Influence of age and type of feed ingredients on apparent and standardized ileal amino acid digestibility in broiler chickens

  • Two experiments were conducted to determine the effects of bird age on apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of amino acids (AA) for 10-d-old Experiment (Exp. 1) and 22-d-old (Exp. 2) male broilers. This study investigated the effects of different broiler ages and feed ingredients on AID and SID of AA in corn and soybean meal (SBM). Four hundred and eighty (age = 7 d; initial body weight [BW] = 173.4 ± 12.65 g) and 192 (age = 18 d; initial BW = 772.2 ± 62.13 g) birds were allocated to three dietary treatments in a randomized complete block design with eight replicate cages per treatment. Two diets were formulated based on corn or SBM as the sole source of AA in the diet. A nitrogen-free diet was also formulated to measure basal endogenous losses of AA. Experimental diets were given for 3 and 4 days in Exps. 1 and 2, respectively. An interaction was observed (p < 0.05) between the age of birds and the type of ingredient for the AID of most AA, except for methionine, valine, cysteine (Cys), and tyrosine; however, the effects of age and type of ingredients were diminished in the SID of AA, except for histidine, isoleucine, leucine (Leu), phenylalanine, alanine (Ala), and glutamic acid (Glu). The AID of AA, except for Leu and Cys and the SID of AA, except for Leu, Ala, Glu, and Pro in SBM were greater (p < 0.05) than in corn. As the age of birds increased from 10 to 22 d, digestibility of all AA increased (p < 0.05), regardless of the expression of AA digestibility (i.e., AID and SID). In conclusion, the AID and SID of AA in both corn and SBM increased with increasing age, and the AID and SID of AA in SBM were greater than in corn.

  • F. velutipes Leu 2 gene (${\beta}-isopropylmalate$ dehydrogenase gene) was used for transformation of P. florida leucine requiring auxotrophic mutant P101. Transformation frequency was very low but the transformed colony can grow on minimal medium very slowly. Transformation was identified by Southern hybridization and reverse transformation into E. coli using chromosome DNA isolated from transformed P. florida.

Oligosaccharide Production by Leuconostoc lactis CCK940 Which Has Glucansucrase Activity (Leuconostoc lactis CCK940의 Glucansucrase 활성에 의한 올리고당 생산 최적화)

  • Glucansucrase is an enzyme classified as a glycoside hydrolase (GH) 70 family, which catalyzes the synthesis of glucooligosaccharides with a low molecular weight using sucrose as a donor of D-glucopyranose and maltose as a carbohydrate acceptor. In this study, glucansucrase-producing lactic acid bacteria strain was isolated from the fermented foods collected in traditional markets, and the optimum conditions for the oligosaccharide production were investigated. The strain CCK940 isolated from Chinese cabbage kimchi was selected as an oligosaccharide-producing strain due to its high glucansucrase activity, with 918.2 mU/mL, and identified as Leuconostoc lactis. The optimum conditions for the production of oligosaccharides using Leu. lactis CCK940 were to adjust the initial pH to 6.0, add 5% (w/v) sucrose and 10% (w/v) maltose as a donor and acceptor molecules, respectively, and feed 5% (w/v) sucrose at 4 and 8 h of cultivation. When Leu. lactis CCK940 was cultured for 12 h at optimum conditions, at least four oligosaccharides with a polymerization degree of 2-4 were produced.

  • This study was undertaken to investigate the effects of [$D-Ala^2$, D-Leu^5$]-enkephalin (DADLE) on the spontaneous activity of medial vestibular nuclear neurons of the rat. Sprague-Dawley rats, aged 14 to 16 days, were anesthetized with ether and decapitated. After enzymatic digestion, the brain stem portion of medial vestibular nuclear neuron was obtained by micropunching. The dissociated neurons were transferred to a recording chamber mounted on an inverted microscope, and spontaneous action potentials were recorded by standard patch-clamp techniques. The spontaneous action potentials were increased by DADLE in 12 cells and decreased in 3 cells. The spike frequency and resting membrane potential of these cells were increased by DADLE. The depth of afterhyperpolarization was not affected by DADLE. The potassium currents were decreased in 20 cells and increased in 5 cells. These results suggest that DADLE increases the neuronal activity of the medial vestibular nuclear neurons by altering resting membrane potential.

