Response Mechanism of 5, 10, 15, 20-tetraphenyl(porphyrinato) Manganase(III) chloride-Based Ion-Selective Membranes (망간포르피린을 함유한 고분자형 이온선택성 막전극의 감응 메카니즘)

  • Response mechanism for the chloride-selective membrane doped with 5, 10, 15, 20-tetraphenyl(porphyrinato)manganase(III) chloride(Mn(TPP)Cl) in PVC/DOS matrix is proposed by examining the visible spectra of the corresponding optode membrane. The visible spectra of Mn(TPP)Cl-doped membrane placed in aqueous solution show that the chloride ligand is easily replaced with water molecule. When other halogen ions, such as $F^-$, $Br^-$ and $I^-$, are added to the sample solution, they replace the water ligand, exhibiting distinctive change in the Soret band of Mn(TPP). On the other hand, bulky anions, such as SCN and salicylate, do not form a bond with the central metal. These results suggests that the potentiometric response of Mn(TPP)-based membrane results either from the ligand exchange (water with halides) at the central metal or from the counter ion exchange (chlorides with bulky lipophilic anions) around the positively charged porphyrin molecule in membrane phase. It was also noted that both hydration enthalpies of anions and their binding constants to Mn(TPP) play critical role in determining the potentiometric selectivity pattern of the membrane.

DHA Enrichment of Salmon Patty using Emulsion Curd and Its Component Characteristics (유화커드를 이용한 연어 패티의 DHA 강화 및 이의 식품성분 특성)

  • To prepare high-quality fast foods using salmon, this study developed a docosahexaenoic acid (DHA)-enriched salmon patty, and compared its quality with soybean oil-added patty and commercial patty. The moisture and lipid contents of the DHA-enriched patty were 61.9% and 7.7%, respectively. The moisture content was lower than in the commercial patty, while the lipid content was higher. The lipophilic and hydrophilic browning indices of the DHA-enriched patty were 0.10 and 0.05, respectively, which were lower than those of the commercial patty. The DHA-enriched patty had 5% less trichloroacetic acid (TCA)-soluble nitrogen than the commercial patty, and it was firmer. There were no differences in the proximate composition, browning indices, TCA-soluble nitrogen content, and hardness between the soybean-added and DHA-enriched patties prepared in this experiment. Examining the fatty acid compositions of the patties, the saturated acid and monoenoic acid compositions decreased in the following order: DHA-enriched patty > soybean oil-added patty > commercial patty, while the polyenoic acid composition increased in that order. The total amino acid content of the DHA-enriched patty was 19.13 g/100 g, which was 32% higher than that of the commercial patty (14.47 g/100 g). No enriching effect of minerals would be expected on eating a 100 g DHA-enriched patty, except for phosphorus. The sensory evaluation indicated that the DHA-enriched patty was superior in color and taste to the commercial patty, while there was no difference in flavor between the patties.

  • This study was carried out in order to investigate oxidative deterioration during dehydration at $40^{\circ}C,\;50^{\circ}C$ and $60^{\circ}C$ of sea mussel and baby clam. Moisture content was decreased with drying temperature and time. Sea mussel was dehydrated more rapidly than baby clam that had Harder muscle tissue. Both samples were not reached to Aw 0.62 in case of 10 hrs drying at $40^{\circ}C$, But it reached within 8 hrs in sea mussel and 10 hrs in baby clam at $50^{\circ}C$, respectively. Even if $60^{\circ}C$ could speed up drying, it caused to form more free fatty acid, peroxide, thiobarbituric acid and brown pigments. Lipophilic brown pigment was 10 times higher than hydrophilic and actively increased in all samples. fluorescence intensity was also increased with drying temperature and time. Particularly, it was higher sea mussel than baby clam more or less.

Stable Liquid Paraffin-in-Water Nanoemulsions Prepared by Phase Inversion Composition Method (조성 상전이 방법으로 제조된 안정한 액상 파라핀-물 나노에멀젼)

  • Oil-in-water nanoemulsions were prepared in the system of water/Span 80-Tween 80/long-chain paraffin oil via the PIC (phase inversion composition) method. With the increase of preparation temperature from $30^{\circ}C$ to $80^{\circ}C$, the diameter of emulsion droplets decreased from 120 nm to 40 nm, proving the formation of nanoemulsions. By varying the HLB (hydrophilic lipophilic balance) of mixed surfactants, we found that there was an optimum HLB around 12.0 ~ 13.0 corresponding to the minimum droplet size. The viscosity of nanoemulsions clearly increased with droplet volume fraction, f, but the droplet size slightly increased. Significantly, at ${\phi}{\leq}0.3$, the size distribution of nanoemulsions kept constant more than 2 months. These results proved that the viscous paraffin oil can hardly be dispersed by the PIC method at $30^{\circ}C$, but the increase in preparation temperature makes it possible for producing monodisperse nanoemulsions. Once the nanoemulsion is produced, the stability against Ostwald ripening is outstanding due to the extremely low solubility of the liquid paraffin oil in the continuous phase. The highly stable nanoemulsions are of great importance in cosmetic applications.

