A Study on the Phase Behavior and Stability of the Polar Oil Emulsion System (Polar Oil계 Emulsion의 상거동과 안정성에 관한 연구)

  • O/W emulsions were prepared by adding water to the solution containing amphiphilic resin and the mixed emulsifier of cetyl alcohol polyoxyethylene(20) sorbitan monooleate. Phase behavoir of these emulsions was studied at various HLB(Hydrophilic Lipophilic Balance) values and temperatures. The polar oil emulsion containing the amphiphilic resin showed improved phase stability at various temperatures. Model compounds which contain one of the functional groups in the amphiphilic resin were used in the polar oil phase in order to study the effect of interaction between the functional group and the emulsifier on the phase stability of emulsion. These model compound emulsions showed the phase stability order of poly(acrylic acid)

  • In this study, the formation potential and the stability of nano-emulsions were evaluated according to the structure of various cosmetic oils in Tween 80/Span 80 system using PIC method at 80 oC LP 70, Isopar H and Pripure 3759 of hydrocarbons were both form a stable nano-emulsion, particle size distribution of about 40 nm. A linear structure of silicone oil formed an unstable emulsion, but cyclic or short chain oil formed was a stable nano-emulsion. In ester oils, the particle size of emulsions increases with increasing molecular weight of oils and a stable nano-emulsion could not be obtained in the molecular weight of about 450. The particle size of the nano-emulsion against required HLB value for calculating in consideration of the lipophilic and hydrophilic oil was smaller in the HLB of 8-10.

  • The anti-oxidative effects of Korean red ginseng extracts (total saponine, water extracts, alcohol extracts, lipophilic extracts), which were administered with the concentration of 200 mg/kg BW, were investigated after the injection of paraquat (1,1-dimethyl-4,4-bipyrimidinium dichloride: PQ) with dosage of 100 mg/kg BW on the peritoneal cavity to 6 weeks of 23~26 gm ICR male mice. The accumulation of hydrogen peroxide ($H_2O$$_2$) on the liver of mouse was lowered only in alcohol extract-treated group (p<0.05). The activity of glutathione peroxidase increased in the mouse treated with lipophilic ginseng extracts. And GSSG level was lowered in all groups, and this might be due to the paraquat ions that might prevent the reaction of GSSG into GSH. But we cannot find any relation between glutathione oxide-reductase activity on Korean ginseng extracts. Finally, the lipid peroxidation (MDA) level was lowest (p<0.01) in the group of mouse treated with water extracts of ginseng.

  • Drug delivery technologies are patent protected formulation technologies that modify drug release profile, absorption, distribution, and elimination for the benefit of improving product efficacy and safety, as well as patient convenience and compliance. The most commonly used transdermal system is the skin patch using various types of technologies. Compared with other method of dosage, it is possible to use for a long term. It is also possible to stop the drug dosage are stop if the drug dosage lead to side effect. Polysaccharide, such as karaya gum and locust bean gum(LBG)/water-soluble chitosan oligomer(WSCO) were selected as base materials of TDS. Also, these polymers were characterized in terms of enhancers, tacrine contents. Among these polysaccharide, the permeation rate of karaya gum matrix was fastest in tacrine such as lipophilic drug in vitro. We used glycerin, PEG 400, and PEG 800 as enhancers. Therefore, transdermal absorption of tacrine could be improved by changing vehicle composition or by using penetration enhancers. Especially it would be anticipated that the high permeation efficacy could be obtained by using vehicle that has enhancing effect for itself and by adding enhancers to it.

Simultaneous analysis and occurrences of six pharmaceuticals in surface water by LC/ESI-MS/MS (LC/ESI-MS/MS를 이용한 하천수 중 잔류 6종 의약물질의 동시분석 및 모니터링)

  • The extraction/clean-up and concentrating of pharmaceuticals from surface water were performed by HLB (Hydrophilic-Lipophilic Balanced) cartridge. The method allows for the simultaneous determination of six pharmaceuticals by HPLC/ESI(+)-MS/MS. Recoveries of the pharmaceutical were between 71.1 to 92.6% (except fenbendazole) and the overall variability of the method was below 11.2% (RSD). The calibration curves for the pharmaceuticals from blank surface water showed good linearities (above $r^2$ = 0.99) in the concentration range of 0.007~1.2 ng/mL. The limit of detection (LOD) and the limit of quantification (LOQ) were 7.2~128.7 pg/mL and 23.8~429.1 pg/mL, respectively. The present analytical method can be useful for monitoring residual pharmaceuticals in surface water and other aquatic samples. High concentrations of iopromide and fenbendazole were detected in a few samples of surface water.

  • The purpose of this study was to investigate the effect of reaction conditions, solvents, and surfactants on the average size and size distribution of silica particles in preparing silica fine powders by sodium silicate. Silica fine particles were synthesized by varying kinds of solvents and surfactants using the emulsion method. Span 20, Span 40, Span 60, and Span 80 were used as nonionic surfactants, Dispersing solvents were n-Hexane, n-Heptane, iso-Octane, and n-Decane of the alkane group. In these experiments, it was known that the optimum dispersion stirring time to form the emulsion of the constant size was around 6 min. The mean sizes of silica particles, at a variety of the dispersion stirring speeds, decreased as the dispersion stirring speed increased. Also, in the case of the solvents, the size of the formed silica particles decreased when the molecular weight of the solvent increased. Lastly, in the case of the surfactants, the mean size of silica particles increased as the hydrophobic lipophilic balance (HLB) value of the surfactant decreased.

