Effect of Fermented Anchovy Extracts on the N-methyl-N'-nitro-N-nitrosoguanidine-Induced Mutagenicities (멸치젓갈추출물이 돌연변이 유발에 미치는 영향)

  • The effects of raw anchovy, salted raw anchovy (20% salt+anchovy), 6- and 12-month fermented anchovy (20% salt added) on the N-methyl-N'-nitro-N-nitrosoguanidine (MNNG)-induced mutagenicities were evaluated using Salmonella assay and the SOS chromotest. The methanol extracts from raw, salted and the fermented anchovy (FA) sample increased the revertants of Salmonella typhimurium TA100 at the level of $0.125{\sim}5\;mg/plate$ in Ames test. The salted raw anchovy extract induced the largest number of the revertants. All the FA extracts had comutagenic effect on the MNNG. Twelve-month FA juice exerted the lowest comutagenic activity among the FA samples. The comutagenicity of 12-month FA was due to the synergistic effect of salt and histidine which teem in FA. Thus the Ames test using histidine requiring mutant, S. typhimurium, is not appropriate to determine the mutagenicity of FA which is rich in histidine. In SOS chromotest using E coli, raw, salted and fermented anchovy extracts did not show any mutagenicity in the absence of MNNG. The raw and fermented anchovy samples blocked the SOS response of E. coli PQ37 induced by MNNG, while raw salted anchovy increased the SOS induction factor. Twelve-month FA juice showed higher antimutagenic effects than 6-month FA samples (both solid and liquid). The ripened (12-month) FA along with raw anchovy in the SOS chromotest exhibited antimutagenic activity.

Analysis of Optimum Condition for Production of an Onionic Vinegar by Two-Step Fermentations (2단계 발효에 의한 양파식초 제조의 최적 조건 검토)

  • This study was carried out to develop a vinegar by an onion juice. Onions are considered to be a promising source of the vinegar because these are rich in sugars, amino acids and various nutrients. An Acetobacter for an acetic acid fermentation was isolated and used from vinegars produced by industrial goods or from matured Kimchi. When supplemented with 2-8% ethanol into an onionic juice medium, the highest production of the acetic acid was observed at 9 days by addition of 4% ethanol. Optimum temperature and aeration for acetic acid production were exhibited at $30^{\circ}C$ and 200 rpm, respectively. A flask containing larger air-contact surface region for fermentation was produced the more acetic acid than that of a test tube. Taken all these together, an optimum condition for the acetic acid fermentation was over 9 days at $30^{\circ}C$, 200 rpm with 5% alcohol and 2% initial acidity. When fermented by the upper condition, the final product contains 5.2% total acidity and less than 1% ethanol. These are suitable for conditions of fruit vinegar notified by the Ministry of Commerce, Industry and Energy.

Effect of Dill and Stevia Hot-Water Extracts on Quality and Sensory Characteristics of Kimchi (딜.스테비아 열수 추출물에 절인 배추김치의 품질 및 관능적 특성)

  • The quality and sensory characteristics of kimchi during fermentation at 15 $^{\circ}C$ were evaluated. Kimchi was prepared with Chineses cabbage which was brined in 10% salt water added dill hot-water extracts(DK) and stevia hot-water extracts(SK) respectively. The pHs and reducing sugar contents of kimchi were decreased with increasing the fermentation time whereas lactic acid(%) was increased. The pH of DK was lower than control, and the lactic acid(%) contents of DK, SK were equal or higher than control in the first stage of fermentation. Also, the reducing sugar contents of DK, SK were lower than control. These results showed that the fermentation of DK and SK were faster than that of control in the first stage of fermentation. But the lactic acid(%) contents of DK, SK were higher than control after the 12th day of fermentation. It showed that it is possible to extend the preservation of kimchi. With increasing fermentation time, the L value of kimchi juice was increased, and the a value was decreased, and the b value was constant. Especially, the a value of DK, SK was higher than control. It meant that DK, SK kept more carotenoids in kimchi than control. On the 6th day of fermentation, the sensory evaluation analyzed by ANOVA followed Duncan's multiple range test showed that the levels of the green smell and the moldy flavor of DK were significantly lower than control(p<0.05) and the levels of the green smell, the green flavor and the moldy smell of SK were significantly lower than control(p<0.05), too. And the level of overall flavor of DK, SK was higher than control. The most preference among experimental groups was SK and then control, DK. The levels of TBARS of DK, SK were generally lower than control, and the level of TBARS of DK was lower than SK.

