Objective : The object of this study was to observe the effects of Lonicerae Flos (LF) aqueous extracts on lipopolysaccharide (LPS)-induced rat acute lung injury. Method : Five different dosages of LF extracts were orally administered once a day for 28 days before LPS treatments, and then all rats were sacrificed after 5 hour-treatment of LPS. Eight groups of 16 rats each were used in the present study. The following parameters caused by LPS treatment were observed ; body weights, lung weights, pulmonary transcapillary albumin transit, arterial gas parameters (pH, $PaO_2$ and $PaCO_2$) bronchoalveolar lavage fluid (BALF) protein lactate dehydrogenase (LDH), and proinflammatory cytokines tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$) contents, total cell numbers, neutrophil and alveolar macrophage ratios, lung malondialdehyde (MDA), myeloperoxidase (MPO), proinflammatory cytokines TNF-${\alpha}$ and IL-$1{\beta}$ contents. In addition, the histopathologic changes were observed in the lung in terms of luminal surface of alveolus, thickness of alveolar septum, number of polymorphonuclear neutrophils. Result : As results of LPS-injection, dramatical increases in lung weights, pulmonary transcapillary albumin transit increases, increases in $PaCO_2$, decreases in pH of arterial blood and $PaO_2$, increases of BALF protein, LDH, TNF-${\alpha}$ and IL-$1{\beta}$ contents, total cells, neutrophil and alveolar macrophage ratios, TNF-${\alpha}$ and IL-$1{\beta}$ contents increases were detected with decreases in LSA and increases of alveolar septum and PMNs numbers, respectively as compared with intact control. These are means that acute lung injuries (resembling acute respiratory distress syndrome) are induced by treatment of LPS mediated by inflammatory responses, oxidative stress and related lipid peroxidation in the present study. However, these LPS-induced acute lung injuries were inhibited by 28 days continuous pretreatment of 250 and 500mg/kg of LF extracts. Because of lower three dosages of LF treated groups, 31.25 and 62.5 and 125mg/kg did not showed any favorable effects as compared with LPS control, the effective dosages of LF in LPS-induced acute lung injuries in the present study, is considered as about 125mg/kg. The effects of 250mg/kg of LF extracts showed almost similar effects with ${\alpha}$-lipoic acid 60mg/kg in preventing LPS-induced acute lung injuries. Conclusion : It seems that LF play a role in protecting the acute respiratory distress syndrome caused by LPS.
혈류를 제한한 트레이닝이란, 특수제작 된 공압식 가압벨트를 사용해 사지의 혈류을 제한한 상태에서 저강도로, 단시간 운동하는 것을 말한다. 이 연구의 목적은 만성질환의 발병 위험성이 높은 직장 비만여성을 대상으로, 낮은 강도의 혈류를 제한한 4주간의 걷기트레이닝이 신체조성 및 성장호르몬, 근손상지표에 어떠한 영향을 미치는 지를 구명하고자 한다. 연구대상자는 BMI $25kg/m^2$와 체지방률 30% 이상의 직장 비만여성 11명으로 하였으며 혈류제한은 공압방식의 탄성벨트를 양쪽의 대퇴부위에 착용하고 압력을 조절하였다. 걷기프로그램은 트레드밀을 이용하여 실시하였으며(4주, 주 3일, 1일 2회), 2분 걷기와 1분 휴식으로 구성하여 총 5세트 반복하였다(4km/h, 5%). 이 연구의 결과에서 4주간의 혈류를 제한한 걷기트레이닝 후, 체중과 BMI, 지방량은 유의하게 감소되었으며(p<.05) 체지방률은 감소하는 경향을 보였으나 통계적 유의수준에는 도달하지 못하였다. 성장호르몬 또한 트레이닝 후, 증가하는 경향을 보였으나 통계적 유의차는 나타나지 않았다. 근손상지표 인 CK(p<.05)와 LDH(p<.05), $K^+$(p<.01)의 농도는 트레이닝 후, 유의하게 증가되었으며 Mb의 농도는 증가하는 경향을 보였지만 통계적 유의수준에는 도달하지 못하였다. 이상의 결과에서, 4주간의 혈류를 제한한 걷기트레이닝은 직장 비만여성들의 각종 만성성인질환 및 대사성 질환의 원인인 비만을 예방하고 치료하는데 효과가 있는 것으로 생각되며 이는 짧은 운동시간에도 불구하고 장시간의 운동 효과를 얻을 수 있는 것으로 사료된다.
