Natural Dyeing of Silk Fabrics with Dansam (Salvia miltiorrhiza Bunge) Extract (단삼 추출액을 이용한 견직물의 천연염색)

  • Dansam (Salvia miltiorrhiza Bunge) is a perennial plant that belongs to the Labiatae family and is characterized by red pigment found in the epidermis of its roots. Research on the dyeability of Dansam extract was performed with repeated trials under various conditions such as temperature, time, and mordant treatment. Silk fabrics were dyed with Dansam extract and mordanted with various agents such as Al, Fe, Cu, and Sn to evaluate the dyeability and functionality of Dansam extract; subsequently, the K/S value and the change of surface color were analyzed using a colorimeter. In addition, the dyeability of dyed fabrics (such as color fastness to washing, dry cleaning fastness, rubbing fastness and light fastness) and functionality (such as antibacterial activity and deodorization) were analyzed. The surface colors of all dyed fabrics were recorded and the results showed the presence of mostly YR series of colors; in addition, a GY color series appeared in the Fe pre-mordants of the silk fabric. The highest K/S values were recorded in Fe post-mordants. The washing fastness were Level 3 and Level 4; in addition, the dry-cleaning fastness showed excellent results at Level 4 and Level 5. The rubbing fastness was satisfactory at Level 3 and Level 4 and the light fastness was satisfactory at Level 4. The results of the measurements (that pertained to the antibacterial activity of fabrics dyed with Dansam extract) showed a 99.9% bacteria reduction rate of Staphylococcus aureus and Klebsiella pneumoniae. In regards to deodorization ability, dyed fabrics were found to have higher deodorization rates than those not dyed. The dyed fabric demonstrated exceptional deodorization qualities.

Antibiotic susceptibility and imaging findings of the causative microorganisms responsible for acute urinary tract infection in children: a five-year single center study

  • Purpose: We studied the differences in the antibiotic susceptibilities of the microorganisms that causeing urinary tract infections (UTI) in children to obtain useful information on appropriate drug selection for childhood UTI. Methods: We retrospectively analyzed the antibiotic susceptibilities of 429 microorganisms isolated from 900 patients diagnosed with UTI in the Department of Pediatrics, Chungbuk National University Hospital, from 2003 to 2008. Results: The most common causative microorganisms for UTI were Escherichia coli (81.4%), Klebsiella pneumoniae (8.4%), Enterobacter spp. (1.7%), and Proteus spp. (0.4%). E. coli showed relatively high susceptibility as compared to imipenem (100%), amikacin (97.7%), aztreonam (97.9%), cefepime (97.7%), and ceftriaxone (97.1%), while it showed relatively low susceptibility to gentamicin (GM) (79.0%), trimethoprim/sulfamethoxazole (TMP/SMX) (68.7%), ampicillin/sulbactam (33.0%), and ampicillin (AMP) (28.6%). There were no significant differences in the image findings for causative microorganisms. Conclusion: Gram-negative organisms showed high susceptibility to amikacin and third-generation cephalosporins, and low susceptibility to AMP, GM, and TMP/SMX. Therefore, the use of AMP or TMP/SMX as the first choice in empirical and prophylactic treatment of childhood UTI in Korea should be reconsidered and investigated further.

Characterization and Identification of Organic Selenium-enriched Bacteria Isolated from Rumen Fluid and Hot Spring Water

  • In the present study, the isolation of selenium (Se)-enriched bacteria from rumen fluid and hot spring water was carried out. Rumen fluid samples were taken from cannulated goats fed a basal diet and the water samples were collected from Selayang hot spring, Selangor-Malaysia. A total number of 140 Se-tolerant isolates were obtained aerobically using an Se-enriched medium and spread plate technique. All the isolates were initially screened for the ability to transform the Se-containing medium to a red-orange culture using a spectrophotometer. Twenty isolates of dark red-orange medium were selected for a screening of the highest Se-containing protein accumulating strains using the dialysis technique and icp.ms to measure the Se content. Four isolates, identified as Enterobacter cloacae (ADS1, ADS7, and ADS11), and Klebsiella pneumoniae (ADS2) from rumen fluid origin, as well as, one isolate from hot spring water (Stenotrophomonas maltophilia (ADS18)), were associated with the highest biomass organic Se-containing protein when grown in a medium enriched with $10{\mu}g/ml$ sodium selenite. In addition, around $50{\mu}g/100{\mu}g$ of the absorbed inorganic Se was accumulated as an organic form. Organic Se-containing protein in all the selected strains showed antioxidant properties in the range of 0.306 to 0.353 Trolox equivalent antioxidant capacity (TEAC) mg/ml. Therefore, these strains may offer a potential source of organic Se due to their Se-tolerant nature and higher biomass organic to inorganic Se ratio.

