Studies on the Lipase Activity and Lipid Components of the Molded Sardine Meal 'Koji' (정어리를 기질로 제조한 Koji의 Lipase활성과 지질성분)

  • The lipase activity of crude enzyme extracted from the molded sardine meal(MSM) 'koji', and lipid classes and fatty acid composition of neutral lipid(NL) and phospholipid (PL) of MSM 'koji' were investigated. The optimum pH and temperature of the crude enzyme was 8.0 and $40^{\circ}C$ using olive oil emulsion as a substrate, but the residual activity depended on pH and temperature. Total lipid contents of the MSM 'koji' consisted of 15.33g/100g NL, 5.45g/100g PL respectively. The major lipid classes of NL were triglyceride ($53.4\%$) and free fatty acid($28.1\%$), that of PL were phosphatidylethanol($32.7\%$), phosphatidylinositol($25.5\%$), phosphatidylcholine($25.1\%$), lyso-phosphatidylcholine($7.3\%$) and sphingomyeline($3.5\%$). The prominent fatty acid fractions of NL were 16:0, 22:6, 18:1 n-9, 18:2n-6 and 20:5, with PL fractions the major fatty acid ratios were 22:6, 16:0, 18:1n-9, 18:2n-6 and 20:5, in the same respective order.

Effects of Kimchi Solvent Fractions on Anti-oxidative Enzyme Activities of Heart, Kidney and Lung of Rabbit Fed a High Cholesterol Diet (김치 용매획분이 고 콜레스테롤 식이를 섭취한 토끼의 심장, 신장 및 폐의 항산화 효소계 활성에 미치는 영향)

  • The effects of dichlorornethane (CH$_2$C$_2$), ethylacetate (EtOAc) or water ($H_2O$) fraction of Korean cabbage kimchi on anti-oxidative enzyme activities of the heart, kidney and lung of rabbit fed 1% cholesterol diet for 16 weeks were studied. The amount of kimchi fraction added to the 100 g of diet was 8.3 mg of CH$_2$C1$_2$,5.6mg of EtOAc, and 221.9 mg of $H_2O$, which are equivalent to 5% of freeze-dried kimchi in the diet. Each group had 6 rabbits and rabbit was housed individually. Lipid peroxide values for the heart was the highest followed by lung and kidney. But the activities of catalase and GSH-px were the lowest in the heart and the highest in the kidney. Activities of anti -oxidative enzymes (catalase, GSH-px, Mn-SOD, Cu, Zn-SOD) of rabbits fed kimhi solvent fractions added diets were lower than those of the control which fed 1% cholesterol diet. Among kimchi groups, CH$_2$Cl$_2$ group showed the lowest (p<0.05) enzymes activities. Lipid accumulation in these organs fed diets with kimchi solvent fractions, especially with CH$_2$C1$_2$fraction, were lower than that of the control. Therefore, production of the lipid free radicals might be suppressed and the activities of anti -oxidative enzymes responsible for removing the free radicals seemed to be lowered in kimch frachoin-fed rabbits.

Studies on the Lipid and Fatty Acid Compositions of Ark-Shell, Anadara broughtonii (피조개 지질 및 지방산 조성에 관한 연구)

  • The present study was designed to analyze the lipid and fatty acid compositions of ark shell, Anadara broughtonii. The crude lipid was extracted by Bligh and Dyer's method, and then fractionated by TLC and quantitatively analyzed by TLC scanner. Lipid extracted from ark shell was fractionated into neutral and polar lipid by column chromatography with silicic acid. The fatty acid composition of lipid fractions were determined by gas liquid chromatography. Total lipid content of ark shell was 0.83% base on wet weight. The content of unsaponifiable matter was 20.19%, and iodine value was 156.13. The main components of total lipids were triglyceride, diglyceride, hydrocarbon, and sterol ester. The fatty acid composition of total lipid chiefly consisted of $C_{17 : 0}$, $C_{16 : 0}$, $C_{18 : 1}$ and $CT_{16 : 1}$. The main fatty acids of neutral lipid were $C_{16 : 0}$, $C_{18 : 1}$, $C_{22 : 1}$, $C_{18 : 0}$ and $C_{16 : 1}$. The major fatty acids of polar lipid were $C_{16 : 0}$, $C_{18 : 2}$, $C_{20 : 5}$ and $C_{22 : 6}$. In total lipid fractionation, saturated acid contents were high in all (SA>MA> PA), in neutral lipid fractionation, menoenoic acid contents were high in all (MA > SA> PA), and in polar lipid fractionation, saturated acid con-tents were high in all (SA> PA> MA).

