cDNA Sequences for Asialoglycoprotein Receptor from Human Fetal Liver

  • The asialoglycoprotein receptor (ASGPR) was the first described mammalian lectin that mediates the specific binding and internalization of galactose/N-acetylgalactosamine-terminating glycoproteins by hepatic parenchymal cells. H1 and H2 are known as essential subunits of the functional ASGPR. There were close similarities in ASGPR H2 subunits between cultured cell line HepG2 and normal human liver cells including identical sequences at both termini. It was therefore expected that there may be some similarities between the subunits from normal liver cells and fetal liver cells. The two subunits of human fetal liver ASGPR. designated FL-H1 and FL-H2. were cloned from cDNA library by peR and the sequences were compared with the known HI and H2 sequences of HepG2, and the H1 sequence of nornal human liver cells. The results showed that FL-H1 was identical to H1 of HepG2. Whereas FL-H2 contains a 15-bp miniexon, but missing 57-bp at the near upstream from the membrane-spanning domain compared to H2 of HepG2 and normal human liver cells indicating that FL-H2 resulted from a differential splicing compared to HepG2 and normal liver cells.

The gene repertoire of Pythium porphyrae (Oomycota) suggests an adapted plant pathogen tackling red algae

  • Pythium porphyrae is responsible for devastating outbreaks in seaweed farms of Pyropia, the most valuable cultivated seaweed worldwide. While the genus Pythium contains many well studied pathogens, the genome of P. porphyrae has yet to be sequenced. Here we report the first available gene repertoire of P. porphyrae and a preliminary analysis of pathogenicity-related genes. Using ab initio detection strategies, similarity based and manual annotation, we found that the P. porphyrae gene repertoire is similar to classical phytopathogenic Pythium species. This includes the absence of expanded RxLR effector family and the detection of classical pathogenicity-related genes like crinklers, glycoside hydrolases, cellulose-binding elicitor lectin-like proteins and elicitins. We additionally compared this dataset to the proteomes of 8 selected Pythium species. While 34% of the predicted proteome appeared specific to P. porphyrae, we could not attribute specific enzymes to the degradation of red algal biomass. Conversely, we detected several cellulases and a cutinase conserved with plant-pathogenic Pythium species. Together with the recent report of P. porphyrae triggering disease symptoms on several plant species in lab-controlled conditions, our findings add weight to the hypothesis that P. porphyrae is a reformed plant pathogen.

  • 렉틴(Lectins)은 세포막의 당단백질의 과당류 말단기에 특이적으로 결합하는 비면역성의 당단백질 혹은 단백질을 말하는데 정상세포와 변형된 세포에서 렉틴반응의 차이가 남으로써 요즘에는 렉틴을 병리학적 진단에 이용 가능하게 되었다. “Loss of contact inhibition”은 세포 악성화의 중요한 특징으로 세포의 악성화는 세포막의 구조의 변화와 관계가 있을 것으로 알려져 왔으며 1989년 후두암에서 PNA의 반응이 양성 반응을 나타낸다고 보고되어 저자들은 7가지의 렉틴을 이용하여 후두암전구증의 하나인 후두각화증에서 이형성(Atypia)의 존재유무에 따른 렉틴 반응의 변화를 알아보고자 본 연구를 시행하여 다음과 같은 결과를 얻었다. 1. 이형성이 없는 단순 후두각화증에서는 Con A와 WGA만이 기저세포에 반응이 있었으며, 후두각화증의 가장 중요한 부위인 극세포층에서는 WGA, RCA-I, PNA, SBA, UEA-I, DBA가 세포질에는 반응이 없이 세포간교에만 염색이 되는것을 관찰할 수 있었고 Con A는 반대로 극세포층의 세포질에 반응을 하였으나 세포간교에는 반응이 없었다. 2. 이형성을 동반한 후두각화증에서, 기저세포에서는 반응의 차이가 없었고 극세포층에서는 UEA-I, RCA-I. WGA, PNA, SBA는 세포간교에서 반응이 나타나지 않았고 세포질에서 양성 반응을 관찰 할 수 있었으며 DBA, Con-A에서는 반응의 차이를 관찰 할 수 없었다. 따라서 후두각화증의 극세포층 세포막에서 UEA-I, RCA-I, WGA, PNA, SBA 반응의 소실은 후두암으로의 진행과 밀접한 관계가 있는 이형성의 존재를 암시한다하겠다.