Enhancing the Alginate Degrading Activity of Streptomyces sp. Strain M3 Alginate Lyase by Mutation (Streptomyces sp. M3 알긴산분해효소의 돌연변이에 의한 활성증대)

  • A polyguluronate-specific lyase from Streptomyces sp. strain M3 has been previously cloned and characterized. In this study, the M3 alginate lyase gene in the pColdI vector was mutated by site-directed mutagenesis and random mutagenesis to enhance the alginate degrading activity. Six mutants were obtained: Ser25Arg, Phe99Leu, Asp142Asn, Val163Ala, Lys191Glu, and Gly194Cys. Phe99Leu and Lys191Glu mutants completely lost their alginate lyase activity, whereas the alginate degrading activity of Gly194Cys mutant increased by nearly 10 fold. The 3-D protein structure of M3 alginate lyase, which was constructed using the Swiss-Model automodeler, was also compared to the crystal structure of another alginate lyase. A mutated glycine residue was positioned between Gly193 and Tyr195 of the C-terminal conserved sequence, YFKAGXYXQ. A phenylalanine residue (at position 99) and a glycine residue (at position 194) mutated in this study were distant from the active site, but the degrading activity was strongly affected by their mutation.

LEU+ loaded APR1400 using accident tolerant fuel cladding for 24-month two-batch fuel management scheme

  • In this work, a 24-month two-batch fuel management strategy for the APR1400 using LEU + has been investigated, where enrichments of 5.9 and 5.2 w/o are utilized in lieu of the conventional 4-5 w/o UO2 fuel. In addition, an Accident Tolerant Fuel (ATF) clad based on the swaging technology is applied to APR1400 fuel assemblies. In this special ATF clad design, both outer and inner SS316 layers protect the conventional zircaloy clad. Erbia (Er2O3) is introduced as a burnable absorber with two-fold goals to lower the critical boron concentration in the long-cycle LEU + loaded core as well as to handle the LEU + fuel in the existing front-end fuel facilities without renewing the license. Two types of fuel assemblies with different loading of gadolinia (Gd2O3) are considered to control both the reactivity and the core radial power distribution. The erbia burnable absorber is uniformly admixed with UO2 in all fuel pins except for the gadolinia-bearing ones. In this study, two core designs were devised with different erbia loading, and core performance and safety parameters were evaluated for each case in comparison with a core design without any burnable absorbers. The core analysis was done using the two-step method. First, cross-sections are generated by the SERPENT 2 Monte Carlo code, and the 3-D neutronic analysis is performed with an in-house multi-physics nodal code KANT.

An investigative study of enrichment reduction impact on the neutron flux in the in-core flux-trap facility of MTR research reactors

  • Research reactors in-core experimental facilities are designed to provide the highest steady state flux for user's irradiation requirements. However, fuel conversion from highly enriched uranium (HEU) to low enriched uranium (LEU) driven by the ongoing effort to diminish proliferation risk, will impact reactor physics parameters. Preserving the reactor capability to produce the needed flux to perform its intended research functions, determines the conversion feasibility. This study investigates the neutron flux in the central experimental facility of two material test reactors (MTR), the IAEA generic10 MW benchmark reactor and the 22 MW s Egyptian Test and Research Reactor (ETRR-2). A 3D full core model with three uranium enrichment of 93%, 45%, and 20% was constructed utilizing the OpenMC particle transport Monte Carlo code. Neutronics calculations were performed for fresh fuel, the beginning of life cycle (BOL) and end of life cycle (EOL) for each of the three enrichments for both the IAEA 10 MW generic reactor and core 1/98 of the ETRR-2 reactor. Criticality calculations of the effective multiplication factor (Keff) were executed for each of the twelve cases; results show a reasonable agreement with published benchmark values for both reactors. The thermal, epithermal and fast neutron fluxes were tallied across the core, utilizing the mesh tally capability of the code and are presented here. The axial flux in the central experimental facility was tallied at 1 cm intervals, for each of the cases; results for IAEA 10 MW show a maximum reduction of 14.32% in the thermal flux of LEU to that of the HEU, at EOL. The reduction of the thermal flux for fresh fuel was between 5.81% and 9.62%, with an average drop of 8.1%. At the BOL the thermal flux showed a larger reduction range of 6.92%-13.58% with an average drop of 10.73%. Furthermore, the fission reaction rate was calculated, results showed an increase in the peak fission rate of the LEU case compared to the HEU case. Results for the ETRR-2 reactor show an average increase of 62.31% in the thermal flux of LEU to that of the HEU due to the effect of spectrum hardening. The fission rate density increased with enrichment, resulting in 34% maximum increase in the HEU case compared to the LEU case at the assemblies surrounding the flux trap.

