  • The purpose of this study was to investigate changes in the amino acid content and physicochemical properties of Cheonggukjang prepared by using various soybean cultivars (Daewon, Deapung, Seadanbeak, and Taekwang) and a functional microorganism (Bacillus subtilis HJ18-9). These soybeans were conventional Cheonggukjang (control) and Cheonggukjang fermented with Bacillus subtilis HJ18-9 (treated). The moisture contents of steamed, control, and treated soybean were 62.45~67.12%, 63.28~67.14%, and 64.50~66.87%; amino-type nitrogen contents were 6.53~24.25 mg%, 27.63~122.09 mg%, and 37.29~133.48 mg%; and ammonia-type nitrogen contents were 26.92~47.95 mg%, 45.45~156.36 mg%, and 28.02~121.13 mg%, respectively. The umami taste associated with several amino acids (aspartic acid and glutamic acid) in Cheonggukjang was lower than that for steamed soybeans, while the bitter taste from amino acids (methionine, valine, isoleucine, leucine, and phenylalanine) was higher than that for steamed soybeans. The result of sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis showed that the molecular weight of steamed soybeans was less than 100 kDa, while control and treated groups showed low molecular weights below 34 kDa, confirming their protein hydrolysis to small molecular weight. These results are information for developing functional fermented soybean paste and diversification using soybean cultivars.

  • Fermented vinegars using pear was compared according to the appled side materials. Quality characteristics of three kinds of the fermented vinegars (pear vinegar, PV; pear black rice vinegar, PBV; pear mint vinegar, PMV) were investigated, which includes pH, total acidity, colors, the contents of sugar, amino acids, total polyphenol, and total flavonoid, ${\alpha}^{\prime}{\alpha}-diphenyl-{\beta}-pycrylhydrazyl$ (DPPH) radical scavenging ability, and sensory evaluation. Brown rice vinegar (BRV) was used as a control. The pH and total acidity of the fermented pear vinegars were significantly different showing the range of 3.17~3.43 and 4.01~5.05%, respectively (p<0.05). The sugar contents of PV and PMV were significantly higher than other vinegars (p<0.05). L, a, and b values were the highest in PV, PBV, and PMV, respectively. Among the four vinegars, the essential amino acids were the highest in PV with the order of lysine, isoleucine, valine, and threonine. Besides, the fermented pear vinegars have many non-essential amino acids such as glutamic acid and aspartic acid. The aspartic acid content was the highest in PV while glutamic acid content was the highest in BRV. The total polyphenol content was the highest in PV while total flavonoid content was the highest in PBV. The DPPH radical scavenging ability (%) was the highest in PV. In sensory evaluation, PBV showed the highest color, taste and overall preference scores. These results show that pear would be desirable to prepare high-quality vinegars and functional foods.

  • The purpose of this paper is to investigate potential anti-inflammatory and anti-oxidant effects of Tenebrio molitor. Macrophage cell response by outside stimulation leads expression of pro-inflammatory cytokines, such as tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin-6 (IL-6), $interleukin-1{\beta}$ ($IL-1{\beta}$), and trigger expression of genes which are affected by inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), resulting in formation of inflammatory factors like nitric oxide (NO) and Prostaglandin $E_2$ (PGE2). Cell viability was determined by MTT assay. In order to investigate anti-inflammatory agents, the inhibitory effects on the production of lipopolysaccharide (LPS)-induced NO in RAW 264.7 cells were examined. T. Molitor significantly decreased the production of NO in a dose-dependent manner, and also reduced the expression of iNOS, a COX-2 protein. As a result, the levels of protein such as $PGE_2$, iNOS, COX-2 and MARKs were significantly reduced compared to non-treated group in T. Molitor water extract (TDW) treated group. Also, antioxidant effect of T. Molitor were investigated using DPPH, ABTS+ and superoxide anion radical scavenging activity tests in cell-free system. Antioxidant activity of T. molitor was found low in the DPPH radical scavenging test while high in the ABTS+ and superoxide anion radical scavenging activity tests. These results show that TDW could be an effective anti-pro-inflammatory and anti-oxidant agent.