Protective Effects of Acetylbergenin against Carbon Tetrachloride Induced Hepatotoxicity in Rats

  • The present study was undertaken to investigate whether or not the hepatoprotective activity of acetylbergenin was superior to bergenin in carbon tetrachloride ($CCl_4$)-intoxicated rat. Acetylbergenin was synthesized by acetylating bergenin, which was isolated from Mallotus japonicus. The hepatoprotective effects of acetylbergenin were examined against $CCl_4$-induced liver damage in rats by means of serum and liver biochemical Indices. Acetylbergenin was administered orally once daily for 7 successive days, then a 0.5 ${m/kg}$ mixture of $CCl_4$in olive oil (1:1) was intraperitoneally injected at 12 h and 36 h after the final administration of acetylbergenin. Pretreatment with acetylbergenin reduced the elevated serum enzymatic activities of alanine/aspartate aminotransferase, sorbitol dehydrogenase and $\gamma$-glutamyltransferase in a dose dependent fashion. Acetylbergenin also prevented the elevation of hepatic malondialdehyde formation and depletion of glutathione content dose dependently in $CCl_4$-intoxicates rats. In addition, the decreased activities of glutathione S-transferase and glutathione reductase were restored to almost normal levels. The results of this study strongly suggest that acetylbergenin n has potent hepatoprotective activity against $CCl_4$-induced hepatic damage in rats by glutathione-mediated detoxification as well as having free radical scavenging activity. In addition, acetylbergenin doses of 50 ${mg/kg}$showed almost the same levels of hepatoprotection activity as 100 ${mg/kg}$ of bergenin, indicating that lipophilic acetylbergenin is more active against the antihepatotoxic effects of $CCl_4$ than those of the much less lipophilic bergenin.

  • Transdermal drug delivery(TDS) offers many important advantages. For instance, it is easy and painless, it protects the active compound from gastric enzymes, and it avoids the hepatic first-pass effect. Also, it is simple to terminate the therapy if any adverse or undesired effect occurs. But skin is a natural barrier, and only a few drugs can penetrate the skin easily and in sufficient quantities to be effective. Therefore, in recent years, numerous studies have been conducted in the area of penetration enhancement. The most commonly used transdermal system is the skin patch using various types of technologies. Compared with other method of dosage, it is possible to use for a long term. It is also possible to stop the drug dosage are stopped if the drug dosage lead to side effect. Polysaccharide, such as xanthan gum and algin were selected as base materials of TDS. Also, these polymers were characterized in terms of enhancers and drug contents. Among these polysaccharide, the permeation rate of Paroxetine such as lipophilic drug was the fastest in xanthan gum matrix in vitro. We used glycerin, PEG400 and PEG800 as enhancers. Since dermis has more water content(hydration) than the stratum corneum, skin permeation rate at steady state was highly influenced when PEG400 was more effective for lipophilic drug. Proper selection of the polymeric materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate.

  • New biological treatments were being developed at a record place, but their potential could be compromised by a significant obstacle: the delivery of these drugs into a body. Pharmaceutical delivery is now nearly as important as product. New systems are being developed, and Drug Delivery Markets Series cover these new systems. Transdermal Delivery System(TDS) is often used as a method of drug dosage into the epidermic skin. An approach used to delivery drugs through the skin for therapeutic use as an alternative to oral, intravascular, subcutaneous and transmucosal routes. Various transdermal drug delivery technologies are described including the use of suitable formulations, carriers and penetration enhancers. The most commonly used transdermal system is the skin patch using various types of technologies. Compared with other methods of dosage, it is possible to use for a long term. It is also possible to stop the drug dosage are stopped if the drug dosage lead to side effect. Polysaccharides, such as karaya gum and glucomannan, were selected as base materials of TDS. Also, these polymers were characterized in terms of enhancers, drug contents. Among these polysaccharide, the permeation rate of karaya gum matrix was fastest in fibric acid(ciprofibrate) such as lipophilic drug in vitro. We used glycerin, PEG400 and PEG800 as enhancers. Since dermis has more water content(hydration) than the stratum corneum, skin permeation rate at steady state was highly influenced when PEG400 was more effective for lipophilic drug. Proper selection of the polymeric materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate. Especially, this result suggests a possible use of polysaccharide gel ointment matrix as a transdermal delivery system of anti-hyperlipoproteinemic agent.

Designs and Syntheses of Oxathiin Carboxanilide Analogues and their Antiviral Activities

  • Syntheses of new analogues of oxathiin carboxanilide (UC84) and their antiviral activities were described. The heterocyclic carboxylic acids including oxathiins (4), thiazines (9) and dithiins (13) in which the methyl was replaced either by lipophilic trifluoromethyl- or bulky phenylgroup were synthesized starting from $\beta$-keto esters (5). Reaction of 4, 9 and 13 with thionyl chloride followed by treatment of the substituted aniline 22 gave the corresponding carboxanilides (24a~24f). The carboxanilides were subjected to Laweson's reagent the corresponding thiocarboxanilides (24g~24k). The antiviral activities of the synthesized compounds against human immunodeficiency virus type 1 (HIV-1), poliovirus type 1 (PV-1 ), coxsackie B virus type 3 (CoxB-3), vesicular stomatitis virus (VSV) and herpes simplex virus type 1 (HSV-1) were presented. The antiviral activity against HIV-1 of dithiin carboxanilide (24e) was similar with that of UC84 (24a). The corresponding thiocarboxanilides (24g~24k) showed higher inhibitory activity against HIV-1 than the carboxanilides (24a, 24b, 24d, 24e). The compounds in which ether the lipophilic trifluorormethyl substituents (24d, 24f, 24i ,24k) or bulky phenyl substituent is present in the heterocyclic compounds showed lower inhibitory activity than that of the methyl substituents is present in the compounds against the HIV-1. But the trifluoromethylated dithiin (24f) showed higher inhibitory activity against PV-1 and CoxB-3 virus than commercial antiviral agents, ribavirin (RV).
