Photochemical/Biophysical Properties of Proteorhodopsin and Anabaena Sensory Rhodopsin in Various Physical Environments (막 단백질인 Proteorhodopsin과 Anabaena Sensory Rhodopsin의 다양한 측정 환경에 따른 광화학/생물리학적 특성)

  • Rhodopsin is a membrane protein with seven transmembrane region which contains a retinal as its chromophore. Although there have been recently reports on various photo-biochemical features of rhodopsins by a wide range of purifying and measurement methods, there was no actual comparison related to the difference of biochemical characteristics according to their physical environment of rhodopsins. First, proteorhodopsin (PR) was found in marine proteobacteria whose function is known for pumping proton using light energy. Second one is Anabaena sensory rhodopsin (Nostoc sp.) PCC7120 (ASR) which belongs to eubacteria acts as sensory regulator since it is co-expressed with transducer 14 kDa in an operon. In this study, we applied two types of rhodopsins (PR and ASR) to various environmental conditions such as in Escherichia coli membranes, membrane in acrylamide gel, in DDM (n-dodecyl-${\beta}$-D-maltopyranoside), OG (octyl-${\beta}$-D-glucopyranoside), and reconstituted with DOPC (1,2-didecanoyl-sn-glycero-3-phosphocholine). According to the light-induced difference spectroscopy, rhodopsins in 0.02% DDM clearly showed photointermediates like M, and O states which respond to the different wavelengths, respectively and showed the best signal/noise ratio. The laser-induced difference spectra showed the fast formation and decay rate of photointermediates in the DDM solubilized samples than gel encapsulated rhodopsin. Each of rhodopsins seemed to be adapted to its surrounding environment.

VISUALIZATION AND MEASUREMENT OF A NARROW-CONE DI GASOLINE SPRAY FOR THE IMPINGEMENT ANALYSIS

  • Wall interactions of direct injection spray were investigated using laser-sheet imaging, shadowgraphy, wetted footprint and phase Doppler interferometry techniques. A narrow-cone high-pressure swirl injector is used to inject iso-octane fuel onto a plate, which has three different impact angles inside a pressurized chamber. Heated air and plate conditions were compared with unheated cases. Injection interval was also varied in the heated case to compare dry- and wet- wall impingement behaviors. High-speed macroscopic Mie-scattering images showed that presence of wall and air temperature has only minor effect on the bulk spray structure and penetration speed for the narrow-cone injector tested. The overall bulk motions of the spray plume and its spatial position at a given time are basically unaffected until a few millimeters before impacting the wall. The surface properties of the impact surface, such as the temperature, the presence of a preexisting liquid film also have a small effect on the amount of wetting or the wetted footprint; however, they have strong influence on what occurs just after impact or after a film is formed. The shadowgraph in particular shows that the plate temperature has a significant effect on vapor phase propagation. Generally, 10-20% faster horizontal vapor phase propagation is observed along the wall at elevated temperature condition. For impingement onto a preexisting film, more splash and evaporation were also observed. Contrary to some preconceptions, there is no significant splashing and droplet rebounding from surfaces that are interposed in the path of the DI gasoline spray, especially for the oblique impact angle cases. There also appears to be a dense spray front consists of large sac spray droplets in the oblique impact angle cases. The bulk of the spray is not impacted on the surface, but rather is deflected by it The microscopic details as depicted by phase Doppler measurements show that the outcome of the droplet impaction events can be significantly influenced. Only droplets at the spray front have high enough Weber numbers for wall impact to wet, splash or rebound. Using the sign of vertical velocity, the time-resolved downward droplets and upward droplets are compared. The Weber number of upward moving droplets, which seldom exceeds unity, also decreases as the impact angle decreases, as the droplets tend to impact less and move along the wall in the deflected spray plume.

Application Study of Dry-ice Pellet Cleaning for Removing Oil Paint and Lacquer of Outdoor Metal Artifacts (Dry-ice Pellet Cleaning 적용 옥외 금속문화재 표면 페인트 및 유성물질 제거방법 비교 연구)

  • Damage of cultural properties happens variously. Particularly, the surface damage of metal cultural properties exposed outdoors lowers historical and art historical value of artifact by artificial damage such as paint, scribbling containing oily material and so on. Therefore, this study compared dry-ice pellet cleaning with poultice when clearing paint and oily material environment-friendly, harmlessly to humans and without damage of artifact. As the result of experiment, when clearing those (paint and oily material) by poultice, oily paint was cleared, but there were spots of metal surface. Also, Lacquer spray wasn't well cleared, and resin came off the surface of artifact. When clearing those by dry-ice pellet cleaning, oil paint was cleanly cleared without surface damage of artifact according to Stereoscopic microscope observation, color-measurement, FT-IR, SEM analysis. Also, lacquer spray seemed to be cleared with the naked eye, but there were minute particles on surface according to the result of SEM observation. Consequently, we could confirm possibility of dry-ice pellet cleaning substitution.

