Lactobacillus rhamnosus CBT-LR5 Improves Lipid Metabolism by Enhancing Vitamin Absorption

  • Probiotics provide a symbiotic relationship and beneficial effects by balancing the human intestinal microbiota. The relationships between microbiota changes and various diseases may predict health abnormalities and diseases. Treatment with vitamins and probiotics is one therapeutic approach. To evaluate the effect of probiotics on vitamin absorption, we chose Lactobacillus rhamnosus CBT-LR5 treatment, which has resistance to vitamin C-inducible toxicity, with vitamins in high-fat diet (HFD)-induced obesity models. CBT-LR5 affected the absorption of micronutrients, such as ionic minerals and water-soluble vitamins. An increase in vitamin C absorption by CBT-LR5 enhanced the antioxidant response in HFD-induced obesity models. Increased vitamin B absorption by CBT-LR5 regulated lipid metabolism in HFD-induced obesity models. These favorable effects of CBT-LR5 on the absorption of vitamins should be investigated as candidate therapeutic target treatments for metabolic diseases.

Protectins Effects of Vitamin E against Immobilization Stress-Induced Oxidative Damage in Rat Brain (스트레스로 인한 뇌조직의 산화적 손상에서 Vitamin E의 방어 효과)

  • The remarkable change of phenomenon induced by stress increase energy metabolism that can induce many reactive oxygen species (ROS) production. ROS can peroxidize cellular macromolecules including lipid and protein. The object of this study was to investigate whether stress may induce cellular damage by producing ROS and whether vitamin E, as a strong lipid-soluble antioxidant, can protect cells against reactive oxygen species produced by noise and immobilization stress in SD rats. The stress group increased 5-hydroxyindole aceti acid (5-HIAA) , one of the stress hormone, in brain tissue and free fatty acid in plasma. Vitamin I treatment had no effect on 5-HIAA but free fatty acid contents decreased with a fortified vitamin I diet. Furthermore, the body weight of vitamin I-treated rats increased more than that of the stress group. Lipid peroxidation and protein degradation as an index of oxidative damage in brain tissue decreased with the use of the fortified vitamin I diet supplement. The results suggest that vitamin I supplements have a protective effect against noise and immobilization stress-induced oxidative damage in brain tissue.

  • It is well known that ascorbic acid (VC) is an important factor for several physiological reactions. In the skin, VC works as an anti-aging agent due to removing of oxidative stress generated by UV irradiation and stimulation of collagen synthesis. Thus, developing more effective VC derivatives is an important issue in creating anti-aging skin care products. Our study succeeded to develop a novel ascorbic acid derivative, ascorbic acid tetra-2-hexyldecanoate (VC-IP), which is a lipid-soluble pro-VC. The purpose of this study was to indicate the effects of VC-IP as pro-VC and anti-aging agent. First, it was examined whether VC-IP is converted to VC in physiological conditions. Since VC was detected from the cell extracts treated with VC-IP, it was indicated that VC-IP is a pro- VC.(omiited)

Effects of Vitamins C and E on Hepatic Drug Metabolizing Function in Nypoxia/Reoxygenation (저산소 및 산소재도입시 vitamin C와 E가 간장 약물대사 기능에 미치는 영향)

  • Liver isolated from 18 hours fasted rats was subjected to $N_2$hypoxia (for 45 min) followed by reoxygenation (for 30 min). The perfusion medium used was Krebs-Henseleit bicarbonate buffer (pH 7.4, $37^{\circ}C$). Vitamin C (0.5 mM) and trolox C (0.5 mM), soluble vitamin E analog, were added to perfusate. Lactate dehydrogenase (LDH), total glutathione, oxidized glutathione, lipid peroxide and drug-metabolizing enzymes were measured. After hypoxia LDH significantly increased but this increase was attenuated by vitamin C and combination of vitamin C and E. Total glutathione and oxidized glutathione in perfusate markedly increased during hypoxia and this increase was inhibited by vitamins C, E and its combination. Similarly; oxidized glutathione and lipid peroxide in liver tissue increased after hypoxia and reoxygenation and this increase was inhibited by vitamin I and combination of vitamin C and E. Hepatic drug metabolizing function (phase I, II) were suppressed during hypoxia but improved during reoxygenation. While vitamins C and E only increased glucuronidation, the combination of vitamin C and E increased the oxidation, glucuronidation and sulfation. Our findings suggest that vitamins C and E synergistically ameliorates hepatocellular damage as indicated by abnormalities in drug metabolizing function during hypoxia/reoxygenation and that this protection is in major part, caused by decreased oxidative stress.