Physiological Characteristics and GABA Production of Lactobacillus plantarum K255 Isolated from Kimchi

  • As a major inhibitory neurotransmitter of the central nervous system in animals, ${\gamma}$-aminobutyric acid (GABA) has several physiological functions, such as anti-hypertensive, diuretic, tranquilizer and anti-stress effects in human. In order to determine strains with high GABA producing ability and glutamate decarboxylase (GAD) activity, 273 bacteria were isolated from various types of Kimchi. Strain K255 contained $386.37{\mu}g/mL$ of GABA in MRS broth containing 1% MSG, $600.63{\mu}g/mL$ of GABA in MRS broth containing 2% MSG and $821.24{\mu}g/mL$ of GABA in MRS broth containing 3% MSG. It showed that K255 had the highest GABA production ability compared to other commercial lactic acid bacteria. K255 was identified as Lactobacillus plantarum based on its API carbohydrate fermentation pattern and 16S rDNA sequence. K255 was investigated for its physiological characteristics. The optimum growth temperature of K255 was $37^{\circ}C$and cultures took 13 h to reach the pH 4.4. K255 showed more sensitive to bacitracin in a comparison of fifteen different antibiotics, and showed most resistance to kanamycin and vancomycin. Moreover, it was comparatively tolerant to bile juice and acid and displayed resistance to Escherichia coli, Salmonella Typhimurium, Staphylococcus aureus, with rates of 30.8%, 29.7%, and 23.4% respectively. These results demonstrate that K255 could be an excellent strain for the production of functional products.

Factors stimulating the Growth of Lactic acid Bacteria in Cucumber Juice (오이에 대(對)한 유산균(乳酸菌) 생육촉진(生育促進) 인자(因子)에 관(關)하여)

  • Addition of one percent cucumber, as a Kimchi component, showed the stimulation of lactic acid fermentation, by reducing the lagphase, and resulted the early matured Kimchi. Alcohol-soluble fraction from cucumber serum accerelated the growth of Lactobacillus plantarum, p-1, p-2 newly isolated strains from cucumber pickle, but had no effect on the growth rate of Streptococus faecalis. The growth promoting activity was inactivated by dialysis against water and heat treatment at $120^{\circ}C$ for 10 minutes. Softness of cucumber tissues enables to extract easily out a substance, which is thought one of the important factors for the stimulation of me growth of Lactic acid bacteria.

Production of Yeast Cell Mass from Waste Brine and Cabbage Waste of Kimchi Factory (김치 공장의 절임 폐수 및 배추 쓰레기를 이용한 효모 배양)

  • The possibility of using waste brine and cabbage waste from kimchi industry as raw materials for the production of yeast cell mass was investigated. Among four strains of osmotolerant yeast, Candida guilliermondii ATCC 6260 showed the best growth in the waste brine containing about 1.0g/L of reducing sugar and 7% to 12% of NaCl. The growth of C. guilliermondii in waste brine was affected slightly between the temperature range of $25^{\circ}C$ to $35^{\circ}C$ and the initial pH of 3 to 6. The NaCl concentration up to 9% was not inhibitory to the growth of C. guilliermondii and the addition of 10mM of ammonium salts or 5mM of potassium phosphate had no effect on the growth. The growth of yeast reduced BOD of the waste brine by 85% within 24hours. When C. guilliermondii was cultured in waste brine added with cabbage juice extracted from waste cabbage, the cell mass was increased significantly.

Study on Frequently Consumed Dishes and Menu Patterns of Middle-aged Housewives for 1 Year (중년 주부의 연중 음식 섭취 및 식단 패턴 조사)

  • This study was conducted to investigate the dishes and menu patterns consumed frequently for 1 year, using estimated record method with 30 housewives. The purpose of this study was to suggest the dishes and menu pattern for the basis data of the nutrition education program and menu development project. The results were following : cooked rice and kimchi had the highest number of frequencies. The dish group (classified by the cooking method) consumed in largest quantities per capita per meal is the rice group (146.5g). Most frequently consumed dish group were the rice (72.7%), kimchi (60.6%), beverages (33.6%), fruits and juice (26.2%), soup (25.1%), stew and casserole (25.1%) in the order. Some dishes in several dish groups had small number of frequencies. Thus they were not included in the frequently consumed dishes list although they are in the top list of their own dish group. The menu had traditional menu pattern based on cooked rice, Korean soup, and kimchi. Most frequently used menu patterns were rice+soup+kimchi, rice+noodle (or Mandu)+kimchi, rice+kimchi, rice+soup+kimchi+seasoned-vegetable in the order. These were very simple menu patterns with only 1~2 kinds of side dish. Therefore, it is necessary for middle-aged housewives to consume menu that the composition of main dish and side dish are adequate. This result can be used as basic data for nutrition education programs in middle-aged housewives.