Effects of Wax Gourd Extracts on Adipocyte Differentiation and Uncoupling Protein Genes(Ucps) Expression in 3T3-Ll Preadipocytes

  • Although various raw plant materials have been demonstrated to exert anti-obesity effects to a greater or lesser extent in both humans and animals when they are used to supplement the diet, it has not been shown extensively that they influence adipocyte cell differentiation involving lipid metabolic gene expressions. Using a well-established 3T3-L1 preadipocyte differentiation system, we decided to look into molecular and cellular event occurring during adipocyte differentiation when raw plant materials aye included in the process, in an effort to demonstrate the potential use of a screening system to define the functions of traditionally well-known materials. To these ends, the effects of ethanol (EtOH) or EtOH/distilled water (DW) extracts of Wax Gourd were examined using cytochemical and molecular analyses to determine whether components of the extracts modulate adipocyte differentiation of 3T3-Ll preadipocytes in vitro. The cytochemical results demonstrated that EtOH or EtOH/DW extracts did not affect lipid accumulation and cell proliferation, although the degree of lipid accumulation was influenced slightly depending on the extract. EtOH extract was highly effective in apoptotic induction during differentiation of 3T3-Ll preadipocytes (p<0.05). Reverse transcription-polymerase chain reaction (RT-PCR) analysis of lipoprotein lipase (LPL), Uncoupling protein (Ucp) 2, 3 and 4 also showed that while LPL expression was not influenced, Ucp2, 3 and 4 were up regulated in the EtOH extract-treated group and down regulated in the EtOH/DW extract-treated group. These changes in gene expressions suggest that the components in different fractions of Wax Gourd extracts may modulate lipid metabolism by either direct or indirect action. Taking these results together, it was concluded that molecular and cellular analyses of adipocyte differentiation involving lipid metabolic genes should facilitate understanding of cellular events occurring during adipocyte differentiation. Furthermore, the experimental scheme and analytical methods used in this study should provide a screening system for the functional study of raw plant materials in obesity research.

Inhibitory Effects of Natural Plant Extracts on ICAM-1/LFA-1 Mediated Adhesion of HL-60 Cells (자생식물 추출물의 세포접착인자 저해활성 검색)

  • Atherosclerosis is a progressive disease characterized by the accumulation of lipids and fibrous elements in the arteries. Monocyter/macrophages are involved in many aspects of the development of atherosclerotic plaques. It is known that the intercellular adhesion molecule-1(ICAM-1) expressed preferentially on endothelial cells of atherosclerotic plaque, promotes local adhesion and transendothelial migration of monocytes, neutrophils, and lymphocytes. Using the human promyelocytic leukemia HL-60 cell line, we investigated the inhibitory effects of methanol extracts of 175 natural plants on ICAM-1/LFA-1 mediated cell adhesion. Eight kinds of methanol extracts of tested plants inhibited PMA-induces homotypic aggregationof HL-60 cells without cytotoxicity at the concentration of $6.25\;{\mu}g/ml$. They were divided two fractions of $CHCI_3$ and $H_2O$ to use solvent partition. Among them, $CHCI_3$ extract $(1.0\;{\mu}g/ml)$ of Saururus chinensis and Chloranthus japonicus singificantly inhibited aggregation of HL-60 cells without cytotoxicity, respectively.

Excessive Dietary Conjugated Linoleic Acid Affects Hepatic Lipid Content and Muscular Fatty Acid Composition in Young Chicks

  • The effects of dietary conjugated linoleic acid (CLA) on lipid concentrations and fatty acid composition of various tissues were studied in young chicks. From 7 days of age, a total of 160 chicks were divided into 4 groups, placed into 4 pens per group (10 birds per pen) and fed one of four experimental diets containing 6% tallow (TO 6%), 4% tallow plus 2% CLA (TO 4%-CLA 2%), 2% tallow plus 4% CLA (TO 2%-CLA 4%) or 6% CLA (CLA 6%) for 3 weeks. There were no significant differences in growth performances and the relative weights of various organs, but relative liver weight of chicks fed dietary CLA at 4 and 6% levels was significantly higher (p<0.05) than that of TO 6% group. The chemical compositions of leg muscle were not affected by CLA feeding. However, hepatic total lipid of chicks fed 6% CLA diet was significantly higher (p<0.05) than those of TO 6% and TO 4%-CLA 2% groups. The concentrations of various lipid fractions in serum were not affected by CLA feeding. With the increase in dietary CLA levels, cis 9-trans 11 CLA, trans 10-cis 12 CLA and total CLA of leg muscle increased linearly. The relative proportions of C18:1 $\omega$ -9 and C20:4 $\omega$-6 fatty acids in the leg muscles of chicks fed the CLA containing diets were significantly lower (p<0.05) than those of TO 6% group. These results indicate that the levels of CLA isomers were increased linearly in dose-dependent manner after feeding of synthetic CLA source. But it was also observed that excessive amount of dietary CLA resulted in the possible adversely effects, such as increase of liver weight, hepatic lipid accumulation and serum GOT level.

