Effects of Squalene on Renal Toxicity Induced by Lead Acetate in Proximal Tubules of the Mice (생쥐 근위세뇨관에서 납에 의한 신장 독성에 대한 스쿠알렌의 효과)

  • To investigate the effect of the squalene against the lead toxicity and recovery of renal failure. Healthy male ICR mice were used for experiment. The activity of nitric oxide (NO) was observed after the intraperitoneal injection in mice. The ultrastructural changes of the kidney were observed after the intraperitoneal injection of lead acetate in mice. The experimental groups were divided into three groups. Group 1 was normal mouse. Group 2 was not treatment with squalene after intraperitoneal contamination of lead acetate (30 mg/kg). And, Group 3 was injected squalene (180 mg/kg) after intraperitoneal contamination of lead acetate. All groups were used to 10 mice. The results were as follow: In the case of the group 2, swelling of the outer membrane and destruction of the inner membrane (cristae) of the mitochondria, dripping of the ribosomes from the rough endoplasmic reticulum were happened at 24 hours and 48 hour. These were gradually reparied after 72 hours. In the case of the group 3, damages of the mitochondria and the rough endoplasmic reticulum were showed less than the group 2 at 24 hours. Especially, after 48 hours, these were almost same as the group 1. In the case of group 2, the level of NO was decreased. However, In the case of group 3, the level of NO was increased more than normal as well as repaired the decreased NO level by Pb (P<0.05). It was concluded that the squalene was the protective and recovery effects for the toxicity of the lead in the renal proximal tubules.

  • Details on the electrochemical formation of lead dioxide from aqueous plumbous ion are studied by measuring current-time behavior with potential step method at a rotating platinum electrode. A cubic law without induction period can be applied to the crystallization of lead dioxide in both acetate and nitrate media. In the course of the mechanistic study, the presence of a soluble intermediate during the nucleation step is clearly observed with a rotating ring-disk electrode. Decrease in the anodic ring current due to the reduction of soluble species formed during the anodic crystallization of lead dioxide at disk is detected.

Histological Study on the Effects of the Green-Tea in Rat Liver Toxicated by Lead (녹차가 납 중독된 흰쥐의 간장에 미치는 형태학적 연구)

  • To investigate the defensive effect of green tea against the lead toxicity, Sprague-Dewley rats (150 gm) were divided into 5 groups; the control group, the group treated with lead for 4 weeks (Group A-1), the group treated with lead and green tea for 4 weeks (Group A-2), the group treated with lead for 8 weeks (Group B-1), and the group treated with lead and green tea for 8 weeks (Group B-2). The lead acetate (500 ppm) was injected two times for one week into the abdomen and green tea solution (3 g/100 ml distilled water) offered freely. In the liver cell, The cristae of mitochondria were enlarged, the rough endoplasmic reticulum (rER) extended and many lysosome observed on the Group A- 1. The cristae of mitochondria were enlarged and many lysosome observed, but nucleus was normal on the Croup A-2. All of observed parts on the Group B-1 deeply wounded by lead than the group A-1. In the Group B-2, the mitochondria and rER were increased in number than the Group B-1 and the nucleus was observed normally.

Protective Effect of Saengshik Supplementation on Lead Induced Toxicity in Rats (생식의 섭취가 납중독 흰쥐의 피해 경감에 미치는 영향에 관한 연구)

  • Lead is a ubiquitous environmental and industrial pollutant that causes a major health concerns. It is known to induce a broad range of physiological, biochemical, and behavioral dysfunctions in laboratory and humans, including hematopoietic system, kidneys, liver, and reproductive system. This study was conducted to investigate the effect of Saengshik supplementation on the lead-induced toxicity in rats. Five week old male Sprague­Dawley rats were randomly assigned to five groups for six weeks as followings: control group (CT), lead acetate treated group (PT), and lead acetate groups administered with three different dosages of Saengshik $(SI2.5-12.5\%,\;S25-25\%,\;and\;S50-50\%).$ Lead acetate (12 mg/rat) was intragastrically administered daily for 6 weeks. The results were summarized as follows; Weight gain and food efficiency ratio were significantly lower (p<0.05) in lead administered group compared with those of the control group. Also, significant lead-induced alteration in blood hemoglobin (HGB), hematocrit (HCT), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), and reticulocyte distribution width (RDW) were observed. In the liver of lead-exposed animals, there was an increase in the lipid peroxidation (MDA) and the level of glutathione (GSH), but superoxiede dismutase (SOD) activity did not change. Lead-exposed animals with $25\%\;and\;50\%$ Saengshik supplementation showed marked improvements in the values of MCH, MCV, and RDW. Also, the level of HCT was significantly increased by $50\%$ Saengshik supplementation. The levels of liver MDA in $12.5\%\;and\;50\%$ Saengshik administered groups and GSH level in $50\%$ Saengshik administered group were significantly decreased compared to the lead administered group. Also, hepatic SOD activity tended to increase in the presence of Saengshik supplementation. Furthermore, the accumulation of lead in liver and kidney was reduced by presence of Saneghshik supplementation. Liver lead concentration was significantly reduced by both $25\%\;and\;50\%$ Saengshik supplementations and kidney lead concentration was significantly reduced by the $25\%$ Saengshik supplementation. These results show that Saengshik may have a protective effect against lead intoxication but the mechanism of their effects remains unclear.

