Ice plant (Mesembryanthemum crystallinum) 추출물의 생리 활성 (Biological Activities of Mesembryanthemum crystallinum (Ice plant) Extract)

  • 본 연구는 ice plant (Mesembryanthemum crystallinum)의 생리활성을 분석하여 기능성 식품으로서의 가능성을 조사하였다. Ice plant는 아프리카와 서유럽, 북미, 남미 그리고 호주에서 서식하는 다육식물로 항당뇨, 항산화, 지질 대사 활성에 효능이 있다고 알려져 있다. Ice plant를 methanol (MCM), ethanol (MCE), 열수(MCHW), 열수 추출 후 methanol (MCHM)로 추출하여, mushroom tyrosinase, pancreatic lipase, DPPH free radical 소거 활성, NO 생성, α-glucosidase에 대한 저해 활성을 측정 하였다. MCM, MCE, MCHW, MCHM 추출물의 추출 수율은 0.37%, 0.33%, 0.5%, 0.07%로 나타났다. MCHW 추출물은 DPPH free radical 소거능에 높은 효과를 보였고, MCM, MCE 추출물은 lipase 저해 활성, α-glucosidase 저해 활성, NO 생성 저해활성을 보였다. 그러나 mushroom tyrosinase 저해 활성은 모든 추출물에서 매우 낮았다. 이러한 결과를 통해 ice plant는 항산화 물질로서 비만, 당뇨, 염증 치료용 기능성 식품으로서의 가능성이 매우 높음을 확인할 수 있었다.

Effects of Alternatively Prepared Meju Methanolic Extracts on Dietary Lipid Digestion

  • We examined the effect of extracts of meju prepared with traditional and standardized methods on pancreatic lipase and the absorption of dietary lipid. Aqueous methanolic (80%, v/v) extracts of meju dose-dependently inhibited the activities of porcine pancreatic lipase. The plasma triglyceride levels in Imprinting Control Region mice after a single oral administration of lipid emulsion containing aqueous methanolic extracts from meju made by the standardized methods were lower than that of the group given a lipid emulsion containing the extracts of meju made by traditional methods. The inhibitory activity of the meju extract on dietary lipid digestion appears to be more closely associated with aglycone forms of phenolic compounds such as free isoflavones than with glycosides, since meju samples with higher total phenolic or free isoflavone content showed the stronger inhibition against pancreatic lipase. Furthermore, the data suggest that meju made using the standardized method, which contains higher levels of total isoflavones relative to traditionally prepared meju, could effectively suppress digestion of dietary lipids and therefore have the potential to help ameliorate hyperlipidemia and obesity.

마디풀 추출물의 Pancreatic Lipase 저해 및 항산화 활성 평가 (Pancreatic Lipase Inhibitory and Antioxidant Activities of Polygonum aviculare Extract)

  • 마디풀을 80% ethanol로 침지 추출하여 얻어진 추출물에 대해 $CHCl_3$, EtOAc 및 n-BuOH로 순차 용매 분획 하였고, 얻어진 결과물에 대하여 DPPH 및 $ABTS^+$ radical 소거능 및 pancreatic lipase 저해활성을 평가하였다. DPPH 라디칼 소거능은 페놀성 화합물의 함량이 상대적으로 높은 EtOAc층의 $100{\mu}g/mL$의 시험농도에서는 $58.1{\pm}2.3%$의 라디칼 소거능을 확인하였고, 마디풀 추출물에 존재하는 페놀성 화합물이 라디칼 소거능과의 연관성을 시사하였다. 또한 $ABTS^+$ 라디칼 소거능은 EtOAc층이 $IC_{50}=2.8{\pm}0.5{\mu}g/mL$. $CHCl_3$층이 $IC_{50}=9.2{\pm}0.5{\mu}g/mL$의 강한 활성이 확인되었고, 강력한 활성물질의 존재가 시사되었다. 또한, pancreatic lipase 저해활성을 측정한 결과, 강한 $ABTS^+$ 라디칼 소거능을 나타낸 EtOAc층과 $CHCl_3$층의 $IC_{50}=1.9{\pm}0.2$. $5.7{\pm}1.3{\mu}g/mL$의 저해율을 나타내었으며 이는 대조군인 orlistat에 비해 다소 약하나 단일물질로 정제할 경우 더욱 강한 효능의 화합물이 존재할 가능성을 시사하였다. 향후 이들 활성물질 동정을 통한 활성 기작에 대한 연구가 필요하며 본 연구결과는 보다 우수한 라디칼 소거능 및 pancreatic lipase 저해능을 가지는 새로운 선도화합물 발굴을 위한 기초자료로 이용될 수 있을 뿐만 아니라 국내에 자생하는 마디풀의 식물 화학적 성분연구에 대한 기초자료로 활용가능하리라 사료된다.