Trend in Study of Biological Pollutants in Indoor Air Quality in Korea (실내공기 중 생물학적 오염물질에 관한 국내 연구 동향)

  • Objectives: Indoor air quality problems in public spaces have recently become a very important issue in South Korea, because many spend a long time every day indoors. In this study, an extensive literature review was performed on the subject of suspended bacteria and fungi in public facilities. The objective of this study was to evaluate the extent and types of suspended bacteria and fungi in indoor air in South Korea. This study will provide direction to the research in the field of indoor air. Methods: Article reviews was conducted using a search engine and the following keywords; 'bacteria', 'total suspended bacteria', 'fungi' and 'indoor air' from published articles since 2000. Results: Most of the places of measurement were schools, kindergartens, childcare centers and general hospitals. The highest concentrations of total suspended bacteria and fungi were found in schools and kindergartens. Some public facilities exceeded the standard of 800 CFU/$m^3$. There were few studies on other suspended bacteria: Gram Negative Bacteria, Staphylococcus and Klebsiella pneumoniae. Conclusions: It appears that more studies with intensive controls are needed. It was found that schools and kindergartens were most susceptible to microbial indoor air pollution. These results may be able to suggest an indoor standard for suspended bacteria and fungi in South Korea.

Protective Effect against Neuronal Cell and Inhibitory Activity against Bacteria of Mulberry Fruit Extracts (오디 추출물의 신경세포 보호활성 및 항균활성)

  • As functional evaluation of mulberry fruits extracts, the protective effect on cerebral cell and antibacterial activities were carried. $1\%$ HCl-MeOH extract showed $37\%$ cytoprotective effect on hydrogen peroxide, also C3G identified mulberry fruits and cyanidin showed $52\%,\;76\%$, respectively, protective effects on oxygen-glucose deprivation (OGD). In the antibacterial activity of mulberry fruit extracts, MeOB-Cheongil extract showed the highest inhibitory activity. Salmonella typhimurium was shown inhibitory rate more than $70\%$ in all treatment groups. Also Klebsiella pneumoniae was shown inhibitory activity in all treatment groups.

Fluorogenic and Chromogenic Assay for Rapid Detection of Escherichia coli and Total Coliform Bacteria (효소발색법을 이용한 대장균 및 총대장균군 신속 검사)

  • We developed the Eco medium for Escherichia coli and total coliforms, which was modified by Violet Red Bile (VRB) medium, and derived the standard curve of exponential phase at $OD_{410}$ by using type strains such as E. coli ATCC11303, Enterobacter cloacae KCTC2361, Klebsiella pneumoniae KCTC2241, and Citrobacter freundii KCTC2359. Also, we used total 93 samples of spring and stream water to compare the detection ability of total coliforms between the method using Eco medium and such as most probable number (MPN), and plate count methods. As a result, the qualitative analysis of E. coli and total coliforms using Eco medium contained ortho-nitrophenyl-$\beta$-galactoside (ONPG) and 4-methylumbelliferyl-$\beta$-D-glucuronide (MUG) was same as those of Korean standard methods (Colilert kit). And the colony forming unit (CFU) detected in Eco medium was similar to those of result from MPN and plate count methods. Moreover, the agreement, sensitivity, and specificity of the developed kit was more than 97.5% in comparison with Colilert kit for 350 samples. Thus, the Eco medium can be used both qualitative and quantitative analysis of E. coli and total coliforms.

The Extract of the Glycyrrhizin from Glycyrrhizae Radix and Antibacterial Activity of the Treated Fabrics (감초로 부터 글리시리진의 추출 및 직물의 항균성)

  • This study investigates the attachment of glycyrrhizin to fabric using an X-ray photoelectron spectrophotometer( XPS). XPS spectra analysis showed that carbon content on treated fabrics with 0.2% glycyrrhizin increased to 2.699% for silk, 2.829% for nylon, 1.505% for cotton, respectively. The results show that glycyrrhizin is absorbed on treated fabrics. The glycyrrhizin extraction method makes radix glycyrrhizae powder 10g treat the first and the second treatment with ethanol, remove impurities on $75^{\circ}C$; subsequently, it is treated for 10 hours with ethanol 75% on $85^{\circ}C$ and lyophilizated. As the result, glycyrrhizin is extracted 1.7g in GL-I, 1.1 g in GL-II. As the result of abstracting glycyrrhizin with two methods, pure glycyrrhizin was abstracted 45.9% in GL-I, 74.9% in GL-II. GL-I, GL-II; in addition, glycyrrhizin( Japan) on TLC plate was separated in Rf 0.6. By GL-II extract method, this experiment obtained glycyrrhizin 15 g treated in a bath ratio set to 1: 100. Silk fabric was treated at $80^{\circ}C$, 60 min. in, nylon fabric $10^{\circ}C$, 70 min., and cotton fabric $30^{\circ}C$, 80 min.; subsequently, silk, nylon, cotton fabrics showed a 99.9% antibacterial activity for Staphylococcus aureus and Klebsiella pneumoniae.