Second Harmonic Rotational Anisotropy of Polycrystalline Fe Films on Glass Substrates (유리 위에 증착된 다결정 Fe 자성박막의 이차조화파 회전 이방성)

  • The surface structure of polycrystalline Fe films of various thicknesses on glass substrates have been studied using a Ti: Sapphire laser at 780 nm. We found that the surface structure possesses C$_s$ crystal structure close to $C_{2v}$ through symmetry consideration. We present one-fold intensity profile with one mirror plane in second harmonic (SH) intensity as a proof of $C_s$ symmetry. $C_s$ and $C_{2v}$ surface symmetries usually appear at the (110) surface of a cubic diamond single crystal [1]. Therefore this surface symmetry would be related to bcc (110) growth orientation coinciding with XRD measurement. Further we treated surface normalized SH asymmetry with various thicknesses. The SH asymmetry increases with increasing of film thickness. From these results, it is observed that the surface structure of thin polycrystalline Fe film below 5 nm is almost isotropic, while in the case of the thicker Fe films, surface structure have $C_s$ symmetry structure close to $C_{2v}$.

Optical Characterizations of TlBr Single Crystals for Radiation Detection Applications

  • Background: TlBr is of considerable technological importance for radiation detection applications where detecting high-energy photons such as X-rays and ${\gamma}$-rays are of prime importance. However, there were few reports on investigating optical properties of TlBr itself for deeper understandings of this material and for making better radiation detection devices. Thus, in this paper, we report on the optical characterizations of TlBr single crystals. Spectroscopic ellipsometry (SE) and photoluminescence (PL) measurements at RT were performed for this work. Materials and Methods: A 2-inch TlBr single crystalline ingot was grown by using the vertical Bridgman furnace. SE measurements were performed at RT within the photon energy range from 1.1 to 6.5 eV. PL measurements were performed at RT by using a home-made PL system equipped with a 266 nm-laser and a spectrometer. Results and Discussion: Dielectric responses from SE analysis were shown to be slightly different among the different samples possibly due to the different structural/optical properties. Also from the PL measurements, it was observed that the peak intensities of the middle samples were significantly higher than those of the other two samples. With the given values for permittivity of free space (${\varepsilon}_0=8.854{\times}10^{-12}F{\cdot}m^{-1}$), thickness (d = 1 mm), and area ($A=10{\times}10mm^2$) of the TlBr sample, capacitances of TlBr were 6.9 pF (at $h{\nu}=3eV$) and 4.4 pF (at $h{\nu}=6eV$), respectively. Conclusion: SE and PL measurement and analysis were performed to characterize TlBr samples from the optical perspective. It was observed that dielectric responses of different TlBr samples were slightly different due to the different material properties. PL measurements showed that the middle sample exhibited much stronger PL emission peaks due to the better material quality. From the SE analysis, optical, dielectric constants were extracted, and calculated capacitances were in the few pF range.

  • The purpose of this study was to measure the changes of regional cerebral blood flow (rCBF) and blood pressure (BP) in rats, following the intravenous injection of Schizandrae fructus water extract. The measurement was continually monitored by laser-doppler flowmeter and pressure tranducer in anesthetized adult Sprague-Dawley rats for 2 hours to 2 hours and a half through the data acquisition system composed of MacLab and Macintosh computer. The result of this experiment was as followed. Schizandrae fructus increased the changes of rCBF in rats significantly. The rCBF of Schizandrae fructus did not change by pretreated propranolol, atropine, L-NNA and indomethacin. But the rCBF of Schizandrae fructus was increased by pretreated methylene blue. Schizandrae fructus decreased the changes of BP, significantly. The BP of Schizandrae fructus did not change by pretreated propranolol, atropine, L-NNA and indomethacin. But the BP of Schizandrae fructus was decreased by pretreated methylene blue. There results indicated that Schizandrae fructus can increase the rCBF and decrease the BP, that is related to guanylyl cyclase activity.

A Study on the Effect of Large Coherent Structures to the Skin Friction by POD Analysis (적합직교분해(POD)기법을 사용한 난류 응집구조 거동에 관한 연구)

  • An experimental study in a recirculating water channel was carried out to investigate the effect of large coherent structures to the skin friction on a flat plate. Particle Image Velocimetry (PIV) technique was used to quantify characteristic features of coherent structures growing to the boundary layer. In the PIV measurement, it is difficult to calculate the friction velocity near the wall region due to laser deflection and uncertainty so that Clauser fitting method at the logarithmic region was adopted to compute the friction velocity and compared with the one directly measured by the dynamometer. With changing the free-stream velocity from 0.5 m/s to 1.0 m/s, the activity of coherent structures in the logarithmic region was increased over three times in terms of Reynolds stress. The flow field was separated by Variable Interval Time Averaging (VITA) technique into the weak and the strong structure case depending on the existence large coherent structures in order to validate its effectiveness. The stream-wise velocity fluctuation was scanned through at the boundary thickness whether it had a large deviation from background flow. With coherent structures connected from near-wall to the boundary layer, mean wall shear stress was higher than that of weak structure case. Proper Orthogonal Decomposition (POD) analysis was also applied to compare the energy budget between them at each free-stream velocity.

