Studies on the mechanism in the induced to unfertilized eggs(male sterility of Silkworm) by protected environment during pupae period (용기의 보호환경에 따른 불수정란(웅성불임잠)의 유발기구에 관한 조사연구)

  • In order to find out effects of the generative power of silk worm moth which have been brought up in the high temperature accommodation at their pupa stage. For this specific study, 9 different kinds of male silk worms have been selected as specimen. All those specimen were brought up in the normal temperature at their larvae stage and after the pupation period they have been accommodated in the condition of high temperature for a certain length in accordance with the study programme. Afterwards, those mlae specimen were copulated with Suwon jam 103${\times}$104 which were all brought up in normal conditions. This study was carried out to find the copulation function as well as the ratio of unfertilized eggs(male sterility test). Results of study have been revealed as follows: 1) Although some differences were observed, male pura which have been brought up in the condition of high temperature shown the low rate of unfertilized eggs rather than those were brought up in the normal conpition. 2) In this group the eclosion(emergency) has been found to be poor rather, than those specimen brought up in normal conditions. 3) The copulation function of Moran, Daedong, J124 and C108 specimen were found to be poor than those of Suwon jam. 4) Fertility rate of Moran, Daedong, J124 and C108 was found to be around 65%. This figure is rather lower than what we normally expect. 5) Unfertilized egg ratio of Moran, Daedong and C108 were found to be around 20% if they were brought up in the condition of high temperature for one day from the time of pupation: 40% at 2 days, and 70% at 3 days duration. More than 3 days treatment has shown no progress in the unfertilized egg ratio. 6) One day's treatment for the pupa at the later stage has shown the unfertilized egg ratio of about 10%; 20% at 2 day's treatment, 35% at 3 day's treatment, 40-60% at 4 day's treatment, more than 60% at 5 day's treatment, and the 70% of fertilized egg ratio was only observed when the treatment days come to 7 days. It was understood that the unfertilized egg ratio was high at the antepupa stage rather than that of post-pupa stage. 7) According to the result of observation the sperm in copulatory pouch and seminal receptacle out of the normal female silk worm which have been copulated with the male brought up in the condition of high temperature at their moth stage. The reproduction system found in the control zone has been found to be normal and the sperm is amountful and active in motion while the sperm found in the treatment to be limited in amount and slow in motion. The observation was made within 5 hours from the copulation. 8) According to the result of observation of sperm of seminal receptacles of the female silkworm moth, and according to that observation of sperm in the seminal receptacle in female silkworm moth, the amount of sperm and mobility in the female moth brought up in high temperature were poor comparing that were brought up in normal temperature zone. Some of them are even found to be no trace of such. 9) Appearance and mosle of the copulatory organ of the male silkworm moth was found to be no anatomical change. 10) Testis of the later pupa stage which was brought up in the high temperature was found to be almost net developed to anucleate sperm and they are degenerated at stage of between maturation division and sperm abnormal stage. Mean time at control zone, the formation of anucleate sperm was already observed.

Comparison of Physicochemical Properties and Analysis of sEquence Structure Relationships of Commercial Dongchongxiacao of Three Species in Korean Market

  • To compare the quality of manufactured goods distributed in the domestic markets, 6 isolates of Dongchongxiacao products, namely, 4 Paecilomyces tenuipes specimens (J2P, 901A, 901B, and 901C), 1 Cordyceps militaris specimen (901D), and 1 Cordyceps sinensis specimen (CI-1), were analyzed for their physicochemical properties and sequence-structure relationships. P. tenuipes (J2P), a kind of Dongchongxiacao, was successfully inoculated on silkworms by percutaneous infection of Rural Development AdminstraionNam et al., 1999); fruiting bodies were then formed on the complete surface of the pupa. Since P. tenuipes (J2P) from silkworm larva was also proved to have remarkable pharmacological activities, it has been produced in bulk and has been successfully sold to buyers in the Korean market. Additionally, imitation products such as 901A, 901B, 901C, 901D, and CI-1 were sold simultaneously, resulting in deterioration of product quality. This research focuses on establishing quality standards to discriminate between the original and imitation products circulating in the market. The products obtained for the experiments included J2P, 901A, 901B, 901C, 901D, and CI-1; proximate analysis was performed for these products. The hosts and methods of conidia inoculation for proliferation of mycelia differed among the products. P. tenuipes (J2P) was proliferated in live silkworm larvae, and dead silkworm pupae were used to produce 901A, 901B, and 901C. On the other hand, 901D was produced on hulled rice medium. Quality analysis of C. sinensis revealed that CI-1, which was imported from China, smelled bad and proved to be a counterfeit with the fruiting body glued to the insect by twigs. The results of the proximate analysis of 901A, 901B, 901C, and 901D were similar to those of J2P with respect to the moisture content. Otherwise, J2P contained higher crude protein than 901A, 901B, 901C, and 901D, but contained very low fat. C. militaris (901D) and C. sinensis (CI-1) had low crude protein content-12.79% and 9.78% respectively-as compared to that of J2P, which was 62.38%. In contrast to the crude ash content of 6.4% in J2P, the crude ash content of CI-1 was 18.51% and this specimen was found to contain many impurities. phylogenetic analysis of P. tenuipes revealed that the sequence similarity of J2P, 901B, and 901C was in the range of 92.3~92.7%. Additionally, differences in the sequences were found at the positions 65 bp, 436 bp, 441 bp, 463 bp, etc.

