and is generated from L-arginine by nitric oxide synthase (NOS). Prostanoids, including prostaglandins and thromboxanes, are generated by the phospholipase $A_2$/cyclooxygenase(COX) pathway, and leukotrienes are generated by the 5-lipoxygenase pathway from arachidonic acid. Prostaglandin E2 recently have been shown to be beneficial in the resolution of tissue injury and inflammation, also has been implicated as an immunosuppressive agent and plasma levels of PGE$_2$ are elevated in patients sustaining thermal injury. Lipoxygenase metabolites from arachidonic acid have been implicated in inflammation, anti-inflammatory activity of the raw materials was evaluated in vitro by the offered inhibition of lipoxygenase.

  • Twenty six compounds (1-26) were isolated from the root barks of Ulmus davidiana var. japonica. The anti-inflammatory activity of the isolated compounds were evaluated agai nst the generation of inflammatory chemical mediators in bone marrow-derived mast cells. Among them, compounds 10, 11, 13, 15 and 19 inhibited not only cyclooxygenase-2 dependent prostaglandin $D_2$ generation but also 5-lipoxygenase dependent leukotrien $C_4$ generation in a concentration-dependent manner. In addition, compounds 11, 12, 13, 15 and 19 also inhibited $\beta$-hexosaminidase release, a marker of mast cell degranulation reaction, from bone marrow-derived mast cell. These results suggest that the anti-inflammatory activity of U. davidiana might in part occur by both the inhibition of eicosanoid generations and the degranulation reaction of mast cells.

  • DA-5018, a new capsaicin derivative, showed potent analgesic effect comparable to that of morphine in various experimental acute pain models. in this study, whether the analgesic mechanism of DA-5018 is related to opiate receptors or prostanoids was investigated. The affinity of DA-5018 for opiate receptor was determined by receptor binding assay. The Ki values of DA-5018 for nonspecific and specific $\mu$, $textsc{k}$, $\delta$-opiate receptor was 299$\pm$8.88, 735$\pm$215, 2930$\pm$ 163, 1550$\pm$813 nM, respectively and DA-5018 exhibited lower affinity than morphine. DA-5018 (10-"~3$\times$10-′M) inhibited electrically-evoked contractions of the guinea ply ileum and rat vas deferens, and these inhibition was not antagonized by naloxone(10 nM), an opiate receptor antagonist. Antagonism of analgesic effect of 7A-5018 by naloxone was examined by tail pinch test. Analgesic action of DA-5018(0.1 ~2 mg/kg, 5.c.) was not antagonized by naloxone(1 mg/rg, i.p.). These results indicate that pharmacological action of DA-5018 is not related with opiate receptor. Cyclooxygenase and 5-lipoxygenase activities in rat peritoneal neutrophil treated with A23187 and arachidonic acid were measured by radioimmunoassay. DA-5018 stimulated the cyclooxygenase activity and the concentration show-ing the two fold increase of activity was 124$\mu$M. DA-5018 slightly inhibited 5-lipoxygenase activity and these results together indicate that analgesic action of 3A-5018 is not mediated through inhibition of cyclooxy genase or lipoxygenase. These results suggest that the analgesic effect of DA-5018 is not due to blocking opiate receptor or to inhibiting the synthesis of prostanoids in the arachidonic acid metabolism pathway.

  • Present study deals with variations of physicochemicals including $\gamma$-aminobutyric acid (GABA), $\gamma$-oryzanol, free sugar, lipoxygenase activity, fat acidity, and germination rate from Korean brown rice cultivars. With increase of storage time and temperature, GABA, $\gamma$-oryzanol, lipoxygenase activity, and fat acidity increased, whereas free sugar and germination rate was reduced. Among cultivars, 'Gopumbyeo' exhibited the highest contents in GABA and $\gamma$-oryzanol during 12 weeks storage at $25^{\circ}C$ (GABA: $28.6{\pm}5.6{\rightarrow}170.4{\pm}4.6\;mg/100\;g$, $\gamma$-oryzanol: $6.1{\pm}0.7{\rightarrow}6.7{\pm}0.4\;mg/g$) and 'Ilpumbyeo' significantly decreased in free sugar during 12 weeks storage at $10^{\circ}C$ ($1,423.7{\rightarrow}1,058.4\;mg/100\;g$). Moreover, 'Taebongbyeo' exhibited the highest quality owing to low lipoxygenase activity, low fat acidity, and high germination rate. In free sugar compositions, sucrose exhibited the highest content (>70%), followed by fructose (>7%), raffinose (>5%), glucose (>3%), and maltose (>2%) during storage. Based on our results, changes of physicochemicals in stored brown rice may be important information in processing food and functional properties.

  • The physicochemical and sensory properties of bean sprouts with different namulkong cultivars were examined. The five namulkong cultivars included were as follows : Nokchaekong, Dawonkong, Seonamkong, Orialtae, and Pungsannamulkong with light green, black, yellow, dark green, and yellow seed coats, respectively. The bean sprouts were cultivated in a lab and were used for analyses. The instrumental textural hardness and lipoxygenase activity of the heads were higher than those of the stems in both fresh and boiled bean sprouts. The 3 minutes of boiling caused a decrease in the hardness and lipoxygenase activity of the heads and the stems. The chlorophyll a and b contents in the heads were higher than those in the stems, and they decreased with boiling. The fresh bean sprouts with Dawonkong, Seonamkong and Pungsannamulkong showed higher acceptability than those with Nokchaekong and Orialtae. The higher acceptability was observed in the boiled and flavored bean sprouts with Seonamkong and Pungsannamulkong.