  • In general, auto parts production assembly line is assembled and produced by automatic mounting by an automated robot. In such a production site, quality problems such as misalignment of parts (doors, trunks, roofs, etc.) to be assembled with the vehicle body or collision between assembly robots and components are often caused. In order to solve such a problem, the quality of parts is manually inspected by using mechanical jig devices outside the automated production line. Automotive inspection technology is the most commonly used field of vision, which includes surface inspection such as mounting hole spacing and defect detection, body panel dents and bends. It is used for guiding, providing location information to the robot controller to adjust the robot's path to improve process productivity and manufacturing flexibility. The most difficult weighing and measuring technology is to calibrate the surface analysis and position and characteristics between parts by storing images of the part to be measured that enters the camera's field of view mounted on the side or top of the part. The problem of the machine vision device applied to the automobile production line is that the lighting conditions inside the factory are severely changed due to various weather changes such as morning-evening, rainy days and sunny days through the exterior window of the assembly production plant. In addition, since the material of the vehicle body parts is a steel sheet, the reflection of light is very severe, which causes a problem in that the quality of the captured image is greatly changed even with a small light change. In this study, the distance between the car body and the door part and the door are acquired by the measuring device combining the laser slit light source and the LED pattern light source. The result is transferred to the joint robot for assembling parts at the optimum position between parts, and the assembly is done at the optimal position by changing the angle and step.

A study on the effects of polymer core gate sizes on thickness shrinkage rate (폴리머코어 게이트 크기 변화가 두께 방향 수축률에 미치는 영향에 대한 연구)

  • In this study, the variation of the shrinkage in the thickness direction of the molded parts according to the gate size of the polymer core fabricated through the 3D printer using the SLS method was studied. The polymer cores are laser sintered and the powder material is nylon base PA2200. The polymer cores have lower heat transfer rate and rigidity than the metal core due to the characteristics of the material. Therefore, the injection molding test conditions are set to minimize the deformation of the core during the injection process. The resin used in the injection molding test is a PP material. The packing condition was set to 80, 90 and 100% of the maximum injection pressure for each gate size. The runner diameter used was ∅3mm, and the gates were fabricated in semicircle shapes with cross sections 1, 2, and 3 ㎟, respectively. Thickness measurement was performed for 10 points at 2.5 mm intervals from the point 2.5 mm away from the gate, and the shrinkage to thickness was measured for each point. The shrinkage rate according to the gate size tends to decrease as the cross-sectional area decreases as the maximum injection pressure increases. The average thickness shrinkage rate was close to 0% when the packing pressure was 90% for the gate area of 1mm2. When the holding pressure was set to 100%, the shrinkage was found to decrease by 3% from the standard dimension due to the over-packing phenomenon. Therefore, the smaller the gate, the more closely the molded dimensions can be molded due to the high pressure generation. It was confirmed that precise packing process control is necessary because over-packing phenomenon may occur.

Diurnal fluctuations of vision in myopes (근시안의 일일 시럭변동에 관한 연구)

  • Subjective and objective visions were measured on young adults(mean 21 yrs, 126 eyes) who were free of any ocular diseases and laser surgery and none wore contact lenses. The aim of this study was to investigate the diurnal variation of vision through subjective and objective measurements. Subjective visual acuity were measured at 5 m three times a day, morning(8:00 AM-10:00 AM), noon(12:00 PM-2:00 PM) and afternoon(4:00 PM-6:00 PM). The instrument used for objective refraction right after visual acuity measurement was Nvision-K 5001(shin-nippon) which unique in being able to disregard subject's accommodation because of its unrestricted viewing conditions. Also, we measured that three times and then calculated the average values. The result showed that an average subjective visual acuity in the morning, noon, afternoon were 0.256(${\pm}0.263$), 0.266(${\pm}0.276$), 0.242(${\pm}0.249$) respectively. Average spherical equivalent power in objective refraction of right eyes showed -3.416 D(${\pm}2.907$), -3.359 D(${\pm}2.735$), -3.297 D(${\pm}2.709$) respectively and dioptric power was decreased from morning to afternoon. Vision changed throughout the day in both subjective and objective measurements nevertheless its variations were statistically insignificant(p<0.05). Therefore it does not seem to matter of time for either visual acuity test or refraction.
