새로운 endo-inulinase 생산 균주의 선발 및 효소의 생산 (Production of a novel endo-inulinase from Arthrobacter sp. S37)

  • 토양으로부터 inulin을 기질로 하여 중합도가 큰 oligo당으로 가수분해하는 새로운 endo형 Inulinase 생산균주로, Arthrobacter sp. S37를 분리, 선발하였다. 본 효소의 생산은 inulin과 돼지감자 추출물에 의해 유도되었으며, 탄소원으로 1.5% 돼지감자 추출물, 유기질소원으로 1.0% yeast extract, 무기 질소원으로 0.5% $NaNO_3$를 사용하였을 때 효소 생산이 가장 많았다. 또한 최적 배양 pH와 온도는 각각 pH 8.0과 $30^{\circ}C$로 나타났다. 최적 배지와 최적 배양 조건하에서 배양한 결과 배양 24시간에 10.8 units/ml로 최대효소생산을 보였다.

열처리에 따른 돼지감자 Methanol 추출물의 항산화 및 α-glucosidase 저해 효과 (Antioxidant Activity and α-Glucosidase Inhibitory Effect of Jerusalem Artichoke (Helianthus tuberosus) Methanol Extracts by Heat Treatment Conditions)

  • This study investigated the changes of antioxidant activity and ${\alpha}$-glucosidase inhibitory effect of Jerusalem artichoke (Helianthus tuberosus) 100% methanol extracts by various heat treatment. The contents of total phenolic and flavonoid compounds in methanol extract tended to increased gradually with the rise of temperature to 180$^{\circ}C$. The maximum yield of gallic acid (51.52 ${\pm}$ 2.17mg/g extract weight) and quercetin (13.39 ${\pm}$ 0.03mg/g extract weight) were obtained with extraction temperature of 180$^{\circ}C$ for 120min. In addition, the improving extraction efficiency resulted in the increased biological activities, such as electronic donation ability (EDA, 90.36${\pm}$ 0.57%), reducing power (Abs 1.14) and ${\alpha}$-glucosidase inhibitory effect (92.14 ${\pm}$ 1.14%). Overall, the results of this study indicate that the optimum conditions for the extraction process were an extraction temperature at 180$^{\circ}C$ for 120 min, and will provide the basis for future research on the improving extraction yield of phenolic and flavonoid compounds.

효모 K. marxianus에 의한 돼지감자 착즙에 대한 에탄올 발효 특성 (Ethanol Fermentation Characteristics of K. marxianus on Jerusalem Artichoke tuber Extract)

  • 돼지감자 착즙의 당농도를 50,80,110,190,250g/l로 하고 K.marxianus의 비증식속도,균체수율,알콜수율등을 발효액주으이 생성알콜농도및 당 소모량의 함수로 구명한 결과 다음의 결론을 얻었다. 생성알콜농도에 대한 비증식속도 및 알콜의 비생성속도의 함수관계는 알콜생성량이 낮을때 선형 함수관계를, 알콜생성량이 높을때는 지수 함수관계를 나타내었다. 균체의 증식은 생성알콜농도가 50g/l이상에서, 그리고 당의 소모량이 100g/l이상에서 잔여 당농도에 무관하게 정지되는 경향을 나타내었다. 비증식속도와 당 소모량사이에는 선형 함수관계를 나타내었으며,알콜 비생성속도와 당소모량사이의 함수관계는 초기당농도가 낮을때 선형 함수관계를 나타내었으나 초기당농도가 높아질수록 지수함수관계로 천이되었다. 균체수율및 알콜수율은 초기당농도에 무관하게 생성알콜농도 25g/l 부근에서 최대치를 나타내었으며, 알콜생성량이 증가함에 따라 초기 당농도가 증가할수록 완만하게 감소하였다. 이상의 함수관계로부터 K.marxianus의 알콜발효에 있어 한계 당농도는 110g/l근방으로, 한계 알콜농도는 50g/l근방인 것으로 추정되었다.