  • Gametogenes, reproductive cycle, first sexual maturity(biological minimum size), sex ratio and larval development of the marsh clam Corbicula japonica were investigated monthly by histological observations. Samples were collected in brackish water of Gokgang stream, Kyungsangbuk-Do, Korea, from August 1997 to July 1998. Sexuality of Corbicula japonica is dioecious and the species are an oviparous clam. The gonads are irregularly arranged from the sub-region of mid-intestinal gland in visceral cavity to reticular connective tissue of foot. The ovary is composed of a number of ovarian sac which are branched arborescent. Oogonia actively proliferate along the germinal epithelium of ovarian sac, in which young oocytes are growing. The testis is composed of a number of testicular tubules, and the epithelium of the tubule has function of germinal epithelium, along which spermatogonia actively proliferate. A great number of undifferentiated mesenchymal tissue and eosinophilic granular cells are abundantly distributed between developing oocytes and spermatocytes in the early developmental stages. With the further development of the ovary and testis these tissue and cells gradually disappear. Then the undifferentiated mesenchymal tissue and eosinophilic granular cells are considered to be related to the growing of the oocytes and spermatocytes. The spawning period is from July to September, and the main spawning occur between July and August when seawater temperatures reach above 22$^{\circ}C$. The reproductive cycle of this species can be divided into five successive stages; early active (February to April), late active (May to July), ripe (June to September), partially spawned (July to September), degenerative (September to October) and resting stage (October to February). Percentages of first sexual maturity of female and male clams ranging in length from 10 mm to 12 mm are over 50% and 100% for clams over 16.0 mm in shell length. Fertilized eggs or Corbicula japonica were 80-90 ${\mu}{\textrm}{m}$ in diameter. In the early embryonic development of C. japonica, the appearance of polar body, trochophore and D-shaped veliger were observed around 40 min., 27 hours and 4 days after spawning, respectively, at a water temperature of 26.5-28.$0^{\circ}C$. The size of larvae of early umbo stage was about 185-210 ${\mu}{\textrm}{m}$ in shell length, 160-180 ${\mu}{\textrm}{m}$ in shell height around 7 days after fertilization. The correlation of relative growth between the culture day (D) and shell length (SL) was expressed by the following simple formula from D-shaped veliger to metamorphosing stage; SL = 13.300D + 209.36($r^2$= 0.9078).

  • Studies were made on the morphology and life cycle of the mite, Pyemotes tritici $Lagr{\acute{e}}z$-Fossot & $Montagn{\acute{e}}$ (Trombidiformes; Pyemotidea) ,parasitic on larvae of cigarette beetle, Lasioderma serricorne F., which is one of the most serious pests of stored tobacco, cigar, and cigarette in Korea. One generation time was $20.9{\pm}0.7$ days out of which $9.5{\pm}0.3$ days were spent for feeding and $10.3{\pm}0.8$ days for reproduction of progenies. A female of this insect-parasitic mite produced $56.7{\pm}6.9$ progenies during her reproduction period. The body size of a newly-laid male or female was $280{\mu}m$ long and $85{\mu}m$ wide. As female of this mite sucked on, their abdomen grew larger and larger to reach $825{\mu}m$ in width and $0.346mm^3$ in volume after $9{\sim}10$ days. By sucking the humor of a host, the abdomen of a female mite became almost a global shape in two days. The increase rate of abdominal width was the biggest on the second or the third day of feeding while the volume of abdomen reached to the largest on $6{\sim}8$ days after feeding. The largest number of the daily young produced on 4-th day after a female began to reproduce.

