Probiotication of Tomato Juice by Lactic Acid Bacteria

  • This study was undertaken to determine the suitability of tomato juice as a raw material for production of probiotic juice by four lactic acid bacteria (Latobacillus acidophilus LA39, Lactobacillus plantarum C3, Lactobacillus casei A4, and Lactobacillus delbrueckii D7). Tomato juice was inoculated with a 24­h-old culture and incubated at $30^{\circ}C$. Changes in pH, acidity, sugar content, and viable cell counts dur­ing fermentation under controlled conditions were measured. The lactic acid cultures reduced the pH to 4.1 or below and increased the acidity to $0.65\%$ or higher, and the viable cell counts (CFU) reached nearly 1.0 to $9.0\times10^9/ml$ after 72 h fermentation. The viable cell counts of the four lactic acid bacteria in the fermented tomato juice ranged from $10^6\;to\;10^8\;CFU/ml$ after 4 weeks of cold storage at $4^{\circ}C$. Pro­biotic tomato juice could serve as a health beverage for vegetarians or consumers who are allergic to dairy products.

Breeding of L(+)-Lactic Acid Producing Strain by Low-Energy Ion Implantation

  • In order to obtain an industrial strain with higher L(+)-lactic acid yield, the wild type strain Rhizopus oryzae PW352 was mutated by means of Nitrogen ions implantation (l5 Kev, $7.8 \times 10^{14} - 2.08 \times 10^{15} ions/Cm^2$ and two mutants RE3303 and RF9052 were isolated. After 36 h shake-flask cultivation, the concentration of L(+)-lactic acid reached 131-136 g/l, the conversion rate of glucose was as high as 86%-90% and the productivity was 3.61 g/l.h. It was almost a 75% increase in lactic acid production compared with the wild type strain. Maximum fermentation temperature of RF9052 was increased to $45^{\circ}C$ from original $36^{\circ}C$. At the same time, the preferred range of fermentation temperature of RF9052 was broadened compared with PW352.

Regulation of Metabolic Flux in Lactobacillus casei for Lactic Acid Production by Overexpressed ldhL Gene with Two-Stage Oxygen Supply Strategy

  • This study describes a novel strategy to regulate the metabolic flux for lactic acid production in Lactobacillus casei. The ldhL gene encoding L-lactate dehydrogenase (L-LDH) was overexpressed in L. casei, and a two-stage oxygen supply strategy (TOS) that maintained a medium oxygen supply level during the early fermentation phase, and a low oxygen supply level in the later phase was carried out. As a consequence, a maximum L-LDH activity of 95.6 U/ml was obtained in the recombinant strain, which was over 4-fold higher than that of the initial strain. Under the TOS for L. casei (pMG-ldhL), the maximum lactic acid concentration of 159.6 g/l was obtained in 36 h, corresponding to a 62.8% increase. The results presented here provide a novel way to regulate the metabolic flux of L. casei for lactic acid production in different fermentation stages, which is available to enhance organic acid production in other strains.

Beneficial Effects of Lactic Acid Bacteria Inoculation on Oat Based Silage in South Korea

  • The objective of the study was to measure the beneficial effects of lactic acid bacteria (LAB) inoculation on the nutritive value of oat silage collected from thirteen regions in the Republic of Korea. The contents of crude protein, acid detergent fiber (ADF), neutral detergent fiber (NDF) and crude ash (CA) were slightly lower in LAB inoculated silage when compared with the control silage, whereas inoculation of LAB resulted in increased total digestible nutrient (TDN). Higher number of LAB, but lower count of yeast and fungi indicated the effectiveness of the LAB inoculation on oat silage fermentation. LAB inoculation resulted in low pH silage, which may prevent undesirable microbial growth. The LAB inoculation promoted lactic acid dominant fermentation with marginal levels of acetic acid and butyric acid in oat silage. These data suggest that the LAB inoculation may preserve oat silage at better quality for ruminant animal production.

