Influence of Dietary n3 Polyunsaturated Fatty Acids on Plasma Lipid-Lowering Effect and Peroxidation Level In Rats (쥐에서 n3계 불포화지방산 식이의 혈장지질 저하효과와 과산화물형성에 미치는 영향)

  • To compare the hypolipidemic effect of n6 linoleic acid n3 $\alpha$-linolenic acid and n3 eicosapentaenoic acid plus docosahexaenoic acid, male Sprague Dawley rats weighing about 450g were fed the experimental diets for 6 weeks which composed of fat at 15%(W/W) level and were different only in dietary PUFA. Dietary fat was corn oil, perilla oil, and fish oil concentrate as a source of n6 linoleic acid, n3 $\alpha$-linolenic acid, and n3 eicosapentaenoic acid+docosahexaenoic acid, respectively. Plasma total Chol and HDL-chol levels were significantly-lower in fish oil group than in corn oil and erilla oil groups. Plasma cholesterol lowering effect of PUFa was in the order of n3 EPA+DHA>n3 $\alpha$-linolenic acid>n6 linoleic acid. Plasma TG was significantly lower in both fish oil and perilla oil groups than in corn oil group. Plasma TG-lowering effect was greater by n3 PUFA (EPA+DHA, $\alpha$-linolenic acid) than by n6 PUFA(linoleic acid). However, there were no significant effects on lipoprotein pattern hemolysis, and the levels of tocopherol and malondialdehyde in plasma and RBC by difference dietary fat with sufficient tocopherol supplement. Liver superoxide dismutase activity was significantly increased in proportion to the degree of fat unsaturation, thereby resulted in the lower level of MDA in fish oil group. In conclusion, fish oil and perilla oil rich in n3 PUFA may have important nutritional applications in the prevention and treatment of atherosclerotic disease.

Anti-Obesity Effects of Kimchi Tablet Composition in Rats Fed High-Fat Diet (김치를 이용한 타블렛 조성물이 고지방식이로 유도된 흰쥐의 비만 억제에 미치는 효과)

  • Anti-obesity effects of kimchi tablet composition (KTC) were investigated with male Sprague-Dawley rats fed high-fat diet for 8 weeks. Fifty rats were classified to 4 groups: normal diet (ND), high fat diet (HFD), HFD+KTC 5% and HFD+KTC 10% for feeding experiments. This research showed that the final weight, weight gain, food efficiency ratio and organ weight were decreased by the addition of KTC compared to those of HFD group. Amount of subcutaneous fat of KTC groups were 13.6% and 21.3%, respectively. The plasma triglyceride concentration of KTC groups was lower than that of high fat diet group. Plasma cholesterol concentration of all four groups were not significantly different. HDL-cholesterol concentration of KTC groups was higher than those of high fat diet group. In addition, triglyceride and cholesterol concentrations in the liver and heart were reduced by the addition of KTC. Moreover, triglyceride concentration of fecal and hepatic HMG-CoA reductase activity were increased in KTC group. These results suggested that KTC might be useful for obesity control and good source of functional kimchi tablet.

Antioxidant potential of Sargassum horneri extracts in the liver of mice with PM-induced asthma (미세먼지 흡입 과민성 천식 마우스의 간 조직에서 괭생이모자반 추출물의 항산화 효능)

  • Particulate matter (PM) causes oxidative stress and can rapidly diffuse from the lung to the blood and accumulate in the liver when inhaled. Natural antioxidants can be used to protect against oxidative stress caused by PM. Sargassum horneri, a brown seaweed, possesses antioxidative activity and is a good source of functional foods. Therefore, this study investigated the antioxidant potential of S. horneri extract (SHE) in the livers of PM-induced asthmatic mice. PM inhalation triggered lipid peroxidation and oxidative stress, and SHE treatment attenuated malondialdehyde in the liver of mice with PM-induced asthma. Furthermore, SHE mitigated the increase in catalase activity. Importantly, SHE reduced the activity of 8-oxoguanine glycosylase (OGG1), a DNA repair enzyme. These results suggest that SHE has antioxidant potential for moderating PM-induced oxidative stress and DNA damage in the liver of asthmatic mice.