Effect of lead on mitochondrial activity in rat tussues (랫드의 조직 mitochondria 활성에 대한 납 투여의 영향)

  • Cardiac, hepatic and renal mitochondria in rats fed lead containing diets were isolated and their activities were studied in terms of NADH oxidation. In normal rats, cardiac and renal mitochondria had similar activities and showed activity values of higher than those in hepatic mitochondria. Cardiac mitochondiral activities in rats fed lead containing diets were increased after 4 weeks of feeding but decreased to activity values close to normal. Renal mitochondrial activities showed a trend of inhibition in all groups fed lead containing diets but were no differenes by feeding periods of 4 and 8 weeks. Feeding of lead containing diets could not be attributed to any changes in the hepatic mitochondrial antivities at experimental doses during 4~8 weeks.

Effects of Lespedeza Caneata ethanol extract on the Liver, Kidneys of Lead administered Mice (야관문(Lespedeza Caneata) Ethanol 추출물이 납 투여한 생쥐의 간장과 콩팥에 미치는 영향)

  • This study determined the effect of Lespedeza Caneata extract on the livers and kidneys of lead-administered mice. The experimental groups were divided into a normal group, Pb 4W group, Pb-LC 4W group, Pb 8W group, and Pb-LC 8W group. The normal group was supplied single distilled water, and the pb group was provided distilled water in which lead acetate was dissolved at 1,000 ppm. The Pb-LC group was provided with lead as drinking water, and the Lespedeza Caneata was orally medicated at a concentration of 500 mg/kg daily. AST, ALT, and BUN enzyme activities and histological experiments on the livers and kidneys resulted in the following conclusions. AST, ALT, and BUN activities increased in the experimental group compared to the normal group and decreased more in the Pb-LC group than the pb group during the same period. The histological results reveal that portions of the livers and kidneys were deformed in the Pb 4W group, and most of the Pb 8W group experienced necrosis and deformation. pb-LC4W and Pb-LC 8W groups experienced less deformation than the Pb 4W and Pb 8W groups. During the same period, the Pb-LC group experienced less histological changes than the Pb group. These results suggest that Lespedeza Caneata extract may have some protective effect on hepatic tissue and renal tissue damage with lead-administered in mice.

Effects of Chitosan on the Rat Nephrotoxicity Induced by Lead (납으로 유발된 흰쥐 신장 독성에 대한 키토산의 효과)

  • The purpose of this study was to evaluate the effect the lead toxicity of chitosan on the rats. A healthy male of Wistar rat was used for experiment. The lead acetate (30 mg/kg, Pb) was injected two times per a week into the abdomen. 0.1% chitosan solution (1 mg/ml) was supplied by feeding ad libitum. After 4 or 8 weeks, we was observed kidney by electron microscope. The experimental groups were divided into two groups. Group 1 was control group which was not treated with chitosan solution after contaminatation of lead acetate. Group 2 was experimental group which was treated with chitosan solution after contamination of lead acetate. All groups were used to 10 rats. The results were as follow: On the proximal convoluted tubules of the group with only Pb, the number of microvillis were decreased and the number of lysosomes were increased on the cytoplasm. The mitochondria and rough endoplasmic reticulum were extended and ribosomes dropped from the rough endoplasmic reticulum. On the proximal convoluted tubules of the group with Pb-chitosan, the shapes and number of microvilli were not changed and the number of lysosomes were decreased. The shapes of mitochondria and endoplasmic reticulum observed not changes. We concluded that chitosan has effect on reducing toxicity of lead in the rat kidney.

  • Vinyl acetate monomer can be polymerized through bulk, solution, emulsion, and suspension polymerization processes. However, in the preparation of PVA from bulk or solution polymerization, there are several technical limitations for obtaining high yield and high molecular weight simultaneously. Thus, the improvement of polymerization method is necessary to prepare the PVA with high yield and high molecular weight because that the difficulty in control of high viscosity and in removal of the heat of polymerization, which might lead to side reactions like branching.   • This study was done to determine the effect of lead acetate on the activities of the hepatic cytosofic xanthine oxidase and aldehyde oxidase which were well known as oxygen free radical generating enzyme in vitro. Lead ion accelerated the formation of lipid peroxide and the increment of xanthine oxidase(type O) activity and the type conversion ratio from xanthine dehydrogenase to xanthine oxidase dose-dependently. But xanthine dehydrogenase(type D) activity was decreased. Aldehyde oxidase activity was not changed by lead ion. These data suggested that lead-induced cellular to)dcity may be concerned partially with xanthine oxidase mediated lipid peroxidation.

Morphological changes of Schwann cells as neurotoxic responses (신경독성에 의한 Schwann 세포의 형태적 변화)

  • The early change observed in lead-induced neurophathy in the rat was Schwann cell swelling. In order to quantify this cell swelling, Schwann cell thickness and major diameter of the nucleus were measured using tranverse section with associated myelinated fiber of sciatic nerves. Group I rats were intoxicated with 0.5% lead acetate in the drinking water for 30 days; group II animals were treated as in group I and then restored to normal laboratory conditions for 30 days; and group III were controls. The results showed that the cell sizes were significantly greater in intoxicated animals, compared with control, and the cell sizes of group II did not differ significantly from control rats.