Carbohydrate, Lipid Inhibitory Activity and Antioxidant Activity of Extracts from Several Economic Resource Plants in Vitro

  • The objective of this study was determined to evaluate ${\alpha}$-amylase, ${\alpha}$-glucosidase, pancreatic lipase inhibition in vitro and DPPH radical scavenging activity of the several Korean resources plants. The ${\alpha}$-amylase inhibitory activity of Salicornia herbacea, Erythronium japonicum (flower) and Phragmites communis (root) in water extract showed relatively high 62.8%, 66.5% and 69.3%, respectively. The ${\alpha}$-amylase inhibitory activity of Citrus junos (pericarp) and Cornus officinalis in methanol extract was found to have an effect with 32.8% in Citrus junos (pericarp) and 60.9% in Cornus officinalis. Corylopsis coreana in both water and methanol extract had the highest ${\alpha}$-glucosidase inhibitory activity of 81.7% and 89.5%, while the extract of Portulaca oleracea, Ficus carica and Citrus junos was not measured ${\alpha}$-glucosidase inhibitory activity at given experiment concentration. Depending on the extraction solvent and the plant species, it was observed that there was a significant difference in ${\alpha}$-glucosidase inhibitory activity. The pancreatic lipase inhibitory activity showed relatively higher in the methanol extract than water extract except pericarp of Citrus junos. The DPPH radical scavenging activity of selected plants was much difference between measured plant species, and showed that the increase was proportional to the concentration. These results suggested that selected plants had the potent biological activity on carbohydrate, lipid Inhibitory activity and antioxidant activity, therefore these plant resources could be a good materials to develop medicinal preparations, nutraceuticals or health functional foods for diabetes or obesity.

소금 첨가에 따른 도라지 발효 특성과 미생물 변화 및 항비만 효능 평가 (Effect of Platycodon grandiflorum Fermentation with Salt on Fermentation Characteristics, Microbial Change and Anti-obesity Activity)

  • Objectives: This study investigated the effect on microbial ecology, fermentation characteristics and anti-obesity of Platycodon grandiflorum (PG) fermentation with salt. Methods: PG was fermented for four weeks with 2.5% salt and the characteristics of fermented PG were performed by measuring pH, total sugar content, viable bacteria number and microbial profiling. Also, we measured total polyphenol, flavonoid and the percent of inhibition of lipase activity and lipid accumulation. Results: Salt added to PG for fermentation had an effect on pH, total sugar, total and the number of lactic acid bacteria. Total sugar and pH were reduced and number of total and lactic acid bacteria were increased after fermentation. The majority of bacteria for fermentation were Lactobacillus plantarum, Leuconostoc psedomesenteroides and Lactococcus lactis subspecies lactis regardless of salt addition. However, microbial compositions were altered by added salt and additional bacteria including Weissella koreensis, W. viridescens, Lactobacillus sakei and Lactobacillus cuvatus were found in fermented PG with salt. Total flavonoid was increased in fermented PG and lipid accumulation on HepG2 cells treated with fermented PG was reduced regardless of salt addition. Moreover, fermented PG without salt suppressed lipase activity. Conclusions: Addition of salt for PG fermentation had influence on fermentation characteristics including pH and sugar content as well as number of bacteria and microbial composition. In addition, fermented PG showed anti-obesity effect by increasing flavonoid content and inhibition of lipase activity and lipid accumulation.

적양파 추출물의 항비만 활성 (Anti-Obesity Effects of Red Onions Extract)

  • 비만은 에너지의 섭취와 소비의 불균형으로 지방조직이 비정상적으로 분화하면서 생기는 대사질환으로 알려져있다. 본 연구에서는 본 연구는 적양파 추출물 처리에 따른 Pancreatic lipase 억제, 지방세포분화 억제 활성을 확인하고자 하였다. 적양파추출물 처리에 따른 활성은 지방세포 분화 및 관련 유전자에 대한 평가는 3T3-L1 지방전구세포를 이용하고 Real-Time PCR을 통하여 확인하였다. 실험 결과, Pancreatic lipase 활성 억제 실험에서 적양파 추출물은 농도 의존적으로 lipase 활성을 억제하였다. 지방세포분화 실험을 수행한 결과, insulin, dexamethasone, 3-isobutyl-1-methylxanthine(MDI)등으로 분화 유도된 3T3-L1 세포에서 적양파 추출물은 지방전구세포의 분화를 억제하였으며 동시에 지방구 형성을 억제하는 것으로 나타났다. 또한, 지방전구세포의 분화 과정과 관련된 C/EBP-α, C/EBP-β, PPAR-γ의 발현을 억제하였다. 본 실험에서 적양파 추출물은 지방분해효소를 억제하며, 지방전구세포 분화와 관련된 유전자 발현을 억제함으로써 지방세포 분화 및 지방구형성을 억제할 수 있는 항비만 소재로의 개발 가능성이 높다고 판단된다.