In Vitro Antioxidant Activity of Alnus firma Extracts (사방오리(Alnus firma) 추출물의 in vitro 항산화 활성)

  • This study evaluated the antioxidant activity of the extract and fractions of Alnus firma. Alnus firma had the highest total phenolic content ($452.80{\pm}7.01{\mu}g$ gallic acid equivalents/mg) in a methanol (MeOH) fraction and the highest total flavonoid content ($112.29{\pm}11.14{\mu}g$ rutin equivalents/mg) and antioxidant capacity ($936.23{\pm}0.07{\mu}g$ ${\alpha}$-tocopherol equivalents/mg) in an ethylacetate (EA) fraction. The antioxidant activities of various solvent extract fractions of Alnus firma were evaluated using various antioxidant assays, including ${\beta}$-carotene-linoleate assay, reducing power assay, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay, metal chelating activity assay, superoxide anion radical scavenging assay, and lipid peroxidation inhibition assay using the ferric thiocyanate method. These activities were compared with those of ascorbic acid, butylated hydroxyanisole (BHA), gallic acid (GA), butylated hydroxytoluene (BHT), and ${\alpha}$-tocopherol. First, at a $250{\mu}g/ml$ concentration, the EA and MeOH fractions of A. firma showed 92.43% and 89.20% DPPH radical scavenging activity, respectively. Second, $50{\mu}g/ml$ of the EA fraction exhibited 72.49% superoxide anion radical scavenging activity, a little greater than the same dose of GA (60.88%). Finally, 0.5 and 1 mg/ml of the EA fraction showed 73.45% and 73.29% inhibition of peroxidation in the ${\beta}$-carotene-linoleic acid system, respectively. The decreasing order of reducing power was EA fraction > n-butanol (BuOH) fraction > dichloromethane (DCM) fraction > n-hexane (HX) fraction. The results obtained in the present study indicated that Alnus firma can be used as an easily accessible potential source of natural antioxidants.

MoJMJD6, a Nuclear Protein, Regulates Conidial Germination and Appressorium Formation at the Early Stage of Pathogenesis in Magnaporthe oryzae

  • In plant-pathogen interactions, Magnaporthe oryzae causes blast disease on more than 50 species of 14 monocot plants, including important crops such as rice, millet, and most 15 recently wheat. M. oryzae is a model fungus for studying plant-microbe interaction, and the main source for fungal pathogenesis in the field. Here we report that MoJMJD6 is required for conidium germination and appressorium formation in M. oryzae. We obtained MoJMJD6 mutants (ΔMojmjd6) using a target gene replacement strategy. The MoJMD6 deletion mutants were delayed for conidium germination, glycogen, and lipid droplets utilization and consequently had decreased virulence. In the ΔMojmjd6 null mutants, global histone methyltransferase modifications (H3K4me3, H3K9me3, H3K27me3, and H3K36me2/3) of the genome were unaffected. Taken together, our results indicated that MoJMJD6 function as a nuclear protein which plays an important role in conidium germination and appressorium formation in the M. oryzae. Our work provides insights into MoJMJD6-mediated regulation in the early stage of pathogenesis in plant fungi.