Natural Dyeing Absorption Properties of Chitosan and Nano Silver Composite Non-Woven Fabrics -Focus on Chrysanthemum Indicum Linn- (키토산/나노실버 복합섬유 혼방 부직포의 천연염색 염착특성 -감국을 중심으로-)

  • This study examines the dyeability, light fastness, washing fastness, and the antibacterial activity of chitosan and nano silver composite non-woven fabrics dyed with an extracted solution from Chrysanthemum Indicum Linn. The results show that an increase in the chitosan and nano silver percentage resulted in an increase in the $a^*$ values and $b^*$ values; however, the $L^*$ values decreased in the undyed condition. ${\Delta}E$ values of chitosan and nano silver composite non-woven fabrics were higher than cotton 100% non-woven fabrics in the dyed condition with an extracted solution from Chrysanthemum Indicum Linn, and mordant treatments influenced the chrominance change. In the dyed condition with an extracted solution from Chrysanthemum Indicum Linn, an increase in the percentage of chitosan and nano silver resulted in an increase of the K/S values. The dyeability of chitosan and nano silver composite non-woven fabrics increased by mordant treatments. The light fastness and washing fastness of the mordanted non-woven fabrics were better than the non-mordanted. For the antibacterial activity, the bacterial reduction rate of chitosan and nano silver composite non-woven fabrics was 99.9% to Staphylococcus aureus and Klebsiella pneumoniae.

Growth Inhibition of Coliform Bacteria in Model System of Naengmyon-Broth by Using Dongchimi-Juice (냉면국물 모델 시스템에서 동치미 국물의 사용에 의한 대장균군 증식 억제)

  • Naengmyon is a Korean buckwheat noodle with chilld broth, and the juice of dongchimi, a Korean radish pickle fermented with wild lactic acid bacteria, has been used as the broth for naengmyon traditionally. The purpose of this study was to demonstrate the inhibitory effect of dongchimi-juice against coliform bacteria in model system of naengmyon-broth. Dongchimi-juice was made from radish juice by the cultivation of lactic acid bacteria which had been isolated from kimchi. Three types of naengmyon-broth, beef-broth type, dongchimi-juice type and the mixed type, were made as model systems, and then the changes in viable cell counts of seven coliform bacteria, Klebsiella planticola Bo2, X. terrigena CO8, K. pneumoniae DOI, K. ozaenae DO4, Enterobacter sp. AO2, Enterobacter sp. CO7, Citrobacter sp. BO7 and Escherichia sp. DO3, which had been added to each type of naengmyon-broth in advance, were investigated during storage at 3$0^{\circ}C$ All coliforms grew rapidly in naengmyon-broth of beef-broth type, while none grew in dongchimi-juice type or in the mixed type. All coliforms died out far more rapidly in dongchimi-juice type than in mixed type. The decreasing slopes of Citrobacter sp. Bo7, K. planticola BO2, X. terrigena CO8 and K. ozaenae DO4 were more steep than those of the rest. It was thought that the preparation method of Korean traditional naengmyon such as dongchimi-naengmyon or Pyongyang style-naengmyon, which uses oxy dongchimi-) juice or the mixture of dongchimi-juice and beef-broth, would be very effective for preventing the growth of coliform bacteria from naengmyon.

Studies on Chronic Mastitis of Dairy Cattle in Kyungnam Province (경남지방(慶南地方)에서 발생(發生)한 젖소의 만성(慢性) 유방염(乳房炎)에 대한 연구(硏究))

  • Some investigations on chronic mastitis in dairy cattle in Kyungnam Province during the year 1982 were conducted with the special reference to the causative agents and their drug resistance. Milk samples from 46 isolated cases of chronic mastitis cattle were investigated bacteriologically and the organisms recovered were examined for their drug susceptibility against the major antibiotics used in this country by the use of disk diffusion susceptibility test. Four major causative agents involved in chronic mastitis were in order of prevalence Staphylococcus aureus (32.6%), Escherichia coli (28.3%), Streptococcus agalactiae (8.7%) and Candida albicans (8.7%). Staph. epidermidis, Streptococcus uberis, Klebsiella pneumoniae and Candida subtropicalis were found to be one of the minor agents. The majority of staphylococcal isolates were highly resistant to the most of antibiotics employed while 8% of them were resistant to gentamicin and 32% to chloramphenicol. The percentages of staphylococcal cultures resistant to penicillin, lincomycin. streptomycin, methicillin, oleandomycin, tetracycline, cephalothin, ampicillin and erythromycin were 100%, 96%, 96%, 92%, 84%, 84%, 80%, 76%, and 64% respectively. Streptococcal isolates were also highly resistant to the majority of the drugs used although 85.7% of them were susceptible to gentamicin. All Escherichia coli isolates were found to be resistant to erythromycin, lincomycin and penicillin while the majority of them were resistant to ampicillin (92.9%), carbenicillin (85.7%), oleandomycin (85.7%), streptomycin(85.7%), kanamycin (78.6%), methicillin (78.6%) and tetracycline (71.4%). The percentages of E. coli cultures resistant to gentamicin, nitrofurantoin, cephalothin and chloramphenicol were 21.4%, 21.4%, 35.7% and 50.0% respectively.