  • Potential mechanisms by which vitamin E improves oxidative stability of myoglobin are documented. The basis by which this lipid-soluble antioxidant, ${\alpha}$-tocopherol, protects water-soluble oxymyoglobin is beginning to be understood. Recent evidence suggests that ${\alpha}$-tocopherol delays the release of prooxidative products of lipid oxidation from biomembranes, which in turn delays oxymyoglobin oxidation and the concomitant loss of desirable beef color. ${\alpha}$, ${\beta}$-Unsaturated aldehydes are one class of lipid oxidation products that enhance oxymyoglobin oxidation in vitro and appear to act by covalently binding to the protein. If ${\alpha}$-tocopherol delays the formation of these reactive aldehydes, then this could inhibit the prooxidative effect of these oxidation products toward oxymyoglobin. Additionally, ${\alpha}$-tocopherol may exert part of its color-stabilizing effect in beef by enhancing the metmyoglobin reduction.

Investigation of Fatty Acids Intake Status and Its Correlation with Body Fat Accumulation in College Students in Gyeongbuk Area (경북지역 일부 대학생들의 식사 중 지방산 섭취양상과 체지방 축적의 상관관계 분석)

  • Relationship between lipid intake and obesity has been well-addressed but recent findings indicated that the type of lipid or composition of lipid in the diet also contributes to body fat accumulation and consequential health outcome. The purpose of this study was to investigate the status of nutrition intake including fatty acids, lipids and lipid soluble nutrients between the obese and lean college students and to analyze the relationship between the intake of certain fatty acids and body fat accumulation. Anthropometric elements including body weight, height, body fat and composition were measured, and dietary recall was conducted on a total of 114 college students. Data showed that total calorie intake and total lipid intake were not significantly different between the obese and lean subjects, in both male and female students. However, male obese subjects ate more amount of plant lipids and palmitic acids (C16:0) from their diet (p<0.05), while female obese subjects consumed more linoleic acids (18:2) and linolenic acids (C18:3) compared to normal subjects (p<0.01). Correlation analysis revealed that the consumption of palmitoleic acid (C16:1) and lipid soluble vitamin D were negatively (p<0.05) correlated with body fat accumulation in all subjects and these findings were supported by simple linear regression analyses for those variables. These results implicate that rather than only considering the amount of lipids, suggesting a proper type of lipids or lipid metabolites can be considered in nutrition counseling or education.

Onion Supplementation Inhibits Lipid Peroxidation and Leukocyte DNA Damage due to Oxidative Stress in High Fat-cholesterol Fed Male Rats

  • The aim of this study was to investigate effects of onion, red onion, or quercetin on plasma antioxidant vitamin, lipid peroxidation, and leukocyte DNA damage in rats fed a high fat-cholesterol diet. Forty SD male rats were assigned to normal control, high fat-cholesterol diet (HF), or HF+5% onion powder, HF+5% red onion powder, or HF+0.0l% quercetin. The HF diet resulted in significantly higher plasma lipid peroxidation which decreased with onion, red onion, or quercetin supplementation. Leukocyte DNA damage induced by HF diet decreased significantly in rats fed onion and red onion, while quercetin supplementation had no effect on preventing leukocyte DNA damage. $H_2O_2$ induced leukocyte DNA damage exhibited a highly significant negative correlation with plasma retinol and tocopherols. These results suggest that onion or red onion powder exerts a protective effect with regard to DNA damage in rats fed HF diet. However, 0.01% quercetin in pure form might not be effective at preventing DNA damage.