Optimal Extraction Conditions of Anti-obesity Lipase Inhibitor from Phellinus linteus and Nutritional Characteristics of the Extracts

  • In an effort to develop novel mushroom-derived anti-obesity nutraceuticals, water and ethanol extracts containing the lipaseinhibitory compound from Phellinus linteus were prepared, and their nutritional components were determined. The optimal conditions for the extraction of P. linteus lipase inhibitor involved the treatment of the fruiting bodies with distilled water at $80^{\circ}C$ for 72 hr and 80% ethanol at $100^{\circ}C$ for 60 hr, respectively. The distilled water extract and ethanol extract contained 10.9% and 6.11% of crude protein, and 0.96% and 15.86% of crude fat, respectively. Additionally, the distilled water extract contained a large quantity of minerals, including 239.5 mg of K, 39.3 mg of Mg, and 39.3 mg of Na. The free amino acid content of the distilled water extracts was also higher than that of the ethanol extracts, and in particular, the distilled water extracts contained 5,139 mg of asparagine, 3,891 mg of tryptophan, 2,598 mg of alanine, and 2,066 mg of serine in 100 g of the distilled water extracts. 100 g of the distilled water and ethanol extracts were found to contain 12.31 g and 8.16 g of malic acid, respectively.

Lipase Inactive Mutant of PLC-γ1 Regulates NGF-induced Neurite Outgrowth Via Enzymatic Activity and Regulation of Cell Cycle Regulatory Proteins

  • Src homology (SH) domains of phospholipase C-$\gamma1$ (PLC-$\gamma1$) impair NGF-mediated PC12 cells differentiation. However, whether the enzymatic activity is also implicated in this process remains elusive. Here, we report that the enzymatic activity of phospholipase C-$\gamma1$ (PLC-$\gamma1$) is at least partially involved to the blockage of neuronal differentiation via an abrogation of MAPK activation, as well as sustained Akt activation. By contrast, Overexpression of WT-PLC-$\gamma1$ exhibited sustained NGF-induced MAPK activation, and triggered transient Akt activation resulting in profound inhibition of neurite outgrowth. However, lipase-inactive mutant (LIM) PLC-$\gamma1$ cells fail to suppress neurite outgrowth, although it contains intact SH domains, specifically enhancing the expression of cyclin D1 and p21 proteins, which regulate the function of retinoblastoma Rb protein. These observations show that the lipase inactive mutant of PLC-$\gamma1$ does not alter NGF-induced neuronal differentiation via enzymatic inability and the modulation of cell cycle regulatory proteins independent on SH3 domain.

In vitro Anti-obesity Effect of 4-hydroxybenzyl Alcohol from Cudrania tricuspidata

  • The present study was investigated on in vitro anti-obesity effect of 4-hydroxybenzyl alcohol from Cudrania tricuspidata. We isolated various compounds from Cudrania tricuspidata. Among these compounds, anti-obesity effects of 4-hydroxybenzyl alcohol was examined by lipase activity assay, cyclic adenosine monophosphate (cAMP)-specific phosphodiesterase type IV (PDE4) activity assay, and citrate synthase activity assay. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts inhibited the enzymatic activities of lipase, PDE4, and citrate synthase. Lipase is known to mediate the hydrolysis of triacylglycerol in adipose tissue and cholesterol esters in other tissue or cells. Also, PDE4 hydrolyses cAMP, a crucial secondary messenger for in metabolic pathways including glucose and lipid metabolism, lipolysis, and thermogenic function. 4-hydroxybenzyl alcohol and Cudrania tricuspidata extracts induced the inhibitory effect against each enzymatic activity on several specific substrates as observed by detection at 405 or 412 nm. These findings might be attributable to the inhibition of adipogenesis, and partial prevention of obesity. In conclusion, these results show that 4-hydroxybenzyl alcohol and Cudrania tricuspidata may be a critical candidate as a natural anti-obesity source.

Pancreatic lipase inhibitory activity of taraxacum officinale in vitro and in vivo

  • Obesity has become a worldwide health problem. Orlistat, an inhibitor of pancreatic lipase, is currently approved as an anti-obesity drug. However, gastrointestinal side effects caused by Orlistat may limit its use. In this study the inhibitory activities of dandelion (Taraxacum officinale) against pancreatic lipase in vitro and in vivo were measured to determine its possible use as a natural anti-obesity agent. The inhibitory activities of the 95% ethanol extract of T. officinale and Orlistat were measured using 4-methylumbelliferyl oleate (4-MU oleate) as a substrate at concentrations of 250, 125, 100, 25, 12.5 and $4\;{\mu}g/ml$. To determine pancreatic lipase inhibitory activity in vivo, mice (n=16) were orally administered with com oil emulsion (5 ml/kg) alone or with the 95% ethanol extract of T. officinale (400 mg/kg) following an overnight fast. Plasma triglyceride levels were measured at 0, 90, 180, and 240 min after treatment and incremental areas under the response curves (AUC) were calculated. The 95% ethanol extract of T. officinale and Orlistat, inhibited, porcine pancreatic lipase activity by 86.3% and 95.7% at a concentration of $250\;{\mu}g/ml$, respectively. T. officinale extract showed dose-dependent inhibition with the $IC_{50}$ of $78.2\;{\mu}g/ml$. A single oral dose of the extract significantly inhibited increases in plasma triglyceride levels at 90 and 180 min and reduced AVC of plasma triglyceride response curve (p<0.05). The results indicate that T. officinale exhibits inhibitory activities against pancreatic lipase in vitro and in vivo. Further studies to elucidate anti-obesity effects of chronic consumption of T. officinale and to identify the active components responsible for inhibitory activity against pancreatic lipase are necessary.