Environment-Friendly Control of Beet Armyworm, Spodoptera exigua (Noctuidae: Lepidoptera) to Reduce Insecticide Use (농약 사용 저감화를 위한 환경 친화적인 파밤나방(Spodoptera exigua)의 방제)

  • For the environment-friendly control of beet armyworm, Spodoptera exigua, in spring onion fields, we have examined an alternative application method. Twenty-five insecticides registered for spring onion were tested for control effect against the beet armyworm in the laboratory, then the best 9 chemical and a single biological insecticides were selected and compared with 2 new isolates of Bacillus thuringiensis in a polyvinyl house. After that, 4 chemical and 3 B. thuringiensis treatments were used in the field for the control of beet armyworm in the spring onion. Two application methods are used: one is triple treatments with the same chemical and the other is alternative application with different chemicals and B. thuringiensis for 7 days intervals. Indoxacarb WP - chlorfluazuron EC - B.t. var. kurstaki CAB141 and indoxacarb WP - methoxyfenozide + spinosad SC - Chlorfluazuron EC - B.t. var. aizawai CAB109, B.t. var. kurstaki CAB141 showed greater than 78% mortality of beet armyworm larvae and greater than 43% damage decrease in spring onions infested by beet armyworm. These results showed that alternative applications had higher control effect than any other applications. It was suggested that alternative applications with microbial biological agents such as B. thuringiensis might minimize the development of insecticide resistance and might be used as the environment-friendly control of the beet armyworm.

Ecological Characteristics of Cotesia glomerata L. (Hymenoptera: Braconidae) and Its Parasitism Rates for Diamondback Moth (Plutella xylostella L.) in a Kimchi Cabbage Field in The Korean Highland Area (배추나비고치벌(Cotesia glomerata L.)의 생태적 특성 및 고랭지 배추밭에서 배추좀나방(Plutella xylostella L.)에 대한 기생률)

  • Cotesia glomerata L., an internal parasitoid wasp, attacks the larvae of both the cabbage white butterfly (Artogeia rapae L.) and the diamondback moth (Plutella xylostella L.). It can be utilized as a natural biological enemy to control these two insect pests in the summer cabbage fields of the Korean highland areas. The developmental response and sex ratio of C. glomerata to various temperatures and its longevity were examined in the laboratory. The egg-to-larva and pupa stages of C. glomerata were 12.1 ± 2.1 and 6.4 ± 1.8 days, respectively, at 20℃, The developmental threshold for egg-to-larva and pupa stages were 7.7 and 8.5℃, respectively. The sex ratios of C. glomerata when reared under various temperatures were 61.0 ± 4.5% at 15℃, 44.2 ± 1.0% at 20℃, and 39.0 ± 2.3% at 25℃, and the incidence of females increased as the temperature decreased. The longevity of C. glomerata when fed a 10% sugar solution was 20.4 ± 0.2 days, while in adults without any feed, the longevity was 3.6 ± 0.1 days. Indoor reared C. glomerata adults were released into cabbage fields from 2007 to 2018, in early August of each year, and the outdoor parasitism rates were surveyed. The parasitism rates were found to increase gradually as the year passed (Y = 0.2696X + 2.8633, R2 = 0.3994). The highest parasitism rate was observed in 2013 at 7.6%, and the lowest was in 2018 at 6.5 %. These results could be used as basic information for biological control of kimchi cabbage pests at highland fields.

Bionomics and a Lana Sampling Method of Coppery Wireworm, Selatosomus puncticollis (Motschulsky), (Coleoptera: Elateridae) in Potato Field (청동방아벌레(Selatosomus puncticollis Motschulsky)의 생태적 특성 및 감자포장내 유충밀도 조사법)

  • The occurrence pattern and bionomics of Selatosomus puncticollis (Motschulsky) were surveyed in three net house (6$\times$70m) in the field and in a laboratory ($20^{\circ}C$, RH$75\%$, L16/DB). Emergence of hibernated adults of S. puncricollis started from early May, reached peak at early June and diminished in late June. Occurrence rate to total number was $42.1\%$ in early June and $31.7\%$ in late May, when most of hibernated adults emerged at these periods. Adults started to oviposit from mid June, and eggs hatched from early July Larvae turned into pupae from mid July and emerged to adults from mid August. Egg-period was 23 days and pupal period was 21 days. It is estimated that preovipositional period and larval period were approximately 10 months and 30 months, respectively. Wireworms were distributed at the soil depth of 10-5 cm, $56.8\%$ in 1997 and $45.8\%$ in 1998. To establish bait techniques to attract wireworms in the soil, six baits: pieces of potato, carrot, and sweet potato, wheat grains, corns, and flour dough wrapped with gauze, were buried at 15 cm of soil depth, and collected after 5 and 10 days. The numbers of wireworms attracted by potato pieces, wheat grains and sweet potato pieces after 10 days were 1.8, 1.6, and 1.4/bait, respectively. Therefore, burying potato pieces at 15 cm of soil depth and collected after 10 days could be recommended as a wireworm baiting technique.