Objectives : This study investigated anti-inflammatory effects of Nardotidis seu Sulculii Concha water extract (NSCE) against restraint-induced stress. Methods : In vivo, NSCE was orally administered to male white mice at concentrations of 250 mg/kg and 500 mg/kg for 3 days, and then restraint-induced stress was induced for 6 hours. The level of liver damage was measured by serum aspartate aminotransferase (AST), alanine aminotransferase (ALT) and lactate dehydrogenase (LDH). The stress-related hormones such as cortisol and corticosterone were measured by ELISA assay. Also, western blot analysis was performed to detect expression of mitogen-activated protein kinase (MAPK) and cyclooxygenase-2 (COX-2) proteins. Pathological changes were observed by hematoxylin and eosin (H&E) staining of the liver tissue, and Immunohistochemical (IHC) staining was performed to examine liver inflammation through macrophage infiltration. Results : The AST, ALT, LDH and the stress related hormones such as cortisol and corticosterone were significantly decreased in the NSCE treated group compared with stress group. In histological analysis, H&E staining of liver tissues did not detect the hepatic injury or damage in all groups. As a result of IHC staining, it was confirmed that infiltration of macrophages was increased in the stress-induced group, but decreased in the group treated with NSCE. The COX-2 and MAPK proteins expression was significantly increased by restraint-induced stress, but these proteins were decreased in the NSCE treated group. Conclusions : These results suggest that NSCE has the anti-inflammatory activity in restraint-induced stress model, and it is believed that NSCE can be used for the prevention of liver inflammation.

Background: Pirarubicin (THP) is an anthracycline antibiotic used to treat various malignancies in humans. The clinical usefulness of THP is unfortunately limited by its dose-related cardiotoxicity. Ginsenoside F1 (GF1) is a metabolite formed when the ginsenosides Re and Rg1 are hydrolyzed. However, the protective effects and underlying mechanisms of GF1 on THP-induced cardiotoxicity remain unclear. Methods: We investigated the anti-apoptotic and anti-oxidative stress effects of GF1 on an in vitro model, using H9c2 cells stimulated by THP, plus trigonelline or AKT inhibitor imidazoquinoxaline (IMQ), as well as an in vivo model using THP-induced cardiotoxicity in rats. Using an enzyme-linked immunosorbent test, the levels of malondialdehyde (MDA), brain natriuretic peptide (BNP), creatine kinase (CK-MB), cardiac troponin (c-TnT), lactate dehydrogenase (LDH), superoxide dismutase (SOD) and glutathione (GSH) were determined. Nuclear factor (erythroid-derived2)-like 2 (Nrf2) and the expression of Nrf2 target genes, including heme oxygenase-1 (HO-1), glutathione-S-transferase (Gst), glutamate-cysteine ligase modifier subunit (GCLM), and expression levels of AKT/Bcl-2 signaling pathway proteins were detected using Western blot analysis. Results: THP-induced myocardial histopathological damage, electrocardiogram (ECG) abnormalities, and cardiac dysfunction were reduced in vivo by GF1. GF1 also decreased MDA, BNP, CK-MB, c-TnT, and LDH levels in the serum, while raising SOD and GSH levels. GF1 boosted Nrf2 nuclear translocation and Nrf2 target gene expression, including HO-1, Gst, and GCLM. Furthermore, GF1 regulated apoptosis by activating AKT/Bcl-2 signaling pathways. Employing Nrf2 inhibitor trigonelline and AKT inhibitor IMQ revealed that GF1 lacked antioxidant and anti-apoptotic effects. Conclusion: In conclusion, GF1 was found to alleviate THP-induced cardiotoxicity via modulating Nrf2 and AKT/Bcl-2 signaling pathways, ultimately alleviating myocardial oxidative stress and apoptosis.
