HPLC를 이용한 감초 추출물의 당 분석법 연구 (The Study of Sugar Analysis in Licorice Extract by HPLC)

  • The study was carried out to develope quantitative analysis method of individual sugars in licorice extract. Individual sugars were analyzed by HPLC equipped with Refractive Index(RI) Detector. R values of sucrose and glucose were 1.0000 and R values of fructose and maltose were 0.9999. Standard calibration curve showed good linearity. Detection limit of sugars was in the range of 1.58 to 3.92 ${\mu}g$. Recovery rate of fructose, glucose, sucrose and maltose was $99.4\~102.2\%,\;92.3\~97.9\%,\;99.4\~102.0\%,\;91.1\~101.0$ respectively. Measure uncertainty was calculated to confirm trust and accuracy of analytical results. Main uncertainty factors were standard purity and HPLC replication injection. In $95\%$ trust level expanded uncertainty of sugars in licorice extract were fructose $1.98\pm0.047,\;glucose\;1.32\pm0.065,\;sucrose\;11.69\pm1.177,\;maltose\;1.06\pm0.042\;g/100\;g$.

감초가 랫드의 배 · 태자 발생에 미치는 영향 (Effects of Licorice on Embryonic and Fetal Development in Rats)

  • The developmental toxicity of water extract of licorice (Glycyrrhiza glabra) was evaluated in rats. Licorice extract (500, 1,000 or 2,000 mg/kg) was dissolved in drinking water and orally administered to male rats from 9 weeks before mating to the day of copulation, and to females from 2 weeks before mating to gestational day 19. On gestational day 20, the animals were sacrificed for Cesarian section, and maternal and fetal abnormalities were examined. Licorice extract neither induce clinical signs, nor affect the body weight gain, feed and water intake, estrous cycle, copulation and fertility rates, blood $17\beta-estradiol$ level and organ weights of dams. Also, the implantation and development including body weights, absorption and death of embryos and fetuses were not influenced by in utero exposure to licorice. In addition, there were no increases in external, visceral and skeletal abnormalities of fetuses. Taken together, it is suggested that no observed adverse effect level of licorice extract is higher than 2,000 mg/kg, and that long-term in utero exposure to licorice might not cause developmental toxicities of embryos and fetuses.

Inhibitory Effects of Licochalcone A and Isoliquiritigenin on Monocyte Adhesion to TNF-$\alpha$-activated Endothelium

  • Numerous natural herbal compounds have been reported to inhibit adhesion and migration of leukocytes to the site of inflammation Licorice extracts, which have been widely used in traditional Chinese medicinal preparation, possess various pharmacological effects. Isoliquiritigenin, a biogenetic precursor of flavonoids with various pharmacological effects, is a natural pigment present in licorice. We attempted to explore whether licorice extracts and isoliquiritigenin mitigate monocyte adhesion to tumor necrosis factor-$\alpha$ (TNF-$\alpha$)-activated human umbilical vein endothelial cells (HUVEC). In addition, it was tested whether the inhibition of monocyte adhesion to the activated HUVEC accompanied a reduction in vascular cell adhesion molecule-l expression(VCAM-l). Dry-roasted licorice extracts in methylene chloride but not in ethanol markedly interfered with THP-l monocyte adhesion to INF-$\alpha$-activated endothelial cells. licochalcone A compound isolated from licorice extract in methylene chloride appeared to modestly inhibit the interaction of THP-l monocytes and activated endothelium. In addition, isoliquiritigenin abolished the monocyte adhesion with attenuating VCAM-l protein expression on HUVEC induced by INF-$\alpha$. These results demonstrated that non-polar components from dry-roasted licorice extracts containing licochalcone A as well as isoliquiritigenin were active in blocking monocyte adhesion to cytokine-activated endothelimn, which appeared to be mediated most likely through the inhibition of VCAM-l expression on HUVEC. Therefore, licorice may hamper initial inflammatory events on the vascular endothelium involving induction of endothelial cell adhesion molecules.

Anti-carcinogenic effects of non-polar components containing licochalcone A in roasted licorice root

  • BACKGROUND/OBJECTIVE: Licorice has been shown to possess cancer chemopreventive effects. However, glycyrrhizin, a major component in licorice, was found to interfere with steroid metabolism and cause edema and hypertension. The roasting process of licorice modifies the chemical composition and converts glycyrrhizin to glycyrrhetinic acid. The purpose of this study was to examine the anti-carcinogenic effects of the ethanol extract of roasted licorice (EERL) and to identify the active compound in EERL. MATERIALS/METHODS: Ethanol and aqueous extracts of roasted and un-roasted licorice were prepared. The active fraction was separated from the methylene chloride (MC)-soluble fraction of EERL and the structure of the purified compound was determined by nuclear magnetic resonance spectroscopy. The anti-carcinogenic effects of licorice extracts and licochalcone A was evaluated using a MTT assay, Western blot, flow cytometry, and two-stage skin carcinogenesis model. RESULTS: EERL was determined to be more potent and efficacious than the ethanol extract of un-roasted licorice in inhibiting the growth of DU145 and MLL prostate cancer cells, as well as HT-29 colon cancer cells. The aqueous extracts of un-roasted and roasted licorice showed minimal effects on cell growth. EERL potently inhibited growth of MCF-7 and MDA-MB-231 breast, B16-F10 melanoma, and A375 and A2058 skin cancer cells, whereas EERL slightly stimulated the growth of normal IEC-6 intestinal epithelial cells and CCD118SK fibroblasts. The MC-soluble fraction was more efficacious than EERL in inhibiting DU145 cell growth. Licochalcone A was isolated from the MC fraction and identified as the active compound of EERL. Both EERL and licochalcone A induced apoptosis of DU145 cells. EERL potently inhibited chemically-induced skin papilloma formation in mice. CONCLUSIONS: Non-polar compounds in EERL exert potent anti-carcinogenic effects, and that roasted rather than un-roasted licorice should be favored as a cancer preventive agent, whether being used as an additive to food or medicine preparations.

