BACKGROUND: Kiwifruit is a fruit tree with relatively small cultivation area in Korea and researches on its soil and physiology are very limited compared to those on cultivar development. Therefore, there are limited information for farmers to cope with the reduction in productivity due to various physiological disorders and premature aging. This study was conducted to investigate the soil and leaf chemical properties, and fruit characteristics, which will be used as basic data for stable kiwifruit orchard soil management. METHODS AND RESULTS: The soil and leaf chemical properties, and fruit characteristics were investigated for two years in 16 kiwifruit orchards growing 'Hayward' (Actinidia deliciosa) in Jeollanam-do and Gyeongsangnam-do. Soil and leaf samples were collected in July and fruit quality was investigated by harvesting fruits about 170 days after full bloom. The average soil chemical properties of kiwi orchards were generally higher than the recommended level, except for pH, and especially, the exchangeable potassium reached about 300% of the recommended level. The proportions of orchards that exceeded the recommended level of soil chemical properties were 63, 31, 100, 69, 94, 88 and 69% for pH, EC, organic content, available phosphate, and exchangeable potassium, calcium and magnesium, respectively. Thirty-three percent of orchards had more than 100 mg/kg of nitrate nitrogen in soil. Available phosphate in soil showed a significantly positive correlation with leaf nitrogen, phosphoric acid and calcium content, but showed a significantly negative correlation with leaf potassium content. The magnesium content in the leaves was significantly correlated with soil pH. The highest fruit weight was observed in about 25 g/kg of leaf nitrogen content which could be attained when plants were grown on the soil containing about 100 mg/kg of nitrate nitrogen content. The average soluble solids content among 16 orchards was 9.58 °Brix at harvest and 13.9 °Brix after ripening, which increased about 45%, and the average fruit weight was about 110 g. CONCLUSION(S): For fruit quality, fruit soluble solids (sugar compounds) content was significantly correlated with leaf potassium content, fruit hardiness with leaf total nitrate, calcium and magnesium, and fruit titratable acidity with leaf magnesium; however, leaf calcium and magnesium negatively affect the soluble solids contents in fruits.
We used a flow cytometer to investigate the variations in endopolyploidy (the frequencies of nuclei with DNA contents equivalent to 4C through 16C) during the short period of the early growing stage in vigorously growing young tissues of maize seedlings. We examined different portions of the root and leaves that had been growing for 7 (day 7) and 13 (day 13) days after germination. Endoreplication showed two opposing phenomena without aging. In one case, the endopolyploidy of the first leaf was higher on day 13 than on day 7. In the latter case, endopolyploidy decreased, as clearly revealed by a comparison of the endopolyploidy of the second leaves and the 160-170 mm portion of the seminal root on days 7 and 13. Endopolyploidy was also lower in the top of the leaf. In roots, endopolyploidy was increased by the exogenous application of abscisic acid for only 1 day. The levels of endopolyploidy increased without an increase in cell size in the roots. These results showed that endoreplication occurs in actively growing and young tissue and that the variation can be induced in the short period examined.

Schizophragma hydrangeoides (S. hydrangeoides) is a vine endogenous to Jeju Island and Ulleungdo, where it grows attached to the foothills and rock surfaces. Previous research has mostly focused on the whitening effect of S. hydrangeoides leaf extract. In this study, we investigated S. hydrangeoides leaf extract further, and detected four phytochemicals in the extract: chlorogenic acid, quercetin-3-O-glucosyl-(1-2)-rhamnoside, quercetin-3-O-xylosyl-(1-2)-rhamnoside, and quercitrin. We pretreated human dermal fibroblast (HDFn) cells with previously established concentrations of the four compounds for 1 h before ultraviolet A (UVA) irradiation. Among the four compounds, quercetin-3-O-glucosyl-(1-2)-rhamnoside (Q-3-GR) best inhibited matrix metalloproteinase-1 (MMP-1) levels. Thus, we investigated the protective effects of Q-3-GR on photoaging and its underlying mechanisms. Q-3-GR significantly reduced MMP-1 production and inhibited MMP-1 protein expression in UVA-irradiated HDFn cells. Furthermore, Q-3-GR increased procollagen type I production and protein expression. Q-3-GR exerted its anti-photoaging effects by downregulating the mitogen-activated protein kinase/ activator protein-1 signaling pathway, and upregulating the transforming growth factor-β/Smad signaling pathway. Thus, S. hydrangeoides leaf-derived Q-3-GR is a potential potent cosmetic ingredient for UV-induced skin aging.