패혈증은 병원성 감염에 의해 여러 장기에 나타나는 전신성 염증 반응으로, 현재로서는 유망한 치료제가 없다. Signal transducer and activator of transcription 3 (STAT3)은 세포 신호전달 전사 인자로서 항염증 및 염증 반응과 관련된 다양한 세포의 생물학적 과정에서 중요한 역할을 한다. Niclosamide는 FDA에서 승인된 구충제로 STAT3 조절에 관여한다고 알려져 있다. C57BL/6 마우스에 복강 주사로 지질 다당체 (lipopolysaccharide, LPS)를 투여해 패혈증을 유발하였고, Niclosamide를 LPS 주사 2시간 후에 경구 투여하였다. 본 연구에서 Niclosamide가 LPS로 유발된 패혈증 모델의 생존률과 폐 손상을 완화시켰고, 혈청 내 interleukin (IL)-6, 종양괴사인자(tumor necrosis factor-α, TNF-α), IL-1β, AST, ALT, LDH 수치를 유의하게 감소시켰다. 또한 폐 조직 면역 블롯을 통해 PI3K, AKT, NF-κB, STAT3 신호 전달 경로가 Niclosamide에 의해 조절되는 것을 확인하였다. Niclosamide는 LPS를 자극한 RAW 264.7 세포주에서 IL-6, TNF-α, IL-1β와 같은 염증성 사이토카인의 발현을 감소시켰으며, 또한 STAT3의 인산화를 감소시켰다. 본 연구를 통해 Niclosamide에 의한 STAT3 조절이 염증 반응을 억제함으로써 패혈증 모델에 대한 새로운 치료 전략을 제시하였다.
본 연구에서는 식물 추출물 혼합 분말의 근육세포 분화 및 산화적 스트레스에 대한 세포 보호 효과를 확인하고자 하였다. 추출물을 최대 1,000 ㎍/mL 농도까지 세포에 처리한 결과, 세포 생존율이 감소하지 않음을 확인되었다. 식물 추출물 혼합분말이 근육세포 분화 인자에 미치는 영향을 확인하기 위해 myogenin과 MHC의 발현 여부를 확인한 결과, 무처리군에 비해 발현이 증가함을 확인하였다. H2O2에 의해 유도된 산화적 스트레스에 대한 세포 보호 효과를 확인한 결과, 식물 추출물 혼합 분말 처리에 의해 H2O2 단독 처리군보다 세포 생존율이 증가하였으며, LDH와 creatine kinase의 활성이 감소하였다. 또한, Bax와 Bcl-2의 발현을 조절하여 caspase-9와 -3 활성화를 억제함을 확인하였다. 이를 통해 식물 추출물 혼합 분말의 근육세포 분화 효과 및 H2O2에 의해 유도된 산화적 스트레스에 대한 세포 보호 효과가 있음을 확인하였다. 따라서 식물 추출물 혼합 분말은 근감소증 개선을 위한 기능성 소재로써 활용이 가능할 것으로 판단되며, 향후 근감소증 개선을 위한 기능성 소재로서의 유효성 확보를 위해서 근감소증 세포 및 동물모델을 이용한 효능 및 기전 분석 연구가 더 필요할 것으로 사료된다.