  • The purpose of this study was to develop the antiallergic and hypoallergic fennented soybean foods without side effect. We manufactured Chungkukjang with addition of herbal (aloe, cinamon, licorice root) extract. Sensory evaluation was performed to evaluate the acceptability by the consumer. Clinical evaluation was performed with 10 atopic dermatitis (AD) patients who showed positive reaction with specific IgE and skin prick test. Cross-over study between nonnal Chungkukjang and Chungkukjang with aloe extract was performed. In sensory evaluation, Chungkukjang with aloe extract obtained best score overall. In clinical evaluation, 7 out of 10 AD patients showed positive reaction to soy-bean and 4 out of 10 AD patients showed positive reaction to normal Chungkukjang. 2 out of 10 AD patients showed positive reaction to Chungkukjang with aloe extract. In conclusion, Chungkukjang could be recommended as functional food with hypoallergic effect. As adding aloe extract to Chungkukjang, hypo allergic effect was increased.

잎새버섯 균사체를 이용한 감초추출발효물의 플라보노이드 생성과 항염 활성 연구 (Production of Flavonoid Compounds and Anti-inflammatory Property of Fermented Licorice Extract with the Basidiomycete Grifola frondosa HB0071)

  • 리퀴리티게닌과 이소리퀴리티게닌은 감초의 주요 플라보노이드 성분이다. 이들 플라보노이드는 수용성 감초 추출물과 ${\beta}$-glucosidase를 생성하는 잎새버섯 HB0071 균사체 발효배양을 통하여 생산하였다. 감초추출물 내 리퀴리티게닌과 이소리퀴리티게닌은 잎새버섯 발효배양 동안 현저히 증가하였다. 이 균주의 ${\beta}$-glucosidase의 활성은 배양 96시간을 기준으로 최고 91.5 mU/mL로 확인되었으며, 감초추출발효물로부터 생성된 리퀴리티게닌과 이소리퀴리티게닌의 함량은 HPLC 분석을 통하여 최대 $568.5{\mu}g/mL$$89.6{\mu}g/mL$로 확인되었다. 본 연구에서는 감초추출물의 잎새버섯 발효 전 후의 시료가 처리된 각질형성세포를 이용하여 자외선 UVB에 조사로 발현된 염증유발인자(COX-2)와 사이토카인(IL-$1{\beta}$, IL-6) 모두 감초추출발효물(FLEx)에서 농도의존적으로 발현이 억제되는 것을 확인하였다. 결론적으로 리퀴리티게닌과 이소리퀴리티게닌의 함량이 증가된 감초추출발효물은 자외선으로부터 손상된 피부 염증반응을 완화시켜줄 것으로 사료된다.

감초의 분획과 Glycyrrhizin이 황소정랑의 Prostaglandin Synthetase활성에 미치는 효과 (The Effects of Licorice Fraction and Glycyrrhizin on Prostaglandin Synthetase Activity of Bull Seminal Vesicle)

  • The investigation aimed to study the effects of methanol fraction of licorice (FM 100) and glycyrrhizin on prostaglandin synthetase activity, in relation to their analgesic effects. Effects of FM 100 and glycyrrhizin on the activity of prostaglandin synthetase extracted from bull seminal vesicles were examined by the modified method of Takeguchi et al. The analgesic effect of FM 100 was tested in mice by the acetic acid writhing method. FM 100 was administered orally to mice. BSV prostaglandin synthetase activity was inhibited significantly by FM 100 in a dose-dependent manner, whereas the activity was slightly inhibited by glycyrrhizin. Statistically significant analgesic effects were also observed with FM 100. The results suggest that analgesic effect of licorice may be due to the inhibition of prostaglandin synthesis.