Biosynthesis of Polymyxins B, E, and P Using Genetically Engineered Polymyxin Synthetases in the Surrogate Host Bacillus subtilis

  • The development of diverse polymyxin derivatives is needed to solve the toxicity and resistance problems of polymyxins. However, no platform has generated polymyxin derivatives by genetically engineering a polymyxin synthetase, which is a nonribosomal peptide synthetase. In this study, we present a two-step approach for the construction of engineered polymyxin synthetases by substituting the adenylation (A) domains of polymyxin A synthetase, which is encoded by the pmxABCDE gene cluster of Paenibacillus polymyxa E681. First, the seventh L-threonine-specific A-domain region in pmxA was substituted with the L-leucine-specific A-domain region obtained from P. polymyxa ATCC21830 to make polymyxin E synthetase, and then the sixth D-leucine-specific A-domain region (A6-D-Leu-domain) was substituted with the D-phenylalanine-specific A-domain region (A6-D-Phe-domain) obtained from P. polymyxa F4 to make polymyxin B synthetase. This step was performed in Escherichia coli on a pmxA-containing fosmid, using the lambda Red recombination system and the sacB gene as a counter-selectable marker. Next, the modified pmxA gene was fused to pmxBCDE on the chromosome of Bacillus subtilis BSK4dA, and the resulting recombinant strains BSK4-PB and BSK4-PE were confirmed to produce polymyxins B and E, respectively. We also succeeded in constructing the B. subtilis BSK4-PP strain, which produces polymyxin P, by singly substituting the A6-D-Leu-domain with the A6-D-Phe-domain. This is the first report in which polymyxin derivatives were generated by genetically engineering polymyxin synthetases. The two recombinant B. subtilis strains will be useful for improving the commercial production of polymyxins B and E, and they will facilitate the generation of novel polymyxin derivatives.

Gamakamide-E, a Strongly Bitter Tasting Cyclic Peptide with a Hydantoin Structure from Cultured Oysters Crassostrea gigas

  • A new cyclic peptide (six-membered amino acid), gamakamide-E (L-Leu-L-Met (SO)-L-Me-Phe-L-Leu-D-Lys-L-Phe), was isolated as a strongly bitter tasting compound from cultured oysters, Crassostrea gigas. The molecular formula of $C_{43}H_{61}N_7O_8S$ was deduced from high resolution fast atom bombardment mass spectrometry (HR FAB-MS) ($[M+H]^+$ m/z 836.4356 ${\Delta}$= -2.4 mmu). Its unique structure including a hydantoin structure was firstly elucidated by nuclear magnetic resonance (NMR) analysis. Stereochemistries of constituent amino acids were determined by chiral high performanced liquid chromatography analysis of natural and synthesized peptides.

Malo-lactic Bacteria in Korean Winery Environment and Their Potential Use in Wine Making (한국내 양조 환경하의 malo-lactic 박테리아의 분포 및 그들의 양조업에의 이용성)

  • Substantial numbers of malo-lactic bacteria were detected in Korean winery environment such as in grape juices, fermenting musts, lees, aging wines, and bottled apple wines. Among 1363 malo-lactic strains isolated from the above habitats, four superior strains were selected and identified as Leuconostoc oenos strain A-25, B-30, C-13 and Lactobacillus plantarum strain D-11. The strain A-25 showed malate utilizing activity almost equivalent to Leu. oenos ML34, a well-known malo-lactic bacterium. Upon experimental vinifications the malo-lactic fermentation efficiency of A-25 was confirmed.
