Triptolide is a compound found in Tripterygium wilfordii and reported to have an anti-inflammatory and anti-oxidant activities. A previous study shows that the dietary supplementation with triptolide increases resistance to environmental stressors, including oxidative stress, heat shock, and ultraviolet irradiation, and extends lifespan in C. elegans. Here, we investigated the underlying mechanisms involved in the lifespan-extending effect of triptolide. The effect of triptolide on age-related diseases, such as diabetes mellitus and Alzheimer's disease, was also examined using animal disease models. The longevity phenotype conferred by triptolide was not observed in the eat-2 mutant, a well-known genetic model of dietary restriction, while there was an additional lifespan extension with triptolide in age-1 and clk-1 mutants. The long lifespan of age-1 mutant is resulted from a reduced insulin/IGF-1-like signaling and the clk-1 mutant lives longer than wild-type due to dysfunction of mitochondrial electron transport chain reaction. The effect of dietary restriction using bacterial dilution on lifespan also overlapped with that of triptolide. The toxicity of high glucose diet or transgenic human amyloid beta gene was significantly suppressed by the supplementation with triptolide. These findings suggest that triptolide can mimic the effect of dietary restriction on lifespan and onset of age-related diseases. We conclude that triptolide can be a strong candidate for the development of dietary restriction mimetics.

This study was conducted to determine the effects of silver thiosulfate (STS) and 1-methylcyclopropene (1-MCP) on flower opening and lifespan of potted Kalanchoe blossfeldiana 'Oriba' for exportation. Ethylene inhibitors, STS and 1-MCP were applied to the kalanchoe plants prior to their export to Japan. STS 0.5 mM with 1% Tween 20 surfactant was directly sprayed (20 mL per plant) to leaves, buds, and flowers and 1-MCP 100 $nL{\cdot}L^{-1}$ was injected into sealed glass chambers containing kalanchoe plants, which were placed on the chambers for 6 hours. After transport to Japan, the plants were immediately transferred to a simulated retail condition room (80 ${\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ for 12 hours of photoperiod at $22^{\circ}C$ and 64% RH) at Toyko University. The numbers of buds, open florets, and wilted florets in the middle inflorescence for each plant were counted right after export, 1 week after export, and 6 weeks after export. The percentages of open florets and wilted florets were calculated from the numbers. STS treatment resulted in 35% more open florets than the control and only 11% of wilted florets at 6 weeks after export to Japan which indicate the extension of lifespan of potted kalanchoe plants. Meanwhile, the plants exposed to 1-MCP before export did not show any significant differences in the numbers of buds and open florets and the percentages of open and wilted florets compared to control plants. In conclusion, STS 0.5 mM treatment strikingly induced better opening florets and lifespan of kalanchoe plants from 1 week to 6 weeks after export than control.
About 50,000 hives of Bombus terrestris are used for pollination of tomato and other economical crops (fruit vegetables and fruit trees) in Korea. Therefore this study was conducted to find out the effective use of B. terrestris and its economic effect in the major beefsteak-tomato cultivation areas from February to August in 2008. The pollinating activities, rate of bite-mark and lifespan of B. terrestris's hive according to all beefsteak-tomato varieties (Super Momotaro, Zeus42, Mascara and Super Sunroad, 500 $m^2$, $9,600{\sim}12,000$plants) were apparent by regular supplement of pollen ($5{\sim}6g$/3 days) to the hive of B. terrestris. Especially, Super Momotaro and Super Sunroad variety were most distinctive in the effects by regular pollen supplement.

The free radical theory of aging suggests that oxidative damage caused by free radicals plays a key role in normal aging. We measured the anti-oxidant activity of acorns and asked whether it can modulate the aging process in Caenorhabditis elegans. Different concentrations of acorn powder were added to culture medium, followed by the monitoring of fertility and survival under oxidative stress. The anti-oxidant activity of 500 mg/L of acorn powder exhibited significant increases in the resistance to oxidative stress in vivo. Acorn powder also significantly extended both the mean and maximum lifespan of C. elegans (the mean lifespan was increased up to 22.4%). The fertility assay indicates the lifespan extension from acorn does not accompany a reduced reproduction, which is common in long-lived mutants. These findings indicate that acorn has a strong antioxidant activity and can induce longevity without the trade-off of reduced reproduction in C. elegans.