표고버섯의 열수 가용성 다당류(PS)와 간 장해물질이며 발암성을 갖는 butter yellow를 흰쥐의 식이에 6주간 투여하여 혈장과 간의 효소활성 및 과산화지질의 함량을 측정한 결과는 다음과 같다. 1. 혈장 중 GOT 활성은 better yellow 첨가군(BO군)보다 PS의 투여군(BP군 : BO+PS)이 비교적 낮은 수준을 나타내었으며 ${\gamma}$-GTP와 GPT 활성에서도 BO군에 비하여 BP군에서 유의하게 낮은 활성을 나타냈다. 2. 간장 중 glutathion peroxidase, catalase 및 lactate dephdregenase 활성 또한 기초식이군(NO군)에 PS를 토여한 군(NP군)은 NO군과 유의한 차이가 없었으나 BP군은 BO군에 비하여 유의하게 낮은 수준이었다. SOD활성은 각 군간에 유사한 수치로 유의적인 차리를 보이지 않았다. 3. 간장의 microsomal 분획분 및 cytosol 분획분 중의 glutathione S-transferase 활성도는 BP군이 BO군보다 유의하게 낮은 수준을 보였으나 better yellow를 첨가하지 않은 NO군에 비하여 상당히 높은 수준을 나타내었다. 4. 간장 내 glutathione과 malondialdehyde의 함량 또한 PS의 투여로 생성이 저하되는 것으로 나타났다. 이상의 결과를 보아 표고버섯의 열수 추출물은 생체조직의 지질의 과산화를 방지하므로써 better yellow에 의한 간장장애의 치료 또는 독성을 완화하는 효과가 있는 것으로 사료된다.
유묘기 보리 습해의 주요 원인은 과습에 의한 근권의 산소 부족이다 따라서 혐기조건에 대한 뿌리와 지상부의 생육 및 혐기적 해당발효 효소 활성의 양적 및 질적 반응을 양액재배를 이용하여 용존산소 1ppm 수준의 혐기조건을 1-7일간 처리한 3엽기 보리 유묘를 재료로 이용하여 분석하였다 보리생육 및 통기조직의 발달은 단기적 혐기조건에 의해 크게 영향을 받지 않았다. 반면 뿌리의 ADH와 LDH의 활성이 현저히 증가하였다. 효소활성 수준의 현저한 증가는 모든 동위효소의 발현 증가에 의해 나타났다. 그러나 지상부의 효소활성과 동위효소 발현 양상은 혐기처리에 의해 변화하지 않았다 이러한 결과는 혐기조건 초기에는 산소부족 조건에서 세포의 pH변화를 안정시키면서 혐기적 해당과정을 통한 에너지 획득 효율을 증가시키기 위할 반응이 우선적으로 급속히 진행되는 것으로 해석된다. 이상의 결과는 보리의 내습성 정도와 혐기적 해당과정의 능력과의 관계에 대한 보다 자세한 연구의 필요성을 제시하는 한편, 내습성 품종 육성효율을 증진시키는데 필요한 내습성 간편검정법 개발에 유용한 정보로 활용될 수 있으리라 생각된다.
The hepatoprotective activity of flavonol glycosides rich fraction (F-2), prepared from 70% alcohol extract of the aerial parts of V calcarata Desf., was evaluated in a rat model with a liver injury induced by daily oral administration of $CCl_4$ (100 mg/kg, b.w) for four weeks. Treatment of the animals with F-2 using a dose of (25 mg/kg, b.w) during the induction of hepatic damage by $CCl_4$ significantly reduced the indices of liver injuries. The hepatoprotective effects of F-2 significantly reduced the elevated levels of the following serum enzymes: alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH). The antioxidant activity of F-2 markedly ameliorated the antioxidant parameters including glutathione (GSH) content, glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), plasma catalase (CAT) and packed erythrocytes glucose-6-phosphate dehydrogenase (G6PDH) to be comparable with normal control levels. In addition, it normalized liver malondialdehyde (MDA) levels and creatinine concentration. Chromatographic purification of F-2 resulted in the isolation of two flavonol glycosides that rarely occur in the plant kingdom, identified as quercetin-3,5-di-O-$\beta$-D-diglucoside (5) and kaempferol-3,5-di-O-$\beta$-D-diglucoside (4) in addition to the three known compounds identified as quercetin-3-O-$\alpha$-L-rhamnosyl- (${\rightarrow}6$)-$\beta$-D-glucoside [rutin, 3], quercetin-3-O-$\beta$-D-glucoside [isoquercitrin, 2] and kaempferol-3-O-$\beta$-D-glucoside [astragalin, 1]. These compounds were identified based on interpretation of their physical, chemical, and spectral data. Moreover, the spectrophotometric estimation of the flavonoids content revealed that the aerial parts of the plant contain an appreciable amount of flavonoids (0.89%) calculated as rutin. The data obtained from this study revealed that the flavonol glycosides of F-2 protect the rat liver from hepatic damage induced by $CCl_4$ through inhibition of lipid peroxidation caused by $CCl_4$ reactive free radicals.
