The Effects of Sedum sarmentosum Bunge Extract using Super Critical Carbon Dioxide on Lipid Metabolism, Lipid Peroxidation and Antioxidation in Carbon Tetrachloride Induced Hepatotoxicity in Rats (초임계 이산화탄소를 이용한 돌나물 추출물이 사염화탄소로 유발된 흰쥐의 지질대사, 지질과산화 및 항산화에 미치는 영향)

  • Extraction of Sedum sarmentosum Bunge by super critical carbon dioxide was operated under $40-50^{\circ}C$and 200-250 atm, thus, yield of extraction was very low as 4 wt%. Rats were administrated with the extract orally once a day for succesive 6 days, followed by treatment with carbon tetrachloride ($CCl_4$) on the sixth day. The activities of aminotransferase, alkaline phosphatase, ${\gamma}$-glutamyl transpeptidase, lactate dehydrogenase and contents of triglyceride, total cholesterol in the extract-pretreated rats were decreased compared to the $CCl_4$controled rats, whereas content of HDL-cholesterol was increased. Especially content of hepatic malondialdehyde (MDA) and atherogenic index (AI) were decreased and HTR was increased in the extract-pretreated rats, and reduced peroxidative liver damage in the $CCl_4$-induced hepatotoxicity rats. In addition, activities of hepatic superoxide dismutase, catalase, glutathione peroxidase in the extract-pretreated rats were significantly decreased compared to the $CCl_4$ controled rats, but the content of glutathione was significantly increased. These results suggest that extract of Sedum sarmentosum Bunge has hepatoprotective effect in the $CCl_4$-intoxicated rats.

Influences of Chitosan, Sericin and Collagen Extract Complexes on the Improvement Actions of Lipid Component in Diabetes (키토산과 세리신 및 콜라겐 추출 혼합물이 당뇨의 혈당 및 지질성분 개선효과)

  • This study was conducted to examine the influences of chitosan, sericin and collagen extract complexes (1:1:1, w/w/w, CSC-F-005) in blood glucose and lipid concentrations in the sera of streptozotocin (STZ, 55 mg/kg BW, IP injection)-induced diabetic rats (SD strain) fed on experimental diets for 5 weeks. The concentrations of blood glucose, total cholesterol, HDL-cholesterol, ratio of HDL-cholesterol concentration to total cholesterol, atherosclerotic index, LDL-cholesterol, free cholesterol, cholesteryl ester, triglyceride (TG) and phospholipid (PL) in serum were effective on the metabolic regulation in diabetic rats. The activities of alkaline phosphatase (ALP) and aminotransferase (AST, ALT) in serum were lower in the extract complexes (CSC-F-005) than in the diabetic rats. The results shown above suggested that CSC-F-005 extract complexes supplementation effectively improvement of blood glucose and lipid components in the serum of STZ-induced diabetic rats.

  • The objective of this study was to assess improvements caused by chitosan, sericin and collagen peptide extract complexes (1:1:1, w/w/w, CSC-F-005) in lipid concentrations in the sera of dyslipidemic rats (SD strain) fed on experimental diets for 5 weeks. Serum concentrations of total cholesterol, HDL-cholesterol, ratio of HDL-cholesterol concentration to total cholesterol, atherosclerotic index, LDL-cholesterol, free cholesterol, cholesteryl ester, triglyceride, phospholipid and blood glucose were effective on the metabolic regulation of dyslipidemic rats. The activities of alkaline phosphatase, aspartate aminotransferase and alanine aminotransferase in serum were remarkably lower in the extract complexes (CSC-F-005) than in the dyslipidemic model. From the above results shows that CSC-F-005 extract complexes were effective on the improvement of the lipid metabolism in sera of dyslipidemic rats.

Effects of Chitosan Oligosaccharide Supplementation on Blood Glucose, Lipid Components and Enzyme Activities in Hyperglycemic Rats (키토산 올리고당이 당뇨성 흰쥐의 혈당과 혈중 지질성분 및 효소활성에 미치는 영향)

  • The principal objective of this study was to assess the effects of chitosan oligosaccharide supplementation on the improvement of blood glucose, lipid components and enzyme activities in the serum of streptozotocin(STZ, 55 mg/kg B.W., I.P. injection)-induced hyperglycemic rats fed on experimental diets for 5 weeks. The concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, free cholesterol, cholesteryl ester ratio, triglycerides(TG) and phospholipids(PL) in serum were remarkably higher in the hyperglycemic group(group BSW) and STZ(I.P.)+chitosan oligosaccharide supplementation group(group ECW) than those in the control group(group BW, basal diet+water). However the concentrations of blood glucose, total cholesterol, atherosclerotic index, LDL, LDL-cholesterol, tree cholesterol, cholesteryl ester ratio, TG and PL in serum were lower in the ECW group than in the BSW group, whereas the ratio of HDL-cholesterol concentration to total cholesterol and HDL-cholesterol concentration in the ECW group were higher than in the BSW group. The activities of alkaline phosphatase(ALP) and aminotransferase(AST, ALT) in serum were lower in the ECW group than in the hyperglycemic BSW group. The results shown above suggested that chitosan oligosaccharide supplementation effectively improves blood glucose, lipid composition and enzyme activities in the sera of STZ-induced hyperglycemic rats.

