A simple and rapid method for detection of single nucleotide variants using tailed primer and HRM analysis

  • Background: Single nucleotide polymorphisms (SNPs) are widely used genetic markers with applications in human disease diagnostics, animal breeding, and evolutionary studies, but existing genotyping methods can be labor-intensive and costly. The aim of this study is to develop a simple and rapid method for identification of a single nucleotide change. Methods: A modified Polymerase Chain Reaction Amplification of Multiple Specific Alleles (PAMSA) and high resolution melt (HRM) analysis was performed to discriminate a bovine polymorphism in the NCAPG gene (rs109570900, 1326T > G). Results: The inclusion of tails in the primers enabled allele discrimination based on PCR product lengths, detected through agarose gel electrophoresis, successfully determining various genotypes, albeit with some time and labor intensity due to the use of relatively costly high-resolution agarose gels. Additionally, high-resolution melt (HRM) analysis with tailed primers effectively distinguished the GG genotype from the TT genotype in bovine muscle cell lines, offering a reliable way to distinguish SNP polymorphisms without the need for time-consuming AS-PCR. Conclusions: Our experiments demonstrated the importance of incorporating unique mismatched bases in the allele-specific primers to prevent cross-amplification by fragmented primers. This efficient and cost-effective method, as presented here, enables genotyping laboratories to analyze SNPs using standard real-time PCR.

For solving this problem, we choose the cellular phone. Nowadays peoples have one cellular phone to one person in korea. Using cellular phone, you can use cellular phone anywhere any time. In this paper, we studied mobile synthesis system using mobile cellular phone. According to use mobile campus system, the university and students can use easy an one to one service in ubiquitous.

A Preliminary Study on the Effectiveness of Cyber Counseling (사이버 상담 활성화를 위한 탐색 연구)

  • The main purpose of this research was to explore the factors that encourage people to use the cyber counseling in resolving difficulties in their lives. Another purpose of this research was to investigate whether cyber counseling really helps people feel better. For this study, a structured survey questionnaire was distributed both on- and off-line, and 429 respondents returned completed survey forms. The major statistical methods used for data analysis were frequency, factor analysis, 1-test and ANOVA. The results of this study revealed that variables such as gender, age, occupation, internet skills, time of internet use, personality, and the awareness of cyber counseling made a difference in people's use of cyber counseling whenever they felt they needed it. However, cyber counseling seemed to fail to increase feelings of well being in people's lives. The implications of these findings are discussed in this article.

  • All the facility, the structure and the rolling stock have limited life so after starting use, we must decide disuse or use when it exeeds the lifetime. Currently the subway which exeeds the lifetime appeared in our country. Structural members of rolling stock structures were occurred corrosion after passage of time and It gives a many effect in the structure strength. In this study, it accomplished a structural analysis which use FEM for the No.2 subway line which operated currently, and it investigated the structural strength change which it follows with dimension change of structural members due to corrosion

  • Recently, OOM(Object-Oriented Method) access method which begins to apply to software development is emphasizing development cost and time reduction through independent software reuse. But because of planning to the exclusion of the lack of knowledge and expansion or performance for development technology many problems are coming out. Therefore, this thesis suggests Effective Object-Oriented Modeling methods considering the plan for higher productivity enhancement of software in the software development life cycle.   • The purpose of this study is to investigate daily lives of the intellectually disabled living in residential facilities. The daily time use method that monitors and records 24 hours of the subject was used for the analysis of daily lives of the institutionalized disabled and verbal interaction patterns within each facility. In particular, the analysis focused on 'frequency of activities' and 'lives', with the method of time budget. Based on the activity category table of the 2004 nationwide Time Use Survey, the analysis aimed to identify differences and similarities between the non-disabled and the disabled in terms of daily time use. The interaction pattern analysis method on grounds of the Flanders Category system(1963) indicates the level, the type, and the full details of interaction between the institutionalized disabled and the living rehabilitation teachers. This study is all the more significant in that it monitored the daily lives of the institutionalized disabled for the first time to allow in-depth understanding about the daily lives of the disabled; analyzed interaction patterns between the institutionalized disabled and living rehabilitation teachers; applied time budget study method, a frequently used one in adjacent fields, and therefore should serve as a valuable source material for future studies.