Dietary Intakes and Food Sources of Total Sugars from Korean National Health and Nutrition Examination Survey 2001-2002 (한국인의 총당류 섭취실태와 급원식품에 대한 연구 -2001년과 2002년도 국민건강영양조사 자료를 이용하여-)

  • This study aimed to assess the total sugar intake for Korean and to identify major food sources contributing to those food components. Korean adults aged over 20 years old from the Korean National Health and Nutrition Examination Survey (KNHANES) 2001 and 2002 were selected. The data were analyzed to obtain nationally and seasonally representative information on the health and nutritional status of the Korean. Forty food groups were used in identifying food sources of total sugar and energy intake. Total sugar contents of foods in the KNHANES data sets were estimated by food code matching technique with Release 18 of the USDA National Nutrient Database for Standard Reference. Sample weighted means, standard errors, and population percentages were calculated using SAS and SUDAAN. The mean total sugar intake of the Korean was 60.3g in 2001, 40.9g in spring 2002, 45.7g in summer 2002, and 52.1g in fall 2002, which were 30-44% of intake of US people. Fresh fruit was identified as the most significant food source for total sugar intake in Korean population in all age groups and all seasons. The next major food sources following fresh fruits were candy/jelly/syrup/honey, coffee/coffee caream, vegetables, Kimchi, soft drinks, milk, fruit juice, cookie/cracker/cake, and vegetable juice/grain juice, which showed similar results through the seasons. While carbonated soft drink was the most significant food sources for total sugar or added sugar intakes for US people. The total sugar intakes were significantly higher in women, higher educational level, and residing in metropolitan area. As intake of total sugar increased, intakes of protein, fiber, calcium, phosphorus, iron, Vit A, B1, B2, C, niacin showed significantly increased, while high intakes of added sugars showed low intakes of some micronutrients in the US people. Percentages of people who consumed nutrients below EAR were less in higher total sugar intake group than in lower intake group. From these results, we can conclude that the food consumption habits including the total sugar intake of Korean people seems relatively good so far. More reliable database of total sugar and added sugar composition tables in public domain should be established in the future, and also more researches about total sugar and added sugar for Koreans should be continued.

Food Behaviors and Food Preferences of the Teenagers in Urban, Fishing, and Rural Areas (일부 농어촌과 도시 청소년들의 식행동 특성 및 식품기호도)

  • The purpose of this study was to investigate the food behaviors and food preferences of teenagers by regional groups. The subjects were composed of 365 third grade students in middle schools from urban, rural and fishing areas in Kyungpook province. The data was collected by using a survey questionnaire. The results of the study are summarized as follows: Eating habits of 21-47% of the subjects, according to the regions, were poor and female students in large city showed the best eating habits while female students in fishing village showed the poorest eating habits. Out of the three regional groups studied, the subjects from the fishing villages had the highest rates of skipping meals and eating between meals. Out of all the people investigated, thirty to forty percent had unbalanced diets, the highest percentage coming from the urban areas and the lowest from the fishing villages. All together an allergic reaction to food was the highest in the fishing villages' subjects and female students were more sensitive compared to the male students. Students from the urban area had higher frequency of symptoms of lactose intolerance. Upon the investigation of the subjects 'food preferences, they preferred boiled rice to any other rice as a staple food. Cha jang myun was the most preferred out of all noodles and breads. It was found that teenagers preferred one course meal such as fried rice to a rice-centered regular meal. Out of all various meat soups, beef soup and beef-rib soup were preferred the most. As for subsidiary foods the most preferred were soybean paste stew and cuttlefish stew which they ate daily. There was a very high preference for fried foods, the most favored being pork cutlet followed by fried vegetables, sweet potatoes, and minced meat. The most preferred cooked vegetables were bean sprouts and seasoned cucumber. Seasoned cubed radish roots were the most preferred kind of kimchi. For desserts there was a very high preference for the following fruits: apples, plums, strawberries, and oranges. For beverages the most popular choice were chocolate drinks followed by orange juice, apple juice, milk and tomato juice. According to these investigations, most teenagers liked daily foods which they are used to. The study showed that on the whole the teenagers had poor eating habits. A problem that needs immediate attention is that female students and students from the fishing villages have a high rate of skipping meals. It is expected that the results of this food preference test of teenagers could be useful in the meal planning of middle school lunch programs in the future.

Characterization and Selection of Lactic Acid Bacteria Producing ${\beta}-Galactosidase$ (${\beta}-Galactosidase$ 생산 유산균 선별 및 특성 조사)

  • This study was carried out to select the lactic acid bacteria producing ${\beta}-galactosidase$ (lactase) and investigate the properties of the ${\beta}-galactosidase$. About 100 strains of lactic acid bacteria showing blue colony on the MRS agar medium containing X-gal were isolated from several kinds of Kimchi. Among them, 2 strains were selected as potential ${\beta}-galactosidase$ producers. The selected strains, ET-1 and LA-12, were identified as Lactobacillus fermentum and L. acidophilus, respectively by the analysis of 16S rDNA sequences. They showed relatively high ${\beta}-galactosidase$ activity and cellular viability. Their ${\beta}-galactosidase$ showed the highest activity at $55^{\circ}C$. And the optimum pHs of the enzymes produced by ET-1 and LA-12 were pH 5.5 and pH 7.0, respectively. They were also highly resistant to artificial gastric juice and bile. Two selected strains showed little change of viable cell number for 3 hr incubation in artificial gastric juice, and maintained the viable cell number at $10^8CFU/ml$ for 24 hr in 0.3% oxgall after incubation for 2 hours in artificial gastric juice. Based on these results, ET-1 and LA-12 are expected to be applied in dairy industry.