Inhibition of Glycation End Products Formation and Antioxidant Activities of Ilex paraguariensis: comparative study of fruit and leaves extracts

  • Objectives: Ilex paraguariensis (Aquifoleaceae) is cultivated to produce "yerba mate". Due to its nutritional, energizing, hypoglycemic and antioxidant effects, it is used in the elaboration of food, pharmaceuticals, and cosmetics. The oxidative stress related to protein glycation and production of advanced glycation end products (AGEs) leads to the development of several diseases. The objective of this work was to compare the antioxidant and anti-AGEs activity of a decoction of fruits (F) and leaves (L). Methods: The antioxidant activity was assayed by the DPPH assay and the inhibition of egg yolk lipid peroxidation (ILP), and anti-AGEs activity, through the inhibition of the formation of fructosamine (IF), β-amyloid (Iβ), protein carbonylation (IC) and AGEs (IA). Polyphenols were quantified by HPLC. Results: Maximum response ± SEM: For F 0.01 ㎍/mL: IF = 42 ± 4%, IC = 17 ± 2% and for 10 ㎍/mL: IA = 38 ± 4%, Iβ = 67 ± 7%. For L 0.1 ㎍/mL: IF = 35 ± 2%, IC = 19 ± 2% and for 100 ㎍/mL: IA = 26 ± 3%, Iβ = 63.04 ± 2%. The DPPH IC50 = 134.8 ± 14 ㎍/mL for F and 34.67 ± 3 ㎍/mL for L. The ILP IC50 = 512.86 ± 50 ㎍/mL for F and 154.8 ± 15 ㎍/mL for L. By HPLC L presented the highest amounts of flavonoids and caffeoylquinic acids. F and L showed strong anti-AGEs activity, affecting the early stages of glycation at low concentrations and the late stages of glycation at high concentrations. The highest activity for both F and L was seen in the IF and Iβ. F presented the highest anti-AGEs potency. L presented the highest antioxidant potency, which was related to the highest content of polyphenols. Conclusion: The fruits of I. paraguariensis could be a source of antioxidant and anti-AGEs compounds to be used with medicinal purposes or as functional food.

Nutrition Components and Physicochemical Properties of Acer termentosum Maxim. Leaf (벌나무 잎의 영양성분 및 이화학 특성)

  • This study examined the nutrient components and physicochemical properties of Acer termentosum Maxim. leaf as a natural health food source. To accomplish this purpose, the general and antioxidative contents of Acer termentosum Maxim leaf were measured. Total contents of carbohydrates, crude protein, crude lipid, and ash were 53.6%, 24.3%, 3.5%, and 3.5%, respectively. Caloric content of Acer termentosum Maxim was 246.5 kcal, while total dietary fiber was 46.7%. Regarding mineral contents, K was the most abundant mineral, followed by Ca, Mg, and P. Therefore, Acer termentosum Maxim is an alkali material. Total phenol contents of the 70% ethanolic extracts of Acer termentosum Maxim was $116.35{\pm}1.4mg\;GAE/g$. Total flavonoid contents of the 70% ethanolic extracts were $20.3{\pm}1.23mg\;RE/g$. The antioxidative activities of Acer termentosum Maxim. were significantly increased in a dose dependent manner on DPPH(1,1-Diphenyl-2-picrylhydrazyl) radical scavenging, ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging, FRAP (ferric reducing antioxidant power) activity, reducing power. It is expected that follow up study of Acer termentosum Maxim through developing processed food and evaluation of their functional properties would provide useful information as a source of functional foods.

Nutritional Characteristics and Some Bioactive Components Contents of Sophorae fructus (괴각(槐角)의 식품영양학적 접근 및 몇 가지 생리활성물질 함량 분석)

