Inflammatory mediator regulation of the Zizyphus jujube leaf fractions in the LPS-stimulated Raw264.7 mouse machrophage (LPS로 염증이 유도된 Raw 264.7 대식세포에서 대추(Zizyphus jujube) 잎 분획물의 염증매개물질 억제)

  • Zizyphus jujube leaf fractions (ZLFs) showed no cytotoxic effects of up to $100{\mu}g/mL$, while the anti-inflammatory effects of ZLFs were analyzed by checking the productions of nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), cyclooxygenase-2 (COX-2), and inflammatory cytokines TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 in the lipopolysaccharide (LPS)-stimulated Raw264.7 macrophage up to the concentration of $100{\mu}g/mL$. ZLFs ($100{\mu}g/mL$) demonstrated a strong anti-inflammatory activity that reduced 61~85% of NO and 71~100% of $PGE_2$ production in the LPS-stimulated Raw264.7 macrophage. Even the low ZLFs concentration of $1{\mu}g/mL$ have reduced NO and $PGE_2$ production by 34~64%. Expressions of COX-2 protein were also effectively inhibited by the ZLFs. Furthermore, the TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 production were significantly suppressed through the treatment of ZLFs at concentrations of 1, 10, and $100{\mu}g/mL$. In the order of the Zizyphus jujube leaf water fraction (ZLWF) < buthanol fraction (ZLBF) < ethyl acetate fraction (ZLEF) showed anti-inflammatory activity. In particular, the ethyl acetate fraction ZLEF at $100{\mu}g/mL$ showed an excellent anti-inflammatory activity by reducing the production of NO, $PGE_2$, COX-2, and inflammatory cytokines (TNF-${\alpha}$, IL-$1{\beta}$, IL-6) in the level of Raw264.7 macrophage without LPS-stimulation or even better. The results of our study suggest the potential of ZLEF for use as an excellent ant-inflammatory inhibiting mediator and may be used as a therapeutic approach to various inflammatory diseases.

Crude Extract of Zizyphi Jujube Semen Protects Kainic Acid-induced Excitotoxicity in Cultured Rat Neuronal Cells

  • Zizypus is one of the herbs widely used in Korea and China due to CNS calming effect. The present study aims to investigate the effect of the methanol extract of Zizyphi Jujube Semen (ZJS) on kainic acid (KA)-induced neurotoxicity in cultured rat cerebellar granule neuron. ZJS, over a concentration range of 0.05 to $5\;{\mu]g/ml$, inhibited KA $(500\;{\mu}M)-induced$ neuronal cell death, which was measured by a trypan blue exclusion test and a 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay. Pretreatment of ZJS $(0.5\;{\mu}g/ml)$ inhibited KA$(50\;{\mu}M)$-induced elevation of cytosolic calcium concentration $([Ca^{2+}]_c)$, which was measured by a fluorescent dye, Fura 2-AM, and generation of reactive oxygen species (ROS). ZJS $(0.5\;{\mu}g/ml)$ inhibited glutamate release into medium induced by KA $(500\;{\mu}M)$, which was measured by HPLC. These results suggest that ZJS prevents KA-induced neuronal cell damage in vitro.

Processing of Powdered Jujube Juice by Spray Drying (분무건조에 의한 분말대추음료의 가공)

  • Spray dried powdered juice was processed from concentrated extract of jujube(Zizyphus jujuba MILLER). Spray drying of the extract solution could not be operated to have powder product by itself over whole concentration range and required addition of some carrier or support material. The concentrated extract of 26$^{\circ}$Bx was combined with carrier material solution to have a final concentration of 30$^{\circ}$Bx, and then spray dried. Proper addition level of carrier solid for physical and flavor quality of the powder product was determined to be 1 : 1 ratio to jujube solid. Combined use of maltose and gum arabic produced the best quality product among the studied carrier materials, which were maltose, dextrin, condensed milk and gum arabic. Enzymatic treatment in extraction process could increase the yield by 13~39%, but hurt the sensory quality of powdered juice. Treatment by 0.5% pectinase(0.05 unit/ml) may be used with lesser quality change for improved yield.