Characterization of Fetal Gonad-Derived Cells by Stem Cell Markers (줄기세포 Marker를 이용한 돼지 태아 생식선 유래 세포의 특성화)

  • In mammals, male and female germline stem cells are derived from primodial germ cells. Despite many efforts to identify stem cells from gonads, there has been little successe to identify germline stem cells yet. In this study, we isolate and characterized porcine germline stem cells using only stem cell markers that are prevalently expressed in various tissues. Gonadal cells derived from both male and female formed colonies and showed AP activities and different lectin binding properties. Pluripotency of germline stem cells was also identified by positive signals against putative stem cells markers such as SSEA-1 and SSEA-3. In addition, nestin was also found in primary gonad cells that have a similar morphology to the AP-positive cells. The nestin expression suggests that the germline stem cells may have similar expression of the prevalent stem cell markers found in other tissues. The demonstration of nestin expression together with pluripotent cell markers calls further investigation of the possible differentiation of nestin-positive cells into neurons.

Characterization of immune gene expression in rock bream (Oplegnathus fasciatus) kidney infected with rock bream iridovirus (RBIV) using microarray

  • Rock bream iridovirus (RBIV) causes high mortality and economic losses in rock bream (Oplegnathus fasciatus) aquaculture industry in Korea. Although, the immune responses of rock bream under RBIV infection have been studied, there is not much information at the different stages of infection (initial, middle and recovery). Gene expression profiling of rock bream under different RBIV infection stages was investigated using a microarray approaches. In total, 5699 and 6557 genes were significantly up- or down-regulated over 2-fold, respectively, upon RBIV infection. These genes were grouped into categories such as innate immune responses, adaptive immune responses, complements, lectin, antibacterial molecule, stress responses, DNA/RNA binding, energy metabolism, transport and cell cycle. Interestingly, hemoglobins (α and β) appears to be important during pathogenesis; it is highly up-regulated at the initial stage and is gradually decreased when the pathogen most likely multiplying and fish begin to die at the middle or later stage. Expression levels were re-elevated at the recovery stage of infection. Among up-regulated genes, interferon-related genes were found to be responsive in most stages of RBIV infection. Moreover, X-linked inhibitor of apoptosis (XIAP)-associated factor 1 (XAF1) expression was high, whereas expression of apoptosis-relate genes were low. In addition, stress responses were highly induced in the virus infection. The cDNA microarray data were validated using quantative real-time PCR. Our results provide novel inslights into the broad immune responses triggered by RBIV at different infection stages.

The Effects of EGEE on Lectins Binding Pattern of Rat Cerebral Cortex in Developmental Phase (발생기 흰쥐 대뇌 피질의 복합당질에 미치는 ethylene glycol monoethyl ether의 영향)

  • This study attempted to investigate the developmental alterations of the cerebral cortex. The effects of EGEE on the developmental cerebral cortex in the prenatal, postnatal and adults were examined by lectin histochemical methods. To investigate the change of sugar residues of glycoconjugates, biotinylated lectins(DBA, SBA, PNA, BSL-1, RCA-1, sWGA, UEA-1, Con A and LCA) were detected with by IHC using ABC kit. In the case of injection of EGEE, the reactions to Con A and LCA were shown in binding phase in the cerebral cortex commonly, and the reactions to PNA, RCA-1 and LCA were shown partially, the number of lectins to be shown reaction were decreased, and there were no reactions to DBA, SBA, BSL-1, RCA-1 and UEA-1. The reaction to Con A was similar to control group during developmental phases. The reaction to LCA was increased in the fetal, suckling, and weanning phases compared with control group. But there were no reactions to SBA and sWGA, the reaction to PNA was decreased in the frontal and occipital cortex and no reaction to sWGA in the fetal phase. There were no reactions to sWGA and PNA in the suckling phase and, no reaction to PNA and sWGA. The reaction to Con A was decreased in the frontal, parietal and occipital cortexes and, the reaction to LCA was decreased in the frontal and occipital cortexes in adult phase. As the results, the effects of EGEE, environmental hormone on the each part of cerebral cortex have shown differences. But, It had deep effect on the differentiation and growth in the cerebral cortex pathologically. In particular, the effect was severe in suckling phase. $Galactosyl-({\beta}-1,3)-N-acetyl-D-galactosamine$,${\beta}-N-acetyl-D-galactosamine$ and ${\beta}-N-acetyl-D-glucosamine$ were decreased while ${\alpha}-D-mannose$ and ${\alpha}-D-glucose$ were increased. It affected the sugar metabloism, and it was severe in fetal and suckling phases.

  • Morphological features of the interaction between the hatching blastocyst and implantation in pig were studied by electron microscopy. The observations extended from late blastocyst stage to the completion of trophoblastic erosion of the epithelium and early decidual transformation of the epithelium and early decidual transformation of the stromal cells. Between day 7 and 17 of pregnancy, blastocysts from 0.3 to 12 mm in diameter were flushed from the uterine horns of Dutch Landrace pigs. On the 7th of development in the pig blastocyst, the blastocyst shedded of the zona pellucida established the tips of microvilli and with bleb-like cytoplasmic protrusions of the epithelial cells. From day 11 on in pig embryo, the bilayered trophoblast undergoes a dramatic phase of elongation so that the initially spherical expanded blastocyst becomes tubular. In pig, close apposition to the uterine wall beg-ins at about 12 $^1$/$_2$ days and then attachment occurred during the afternoon of the 16th or 18th day post coitum. At this stage, embryonic loss compared with corpus luteum number is up to 40% of ovulated oocytes. Therefore, the implantation failture of these embryos may be mainly caused by morphological abnormality and failture of zona shedding.

