Effect of Nepalese Pseudo Ginseng Components on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid (Nepal산 Pseudo Ginseng 성분이 암독소 호르몬-난의 체지방 분해작용에 미치는 영향)

  • This study was divised to observe an Inhibitory effect toward a lipolytic action of toxohormone-L from large root and small root Nepal pseudo ginseng (NPG ; Nepal products) components by water extract and ethanol precipitate in vitro. Toxohormone-L is known to be a lipolytic factor that was partially purified from the ascites fluid of Sarcoma 180-bearing mice and of patients with hepatoma. The inhibitory effect that inhibited the lipolytic action of toxohormone-L by ethanol Precipitate component of large root NPG (mean 55.5%) was higher (mean 1.37 times) than that of water extract component in final reaction concentration of 500 and 1, 000ug/ml, on the other side inhibitory effect of water extract component in small root NPG (mean 55.5%) was higer (mean 1.14 times) than that of ethanol precipitate component. In a way inhibitory effect of precipitate component In large root NPG(47.6%), when final reaction concentration of sample were 1, 000ug/ml, was about 40% lower than that of Korean red ginseng.

  • This study was divised to observe an inhibitory toward a lipolytic action of toxohormone-L from large root and small root Nepal pseudo ginseng(NPG ; Nepal products) components by water extract and ethanol precipitate in vitro. The inhibitory effect that inhibited the lipolytic action of toxohormone-L by ethanol precipitate component of large root NPG(mean 46.8%) was higher (mean 1.8 times) than that of water extract component in final reaction concentration ,5001g1m1, on the other side inhibitory effect of water extract component in small root NPG(mean 43.9%) was higher(mean 1. 2 times) than that of ethano1 precipitate component, respectively. In a way inhibitory effect of ethanol precipitate component in large root NPG(47.6%), when final reaction concentration of sample were 1,000 U g/ml, was about 4095 lower than that of Korean red ginseng, respectively.

  • This study was devised to observe the inhibitory effect of 7 kinds of the acidic polysaccharide fraction(PGI, PGa, PG3, PGa, PGs, PG6 and PG7) from Korean white ginseng on a lipolytic action of Toxohormone-L. Toxohormone-L is a lipolytic factor, found in ascites fluid of .sarcoma -180 bearing mice and of patients with hepatoma. A substace that inhibited the lipolytic action of Toxohormone-L was isolated from white ginseng powder. This substance was an acidic polysaccharides. In vitro test showed that the inhibitory effect of PGs fraction of the lipolysis by Toxohormone-L was highest percent among other treatments at concentration of 50, 10, 200, 500 and 1,000 Ug/ml of reaction mixture. And total inhibitory activity(units) of PGI and PG4 was highest among other treatments at the same concentration and that of 10 Ug/ml.

  • This study was devised to observe an inhibitory reaction toward an lipolytic action of toxohormone-L from KEUMSUNYEON powder. Toxohormone-L is known to be a lipolytic factor that was purified from the ascites fluid of sarcoma 180-bearing mice and of patients with hepatoma. KEUMSUNYEON powder was found to inhibit toxohormone-L induced lipolysis at its concentration of 0.5 mg/ml.

  • Lipolytic characteristics of candia cylindracea lipase was studied by various triacylglycerols with different fatty acyl chains as substrate. The substrate was emulsified with gum arabic and the rate of hydrolysis was determined by pH stat method. The effects of gum concentration, pH, temperature, and $Ca^{2+}$ ion on the enzyme activities were examined. The results show that the effect of these factors are markedly depending on the structurla nature of substrates. The triolein was the best substrate among tested. Present study demonstrates that for characterization of lipolytic enzymes, it is critically important to select proper substrate and activator.r.

  • This study was performed to investigate the lipolytic effects of Portulaca oleracea L. extract in 3T3-L1 adipocytes. The Portulaca oleracea L. was extracted with extrusion method using twin-screw extruder under $58{\sim}60rpm$ screw speed, $4{\sim}5kg/hr$ feed rate, $140^{\circ}C$ extrusion temperature. The lipolytic action of Portulaca oleracea L. extract was estimated by measuring the amount of glycerol and free fatty acids (FFA) released from 3T3-L1 adipocytes and by measuring the cellular lipid content in 3T3-L1 adipocytes. The hormone sensitive lipase (HSL) mRNA level was analyzed using quantitative real-time PCR. The Portulaca oleracea L. extract at 1 to $100{\mu}g/ml$ suppressed lipid accumulation. The release of glycerol and FFA into the medium, and the mRNA level of HSL were significantly increased by the addition of Portulaca oleracea L. extract at dose-dependent manner. In conclusion, the Portulaca oleracea L. extract was suggested to have the lipolytic effect through release of lipolytic products (FFA and glycerol) of triacylglyceride to the culture medium and suppression of lipid accumulation via up-regulation of HSL gene expression in 3T3-L1 adipocytes.

  • Polyphenol profiles, physicochemical properties, antioxidant activities, and inhibitory effect of adipocyte differentiation of Houttuynia cordata fermented with Lactobacillus brevis B84 were evaluated. Six polyphenols were characterized for this plant by using high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS), and the results were compared with total phenolic content by a spectrophotometric method. The total amount of the identified polyphenols was lower than that determined by the spectrophotometric method. However, the fermentation process influenced polyphenol composition such as content of vanillic acid and caffeic acid. The phytochemical profiles were evaluated by high-performance liquid chromatography with UV and electrospray ionization mass spectrometry detection ($HPLC-DAD-ESI-MS^n$). Total sugar and reducing sugar contents decreased after fermentation. Antioxidant activities such as DPPH, ABTS, and superoxide anion radical scavenging and reducing power were evaluated to compare the beneficial effect after fermentation. Fermented H. cordata increased the lipolytic effect in 3T3-L1 adipocytes. Overall, the results indicate that the fermentation of H. cordata with L. brevis B84 produces changes of phenolic compounds, antioxidant activity, and lipolytic effect.

Effect of Korean Red Ginseng Component on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid (고려홍삼 성분이 암독소 호르몬-L의 체지방 분해작용에 미치는 영향)

  • This study was devised to observe the inhibitory effects of 3 kinds of petroleum ether extracts (percolation by petroleum ether) from Korean red ginseng, Chinese red ginseng and American white ginseng on a lipolytic action of Toxohormone-L which has been known as lipolytic and anorexigenic factors. Toxohormone-L was obtained by partial purification of the ascites fluid from mice which had been Inoculated with sarcoma-180. The yields of petroleum ether extract from Korean red ginseng, Chinese red ginseng and American white ginseng were 0.64, 0.47 and 0.58 and respectively, indicating that the yield of Korean red ginseng was the highest. In vitro, at the concentration of 2 mg /ml, the inhibition rate of lipolysis by the petroleum ether extract of Korean red ginseng, Chinese red ginseng and American white ginseng were 55.1, 50.0 and 44.9% respectively, and the total inhibitory activity per gram of ginseng material were 18, 12 and 13 unit respectively, indicating that the Korean red ginseng was the most effective in the inhibition of the lipolysis.