본 연구에서는 예로부터 민간에서 사용되어진 약초인 토인삼 추출물의 피부보호 효과를 측정하기위해 항산화, 자외선에 의해 유도된 matrix metalloproteinases-1 (MMP-1) 발현저해효과, 자외선에 대한 세포보호효과, 노화세포를 이용한 senescence-associated ${\beta}$-galactosidase (SA-${\beta}$-gal) 활성을 사람 섬유아세포(HDF)를 이용하여 확인하였다. 그 결과, 토인삼 추출물의 free radical과 superoxide radical 소거효과는 처리농도가 증가함에 따라 농도 의존적으로 나타났으며, 토인삼 잎, 줄기 추출물(LSE) 500 ${\mu}g/mL$에서 98.45%와 97.01%의 DPPH와 superoxide radical을 소거하여 우수한 항산화 효과를 나타내었다. 토인삼 잎, 줄기 추출물(LSE)의 MMP-1의 발현 저해효과는 섬유아세포에서 UVA조사 실험에서 우수하게 나타났으며, UVB 조사에 의한 세포보호 효과도 우수하게 나타났다. 또한 노화세포를 이용한 SA-${\beta}$-gal활성은 토인삼 잎, 줄기 추출물(LSE)을 처리하였을 시 염색된 세포의 수가 감소하여 세포내 senescence를 억제하는 것을 확인할 수 있었다. 이상의 결과들을 종합해 보면, 토인삼의 지상부분인 잎, 줄기 추출물(LSE)은 항노화 및 항산화제로서의 우수한 특성으로 피부손상에 의한 산화적 스트레스에 대응하는 새로운 기능성 소재로서의 가능성을 가지는 것으로 평가된다.
본 연구에서는 석류의 항산화 및 항염 효과를 평가하고, 이를 통해 석류 잎 추출물이 항노화 화장품 소재로서 활용될 가능성을 확인하고자, 석류 잎 에탄올 추출물의 에틸아세테이트 분획물(Ethyl acetate fraction of ethanolic extract of Punica granatum leaf : EFP)의 총 폴리페놀 및 플라보노이드 함량, 항염 및 항산화 활성을 평가하여, 석류 잎의 유익한 성분과 피부 개선 가능성을 탐구하였다. 실험 결과 EFP의 총 폴리페놀 함량은 871.6±16.3 mg gallic acid/g, 플라보노이드 함량은 36.6±0.3 mg quercetin/g으로 나타났다. ABTS 라디칼 소거활성을 평가에서는 EFP가 농도 의존적으로 항산화능을 보였고, EC50 수치는 24.62±0.48㎍/mL로 나타났다. 피부 세포독성 실험에서 EFP는 50㎍/mL 이하 농도에서 높은 세포생존율을 보였으며, 이는 세포 독성이 거의 없음을 시사한다. NO 생성 억제 실험에서는 EFP가 낮은 농도에서도 효과적으로 NO 생성을 억제하였으며, 6㎍/mL 농도에서 거의 완전히 억제되었다. 이러한 결과는 항산화와 항염 효과를 지닌 천연 화장품 소재로 활용 가능성을 기대할 수 있다.
국내산 녹차의 채엽시기가 녹차 중의 카테킨, 카페인, 아미노산 및 무기질 함량과 조성에 미치는 영향에 대하여 조사하였다. 녹차의 총 카테킨 함량은 40.61-52.04mg/g으로 채엽시기가 늦을수록 증가하였으며, 카테킨 조성은 채엽시기에 관계없이 EGC, EGCg, EC, CC, ECg, C, GCg 및 Cg의 순으로 함유되어 있음을 알 수 있었다. 카테킨류 중 기능성 작용이 우수한 EGC 및 EGCg는 전체의 50% 이상을 차지하였고 온화한 떫은맛으로 후미에 감미를 남기는 유리형 카테킨인 EC와 EGC는 전체의 53.21%에서 57.09%를 차지하였다. 카페인 함량은 4월차 17.56mg/g, 5월차 15.71mg/g, 6월차 14.92mg/g 및 7월차 14.61mg/g으로 채엽시기가 늦을수록 감소하는 경향을 나타냈다. 총 아미노산 함량은 4월차 45.84mg/g, 5월차 22.64mg/g, 6월차 12.22mg/g 및 7월차 10.45mg/g으로 채엽시기가 늦을수록 감소하였으며 특히, 중요한 생리적 기능을 갖는 theanine의 감소 폭이 큰 것으로 타났다. 무기질 함량은 채엽시기가 늦어질수록 Fe, Mn 및 Mg는 증가하였으며, Cu는 감소함을 알 수 있었다.
성체식물기에 잎도열병에 대해 저항성인 벼 품종 선발을 위해 한국에서 재배되거나 육종재배로 쓰이고 있는 32개의 벼 품종을 도열병검정 못자리에 공시하였다. 생육시기가 다른 각 잎에 자연감염된 발병율을 토대로 비교할 때, 16개의 통일형 품종들은 모두 고도 저항성이었으나 16개의 일본형 품종들은 잎도열병에 대해 다양한 저항성정도를 나타내었다. 잎도열병에 대한 저항성정도는 품종에 따라 다르지만, 식물체가 성숙해 갈수록 양적저항성정도는 모든 품종에서 증가하였다. 감수성에서 저항성으로 바귀는 엽위는 품종에 따라 달랐다. 도열병균에 대한 품종의 감수성반응이 상위엽에서 저항성반응으로 뚜렷하게 바뀌었다. 잎도열병에 대한 양적 저항성정도가 큰 품종일수록 하위엽에서 저항성반응을 나타내었다. 생육후기에 도열병감염을 급격히 감소시키는 품종인 아까바레, 팔금, 진흥, 올찰, 도봉 및 은하는 포장에서 잎도열병에 대해 성체식물저항성을 보였다. 성체식물저항성품종들은 식물체가 성숙해감에 따라 최적, 질적으로 잎도열병에 대해 저항성을 나타내었다.