  • Higher and uniform germination should be necessary because many commercial pepper (Capsicum annum L.) seedlings were nowadays sold to farmer. The experiment was carried out to determine the effect of its cultivars (Daemyung; Wanggochu), GA$_3$ (concentration; period), light quality (red; far-red; blue; dark) during GA$_3$imbibition, and germination temperature (GT: 25 or 15$^{\circ}C$ constant; 25/15$^{\circ}C$ alternating) on the rate of germination done under incandescent lamps until 9 days after sowing. Final seed germination was not different between GA$_3$0 to 1.0 mM concentrations but the elapsed days to 50% germination $(T_{50})$ were more reduced by GA$_3$ treatment than water imbibition. Under $25^{\circ}C$ constant germination temperature, earlier germination was enhanced by GA$_3$treatment showing the lowest rate at darkness, although the final germination rates of water imbibition and GA$_3$ treatments were same. The final germination rates of alternating and 25 $^{\circ}C$ constant GT in cv. Daemyng was also equal, while the germination rates of $25^{\circ}C$ and 15$^{\circ}C$ constant GT were the highest and the lowest regardless of cultivars. There was no difference between light quality treatments impelled during GA$_3$ imbibition when light treated seeds were germinated at alternating and $25^{\circ}C$ constant GT. At 15$^{\circ}C$ constant GT, however, red light or dark treatment during GA$_3$imbibition increased the germination rate since 5 days after sowing.

  • The study was performed to explore the molecular changes in the vegetative stage (3-and 5-leaf) of sorghum under waterlogging stress. A total of 74 differentially expressed protein spots were analyzed using LTQ-FT-ICR MS. Among them, 12 proteins were up-regulated and 3 proteins were down-regulated. Mass spectrometry (MS) results showed that about 50% of the proteins involved in various metabolic processes. The level of protein expression of malate dehydrogenase and glyceraldehyde-3-phosphate dehydrogenase related to carbohydrate metabolic process increased in both 3 and 5-leaf stage under waterlogging stress. These proteins are known to function as antistress agents against waterlogging stress. The expression of oxygen-evolving enhancer protein 1 protein related to photosynthesis was slightly increased in the treated group than in the control group, however the expression level was increased in the 5-leaf stage compared to the 3-leaf stage. Probable phospholipid hydroperoxide glutathione peroxidase protein and superoxide dismutase protein related to response to oxidative stress showed the highest expression level in 5-leaf stage treatment. This suggests that the production of reactive oxygen species by the waterlogging stress was the most abundant in the 5-leaf treatment group, and the expression of the antioxidant defense protein was increased.

  • This study presents a comprehensive account of the flora of hydrophytes and hygrophytes on Jeju Island. This approach aims to contribute to fundamental information about the conservation of plant diversity in wetlands. Field surveys were carried out from May of 2010 to September of 2014 in various types of wetlands on Jeju Island. A total of 189 taxa, including 52 families, 98 genera, 181 species, 6 varieties, 2 forma and 189 taxa, were found. Among them, 99 taxa (38 families, 60 genera, 96 species and 3 varieties) were hydrophytes and 90 taxa (23 families, 45 genera, 85 species, 3 varieties and 2 forma) were hygrophytes. In addition, Eleocharis${\times}$yezoensis H. Hara (Cyperaceae) was newly added to the flora of Korea. The protected wild plants designated by the Ministry of the Environment were 5 taxa, including Brasenia schreberi J. F. Gmel. and Dysophylla yatabeana Makino. The endangered species found included one Critically Endangered Species (i.e., Mankyua chejuense B.-Y. Sun, M. H. Kim & C. H. Kim), 5 Endangered Species (e.g., Lysimachia leucantha Miq.) and 7 Vulnerable Species (e.g., Tillaea aquatica L.). The floristic regional indicator plants found in this area were 44 taxa comprising 10 taxa of grade V, 7 taxa grade IV, 15 taxa of grade III, 5 taxa of grade II, and 7 taxa of grade I. The identified naturalized plants were 10 taxa; 8 families, 8 genera, 9 species and 1 variety. In the wetlands on Jeju Island, high plant species diversity was shown, and a number of rare plants and phytogeographically important plants were found. For these reasons, we consider that the wetlands on Jeju Island are very important areas from a botanical point of view. It is necessary to establish specific protection and maintenance policies in an effort to conserve these wetlands and species.