Studies of the Cordyceps militaris Extract Administration on the Metabolic Enzyme Activities in Hypercholesterolemia (동충하초 엑스의 고콜레스테롤혈증 대사효소 활성 변동에 관한 효과)

  • The objective of this study was to investigate the effects of the feeding Cordyceps militaris extract on the improvement of the free fatty acid, lipid peroxide, creatinine and enzyme (creatine phosphokinase; CPK, lactate dehydrogenase; LDH, alkaline phosphatase; ALP, lecithin cholesterol acyltransferase; LCAT) activities in the sera of dietary hypercholesterolemic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of free fatty acid, lipid peroxide and CPK, LDH, ALP activities in sera were fairly reduced in the Cordyceps militaris extract administration group (CHE) than in the hypercholesterolemic diet group (CHD). However, no significance was found in the effect of an creatinine concentration among the groups. The LCAT activity in serum was increased in the Cordyceps militaris extract administration (CHE) than in the hypercholesterolemic diet group (CHD). From these results, Cordyceps militaris extracts were effective on the improvement of the lipid components and metabolic enzyme activities in sera of dietary hypercholesterolemic rats.

Ultrastructural and Cytochemical Studies on the Endometrial Surface Epithelial Cells of Guinea Pig During Estrous Cycle (發情週期에 따른 Guinea Pig의 子宮內膜 表層上皮細胞의 微細構造 및 細胞化學的 硏究)

  • Cyclical changes in the fine structures of the surface epithelial, stroma and glandular cells of guinea pig endometrium during the estrous cycle were studied by transmission and scanning electron microscopy. Cytochemical studies were made in order to investigate the ultrastructural localization of the acid phosphatase, alkaline phosphatase and ATPase in these cells. The results obtained are as follows: 1. The endometrial surface epithelium was pseudostratified columnar during estrus and meterstrus, and simple columnar during proestrus and diestrus. The characteristic features observed in these cells include increased nucleocytoplasmic ratio at proestrus, elongated shapes of both the nucleus and the entire cell, increased volume of the cytoplasm and cytoplasmic bulding into the lumen during estrus, and smaller surface epithelial cells during metestrus. 2. In the cytoplasm of surface epithelial cells, the numbers of mitochondria and free ribosomes were increased, and rough endoplasmic reticulum and Golgi complex appeared during estrus, and the degenerated cells, lipid droplets, multilamellated bodies and lysosomes appeared during diestrus. 3. During estrus, scanning electron microscopic observations of endometrial surface showed a regular arrangement with polygonal outlines of epithelial cells, distinct intercellular border, and bulged surface into the lumen, whereas flat surface and indistinct cell border were characteristic during meterstrus and diestrus. 4. Microvilli which aligned on the surface were longer and most abundant during estrus while short and aparse during other phases. 5. Cytochemical studies indicated that during metestrus acid phosphatase activities were localized in the microvilli and vacuoles, and alkaline phosphatase activities were significant around luminal surface and lateral cell membrane in the surface epithelial cells. ATPase activities were present on the microvilli and cell membrane during proestrus and estrus.

  • Pretreatment of acidic polysaccharide of Korean red ginseng (AcPS) for two weeks remarkably lowered the elevated content of lipid peroxide and levels of aminotransferases, sorbitol dehydrogenase, ${\gamma}$-glutamyltransferase, alkaline phosphatase and lactate dehydrogenase in liver intoxicated by acetaminophen (AA) . Pretreatments of AcPS also strengthen the liver function of glutathione related detoxication system indicated by glutathione contents and activities of glutathione S-transferase and glutathione reeducates which were affected by AA treatments. Activity of ${\gamma}$-glutamylcysteine syntheses was not changed by AcPS pretreatment whereas the activity of flu tathione reeducates was increased significantly. These results collectively indicate that the treatments of AcPS can promote the metabolism of lipid and reduce the production of peroxide in acetaminophen-intoxicated animals.

Hepatoprotective Effect and Antioxidant Role of Caesalpinia bonducella on Paracetamol-induced Hepatic Damage in Rats

  • The hepatoprotective effect of methanol extract of leaves of Caesalpinia bonducella was studied by means of paracetamol induced liver damage in rats. The degree of protection was measured by using biochemical parameters such as serum transaminase (SGPT and SGOT), alkaline phosphatase (ALP), bilirubin, and total protein. Further, the effects of the extract on lipid peroxidation (LPO), glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT) were estimated. The methanol extract of C. bonducella (MECB) (50,100 and 200 mg/kg) produced significant (P<0.01) hepatoprotective effect by decreasing the activity of serum enzymes, bilirubin, and lipid peroxidation, while it significantly increased increased the levels of GSH, SOD, CAT, and protein in a dose dependent manner. The effects of MECB were comparable to that of standard drug Silymarin. However, at a lower dose (25 mg/kg) it could not restore the deleterious effect produced by paracetamol. The results indicate that Caesalpinia bonducella had antioxidant and hepatoprotective effects.

Extracts of Codonopsis lanceolata Decrease $CCl_4$-Induced Hepatotoxicity on High Fat Diet in Rats

  • This study was designed to investigate the effect of Codonopsis lanceolata extracts on carbon tetrachloride (CCl₄)­induced hepatotoxicity in rats fed high, medium or low fat diet. The liver weight was increased not by the level of fat in diet but by administration of CCl₄. Activities of serum aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by CCl₄, the activities of AST and alanine aminotransferase (ALT) were significantly decreased in case of CCl₄ treatment with C. lanceolata extracts. In high fat diet with CCl₄ administration, the content of total lipid in liver was significantly increased, which was significantly decreased by the administration of C. lanceolata extracts. Administration of C. lanceolata water extracts remarkably prevented the tissue injury such as lipid degeneration, infiltration of inflanunatory cells and necrosis caused by CCl₄, but those methanol extracts did not affect histopathological changes of the liver as water extract. The data suggest that intake of C. lanceolata extracts would prevent lipid accumulation and hepatotoxicity induced by high fat diet and CCl₄ abministratim, and the water extract of C. lanceolata is supposed to be more effective than those methanol extract.