  • In this study, noble strains of lactic acid bacteria were isolated and identified by genetic analysis of 16s rRNA. Also, pH-dependent growth curve, cholesterol assimilation ability and sugar production efficiency were measured. Lactic acid bacteria were identified to inhabit in the milks from various animals. Results of sequence analysis showed that there were differences in 16S rRNA sequence among strains and part of gene deletion was also recognized. Growth rates were varied, too, depending on the pH of the medium. Lactobacillus rhamnosus LOCK908 isolated from cow milk showed the highest growth rate and high cholesterol assimilation ability. Results of sugar fermentation tests were relatively consistent with the sequencing results. So, we propose newly isolated Lactobacillus rhamnosus LOCK908 as useful candidate for a starter of fermented beverage and probiotics. Results of this study will contribute to the isolation and identification of noble Lactic acid bacteria and to the public health.

뽕잎의 당침 및 유산발효에 의한 추출물의 기능성 품질 특성 (Functional quality characteristics of extracts by sugar-leaching and lactic acid fermentation of mulberry leaves (Morus alba L.))

  • 이 연구는 뽕잎을 전통방식의 당침 추출액과 Lactobacillus plantarum TO- 2100 유산균을 이용한 유산발효 추출액의 기능성 품질 특성을 조사하였다. 당침 추출액은 12주 동안 추출하였으며, 유산발효 추출액은 8주간 추출하였다. 당침 추출액 및 유산발효 추출액의 수율은 각각 27% 와 166% 였으며, 당 함량은 각각 43 $^{\circ}Brix$와 33 $^{\circ}Brix$였다. pH와 총 산도는 4.6, 0.45% 와 3.6, 1.17%였다. 유산발효 추출액의 유산균 생균수는 $1.2{\times}10^{10}$ $CFU/m{\ell}$으로 당침 추출액의 $2.8{\times}10^2$ $CFU/m{\ell}$ 보다 월등히 많았다. 유산발효 추출액이 amylase, cellulase, pectinase, protease 등 분해효소의 활성을 보인 반면, 당침 추출액은 분해효소의 활성을 나타내지 않았다. 유기산의 경우 malic acid, acetic acid, citric acid의 함량은 당침 추출액이 높은 반면, lactic acid 의 함량은 유산발효 추출액이 2.6배 높았다. 당침 추출액의 유리당은 glucose (200.93mg/g), fructose (236.32mg/g), sucrose (18.41mg/g) 등 GI값이 높은 단당으로 구성된 반면, 유산발효 추출액에서는 유리당이 검출되지 않았다. 기능성 성분인 polyphenol, anthocyanin 및 당질대사 저해물질인 piperidine alkaloid의 함량 또한 유산발효 추출액이 당침 추출액보다 높았다. 당질대사의 주 효소인 ${\alpha}$-glycosidase의 활성을 당침 추출액이 3.4% 저해한 반면 유산발효 추출액은 16.2% 저해 활성을 보였다. 이 결과로부터 전통방식의 당침 추출법에 비해 유산발효 추출법이 뽕잎의 수율 향상 및 기능성 성분의 함량 증진에 더 효과적이며, 이로 인한 대사기능장애의 예방 및 기능증진에 더 유효할 것으로 사료된다.

예비발효 및 압출조리 전처리가 쌀-대두분 혼합액의 유산균 발효에 미치는 영향 (Effects of Prefermentation and Extrusion Cooking on the Lactic Fermentation of Rice-Soybean Based Beverage)

  • 쌀을 기질로 하는 유산균 음료 발효에서 Bacillus와 효모의 혼합배양을 이용한 고체상태 예비발효와 extruder를 이용한 압출조리 전처리가 유산균의 생육을 증진하는 효과에 대하여 검토하였다. Bacillus laevolacticus와 Saccharomyces cerevisiae의 혼합배양을 쌀과 탈지 대두분의 혼합물에 접종하여 고체 상태로 $45^{\circ}C$에서 배양한 후 자가발열형 단일축 압출성형기를 통하여 처리함으로서 살균과 조직의 변화를 도모하였다. 이렇게 처리된 물질을 분산액으로 만들어 Lactobacillus plantarum과 Leuconostoc mesenteroides 혼합배양을 접종하여 유산발효시켰다. 예비발효와 압출조리에 의하여 유산균의 증식속도와 산생성이 증가하였으며 가용성 고형분의 함량이 크게 증가하였고 분산안정성이 향상되었고 관능적 기호도도 증가하였다.

