Zeaxanthin is an important pigment in the photo-protection mechanism of microalgae. However, zeaxanthin epoxidase, an enzyme involved in the accumulation and conversion of zeaxanthin, has not been extensively studied in microalgae. In this work, we report the expression pattern of zeaxanthin epoxidase in Dunaliella tertiolecta (DtZEP) at different light and diverse salinity conditions. To confirm the responsiveness to light conditions, the ZEP expression pattern was investigated in photoperiodic (16 h of light and 8 h of dark) and continuous (24 h of light and 0 h of dark) light conditions. mRNA expression levels in photoperiodic conditions fluctuated along with the light/dark cycle, whereas those in continuous light remained unchanged. In varying salinity conditions, the highest mRNA and protein levels were detected in cells cultured in 1.5 M NaCl, and ZEP expression levels in cells shifted from 0.6 M NaCl to 1.5 M NaCl increased gradually. These results show that mRNA expression of DtZEP responds rapidly to the light/dark cycle or increased salinity, whereas changes in protein synthesis do not occur within a short period. Taken together, we show that DtZEP gene expression responds rapidly to light irradiation and hyperosmotic stress. In addition, ZEP expression patterns in light or salinity conditions are similar to those of higher plants, even though the habitat of D. tertiolecta is different.
This study was conducted with using chlorophyll a fluorescence (indicated as photosynthetic activity) to examine the toxic effect of 96 h exposure of heavy metals and herbicides on the benthic diatom Nitzschia sp. which was isolated from pristine sediment in Pamquat Harbour, Nova Scotia, Canada. Samples of benthic diatom were exposed to 0, 0.01, 0.1 and 1 mg L–1 of copper, 0, 1, 10 and 100 mg L–1 of chrome (VI), 0, 2.45, 24.5 and 245 mg L–1 of paraquat dichloride, and 0, 4.37, 43.7 and 437 mg L–1 of alachlor during 96 hours. The effective quantum yield of photochemistry (ΔF/Fm’) was evaluated by subjecting light acclimated samples to saturating pulses of light using a pulse amplitude modulated (PAM) fluorometer. The impact of heavy metals on Nitzschia sp. photosynthesis was not severe in < 1 mg L–1 but in the high concentrations (> 1 mg L–1) clearly increased toxic stress during 96 h. Herbicides had a limited impact during the exposure period but clearly increased stress on the benthic diatom with increasing concentrations. Acute response of Nitzschia sp. to selected heavy metals and herbicides was characterized, and the capacity of a benthic diatom to tolerate and recover from toxic stress was assessed.

Light guide plate is one of most important components which are composed of back light unit, affecting the quality and performance of LCD. Average brightness and uniformity are especially key factors for designing the light guide unit. These qualities are affected and controlled by the pattern being attached to the back of light guide unit. In order to obtain high brightness and uniformity the optimized pattern design is adopted for LGP. In this study, optimized molding condition for LGP with 0.4 mm thickness was obtained by using the Moldflow simulation software and the optimized pattern for better brightness uniformity was designed for the thickness of the 0.4 mm by trial and error method. The brightness was measured for the different LGP thicknesses and the residual stress analysis was performed for 0.4 mmthickness by the photoelasticity and the results are compared with 0.5 mm, 0.6 mm thickness.

In trench excavation, essential factor of earth-retaining temporary work structure should be easy taking to pieces and movement, and dead weight must be less. This paper studies about the light weight material and application as earth-retaining structure to prevent the slope failure of sand soil ground caused by the variation of groundwater level in trench excavation. That is, light weight earth-retaining structural is proposed and a simulation with FEM on application of proposed structural in sandy soil is presented. The results are summarized as follows; (1) The study proposed FRP H-shaped pannel for the light weight member, and also presented estimation method about stability. (2) Mechanical property (bending moment, shear force, axial force, displacement) were changed according to groundwater level, but these values had been within enough safety rate and allowable stress. Therefore, proposed light weight pannel with FRP is available for bracing structure in trench excavation.

Recently advantages joining dissimiliar materials and light weight material techniques have led to increasing use of structural adhesives in the various industries. Stress singulartiy occurs at the interface edges of adhesively bonded dissimilar materials. So it is required to analyze its stress singularity at the interface edges of adhesively bonded joints indissimilar materials. In this paper, the analysis method of stress singularity is studied in detail. Also, effects of the stress singularity at the interface edge of adhesively bonded scarf joints in combinations of dissimilar materials are investigated by using 2-dimensional elastic program of boundary element method. As the results, the strength evaluation method of adhesively bonded dissimilar materials using the stress singularity factor, $\Gamma$,is very useful. The fracture criterion, method of strength evaluation and prediction of fracture strength by the stress singularity factor on the adhesively bonded dissimilar materials are proposed.