  • The infraspecific taxa of Adonis amurensis Regel et Radde are currently known to include one subspecies (subsp. nanus Y. Lee), two varieties (var. ramosa Makino, and var. dissectipetalis Y. Lee) and two forms (for. argentatus Y. Lee, and for. viridescensicalyx Y. Lee). Among them, the remaining three taxa except for subsp. nanus and var. dissectipetalis are unified with A. amurensis. Therefore, we examined the morphological characteristics and geographical distribution of A. amurensis in effort to elucidate the taxonomic status of subsp. nanus and var. dissectipetalis. After the subsp. nanus samples had been transplanted from their type locality, i.e. highland at more than 1,000 m above sea level, to a lowland habitat, we measured the samples' morphological characteristics. The range of values we recorded of the plant height and floral diameter from the subsp. nanus samples were within the parameters used to characterize A. amurensis. Accordingly, subsp. nanus has been characterized as an ecophene of A. amurensis. It is considered as a synonym of A. amurensis. Among the populations of A. amurensis (sensu Y.Lee, author), those with highly dissected petals at their apices have previously been described as var. dissectipetalis. However, this characteristic has been observed together with obtuse or rounded petal apices not only in one population but also in a single individual. However, populations with these characteristics belong to A. pseudoamurensis due to the simultaneous development of leaves and flowers, branched stems, acute leaf apices, and petals that are shorter than sepals. Therefore, we suggest that A. amurensis var. dissectipetalis be unified with A. pseudoamurensis. Very useful characteristics for distinguishing A. amurensis from its relative species are anthesis, the presence or absence of branches, the development of scales into normal leaves, the relative length of the sepals and petals, and the position of the stipules. A. amurensis is geographically restricted to E. Asia, including E. Siberia, and especially on the Korean Peninsula, it is distributed throughout the northern provinces of Gyeonggi province and in Gangwon province, Gyeongbuk province (Mt. Bohyeon-san) and Jeonbuk province (Mt Jeoksang-san) along Baekdudaegan mountain.

  • This study examined the effect of Momordica charantia L. (bitter melon: BM) on lipid and hepatic antioxidative enzyme levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats by injection of streptozotocin (STZ), and rats were fed for 4 weeks with experimental groups divided into four groups: a normal control group, STZ-control and STZ-BM 5% & STZ-BM 10% treated groups. Levels of free fatty acids (FFA), high-density lipoprotein cholesterol (HDL-chol), triglycerides (TG) in plasma and malondialdehyde (MDA) & protein in liver, catalase (CAT), superoxide dismutase (SOD), glutathione-S-transferase (GST), and xanthine oxidase (XOD) were measured in liver cytosol. Level of HDL-chol significantly increased in the STZ-BM 5% diabetic group. TG & FFA levels were significantly higher in all diabetic groups compared to the control group. MDA and protein levels were significantly higher in the STZ-BM 5% group compared to all other experimental group. CAT level was higher in the supplementary group with BM compared to the STZ-control group, although the difference was not significantly different. SOD level was not significant in any experimental groups. GST level was significantly higher in the BM-treated groups compared to the STZ-control group. XOD level was significantly lower in the BM 5% group and significantly decreased in all experimental groups. These results show that supplementation of BM fruit powder may have beneficial effects on diabetic complications and damage caused by oxidative stress.

  • Pound cakes were prepared with Citrus mandarin powder(CMP) cultivated in JeJu Island, Korea. The impact of CMP amount level, which was incorporated into wheat flour by the ration of 0, 5, 10, 15, and 20% based on a flour weight, on the rheology and sensory profile of the pound cakes was measured. Moisture content of 13.70%, crude protein 5.12%, crude lipid 1.30%, crud ash 1.92%, respectively. Also evaluation was performed on the changes in physicochemical properties of the pound cakes during storage at 4 and $30^{\circ}C$. According to the amylogram, gelatinization temperature of the control dough was $63.35^{\circ}C$ and those of the dough with CMP were $63.85{\sim}66.55^{\circ}C$. Maximum viscosity of the dough was 686 B.U in the control, those were 575 B.U, 553 B.U, 504 B.U and 401 B.U in the dough with 5, 10, 15, and 20% CMP, respectively. The retrogradation degree(setback value) of CMP dough was $31{\sim}57%$ lower than that of the control dough under the same conditions. Water holding capacity of pound cake was increased gradually in proportion to the amount of CMP. The CMP addition decreased the brightuess(L) of pound cakes but increased redness(a) and yellowness(b). Hardness of pound cakes was significantly increased by CMP addition, while springiness, adhesiveness and cohesiveness were decreased. Based on sensory evaluation, pound cakes added with CMP were not significantly different in color and texture, while that of 10% CMP was significantly high in taste, flavor, and overall preferences, compared to the control. pH of pound cake with CMP was decreased during storage, showing that pH of CMP samples was lower than the control. Titrated acidity of pound cake with CMP was increased rapidly from storage for 10 days, which the changes in degree was fast in accordance with CMP amount. The Hunter's color value of pound cake with CMP was decreased, as the storage time proceeded. In the samples prepared with CMP, the firmness, adhesiveness, gumminess and chewiness was increased as the storage time proceeded, while springiness and cohesiveness was decreased.
