Fertilizer and Row-spacing Effects on Growth and Yields in Amorphophallus konjac K. (구약감자 수집종들의 시비 및 재식거리에 따른 생육 및 수량반응)

  • This study was conducted to select an excellent goods and to estabilish the plant density of Amorphohpallus konjac. K about 3 local varieties and 2 introduced varieties through each fertilizer level. 1. Plant height and leaf width were increased according to increasing fertilizer level, and tuber yield was the highest at N-P$_2$ $O_{5}$ -K$_2$ O (14-10-14kg) per 10a as 390kg in Geumsan. 2. The more tuber size was large, the more yields per plant due to increasing the plant height and leaf width at low densities, while yields per 10a was decreased. 3. Among 5 varieties used, Jechon cultivar was good as a propagating variety because of increasing number of tuber and bulblet and Japan cultivar was high tuber yield per 10a.

  • This study was carried out to obtain a basic informations for the improvement of human health and the development of variety through analysis of inorganic compounds, contents of amino acids and saccharids to three elephant- foots (Amporphophallus Konjac. K) collected from Kumsan, Japan and China. Mannan as a carbohydrate of major component of an elephant-food was slightly high in Kumsan than that of Japan and China variety. Degree of viscosity of an elephant-foot depends upon the soluble free sugar contents and amino acid, contents of these free sugars were high in the order of Kumsan, China and Japan elephant-foot powder. Results of analysis of inorganic compounds to an elephant-foot used; Kumsan variety was shown to be high than those of China and Japan, especially. K, P and Fe in Kumsan variety were high, while Na and Ca were appeared to be high in China elephant-foot.

Characterization of the Bacillus licheniformis WL-12 Mannanase from a Recombinant Escherichia coli (재조합 대장균으로부터 생산된 Bacillus licheniformis WL-12의 Mannanase 특성)

  • A gene encoding the mannanase of Bacillus licheniformis WL-12, which had been isolated from Korean soybean paste, was cloned into Escherichia coli and nucleotide sequence of the mannanase gene was subsequently determined. The mannanase gene consisted of 1,080 nucleotides encoding a polypeptide of 360 amino acid residues. The deduced amino acid sequence was identical to that of putative mannanase from B. liceniformis DSM13 belonging to GH family 26. The mannanase was partially purified from cell-free extract of the recombinant Escherichia coli carrying a WL-12 mannanase gene by ammonium sulfate fractionation and DEAE-Sepharose column chromatography. Optimal conditions for the partially purified enzyme occurred at pH 6.0 and $65^{\circ}C$. The enzyme showed higher activity on locust bean gum (LBG) galactomannan and konjac glucomannan than on guar gum galactomannan. The predominant products resulting from the mannanase hydrolysis were mannose, mannobiose and mannotriose for LBG or mannooligosaccharides. The enzyme could hydrolyze mannooligosaccharides larger than mannobiose.

Properties of a Bacillus licheniformis Cellulase Produced by Recombinant Escherichia coli (대장균으로부터 생산된 Bacillus licheniformis WL-12의 Cellulase 특성)

  • Carboxymethyl celluase (cellulase) was purified from cell-free extract of the recombinant Escherichia coli carrying a Bacillus licheniformis WL-12 cellulase gene by DEAE-Sepharose and phenyl-Sepharose column chromatography with specific activity of 163 U/mg protein. The molecular mass of the purified enzyme was estimated to be approximately 49.5 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The enzyme had a pH optimum at 5.5 and a temperature optimum at $55^{\circ}C$. The activity of the enzyme was completely inhibited by SDS (5 mM), and slightly enhanced by $Cu^{2+}$ (5 mM). The cellulase was active on CMC, konjac, barely glucan and lichenan, while it did not exhibit activity towards xylan, locust bean gum, and p-nitrophenyl-$\beta$-glucopyranoside. The predominant products resulting from the cellulase hydrolysis were cellobiose and cellotriose for cellooligosaccharides including cellotriose, cellotetraose and cellopentaose. The enzyme could hydrolyze cellooligosaccharides larger than cellobiose.