Effect of Korean Red Ginseng Powder on the Lipid Concentrations and Tissue Lipid Peroxidation in the Rats Fed High Fat Diet (고지방급여 흰쥐의 혈청과 간의 지질 농도 및 조직 과산화지질 농도에 미치는 홍삼분말의 영향)

  • Effect of Korean red ginseng (KRG) on the level of serum and liver lipids and lipid peroxidation was investigated in the rats fed high fat diet. Content of serum total cholesterol was significantly decreased (P<0.05) in KRG I group and KRG II group. Content of HDL-cholesterol was significantly increased by 69.75% and 39.15% in KRG I and KRG II group compared to control group, respectively. Atherogenic index (hi) was also significantly decreased by 74.76% and 37.38% in KRG I and KRG II groups compared to control group, respectively. Serum triglyceride content was significantly decreased (p<0.05) in only KRG II group. Antioxidative activity of KRG on the lipid peroxidation of serum and tissues in rats was also studied in vivo by measuring the formation of thiobarbituric acid reactive substances (TBARS). Contents of TBARS in the serum of both KRG groups were significantly decreased (p<0.05) and that of nonheme iron in serum was significantly increased (p<0.05) in a dose-dependent manner, which suggested that lipid peroxidation contents are inversely correlated with serum nonheme iron content. Content of TBARS in liver was significantly decreased (p<0.05) in KRG I and KRG II groups, without any influence in other tissues. Content of TBIARS in liver microsomal fractions stimulated by Fe$^{2+}$/ascorbate was significantly decreased (p<0.05) in KRG I and KRG II groups, whereas this observation did not occur in liver mitochondrial fractions. When the effect of KRG on TBARS content in the liver fractions of homogenates, microsomes, and mitochodria stimulated by Fe$^{2+}$/ascorbate was tested in vitro experimental model, TBARS of liver three fractions was significantly decreased at 6 mg/mL KRG compared with those of control. These results suggested that KRG powder have hypocholesterolemic effect as well as antioxidative effect in the serum and liver of the rats fed high fat diet.

Effects of Alismatis Rhizoma Extracts on Lipid Components in Experimental Liver Injury (실험적 간장해에 의한 지질 함량 변동에 대한 택사 추출액의 영향)

  • The protective and therapeutic effects of Alismatis Rhizoma extracts on rat liver injury induced by carcinogenic polycyclic hydrocarbon, benzo(a)pyrene were investigated in rats. Serum and liver triglyceride, total-cholesterol level and serum HDL-cholesterol content were measured. Alismatis Rhizoma extracts inhibited the enhanced production of lipid in chemically induced liver injury. Among 4 fractions, CHCh fraction revealed the most powerful liver-protective effect. Our results strongly suggest that Alismatis Rhizoma be a promising liver-protective agent.

Two p-Terphenyls from Mushroom Paxillus panuoides with Free Radical Scavenging Activity

  • As part of a continuing investigation to identify free radical scavengers from the fruit bodies of basidiomycetes, we isolated two p-terphenyl compounds, designated as PAl and PA2l, from cethanolic extract of the fruit body of Pasillus panuoides. The methanolic extract was processed by ethyl acetate extraction and silica gel column chromatography to yield two active fraction. PAl was obtained from one of the fractions through Sephadex LH-20 and silica gel column chromatographies and reverse-phase HPLC. The other fraction was purified by Sephadex LH-20 and reverse-phase column chromatographies to produce PA2. The compounds PA1 and PA2 were identified as leucomentin-4 and leucomentin-2, respectively, on the basis of various spectroscopic analyses. These compounds exhibited strong inhibitory activities against lipid peroxidation in rat liver microsomes with IC_{50}$ values of 0.10 and $0.06{\;}\mu\textrm{g}ml$, respectively.