Structural light weight aggregate concrete are made with both coarse and fine light weight aggregates, but it is common with the high strength concrete to replace all or part with normal weight sand be called class 1 structural light weight aggregate concrete. Fire resistance of structural light weight aggregate concrete are determined by properties of high water content ratio and explosive spalling. Especially, structural light weight aggregate concrete is occurred serious fire performance deterioration by explosive spalling stem from thermal stress and water vapor pressure. This study is concerned with experimentally investigating fire resistance of class 1 structural light weight concrete. From the test result, class 1 structural light weight concrete is happened explosive spalling. The decrease of cross section caused by explosive spalling made sharp increasing gradient of inner temperature.

This study investigated the effects of light intensity on growth performance, blood components, eye condition, and carcass characteristics of broiler chickens. Three hundred and fifty-two 1-day-old male chicks were assigned to one of four treatments (four repetitions per treatment, 22 birds per repetition) and reared in a floor pen for 5 weeks. From the second week, chicks were reared under four different levels of light intensity (5, 20, 35, and 50 lx) and the lighting duration was maintained at 18-hours light : 6-hours dark (18L : 6D). The feed intake and body weight were measured weekly. At 35 days of age, 12 birds per treatment were randomly selected for blood sampling, eye measurement, and carcass analysis. There were no significant differences in body weight gain, feed intake, and feed conversion ratio among treatments. Triglyceride levels in the serum were significantly higher in the 5 lx treatment, and creatinine was significantly lower in the 5 lx treatment (p < 0.05). The heterophil : lymphocyte ratios decreased significantly as light intensity increased (p < 0.05); however, other blood cell compositions were not affected by light intensity. Interleukin-6 content was significantly higher in the 5 lx treatment than in other treatments (p < 0.05), but the content of tumor necrosis factor-α was not significantly different among treatments. Serum corticosterone concentration was significantly higher at 5 lx than at 20, 35, and 50 lx (p < 0.05). The corneal diameter was the highest in 5 lx treatment (p < 0.05), and tended to increase as the light intensity decreased. Other eye parameters were not significantly different among treatments, but displayed a tendency to increase as the light intensity decreased. Carcass yield and part yields were not affected by light intensity. Meat quality parameters (pH, color, cooking loss, and water-holding capacity) did not show significant difference among the treatments. The results indicate that a light intensity of 5 lx may increase physiological stress or have a negative effect on broiler welfare, even if the performance and carcass characteristics are not affected. Therefore, a light intensity of 20 lx or above is recommended considering both the growth performance and welfare of broilers.
An Arabidopsis hy4 mutant that is specifically impaired in its ability to undergo blue light dependent photomorphogenesis was used to identify cryptochrome 1 signaling-related components. Proteomic analysis revealed about 205 differentially expressed protein spots in the blue light-irradiated hy4 mutant compared to the wild-type. The proteins corresponding to 28 up-regulated and 33 down-regulated spots were identified. Obvious morphological changes in the hy4 mutant were closely related to the expression of various transcription factors. Our findings suggest that blue light signals may be involved in many cellular processes including disease resistance and stress responses.

Stress fiber (SF) alteration is mediated by cellular receptors, which, upon interaction with the extracellular counterpart, signal to the actin cytoskeleton for remodeling. This association is mediated by a variety of scaffold and signaling factors, which control the mechanical and signaling activities of the interaction site. The heterotrimeric transmembrane lymphotoxin α1β2 (LTα1β2), a member of the tumor necrosis factor (TNF) family of cytokines, including soluble homotrimeric lymphotoxin (LT α), plays an important role in lymphoid tissue architecture. Ligation between LTα1β2 and the lymphotoxin β receptor (LTβR) activates signal-cascade in fibroblastic reticular cells (FRCs). We found LTβR stimulation using an agonistic anti-LTβR antibody alone or combined with LTα or TNFα induced changes in the actin and plasticity of cells. To clarify the involvement of myosin underlying the alteration, we analyzed the effect of myosin light chain kinase (MLCK) with an MLCK inhibitor (ML7), the phosphorylation level of myosin light chains (MLC), and the level of phospho-myosin phosphatase target subunit 1 (MYPT1) after treatment with an agonistic anti-LTβR antibody for cytoskeleton reorganization in FRCs. The inhibition of MLCK activity induced changes in the actin cytoskeleton organization and cell morphology in FRC. In addition, we showed the phosphorylation of MLC and MYPT1 was reduced by LTβR stimulation in cells. A DNA chip revealed the LTβR stimulation of FRC down-regulated transcripts of myosin and actin components. Collectively, these results suggest LTβR stimulation is linked to myosin regarding SF alteration in FRC.