In Vitro Antioxidant Activity Profiles of ${\beta}$-Glucans Isolated from Yeast Saccharomyces cerevisiae and Mutant Saccharomyces cerevisiae IS2

  • To explore the possible usefulness of ${\beta}$-glucans as natural antioxidants, the antioxidant profiles of ${\beta}$-glucan, extracted from Saccharomyces cerevisiae KCTC 7911, and water soluble and insoluble mutant ${\beta}$-glucan, isolated from yeast mutant S. cerevisiae IS2, were examined by five different in vitro evaluation methods: lipid peroxidation value (POV), nitric oxide (NO), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, reducing power, and ${\beta}$-carotene diffusion assay. The antioxidant activities of all ${\beta}$-glucans evaluated in POV test were comparable to or better than that of the known antioxidant, vitamin C. Remarkably, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan possessed 2.5-fold more potent activity than vitamin C at a dosage of 2 mg. Although vitamin C showed 100-fold greater activity than all ${\beta}$-glucans in NO and DPPH tests for measuring the radical scavenging capacity, all ${\beta}$-glucans revealed higher radical scavenging activity than the known radical scavenger, N-acetyl-L-cysteine (NAC), in DPPH test. The water insoluble mutant ${\beta}$-glucan had 2.6- and 5-fold greater antioxidative activity than water soluble ${\beta}$-glucan in NO and DPPH tests, respectively, showing that all ${\beta}$-glucans were able to scavenge radicals such as NO or DPPH. While all ${\beta}$-glucans revealed lower antioxidant profiles than vitamin C in both reducing power activity and ${\beta}$-carotene agar diffusion assay, the ${\beta}$-glucan and water insoluble mutant ${\beta}$-glucan did show a marginal reducing power activity as well as a considerable ${\beta}$-carotene agar diffusion activity. These results confirmed the potential usefulness of these ${\beta}$-glucans as natural antioxidants.

Variation in Chemical Components of Korean Green Tea (Camellia sinensis L.) Resulted from Developing Stages and Processing Recipe (한국산 녹차의 채엽시기 및 제조법에 따른 화학성분 변이)

  • In this study physicochemical characteristics such as crude lipid, crude protein and total sugar contents have been analyzed with samples taken at different developing stages and growing sites, Bongsan-ri(steam-roasted green tea) and Buchun-ri(roasted green tea) area. Leaf area of new leaf was larger in Buchun-ri(7.23cm$^2$) than in Bongsan-ri(6.93cm$^2$). Variation of leaf area by the developing stages was the largest at between stage II (May 5) and stage III(May 18). Dry matters ratio of tea leaf were 26.3% in Bongsan-ri and 26.6% in Buchun-ri. Water content, ash, water-soluble matters, total sugar content of green tea sampled in Bongsan-ri and Buchun-ri were 5.6, 6.1, 33.1, 4.6 and 6.2, 5.8, 35.2, 2.8%, respectivley. Crude lipid, total nitrogen and water-soluble protein, inorganic matters content of green tea sampled in Bongsan-ri and Buchun-ri were 2.8, 5.1, 1.1, 2.56 and 2.2, 5.7, 1.0, 2.34%, respectively. At the later developing stages crude lipid, total sugar contents were increased, whereas total nitrogen had low content. Vitamin C, Vitamin E and Vitamin A content of green tea produced in Bongsan-ri and Buchun-ri area were 413mg%, 71.8mg%, 32.15IU and 311mg%, 52.3mg%, 25.68IU, respectively. Vitamin C and Vitamin E content were increased toward the later sampling stages. Water-soluble matters and water-soluble protein content of green tea prepared by the steam-roasted method and the roasted method were increased by 0.11, 0.13 and 0.09, 0.08%, respectively, compared to unroasted check. Crude lipid content of green tea prepared by the steam-roasted method and the roasted method were decreased by 0.13 and 0.24%, respectively. Total nitrogen content of green tea prepared by the steam-roasted method and the roasted method was changed 0.019 and 0.036%, respectively and total nitrogen content showed minute different between two methods.

  • Vitamin D plays a vital role in maintaining blood concentrations of Ca and P. In lipid metabolism, vitamin D is also known to negatively affect the development of adipocytes, but it has received little attention with regard to improve marbling score in beef cattle via restricting its supply. Recently, the method of feeding fat soluble vitamins(including vitamin D)-deficient diets in Korean native cattle (Hanwoo) has been spread by feed companies and TMR plants. But proper guidelines in feeding vitamin D-deficient diets should be provided, otherwise it can adversely affect the health of cattle. To maximize marbling score, a controlled level of vitamin D in blood needs to be maintained and the proper period for feeding vitamin D-deficient diets should be provided. This review was conducted to highlight the regulatory effects of vitamins D on adipocyte differentiation, and provide information on improving beef quality grades in relation to feeding vitamin D-deficient diets in beef cattle.