Chemical Composition of Some Tropical Foliage Species and Their Intake and Digestibility by Goats

  • The chemical composition and water extractable dry matter (DM) of foliages from Erythrina (Erythrina variegata), Fig (Ficus racemosa), Jackfruit (Artocarpus heterophyllus), Jujube (Ziziphus jujuba Mill), Kapok (Ceiba pentandra) and Mango (Mangifera indica) and the feed intake, digestibility and N retention when feeding these foliages were studied in two experiments. In Experiment 1, 12 male goats, 3.5 months old and weighing 14.6 kg, were randomly allocated to a diet consisting of one of the foliages in a change-over design with three periods. The foliages were offered ad libitum at the level of 130% of the average daily feed intake. The Erythrina foliage had a low content of DM and condensed tannins (CT) and a high concentration of crude protein (CP) in leaves plus petioles (193 g/kg DM) and stem, while the Mango foliage had a low CP (69 g/kg DM) and high DM content. The other foliages were intermediate. High content of CT was found in the leaves plus petioles of Jackfruit foliage and in the stem of Fig and Mango foliage. There was a difference in feed intake, nutrient intake, apparent digestibility and N retention between the foliages, with Erythrina, Jackfruit and Kapok foliage being significantly higher in these parameters than Fig, Jujube and Mango foliage. The water extractable DM could be used to estimate N retention, but not DM digestibility in this study. In Experiment 2, 4 male goats weighing 13.4 kg and 6 months old were allocated to a 4${\times}$4 Latin square design. The treatments were: water spinach ad libitum and Fig, Jujube or Mango foliage ad libitum +0.5% of BW as water spinach DM. Feed intake, apparent digestibility and N retention were not significantly different among the foliage diets, but higher than for water spinach alone (p<0.05). Supplementation with water spinach to a diet consisting of low quality foliages such as Fig, Jujube and Mango, increased DM and CP intake, apparent digestibility and N retention, compared to feeding these foliages as sole feeds.

Modified Atmosphere Packaging of Dry Jujube (건조 대추의 변형기체포장)

  • Effect of modified atmosphere packaging conditions on quality changes of dry jujube was investigated. Dry jujubes with moisture content of 26.7% were packaged in PET/Al/PE film pouches with modified atmospheres. The tested packages include those with normal air, vacuum, CO2 flushing, N2 flushing and O2 scavenger. Packages were stored at 25$^{\circ}C$ for 6 months, during which ascorbic acid concentration, browning level, titratable acidity and surface color were measured. Generally modified atmosphere packages could improve quality retention of dry jujubes except that vacuum package resulted in large surface color change. CO2-flushed package showed the best quality retention of high ascorbic acid content and low browning during 112 days, but caused high amounts of ascorbic acid destruction and browning in longer storage. In all the modified atmosphere packages titratable acidity reached a maximum followed by decline and subsequent rise, while it increased linearly with time in normal air package. Considering ascorbic acid retention browning level and surface color changes during 112 days, the packages of CO2 flushing and N2 flushing were better than others.

Effects of Zizyphus jujuba var. boeunesis Extracts on the Growth of Intestinal Microflora and Its Antioxidant Activities (대추 추출물이 장내 미생물의 생육에 미치는 영향 및 항산화 활성)

  • This study was carried out to investigate the effects of jujube extracts on intestinal microflora, along with their antioxidant activities, according to extraction method. The antimicrobial activities of the extracts were measured using the agar diffusion method with a jujube extract concentration of 50 mg/mL. Neither the first nor second jujube extracts were inhibitory against the tested intestinal bacteria. However, water extracts of jujube significantly enhanced the growth of lactic acid bacteria, especially Bifidobacterium bifidum and Bifidobacterium adolescentis. Total phenol compounds and flavonoid compounds were higher in the 1st than in the 2nd water extracts. The EDA values of both water and ethanol extracts increased in proportion to the extract concentration. The 1st water extract showed the highest value among all the others, which was 85.60% at the concentration of 0.05 mg/mL. Furthermore, the 1st water extract showed stronger antioxidant activity than the other samples with an activity of 679.91 mg AA eq/g. These results support the potential use of jujube water extracts as a functional food component and a valuable resource for the development of nutraceutical foods, to increase the growth of Bifidobacterium spp. in the human intestine.

Processing and Reduction Factors of Pesticide Residues in Chinese Matrimony Vine and Jujube by Drying (구기자와 대추 중 잔류농약의 건조에 따른 가공계수 및 감소계수)

  • This study was carried out to elucidate changes in concentration of pesticide residues in Chinese matrimony vine and jujube during drying. Test pesticides, triadimefon and triforine for chinese matrimony vine and methoxyfenozide and thiacloprid for jujube, which are commonly used for the crops in Korea, were treated to the crops by spraying and dipping. The pesticide residues in both fresh and dried Chinese matrimony vine and jujube were analyzed by using a GLC-ECD and an HPLC-DAD. Processing factors of the pesticides in Chinese matrimony vine and jujube by drying ranged from 1.41 to 2.28 and from 1.50 to 4.20, respectively. And reduction factors of the pesticides in the crops ranged from 0.29 to 0.47 and from 0.68 to 0.98, respectively. These results indicate that concentrations of the pesticides in the test crops increased after drying, while amounts of pesticides in them were diminished by drying.