Status of Consumption and Consumer Satisfaction of Dietary Fiber Supplements among Adults Living in Seoul and Gyeonggi Province (서울.경기지역 성인의 식이섬유 보충용 제품의 섭취 현황과 소비자 만족도)

  • The aim of this study was to evaluate the status of consumption and consumer satisfaction of dietary fiber supplements. The survey was conducted using a self-recorded questionnaire developed to meet the purpose of this study. The characteristics of the subjects who have taken dietary fiber supplements in the past are as follows: distributions of socioeconomic variables showed that the highest-ranked group(the group that reported the highest consumption of dietary fiber supplements) was professionals with a college degree or higher degree. In addition, the highest-ranked group were the most healthy, nonsmokers and reported drinking habits of '1~2 times/week'. Regarding the health status of those surveyed, digestive disease and constipation were the most common complaints. The majority responded with 'health concerns' to the question that asked the reason for their purchase of the dietary fiber supplements. Three major places respondents could find dietary fiber supplements were 'through the internet', 'the market', 'the pharmacy' in descending order. The compositions of dietary fiber products varied. 'konjac', 'cellulose', 'psyllium husks' were the most popular components consumed by the subjects. The average duration of product consumption was less than one month. In terms of satisfaction levels(satisfaction was ranked from 1 to 5), 'taste' and 'type' scored the highest in satisfaction levels while 'price' ranked the lowest. When subjects were asked about self-awareness regarding the physiological efficacy of dietary fiber supplements, the responses were positive in 'bowel movement' and 'weight loss'. Further research is required to improve the quality of dietary fiber supplements in order to assist consumers in selecting the appropriate products.

Konjac Glucomannan Derived Carbon Aerogels for Multifunctional Applications

  • Environmental and energy issues have always been a hot topic of global research. Oil leakage has caused great damage to the environment, affecting a wide area and it is difficult to clean up. In most cases, carbon-based adsorbents are typically utilized to remove oil spills because of their economic benefits and high adsorbent efficiency. At the same time, its excellent material properties can also be used for the preparation of supercapacitors. In this paper, the carbon aerogels were prepared by the one-step method. The prepared materials endowed a 3D network structure with a huge number of micropores and mesoporous, and the material is light-weight, stable, hydrophobic and has affinity for oil (17.02 g/g) to the KGM carbon aerogel. Through the physicchemical characterization, the KGM carbon aerogel shows specific surface area is $689m^2/g$, high water contact angle ($136.64^{\circ}$) and excellent reusability (more than 15 cycle times). In addition, we also discussed the electrochemical properties of the material and obtained the specific electrical capacity of 139 F/g under the condition of 1 A/g.

The Relationship between Viscosity of Soluble Dietary Fiber and their Hypoglycemic Effects (수용성 식이섬유소의 점도가 혈당강하에 미치는 영향)

  • Experiments were carried out in humans to assess the relationship between viscosity and post-prandial glucose response of soluble fibers. Eight(3 male and 5 female) healthy individuals were tested for their glucose response after taking control meals or 3 test fiber meals of different viscosities. Meal viscosity of the test food was adjusted to be between 200 and 70, 000 cps. There was a significant increase in post-prandial glucose response(p=0.01) in control meals by solely increasing the volume of water. Fiber with the highest viscosity konjac mannan demonstrated the lowest relative glucose response(70.1$\pm$6.6), followed by medium viscosity xanthan (79.3$\pm$8.7)and low viscosity psyllium (86.3$\pm$10.5). The difference is significant at the level of p<0.05. However, no significant difference in relative glucose response of the same fiber was found when the 4 levels of water were added to make different meal viscosity of each fiber (p=0.476). This result suggested that hypoglycemic action of soluble dietary fibers is related to fiber viscosity rather than meal viscosity.

A Study on the Rheological properties of Glucomannan (Glucomannan 의 유변학적 성질에 관한 연구)