배추의 소금절임시 유기산 첨가가 김치 숙성에 미치는 영향 (Effect of Organic Acids Addition during Salting on the Fermentation of Kimchi)

  • 김치의 초발산도를 조절함으로써 담금초기부터 산미를 부여함과 동시에 보존성을 증대시킬 목적으로 acetic acid(A), citric acid(C), lactic acid(L), A+C+L(ACL) 및 formic acid+acetic acid+fumalic acid+malic acid+citric acid 0.2 - 0.3%를 함유하는 10% 소금물에서 절임하여 김치를 숙성시키면서 품질에 미치는 영향을 조사하였다. 유기산 첨가군중 특히 citric acid 첨가군은 pH의 저하와 산도의 증가율이 낮았으며, 총균수와 젖산 균수도 적었다. 그리고 경도가 가장 높고, 씹힘성은 낮았다. 관능검사 결과에 있어서도 첨가군은 담금 초기부터 적당한 산미를 부여한 반면, 숙성 후기에는 산의 생성량이 적어 타 처리군에 비하여 낮은 산미를 띠었으며, pH, 산도, 조직감 및 관능검사를 통하여 종합적으로 평가한 결과 1$0^{\circ}C$에서의 가식기간이 약 5일 이상 연장되었다.

  • 식품 매개 병원균에 의한 문제는 식품산업뿐 아니라 세계 공공 보건에서도 문제가 된다. 최근 몇 년 간, 발효기술은 식품 내 병원성 미생물의 불활성화 및 이를 조절하기 위한 값 싸고 안전한 방법이라는 것이 밝혀졌다. 유산균 발효는 병원성 세균 및 바이러스에 대해 유의적인 항균효과를 갖는 과학적 증거를 보였다. 유기산, 박테리오신 및 과산화수소와 같은 유산균 대사체는 식품 매개 병원균에 대해 악영향을 미치고 이는 이들의 저해작용으로 이어진다. 이 화합물들은 물리적 결함만을 야기하는 것이 아니라 병원균의 유전자 발현에 대해서도 유의적인 저해 효과를 나타낸다. 게다가, 식품 내 유산균의 존재는 병원균에 대해 영양적인 경쟁을 제공하며 모든 요인이 그 성장을 억제한다. 본 연구는 유산균의 항균력, 분자생물학적 메커니즘 및 식품 매개 병원균의 불활성화를 위한 응용에 대하여 우리의 현 지식을 검토한다.

Effect of Lactic Acid Bacteria Inoculation on Fermentation Characteristics of Whole Crop Barley Silage

  • An experiment was carried out to determine the homofermentative activity of Lactobacillus plantarum KCC-10 and KCC-19 on the ensiling of whole crop barley (WCB). The crude protein in the silages was slightly higher in the KCC-10 and KCC-19 treatments compared to the control, but there was no significant difference between the two inoculant-treated silages. Nutrient parameters such as acid detergent fiber, neutral detergent fiber and in vitro dry matter digestibility in L. plantarum KCC-10 and KCC-19 treated silages did not differ from those in the control silage. The lactic acid content increased in KCC-10 and KCC-19 treated silage when compared with the control silage but the contents of acetic acid and butyric acid produced in KCC-10 and KCC-19 treated silages were similar with the control silage. Further, the number of lactic acid bacteria (LAB) in KCC-10 treated silage demonstrated a significant increase when compared to the control. Especially, KCC-19 treated silage showed greater lactic acid bacterial growth potential. Other microbes such as yeast and fungi were not detected in KCC-10 and KCC-19 treated WCB silages. Hence, this study suggests that the addition of L. Plantarum KCC-10 and KCC-19 to the WCB silage can improve fermentation quality for the production of high-quality silage.