본 연구는 싸리나무 잎에서 분리한 flavonoid 화합물인 eriodictyol의 항산화 활성을 평가하기 위해 hydrogen peroxide로 산화적 스트레스를 유도한 HepG2 세포에서 eriodictyol 화합물의 처리가 SOD-1, SOD-2, CAT 및 GPx의 유전자 발현에 미치는 영향을 분석하였으며, 또한 간 기능 지표효소인 GOT, LDH 및 GGT 활성을 분석하였다. 그리고 세포 내 활성산소종 생성 억제 효능을 분석하기 위하여 DCFH-DA assay를 실시하여 eriodictyol 화합물의 기능성 소재로서의 가능성을 알아보고자 본 실험을 하였다. Eriodictyol 화합물의 세포독성을 확인하기 위하여 HepG2 세포주를 이용하여 실시한 결과 eriodictyol 화합물을 $10{\sim}50{\mu}g/mL$의 농도로 처리한 모든 실험군에서 약 98% 이상의 세포생존율을 나타내었다. 항산화 효소 유전자 발현량을 통한 산화스트레스 억제 효과를 분석하기 위하여 HepG2 세포주에 hydrogen peroxide를 처리하여 산화스트레스가 증가시킨 조건에서 eriodictyol 화합물을 처리하여 SOD-1, SOD-2, CAT 및 GPx 발현량을 분석한 결과 eriodictyol 화합물의 처리 농도가 증가할수록 hydrogen peroxide 처리에 의해 감소한 SOD-1, SOD-2, CAT 및 GPx 발현량이 유의적으로 증가하는 것을 확인할 수 있었다. 간 기능 지표효소 활성을 측정하기 위해 GOT, LDH 및 GGT 활성을 분석한 결과 hydrogen peroxide로 단독 처리한 대조군과 eriodictyol 화합물을 처리한 군을 비교하였을 때 eriodictyol 화합물을 처리한 군에서 hydrogen peroxide 처리에 의해 증가한 GOT, LDH 및 GGT 활성이 유의적으로 감소하였다. HepG2 세포주에 eriodictyol 화합물을 처리하여 세포 내 활성산소종 생성에 미치는 영향을 DCFH-DA assay로 확인한 결과 eriodictyol 화합물의 농도가 증가함에 따라 세포내 활성산소종의 생성을 억제하는 것을 확인할 수 있었다. 따라서 본 실험을 통하여 eriodictyol 화합물은 산화적 스트레스로부터 항산화 효소 활성을 증가시키며, 활성산소종의 생성을 억제하는 효과를 확인할 수 있었다. 또한 간 기능 지표효소의 활성을 감소시켜 세포 보호 효과를 나타내어 항산화 활성 및 세포 보호 효과를 나타내는 기능성 소재로써 이용 가능성이 높을 것으로 판단되며, 후속연구를 통해 싸리나무 유래 eriodictyol 화합물의 세포 내 항산화 단백질 발현에 미치는 영향 및 동물실험을 통한 항산화 효과를 검증하는 것이 필요할 것으로 생각된다.

제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)
① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을
원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만
이 경우 그 내용을 공지합니다.
제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