Transcriptomic Profile in Pear Leave with Resistance Against Venturia nashicola Infection (배 검은별무늬병 감염과 저항성 방어반응 연관 전사체 프로파일)

  • The molecular understanding of resistance and susceptibility of host plants to scab, a most threatful disease to pome fruit production worldwide, is very limited. Comparing resistant line '93-3-98' to susceptible one 'Sweet Skin' at seven time points of 0, 0.5, 1, 2, 3, 4, 8 days post inoculation, RNA-sequencing data derived from infected and mock-inoculated young leaves were analyzed to evaluate the tolerant response and to mine candidate genes of pear to the scab pathogen Venturia nashicola. Analysis of the mapped reads showed that the infection of V. nashicola led to significant differential expression of 17,827 transcripts with more than 3-fold change in the seven pairs of libraries, of which 9,672 (54%) are up- and 8,155(46%) are down-regulated. These included mainly receptor (NB-ARC domains-containing, CC-NBS-LRR, TIR-NBS-LRR, seven transmembrane MLO family protein) and transcription factor (ethylene responsive element binding, WRKY DNA-binding protein) related gene. An arsenal of defense response of highly resistant pear accessions derived from European pear was probably supposed no sooner had V. nashicola infected its host than host genes related to disease suppression like Polyketide cyclase/dehydrase and lipid transport protein, WRKY family transcription factor, lectin protein kinase, cystein-rich RLK, calcium-dependent phospholipid-binding copine protein were greatly boosted and eradicated cascade reaction induced by pathogen within 24 hours. To identify transcripts specifically expressed in response to V. nashicola, RT-PCRs were conducted and compare to the expression patterns of seven cultivars with a range of highly resistant to highly susceptible symptom. A DEG belonging to the PR protein family genes that were higher expressed in response to V. nashicola suggesting extraordinary role in the resistance response were led to the identification. This study provides the first transcriptional profile by RNA-seq of the host plant during scab disease and insights into the response of tolerant pear plants to V. nashicola.

The Effect of Pretreatment with Various Mutagens on Glycoconjugates of Plasma Membrane in HeLa Cells (HeLa 세포에서 원형질 막의 glycoconjugate에 대한 수종 돌연변이원의 전처리 효과)

  • The present investigation has been performed to elucidate the effect of pretreatment with low dose of ultraviolet radiation (UV), ethyl methansulfonate (EMS), and bleomycin (BLM) on cell survival and lectin-binding glycoconjugates of plasma membrane in HeLa cells treated with mutagen. The percentage of survival of cells pretreated with 1 mM EMS following treatment with 10 mM EMS was higher than that of cells treated with 10 mM EMS alone. Wheat germ agglutinin (WGA) staining intensity of cells pretreated with 1 mM EMS and subsequently treated with 10 mM EMS was stronger than that of cells treated with 10 mM EMS alone. But, succinylated wheat germ agglutinin (sWGA) staining intensity of cells pretreated with 1 mM EMS and subsequently treated with 10 mM EMS was similar to that of cells treated with 10 mM EMS alone. These results suggest that the acquired resistance to EMS is related to the glycoconjugates containing sialic acid of plasma membrane involved in multidrug resistance or adaptive response in HeLa cells.

Histopathological changes of epithelium following the exposure of N-methyl-N-nitro-N-nitrosoguanidine in Manila clam, Ruditapes philippinarum (N-methyl-N-nitro-N-nitrosoguanidine 노출에 따른 바지락 (Ruditapesphilippinarum) 상피조직의 병리조직학적 반응)

  • To understand responses of bivalve tissues to various injurious agents, histopathological observation was done in the Manila clam, Ruditapes hilippinarum, during 240 hours after an intramuscular injection of N-methyl-N-nitro-N-nitrosoguanidine (MNNG). Some histopathological changes were observed with the epithelia of stomach, midgut, hindgut, digestive diverticula and gonads after injecting MNNG. The earliest degenerative changes were recognized in the epithelium of midgut 12 hours after the injection. Partial or complete, epithelial destruction was constantly accompanied by the massive infiltration of a mononuclear, necrotic cell group including hemocytes. At 144 hours later, gonads showed the necrotic desquamation of their germinal epithelia. Nearly all of the infiltrated cells within the destructive epithelium were suggested to be hemocytes, from the binding property with a hemocyte-specific lectin, Ricinus communis (RCA-1). From these results it was concluded that the epithelium of digestive system is vulnerable to parenteral MNNG and hemocytes might be deeply involved in this MNNG-induced destruction of the digestive epithelium.