Study on Practical Micropropagation of Juiube Cultivars through Axillary Bud Culture (액아배양(腋芽培養)에 의한 신품종(新品種) 대추나무의 실용적(實用的)인 대량증식(大量增殖)에 관(關)한 연구(硏究))

  • This study was conducted to establish practical micropropagation of jujube cultivars ('Geumsumg', 'Bokjo') by axillary bud culture. The results are summerized as follows : 1. Addition of activated charcoal to half-strength Murashige and Skoog(MS) medium supplemented with 0.5mg/l benzylaminopurine(BAP) enhanced shoot and root growth. At 500mg/l activated charcoal level 'Geumsung' showed best result, and shoot length and the number of multiple shoot were 6.4cm and 10.0, respectively. At 1,000mg/l activated charcoal level 'Bokjo showed best result, and shoot length and the number of multiple shoot were 7.5cm and 12.4, respectively. 2. As indole-3-butyric acid(IBA) concentration increased, rooting and callus growth of microshoot were enhanced. The optimum IBA concentration for shoot elongation and multiplication was 1.0mg/l. 3. Growth responses of shoot-tip and axillary bud segments between two jujube cultivars were different. 'Geumsung' showed that axillary bud explants were about twice better than shoot-tip explants for shoot multiplication, but 'Bokjo' showed that shoot-tip explants mere better than axillary bud explants for shoot elongation and multiplication. 4. In acclimatization processes of plantlets produced in vitro, the survival of plantlets with only root primordia in soil medium was better than that of plantlets with several routs resulting in 97.8%. 5. In cutting of in vitro-derived microshoot, paclobutrazol was more effective than IBA, naphth-aleneacetic acid(NAA) and $Rooton^{(R)}$ in rooting and root growth.

Optimization of Extraction Conditions for Dried Jujube by Response Surface Methodology (반응표면분석에 의한 건대추의 추출조건 최적화)

  • Extraction characteristics of dried jujube and functional properties of corresponding extract were monitored by response surface methodology. Maximum extraction yield of 53.69% was obtained at extraction temperature of $50.35^{\circ}C$, extraction time of 16.69 hr, and ethanol concentration of 72.88%. At extraction temperature, extraction time, and ethanol concentration of $45.80^{\circ}C$, 15.47 hr, and 73.12%, respectively, maximum cyclic adenosine monophosphate content was 8.20 mg/100 g. Maximum total polyphenol content was 18.85 mg/g at extraction temperature, extraction time, and ethanol concentration of $64.91^{\circ}C$, 20.84 hr, and 66.91%, respectively. Maximum total flavonoid content was 0.48 mg/g at extraction temperature, extraction time, and ethanol concentration of $57.36^{\circ}C$, 15.14 hr, and 71.08%, respectively. $IC_{50}$ value of electron donating ability showed maximum level of 32.34 mg/mL at extraction temperature of $48.46^{\circ}C$, extraction time of 19.25 hr, and ethanol concentration of 65.36%. Maximum ascorbic acid equivalent antioxidant capacity was 3.58 mg ascorbic acid equivalent per gram sample at extraction temperature, extraction time, and ethanol concentration of $56.09^{\circ}C$, 21.86 hr, and 65.36%, respectively.

Effect of Packaging Conditions on Keeping Quality of Fresh Jujube (포장조건에 따른 생대추의 저장성)

  • Two hundred grams of fresh jujubes at whitish green maturity was packaged in the film bags of different gas permeabilities and stored at 0 and $5^{\circ}C$. Through the storage package atmosphere and jujube quality were monitored. Package of $30\;{\mu}m\;CPP$ film resulted in anaerobic conditions of $O_2$ content below 1% and $CO_2$ content above 15%, and caused off-flavor at both temperatures within 4 weeks. Package of $60\;{\mu}m\;LDPE$ film also showed potential risk of anaerobic condition development within 5 weeks, because of high $CO_2$ and/or low $O_2$ concentration at 0 and $5^{\circ}C$. Microperforated film packages could preserve quality of jujubes for 10 and 7 weeks at 0 and $5^{\circ}C$, respectively, which were significantly longer than shelf lives for perforated air pack and hermetically sealed packages.