The short-lived nematode Caenorhabditis elegans has been used as a model organism for many studies, including lifespan extension. To screen common seaweeds for natural anti-aging agents, the lifespan of C. elegans (N2 wild-type strain) was measured by its hatch rate, growth rate, survival rate, chemotaxis, brood size, and egg-laying time after exposure to nematode growth medium (NGM) containing seaweed extracts. Approximately 30 animals synchronized at the first larval stage were incubated until they reached their adult stages before laying their eggs and were transferred to fresh NGM every 3 days. We also identified the major active compound from the seaweed by gas chromatography-mass spectrometry and tested its optimal dose for longevity. Of 13 common seaweed species, an ethanol extract of the brown seaweed Hizikia fusiformis showed the greatest effect on hatching, growth, and survival rates. The lifespan of C. elegans was significantly expanded 1.54-fold and 1.23-fold in the presence of the ethanol extract (0.05 mg/ml) and the main active component, fucosterol (0.05 mg/ml), respectively. Exposure to the ethanol extract also increased chemotaxis 1.13-fold, decreased brood size 0.74-fold, and shortened egg-laying time 0.96-fold. These results suggest that the aquaculturable H. fusiformis may be a promising source of a diet supplement to support health care.

BACKGROUND/OBJECTIVES: Acanthopanax sessiliflorus is a native Korean plant and used as a traditional medicine or an ingredient in many Korean foods. The free radical theory of aging suggests that cellular oxidative stress caused by free radicals is the main cause of aging. Free radicals can be removed by cellular anti-oxidants. MATERIALS/METHODS: Here, we examined the anti-oxidant activity of Acanthopanax sessiliflorus extract both in vitro and in vivo. Survival of nematode C. elegans under stress conditions was also compared between control and Acanthopanax sessiliflorus extract-treated groups. Then, anti-aging effect of Acanthopanax sessiliflorus extract was monitored in C. elegans. RESULTS: Stem extract significantly reduced oxidative DNA damage in lymphocyte, which was not observed by leaves or root extract. Survival of C. elegans under oxidative-stress conditions was significantly enhanced by Acanthopanax sessiliflorus stem extract. In addition, Acanthopanax sessiliflorus stem increased resistance to other environmental stresses, including heat shock and ultraviolet irradiation. Treatment with Acanthopanax sessiliflorus stem extract significantly extended both mean and maximum lifespan in C. elegans. However, fertility was not affected by Acanthopanax sessiliflorus stem. CONCLUSION: Different parts of Acanthopanax sessiliflorus have different bioactivities and stem extract have strong anti-oxidant activity in both rat lymphocytes and C. elegans, and conferred a longevity phenotype without reduced reproduction in C. elegans, which provides conclusive evidence to support the free radical theory of aging.

A high-pressure, high-pressure drop control valve, which transforms the power transfer of a system by reducing the inlet pressure of 345bartothe outlet pressure of 112bar, is a fundamental component in an offshore plant process. With the increasingly growing market share of the maritime industry, this valve has been expected to be a high-value-added product. This study not only analyzes the relation between pressure drop and fluid velocity in a trim by using fluid analysis, but also examines the possibility of cavitation in a valve in addition to the plot for the extension of lifespan. Based on the analysis results, the design and production method of the valve are established, and accordingly, performance evaluation is carried out. It is demonstrated that the pressure drop from 345bar to 112bar is more feasible in the presence of the trim, which can induce a continuous and diminutive pressure drop in order to prevent cavitation in a high-pressure drop control valve. Furthermore, despite the fluid velocity near a seatring being found to be over 30m/s, the lifespan of the valve is determined to be adequate considering the operation condition of a prototype valve of 80%.

Second, since it has the characteristic of sustainability, the application of manufacturing process and materials, extension of product life, conversion of original material's function, etc. can be sustainable. Third, as it's a trait of scarcity, all products may be produced by hand, and it can have the specialty which the original materials have. Fourth, since it has an eco-friendly trait, even while saving the original materials, the aesthetic needs could be met according to the consumers' continuous demand.
Muscle atrophy is characterized by a decrease in the mass of the muscle. With an increase in life expectancy and chronic illnesses, the incidence of muscle atrophy is increasing and the quality of life of patients is decreasing. Thus, reducing muscle atrophy is of high clinical and socio-economic importance. Mistletoe is a semi-parasitic plant that has been used as a traditional medicine in many countries to treat various human illnesses. It has been reported that Korean mistletoe extract (KME) has diverse biological functions including anti-tumor, anti-oxidant, anti-diabetic, anti-obesity properties, and extension of lifespan. Especially, we have recently reported that KME improves exercise endurance in mice, indicating its beneficial roles in enhancing the capacity of skeletal muscle. In this study, we investigated whether KME could activate the signaling pathway related to protein synthesis in a mouse model of muscle atrophy. Interestingly, KME efficiently activated the Akt/mTOR pathway, and Akt and mTOR are important signaling hub molecules for the acceleration of protein synthesis in muscle cells. In addition, KME also increased the activity of S6 kinase which is involved in the regulation of muscle cell size. Moreover, the ERK activity, required for transcription of ribosomal RNA for protein synthesis, was also enhanced in KME-treated mouse muscle. These data support the idea that KME increases muscle mass via increased protein synthesis. Our findings also suggest that Korean mistletoe might be a promising candidate for the development of functional foods that are beneficial for preventing muscle atrophy.