Trends in the Workday Leisure-Time of Dual-Income Parents with Preschool Children(2004-2019) (미취학자녀를 둔 맞벌이부모의 평일여가에서 나타난 변화(2004-2019))

  • The purpose of this study was to examine the workday leisure time of dual-income fathers and mothers with preschool children. Data were gathered from the Statistics Korea Life Time Surveys from 2004(y1) and 2019(y2) (n=2,697). Life time was composed of three types(inevitable time, compulsory time, and leisure time) and leisure time was categorized into 11 activities(voluntarism, participation, social relationships, religion, rituals, culture and tourism, sports, game and play, media, rest, and others) to investigate gender differences and time effects. The major results were as follows: First, both working fathers and mothers with preschool children spent less inevitable time, more compulsory time, and less leisure time, than other adult group. Second, fathers spent more time on 10 leisure activities except religion in y1, and on all activities than mothers in y2. In y2, less time on three activities(social relationships, rituals, and media) for both fathers and mothers, less time on religion for mothers only, more time on two activities(participation, game and play) for both groups were spent than y1. Third, in the regression analysis of fathers' and mothers' leisure time on three leisure activities(sports, game and play, and media) with a distinct pattern from leisure time for adult group between y1 and y2, socioeconomic variables and lifetime variables showed different effects by gender and waves. Based on the results, family policy and programs to improve the work-life balances of working fathers and mothers discussed.

Computer Simulation: A Hybrid Model for Traffic Signal Optimisation

  • With the increasing number of vehicles in use in our daily life and the rise of traffic congestion problems, many methods and models have been developed for real time optimisation of traffic lights. Nevertheless, most methods which consider real time physical queue sizes of vehicles waiting for green lights overestimate the optimal cycle length for such real traffic control. This paper deals with the development of a generic hybrid model describing both physical traffic flows and control of signalised intersections. The firing times assigned to the transitions of the control part are considered dynamic and are calculated by a simplified optimisation method. This method is based on splitting green times proportionally to the predicted queue sizes through input links for each new cycle time. The proposed model can be easily translated into a control code for implementation in a real time control system.

  • It is imperative to study the hydrolysis of urea in high saline-sodic condition of a newly reclaimed tidal land in order to overcome the problems associated with use of urea fertilizer. The methodology adopted in this study tried to get a convenient way of estimating rate for N transformation needed in N fate and transport studies by reviewing pH and salt contents which can affect the microbial activity which is closely related to the rate of urea hydrolysis. The hydrolysis of urea over time follows first-order kinetics and soil urease activity in reclaimed soils will be represented by Michaelis-Menten-type kinetics. However, high pH and less microorganisms may delay the hydrolysis of urea due to decrease in urease activity with increasing pH. Therefore, the rate of urea hydrolysis should adopt $V_{max}$ referring enzyme activity ($E_0$) accounting for urease concentration which is indicative for urea hydrolysis, especially in a high saline and sodic soils.

  • The environment is the important resource to use as well as to conserve for the development. Since 1990s, it has accentuated the importance of the environment and tried to conserve the environment in many areas. Among many area, the building industry has played a role to destruct the environment for providing the living decent space and brought the environmental affect. Therefore, the building industry should endeavor to mitigate the environmental affect. In building life cycle, it consumes the energy and natural resources. But it is difficult to evaluate the environmental affect during the whole building life. It is impossible to calculate the environmental affect without the much time and effort. Therefore. it requires the easy and handing program to calculate the affect to environment in each building life cycle. Until now, many program for assessing the affect to the environment have been developed and limitedly used because of the assessment scope and contents. In this paper, it aimed at developing the assessment program of LCA, focused on the energy consumption and carbon-dioxide emission compared with the domestic and foreign concerned programs. The developed program is divided into 3 areas; construction, maintenance, demolition.