Construction of a Transgenic Tobacco Expressing a Polydnaviral Cystatin (폴리드나바이러스 유래 시스타틴 유전자 발현 형질전환 담배 제작)

  • CpBV (Cotesia plutellae bracovirus) is a polydnavirus and encodes a cystatin (CpBV-CST1) gene. Its overexpression suppresses insect immunity and alters insect developmental processes. This study aimed to construct a genetically modified (GM) tobacco to further explore the physiological function of the viral cystatin and to apply to control insect pests. To this end, the transgenic tobacco lines were screened in expression of the target gene and assessed in insecticidal activity. A recombinant vector (pBI121-CST) was prepared and used to transform a bacterium, Agrobacterium tumefasciens. The transformed bacteria were used to generate transgenic tobacco lines, which were induced to grow callus and resulted in about 92% of shoot regeneration. The regenerated plants were screened by PCR analysis to confirm the insertion of the target gene in the plant genome. In addition, the expression of the target gene was assessed in the regenerated plants by quantitative real-time PCR (qRT-PCR). The qRT-PCR analysis showed that the transgenic line plant expressed the target gene about 17 times more than the control tobacco, indicating a stable insertion and expression of the target gene in the transgenic tobacco line. The insecticidal activity was then analyzed using the screened transgenic tobacco lines against the teneral 1st instar larvae of the oriental tobacco budworm, Helicoverpa assulta. Though there was a variation in the insecticidal efficacy among transgenic lines, T9 and T12 lines exhibited more than 95% mortality at 7 days after feeding treatment. These results suggest that CpBV-CST1 is a useful genetic resource to be used to generate GM crop against insect pests.

Development of an Emergence Model for Overwintering Eggs of Metcalfa pruinosa (Hemiptera: Flatidae) (미국선녀벌레(Metcalfa pruinosa) (Hemiptera: Flatidae) 월동난 부화 예측 모델 개발)

  • The temperature-dependent development of Metcalfa pruinosa overwintering eggs was investigated at ten constant temperatures (12.5, 15, 17.5, 20, 22.5, 25, 27.5, 30, 32.5, and $35{\pm}1^{\circ}C$, Relative Humidity 20~30%). All individuals collected before April 13, 2012 failed to develop into first instar larvae. In contrast, some individuals that were collected on April 11, 2013 successfully developed when reared under $20{\sim}32.5^{\circ}C$ temperature regimes. The developmental duration was shortest at $30^{\circ}C$ (13.3 days) and longest at $15^{\circ}C$ (49.6 days) in the fourth collected colony (April 26 2013). Developmental duration decreased with increasing temperature up to $30^{\circ}C$ and development was retarded at high-temperature regimes ($32.5^{\circ}C$). The lower developmental threshold was $10.1^{\circ}C$ and the thermal constant required to complete egg overwintering was 252DD. The Lactin 2 model provided the best statistical description of the relationship between temperature and the developmental rate of M. pruinosa overwintering eggs ($r^2=0.99$). The distribution of the developmental completion of overwintering eggs was well described by the 2-parameter Weibull function ($r^2=0.92$) based on the standardized development duration. However, the estimated cumulative 50% spring emergence dates of overwintering eggs were best predicted by poikilotherm rate model combined with the 2-parameter Weibull model (average difference of 1.7days between observed and estimated dates).

Dipteran Comparison on Carcasses by Decomposition at Different Abandoned Site (사체의 유기장소별 부패진행에 따른 발생 파리류 비교)

  • Because insect visitors was different depending on decomposition stage, dipterans occurred on chicken carcasses depending on postmortem intervals were compared by abandoned sites and decomposition from a serial study on arthropod occurrence from carcasses as forensic indicator. Species occurrence and decomposition of carcasses were different depending on abandoned sites (forest hill, open field, stream, greenhouse and roof), seasons (spring, summer, fall and winter), and situation (burying and non-burying). Development of carcass decomposition was faster at all sites in summer, carcass in greenhouse in winter, and unburied carcasses. Although visiting time of blow flies (Calliphoridae) (Phaenicia sericata, Lucilia illustris, Lucilia sp., Chrysomyia pinguis, and Chrysomyia megacephala) was different depending on abandoned season, their adults were generally collected from carcasses within 2 days. However, there were no visited flies at fresh stage of buried carcasses. The flesh flies (Sarcophagidae) were collected from all sites and seasons, and much faster from unburied carcasses than buried carcasses. Those were collected earlier from carcasses in greenhouse than other sites and occurrence was also shorter. In greenhouse, occurrence time of flesh flies were different depending on season; spring and summer - from fresh to active decay stage, fall - fresh to active decay stage, and winter - advanced decay to remains stage. Calliphora lata, Tricerotopyga calliphoroides, and Aldrichana grahami were dominant species and occurrences were different from other flies. These flies were active mainly from fresh to active stage. Larvae of sarcophagid flies were occurred earlier than those of calliphorid flies on buried carcasses.