오래 전부터 성분에 대한 확실한 분석은 없었으나 식용과 약용으로 이용되어 온 소리쟁이(Rumex crisps.)의 항 산화 물질 및 활성에 대해 시험하였다. 항 산화 활성 측정은 DPPH 법과 효소 활성 검사법을 이용하였고, Rumex crispus, Rumex acetoceae와 Rumex nipponicus의 항산화력을 DPPH 법에 의해 측정한 결과 3종류 모두 항산화력이 뿌리>잎>줄기의 순으로 높게 나타났다. 뿌리 추출물에 항산화력은 50% 억제 율이 Rumex crispus ; 6.1 ug/ml, Rumex nippponicus ; 9.8 ug/mL, Rumex acetoceae ; 25.3 ug/mL의 순으로 활성이 높게 나타났다. 잎 추출물 대한 $IC_{50}$/값이 Rumex acetoceae ; 31.5 ug/mL, Rumex nippponicus ; 59.1 ug/mL, Rumex crispus ; 68.8 ug/mL의 순으로 항산화활성이 높게 나타났다. Rumex crispus에서 분리 동정된 주요 페놀성 항 산화 물질은 2, 6-Dichloro-4-nitrophenol, 2-Isopropyl-5-methylphenol은 잎과 뿌리 추출물 모두에서 확인되었고, 4-Vinyl-2-methoxy-phenol과 2, 3-Dihydro-benzofuran 2종은 잎 추출물에서만 동정되었다. 동정된 각 물질의 분자량은 2, 6-Dichloro-4-introphenol ; 206.95, 2-Isopropyl-5-methylphenol ; 150.10, 4-Vinyl-2-methoxy-phenol ; 150.07, 2, 3-Dihydro-benzofuran ; 120.06이 모두 120~206 범위 안에 속하는 低分子化合物質이였다. Rumex crispus의 callus와 생체식물의 효소활성 측정 결과 POD 비활성 (IU/mg protein)은 줄기가 0.44 IU/mg protein으로 높은 활성을 나타내었으나 callus와 잎 부분의 활성은 모두 낮았고, SOD비활성 (IU/mg protein)은 줄기부분이 24.4 IU/mg protein으로 가장 높았다. POD isoenzyme pattern을 분석한 결과 뿌리와 callus에 3개의 isoenzyme, 잎과 줄기 부분에서는 2개의 isoenzyme이 발견되었다. Rumex SOD isoenzyme pattern 분석결과는 뿌리와 Callus에서만 1개의 Isoenzyme 이 나타났다.
Morus alba has been well known for its tonic effect or preventing of hypertension, aging, or diabetes mellitus in Asian countries. To evaluate the its effect on hair growth promotion, we performed the hair regrowth experiment with leaf, branch, root and fruit extract of Morus alba on animal model of C57BL/6 mouse. The hair regrowth effect was remarkable in the mouse treated branch and root extract (with what %), however, little effect on hair regrowth appeared on the mouse treated with leaf extract. Morus alba extract displayed antibacterial effects on Bacillus subtilis, Staphylococcus aureus, Pseudomonas and Candida albicans. These results indicated that branch and root extract of Morus alba could be applicable for hair regrowth or prevention of hair loss in human.

Muscle atrophy, which is defined as a decrease in muscle mass and strength, is caused by an imbalance between the anabolism and catabolism of muscle proteins. Thus, modulating the homeostasis between muscle protein synthesis and degradation represents an efficient treatment approach for this condition. In the present study, the protective effects against muscle atrophy of ethanol extracts of Morus alba L. (MA) and Angelica keiskei Koidz. (AK) leaves and their mixtures (MIX) were evaluated in vitro and in vivo. Our results showed that MIX increased 5-aminoimidazole-4-carboxamide ribonucleotide-induced C2C12 myotube thinning, and enhanced soleus and gastrocnemius muscle thickness compared to each extract alone in dexamethasone-induced muscle atrophy Sprague Dawley rats. In addition, although MA and AK substantially improved grip strength and histological changes for dexamethasone-induced muscle atrophy in vivo, the efficacy was superior in the MIX-treated group. Moreover, MIX further increased the expression levels of myogenic factors (MyoD and myogenin) and decreased the expression levels of E3 ubiquitin ligases (atrogin-1 and muscle-specific RING finger protein-1) in vitro and in vivo compared to the MA- and AK-alone treatment groups. Furthermore, MIX increased the levels of phosphorylated phosphoinositide 3-kinase (PI3K), protein kinase B (Akt), and mammalian target of rapamycin (mTOR) that were reduced by dexamethasone, and downregulated the expression of forkhead box O3 (FoxO3a) induced by dexamethasone. These results suggest that MIX has a protective effect against muscle atrophy by enhancing muscle protein anabolism through the activation of the PI3K/Akt/mTOR signaling pathway and attenuating catabolism through the inhibition of FoxO3a.