화장품 중 유용성감초추출물의 유통기한 예측을 위한 가속수명 시험연구 (A Study on the Accelerated Life Test for the Estimation of Licorice Durability in Cosmetics)

  • 유용성감초추출물(oil soluble licorice extract)은 미백에 효능이 있는 원료로서, 미백 효과가 최적으로 유지되는 유통기한을 알아야 할 필요가 있다. 실온 상태에서의 화장품 유통기한을 측정하는 것이 제품의 개발 기간에 비해 상대적으로 길기 때문에 가혹 조건하에서의 성분 변화 실험을 통해 실제 유통기한을 예측하고자 하였다. 이를 위해 유용성감초추출물의 수명과 가속온도 스트레스와의 관계를 아레니우스식에 의해 분석하였다. 가속 온도 스트레스 설정 시 화장품 제형이 변하지 않는 온도 범위내인 50, 55 및 $60^{\circ}C$로 설정하였고, 이 구간 내에서 일정 스트레스 부과 방식을 설정하여 실험을 수행하였다. 본 연구에서 화장품 중 유용성감초추출물의 수명은 초기함량의 10%가 감소되는 시간으로 정의하였으며, 각 온도에서의 유용성감초추출물의 수명은 $50^{\circ}C$에서 580 h, $55^{\circ}C$에서 319 h, $60^{\circ}C$에서 166 h로 측정되었다. 실험 결과값을 아레니우스 모델에 적용하여 유용성감초 추출물의 수명과 온도 사이의 수명 관계식[log(수명)=-35.0243+1.15322$\times$(11604.83/온도)]을 도출하였고, 이 식에 의해 $25^{\circ}C$에서 유용성감초추출물의 수명은 26 개월로 예측할 수 있었다. 예측 결과는 유용성감초추출물을 실온에서 측정한 결과로 검증하였으며 95% 신뢰수준에서 수명 예측값과 실제값은 유의하지 않았다. 이 연구 방법은 주름 및 미백개선용 기능성 화장품들의 유통기한을 빠르고 정확하게 예측하는데 기여할 것으로 기대된다.

Oral administration of Jinan Red Ginseng and licorice extract mixtures ameliorates nonalcoholic steatohepatitis by modulating lipogenesis

  • Background: Nonalcoholic steatohepatitis (NASH) is one of the main chronic liver diseases. NASH is identified by lipid accumulation, inflammation, and fibrosis. Jinan Red Ginseng (JRG) and licorice have been widely used because of their anti-inflammatory and hepatoprotective effects. Hence, this study assessed JRG and licorice extract mixtures' effects on NASH progression. Methods: Palmitic acid (PA) and the western diet (WD) plus, high glucose-fructose water were used to induce in vitro and in vivo NASH. Mice were orally administered with JRG-single (JRG-S) and JRG-mixtures (JRG-M; JRG-S + licorice) at 0, 50, 100, 200 or 400 mg/kg/day once a day during the last half-period of diet feeding. Results: JRG-S and JRG-M reduced NASH-related pathologies in WD-fed mice. JRG-S and JRG-M consistently decreased the mRNA level of genes related with inflammation, fibrosis, and lipid metabolism. The treatment of JRG-S and JRG-M also diminished the SREBP-1c protein levels and the p-AMPK/AMPK ratio. The FAS protein levels were decreased by JRG-M treatment both in vivo and in vitro but not JRG-S. Conclusion: JRG-M effectively reduced lipogenesis by modulating AMPK downstream signaling. Our findings suggest that this mixture can be used as a prophylactic or therapeutic alternative for the remedy of NASH.

건조방법과 향신료 추출물 첨가가 육포의 품질특성에 미치는 영향 (The Effects of Drying Method and Spice Extracts Added to Beef Jerky on the Quality Characteristics of Beef Jerky)

  • To develope Hanbang beef jerky as health food, six kinds of beef jerky were prepared by adding sugar (A), licorice (B) and three kinds of spice extracts (clove: C, fennel fruit: D and Chungyang green pepper extract: E) and mixture of all extracts (F). The effects of the drying method and added spice extracts on the quality characteristics of the beef jerky were evaluated. In general, the air-blast dried beef jerky contained $31{\sim}33%$ moisture, $50.0{\sim}51.2%$ crude protein, $7.2{\sim}7.8%$ crude lipid and $3.0{\sim}3.3%$ crude ash. For the mineral content of the air-blast dried jerky, the most prevalent mineral was Na $(1540.08{\sim}1838.17$ mg%), followed by K, P, Mg, and Ca. The Ca content of the beef jerky was highest in the mixed extract group (88.53 mg%), and the lowest content was in sugar-added group (53.12 mg%). For the color properties, the L-value (lightness), a-value (redness) and b-value (yellowness) were higher in the air-blast dried beef jerky than in the hot air dried samples. The drying methods showed their greatest affect on the redness (a) for all six jerkies (p<0.001). For the preference by sensory evaluation, the beef jerky samples with added sugar (A) and licorice extract (B) had significantly higher scores than the beef jerky samples with the added spice extracts, for both of air-blast drying and hot air drying (p<0.05). Preference for the air-blast dried beef jerkies with added clove (C) and fennel fruits (D) were significantly higher, in terms of taste, color, softness and aftertaste as compared to the respective hot air dried jerkies (p<0.01). Considering all the obtained results, we concluded that licorice and spice extracts can be used as natural preservatives in the development of health foods and the air-blast drying method is recommended to improve the quality characteristics of beef jerky.