  • The purpose of this study is to determine the possibility of using Sophorae fructus as natural health food source. To accomplish this purpose, the contents of general and antioxidative nutrients of Sophora fructus were measured. The contents of carbohydrate, crude protein, crude lipid and ash are 75.9%, 16.4%, 2.41%, and 5.2%, respectively, while the calories of Sophora fructus was 337.3 kcal. Total dietary fiber was 15.07% of total carbohydrates. The percentages of water soluble dietary fiber to insoluble dietary fiber were 1.09% and 10.36%, respectively. The protein was composed of a total of 18 different kinds of amino acids. The contents of essential and non-essential amino acids were 2,310.91 mg and 5,218.52 mg. The K was the largest mineral followed by Ca, P and Mg, which means Sophorae fructus is alkali material. The contents of saturated fatty acids, monounsaturated fatty acids and polyunsaturated fatty acids were 24.94%, 32.40%, and 32.86%, respectively. Therefore, the amount of the total unsaturated fatty acid was higher than that of any other plant. The content of vitamin C in Sophorae fructus was higher than that of any other plant, which suggests that it could increase blood elasticity. The content of rutin, which is responsible for capillary vessel permeability, was 1.78%. The contents of water soluble antioxidative materials in 1 mL of water-extracted Sophorae fructus were $4.95\;{\mu}g$ which is comparable to 1,560.96 mmol of vitamin C in antioxidant effect. The general nutrients and other antioxidatant bioactive materials in Sophorae fructus were also potential materials for good health food. It is expected that a follow up study on Sophorae fructus through developing processed food and evaluation of their functional properties would provide useful information as a source of medicinal foods.

Composition of Amino Acids, Minerals, and Heavy Metals in Differently Cooked Laver (Porphyra tenera) (김(Porphyra tenera)의 조리방법에 따른 아미노산, 무기질, 중금속 함량 분석)

  • Laver, a red algae belonging to the genus Porphyra tenera, is one of the most widely consumed edible seaweeds in Korea, China, and Japan. Lavers are usually consumed in dried, roasted, and seasoned forms to improve their palatability. We evaluated the composition of amino acids, minerals, and trace heavy metals in these three differently cooked forms of laver. The moisture and ash contents of three differently cooked lavers ranged from 1.49~9.69% and 6.07~10.31%, respectively. The crude protein and lipid content ranged from 17.24~36.88% and 0.52~42.42%, respectively. Dried laver was found to be a good source of amino acids such as taurine, alanine, and glutamic acid (871.10 mg, 833.53 mg, and 719.77 mg per 100 g dry weight, respectively). Laver was a good source of macro and micro minerals such as K, Ca, Mg, Na, P, I, and Fe, although laver more extensively cooked (roasted and seasoned) contained less minerals compared to the dried form. Mercury levels in the three differently cooked forms of laver were all less than 100 ng/g dry weight (the limit of detection with our methodology). The levels of arsenic were the most abundant elements in the differently cooked laver. There was a clear variation, depending on the cooking process, in terms of amino acid, mineral, and trace metal contents of laver.

Food Component Characteristics of Red-tanner Crab (Chionoecetes japonicus) Paste as Food Processing Source (식품가공소재로서 게 페이스트의 식품성분 특성)

  • The objective of this study was to investigate the components and nutritional quality of red-tanner crab (Chionoecetes japonicus) paste in order to explore possibility for food processing source such as surimi gel containing crab paste. Yield of crab paste was $30\%$ from whole body after crushing and dehydrating. Crude protein contents $(9.5\%)$ of crab paste was lower than that $(13.1\%)$ of crab muscle, but fat $(0.5\%)$ and ash contents $(8.0\%)$ of paste were higher than $0.2\%\;and\;1.3\%$ of crab muscle, respectively. Volatile basic nitrogen (VBN) content of the crab paste was lower than those of the edible parts. Total amino acid content (9,497mg/l00g) of paste was lower than that (12,980mg/100g) of muscle. Aspartic acid, glutamic acid, lysine and leucine were the predominant amino acids in the protein fraction. The calcium content (6,539mg/l00g) was higher than those of phosphorus (579mg/100g), and potassium (793mg/100g) while manganese and iron were present in trace amounts. Major fatty acids of total lipid were 16:0, 18:1n-9, 20:5n-3 and 22:6n-3, and no difference of composition between paste and muscle. Sensory evaluation showed that scores of color and flavor of $15\%$ substituted surimi gel increased significantly when compared to surimi gel without crab paste (p<0.05). From the above results, the addition of crab paste enhanced nutrition and functionality of surimi gel.