  • Glucomannan(G.M.)은 Amorphophallus Konjac C. Koch의 tuber로부터 분리되었고. 이 G.M.은 다시 침전제로 메탄올을 사용하여 4단계로 분별되었다.(F.1, F.2, F.3, F.4,). 각분 별물에 비하여 직선으로부터 벗어남을 보였다. Low shear viscometer로 G.M. 용액의 viscosity를 측정하였고 농도와 zero shear specific viscosity의 logarithm을 도시한 결과 inflection point를 나타내었다. 이것은 G.M. 분자들의 coil overlap의 시작에서 기인한 것이 고 묽은 용액에서 진한 용액으로의 전이행동은 임계농도. C*=4/[η]에서 일어났고 이때의 zero shear specific viscosity는 10을 나타내었다. 또한 specific viscosity는 묽은 용액에대해 서는 C14로써 변화하였고 진한 요액에서는 C3.0으로변화하였다. G.M.의 고체상태에 대한 유 전성($\varepsilon$',$\varepsilon$")과 점탄성(C',C")계수들을 액체질소 온도에서부터 15$0^{\circ}C$ 온도범위에 걸쳐 4단계로 film을 건조시키면서 10Hz에서 측정하였다. G.M. film의 유전성과 점탄성의 허수부 분은 ($\varepsilon$", C"), -10$0^{\circ}C$에서 peak를 나타내었고 이 peak는 hydroxy methyl 기들의 회전 운동에서 생겨난 것이다. 건조시키지 않은 상태의 G.M. film의 유전성과 점탄성의 허수부분 의 값들은 -5$0^{\circ}C$에서 물 분자의 운동에 의하여 생긴 peak를 보였다.

A Study on the Rheological Properties of glucomannan(II) (Glucomannan의 유변학적 성질에 관한연구 (II))

  • GM 은 Amorphopallus Konjac K. Koch의 tuber의 주성분으로알카리 응고제를 첨가 함에 따라 열적으로 안정한 gel을 형성한다. GM에 침전제로 메탄올을 사용하여 4단계로 분 별하였고 각 분별물(F.1, F.2, F.3, F.4)들은 Mark-Houwink 식을 이용하여 분자량을 산출하 였다. Ubbelohde를 이용한 농도에 따른 환산점도로부터 고유 점성도값을 구한 결과 분자량 이 높은 분별물이 분자량이 낮은 분별물에 비하여 직선으로부터 벗어남을 알수 있었다. 또 한 고유점성도는 분자량이 증가할수록 증가하였고, 분자량 분포 범위가 broad하게 나타났다. GM용액의 frequency와 shear rate에 따른 viscosity를 측정한결과 낮은 frequency에서는 loss modulus(G")가 storage modulus(G")보다 큰 값을 가졌으나 높은 frequency에서는 G '가 G"보다 크게 나타났다. 이 현상은 분자들의 얽힘에 관한 시간 의존도로써 설명할수 있다. 즉높은 frequency에서는 탄소 사슬간의 얽힘들이 진동기간내에 분자결합이 끊어지기 에 충분한 시간을 갖지 못하여 그 조직이 교차 결합을 한 gel처럼 행동하게 되고 한편 낮은 frequency에서는 탄소사슬들의 얽힘이 풀리고 흐르기에 충분한 시간을 갖게된다. Shear rate에 따른 viscosity값과 frequency에 대한 dynamic viscosity값은 거의 유사한 변화를 나 타내었다.

Characterization of two β-mannanases from Cellulosimicrobium sp. YB-43 (Cellulosimicrobium sp. YB-43에 의해 생산되는 2종류 β-mannanase의 특성분석)

  • A bacterial strain producing extracellular mannanases was isolated from soil of chestnut tree farm located in Gongju city of Korea by enrichment culture using Avicel as a carbon source. 16S rDNA sequence of the isolate YB-43 was highly homologous to those of genus Cellulosimicrobium strains with sequence similarities of above 99.6%. Mannanase productivity was significantly increased when the Cellulosimicrobium sp. YB-43 was grown in the presence of locust bean gum (LBG) or konjac. The mannanases were partially purified to be mannanase A (ManA) and mannanase C (ManC) by DEAE-Sepharose column and Q-Sepharose column chromatography from the culture filtrate of Cellulosimicrobium sp. YB-43 grown in LB medium supplemented with 0.7% LBG for 24 h. The partially purified ManA showed the highest activity at $55^{\circ}C$ and pH 6.5, while ManC activity was optimal at $65^{\circ}C$ and pH 7.5. ManA was stable up to $40^{\circ}C$ for 1 h, but ManC activity decreased significantly even after 1 h at $20^{\circ}C$. ManA and ManC showed difference from each other according to their substrate specificities and predominant products resulting from the mannanase hydrolysis for mannooligosaccharides. As a result, Cellulosimicrobium sp. YB-43 was found to produce two different kinds of mannanases.