Effects of Red Ginseng Component on the Antioxidative Enzymes Activities and Lipid Peroxidation in the Liver of Mice (홍삼 추출물 투여가 생쥐간에서 항산화 효소 활성과 지질과산화에 미치는 효과)

  • The effects of each component (water extracts, alcohol extracts, lipophillic extracts, total saponin, panaxadiol, panaxatriol) of red ginseng on the antioxidative enzyme activities were investigated in the liver in order to screen antioxidative components of red ginseng. 20∼25g ICR mouse which were pretreated with 50 mg/kg body weight of red ginseng component for 15 days. The ability of red ginseng component to protect against oxidative damage to the mouse liver was examined by determining the level of lipid peroxidation (MDA), hydroperoxide (H$_2$O$_2$) and the activities of superoxide dismutase (SOD), catalase. The hepatic total-SOD activity was highest in lipophilic extracts group and panaxadiol group next (p<0.01). The content of hepatic hydroperoxide was lowest in the order of panaxatriol group and alcohol extracts group (p < 0.01). The hepatic catalase activity in the liver was highest in order of lipophillic extracts group (p <0.01) and total saponin group (p<0.05). Finally the lipid peroxidation (MDA) level was lowest in lipophillic extracts group, alcohol extracts group and panaxadiol next (p <0.01). In conclusion, the order of effectiveness of antioxidants was to be lipophillic extracts>panaxadiol >total saponins.

  • In this study, the effects of cosurfactant on phase equilibrium and dynamic behavior were studied in systems containing nonylphenol ethoxylate (NP) surfactant solutions and nonpolar hydrocarbon oils. All the cosurfactants used during this study such as n-pentanol, n-octanol and n-decanol acted as a hydrophobic additive and the hydrophobic effect was found to increase with both increases in chain length and amount of addition of a cosurfactant. Dynamic behavior studies under hydrophilic conditions showed that the solubilization of hydrocarbon oil by NP micellar solution is controlled by an interface-controlled mechanism rather than a diffusion-controlled mechanism. Both spontaneous emulsification of water into oil phase and expansion of oil drop were observed under lipophilic conditions because of diffusion of surfactant and water into oil phase. Under conditions of a three phase region including a middle-phase microemulsion, both rapid solubilization and emulsification of oil into aqueous solutions were found mainly due to the existence of ultralow interfacial tension.

Effect of Partial Replacement of Wheat Flour with High Quality Cassava Flour on the Chemical Composition, Antioxidant Activity, Sensory Quality, and Microbial Quality of Bread

  • In the current study, wheat flour was mixed with high quality cassava flour (HQCF) in several ratios: 90:10, 80:20, 70:30, and 60:40, and used to prepare 10%, 20%, 30%, and 40% National Root Crops Research Institute (NRCRI) cassava bread, respectively. 100% wheat bread was prepared as a control (100% wheat bread). Five bread samples were prepared per group. Antioxidant assays [i.e., 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) scavenging assay, reducing power assay] revealed that the bread samples had considerable antioxidant capacities. Substitution of wheat flour with HQCF at various concentrations resulted in dose dependent decreases in the mineral and protein contents of the resulting bread samples. The crude fiber content of the bread samples was minimal, while the carbohydrate content of the bread samples ranged from 43.86% to 48.64%. A 20% substitution of wheat flour with HQCF yielded bread samples with a general acceptability that was comparable to that of 100% wheat bread. The mean bacteria counts of the bread samples ranged from $2.0{\times}10^3CFU/mL$ to $1.4{\times}10^4CFU/mL$, while the fungal counts ranged from 0 CFU/mL to $3{\times}10^3CFU/mL$. There was a positive correlation between the DPPH antioxidant activities and the reducing powers of the bread samples ($R^2=0.871$) and a positive correlation between the DPPH antioxidant activities and the flavonoid contents of the bread samples ($R^2=0.487$). The higher microbial load of the NRCRI cassava bread samples indicates that these bread samples may have a shorter shelf life than the 100% wheat bread. The significant positive correlation between total flavonoid content and reducing power ($R^2=0.750$) suggests that the flavonoids present in the lipophilic fractions of the bread samples could be responsible for the reductive capacities of the bread samples.

The Effect on Anti-oxidative Activity and Increasing Extraction Yield of Aralia elata Cortex by Gamma Irradiation (감마선 조사에 의한 총목피(Aralia elata Cortex)의 추출수율 증대 및 항산화 효과)

  • This study was designed to investigate the effects on anti-oxidative activities and increasing extraction yield of Aralia elata Cortex by gamma irradiation. Electron spin resonance (ESR) analysis as physical techniques for irradiation identification of Aralia elata showed that a pair of peak appeared on a space of 6.0 mT at the left and right of symmetric unspecific central ESR spectrums, confirming that the plant was gamma-irradiated. The optimum extracting conditions for preparing gamma irradiated samples from Aralia elata Cortex were to extract with 50% ethanol for 15 hrs after 10 kGy irradiation. DPPH scavenging activity and ABTS radical cation inhibitory activity of the water and 50% ethanol extracts from non irradiated and irradiated Aralia elata Cortex was very high as over 80% and 98%, respectively, at tested low concentration of $50{\mu}g/mL$. Antioxidant protection factor (PF) as anti-oxidation indicator of lipophilic compounds showed a very high level of activity as 2.18~2.78 PF. As for TBARs, water and ethanol extracts showed high level. Increase of TBARs inhibitory activity of water extracts was not shown by gamma-ray irradiation but ethanol extracts showed slight increasement of TBARs inhibitory activity with 10 kGy gamma-ray irradiation. These results shown confirmed increasement of extraction yield for phenolic compounds and anti-oxidative activity from Aralia elata. Thus, the treatment of gamma-irradiation can be used a way to amplify a solubility for biological active compounds and anti-oxidative activity in plants.

Physicochemical Quality and Luminescence Characteristics of Gamma-irradiated Dried Fish Products (감마선 조사에 따른 건포류의 이화학적 품질 및 발광 특성)

  • Dried fish products, such as seasoned cuttlefish, anchovy, and filefish, were analyzed to investigate the physicochemical quality and luminescence properties following gamma-irradiation. The overall color difference slightly increased in all irradiated samples, showing significant changes in Hunter's L, a, and b color values. Cuttlefish and filefish showed higher values in hydrophilic browning, whereas anchovy showed a higher value in lipophilic browning. The brown color intensity of filefish showed a dose-dependent increase. The thiobarbituric acid (TBA) value significantly increased in irradiated samples of cuttlefish and anchovy, which have relatively high lipid contents. Irradiated samples, except anchovy, were not suitable for screening using photostimulated luminescence (PSL) because only negative or intermediate values were observed. Thermoluminescence (TL) measurements were suitable for all fish samples, i.e., irradiation-specific glow curves were obtained, signal intensity increased, and the TL ratio fulfilled the criterion. There was no significant difference among the irradiated samples with regard to the sensory properties.

Evaluation of MiR-34 Family and DNA Methyltransferases 1, 3A, 3B Gene Expression Levels in Hepatocellular Carcinoma Following Treatment with Dendrosomal Nanocurcumin

  • Hepatocellular carcinoma (HCC) is the most common primary malignancy of the liver making up more than 80 percent of cases. It is known to be the sixth most prevalent cancer and the third most frequent cause of cancer related death worldwide. Epigenetic regulation constitutes an important mechanism by which dietary components can selectively activate or inactivate target gene expression. The miR-34 family members including mir-34a, mir-34b and mir-34c are tumor suppressor micro RNAs, which are expressed in the majority of normal tissues. Several studies have indicated silencing of miR-34 expression via DNA methylation in multiple types of cancers. Bioactive nutrients like curcumin (Cur) have excellent anticarcinogenic activity and minimal toxic manifestations in biological systems. This compound has recently been determined to induce epigenetic changes. However, Cur is lipophilic and has a poor systemic bioavailability and poor absorption. Its bioavailability is increased through employing dendrosome nanoparticles. The aim of the current study was to investigate the effect of dendrosomal nanocurcumin (DNC) on expression of mir-34 family members in two HCC cell lines, HepG2 and Huh7. We performed the MTT assay to evaluate DNC and dendrosome effects on cell viability. The ability of DNC to alter expression of the mir-34 family and DNA methyltransferases (DNMT1, DNMT3A and 3B) was evaluated using semi-quantitative and quantitative PCR. We observed the entrance of DNC into HepG2 and Huh7 cells. Gene expression assays indicated that DNC treatment upregulated mir34a, mir34b and mir34c expression (P<0.05) as well as downregulated DNMT1, DNMT3A and DNMT3B expression (P<0.05) in both HepG2 and Huh7 cell lines. DNC also reduced viability of Huh7 and HepG2 cells through restoration of miR-34s expression. We showed that DNC could awaken the epigenetically silenced miR-34 family by downregulation of DNMTs. Our findings suggest that DNC has potential in epigenetic therapy of HCC.