Bio-control of Stem Rot in Jerusalem Artichoke (Helianthus tuberosus L.) in Field Conditions

  • Stem rot is a serious disease in Jerusalem artichoke (JA). To reduce the impact of this disease on yield and quality farmers often use fungicides, but this control method can be expensive and leave chemical residues. The objective of this study was to evaluate the efficacy of two biological control agents, Trichoderma harzianum T9 and Bacillus firmus BSR032 for control of Sclerotium rolfsii under field conditions. Four accessions of JA (HEL246, HEL65, JA47, and JA12) were treated or notreated with T. harzianum T9 and B. firmus BSR032 in a 4 × 2 × 2 factorial experiment in two fields (environments), one unfertilized and one fertilized. Plants were inoculated with S. rolfsii and disease was evaluated at 3-day intervals for 46 days. T. harzianum T9 and B. firmus BSR032 reduced disease incidence by 48% and 49%, respectively, whereas T. harzianum T9 + B. firmus BSR032 reduced disease incidence by 37%. The efficacy of T. harzianum T9 and B. firmus BSR032 for control of S. rolfsii was dependent on environments and genotypes. The expression of host plant resistance also depended on the environment. However, HEL246 showed consistently low disease incidence and severity index in both environments (fertilized and unfertilized). Individually, T. harzianum T9, B. firmus BSR032, or host plant resistance control stem rot caused by S. rolfsii in JA. However, no combination of these treatments provided more effective control than each alone.

명월초, 여주 및 울금을 포함한 돼지감자 복합물의 항산화 및 AMPK 활성화를 통한 항당뇨 활성 (Antioxidant and Antidiabetic Activities of Jerusalem Artichoke Composites Containing Gynura procumbens, Momordica charantia, and Curcuma longa via AMPK Activation)

  • 명월초, 여주 및 울금의 물 및 70% 에탄올 추출물에서 항산화 및 ${\alpha}$-glucosidase 저해활성을 측정하였으며, 돼지감자 농축액에 이들 식물류의 에탄올 추출물을 첨가한 5종의 돼지감자 복합물(JA1~5)에서 항산화 및 항당뇨 활성을 비교하였다. 명월초, 여주 및 울금의 에탄올 추출물은 물 추출물에 비해 총 페놀 및 플라보노이드 함량이 많았다. DPPH, ABTS 라디칼 소거활성 및 FRAP법에 의한 환원력은 총 페놀 및 플라보노이드 함량에 의존적이었으며, 특히 명월초와 울금의 에탄올 추출물은 항산화 활성이 비슷한 수준이었다. ${\alpha}$-Glucosidase 저해활성은 에탄올 추출물의 $2,000{\mu}g/ml$ 농도에서 50% 이상이었다. 5종의 복합물은 돼지감자 농축액에 비해 총 페놀 및 플라보노이드 함량이 많았다. 또한 항산화 활성 및 ${\alpha}$-glucosidase 저해활성도 복합물에서 모두 증가되었으며, 특히 JA1의 활성이 유의적으로 높았다. 돼지감자 복합물은 인체 정상 간세포주인 Chang세포에 대해 세포독성이 없었으며, Chang세포에서 AMPK 활성화 및 근육세포인 C2C12세포에서 세포 내 당 유입을 유의적으로 증가시켰다. 이로써 명월초, 여주 및 울금의 추출물이 혼합된 돼지감자 복합물은 항산화 및 항당뇨 활성 증대에 효과적인 것으로 판단된다.

Characterization of Cyclofructans from Inulin by Saccharomyces cerevisiae Strain Displaying Cell-Surface Cycloinulooligosaccharide Fructanotransferase

  • The cycloinulooligosaccharide fructanotransferase (CFTase) gene (cft) from Paenibacillus macerans (GenBank access code AF222787) was expressed on the cell surface of Saccharomyces cerevisiae by fusing with Aga2p linked to the membrane-anchored protein Aga1p. The surface display of CFTase was confirmed by immunofluorescence microscopy and enzymatic assay. The optimized reaction conditions of surface-displayed CFTase were as follows; pH, 8.0; temperature, $50^{\circ}C$; enzyme amount, 30 milliunit; substrate concentration, 5%; inulin source, Jerusalem artichoke. As a result of the reaction with inulin, cycloinulohexaose was produced as a major product along with cycloinuloheptaose and cycloinulooctaose as minor products.

수성이상계 에탄올 추출발효 조건의 최적화에 관한 연구 (Optimization of Conditions for Extractive Ethanol Fermentation in an Aqueous Two Phase System)

  • This study was undertaken with objective of optimizing the conditions of fermentation in an aqueous two-phase system which is composed of polyethylene glycol (PEG) 20000 and crude dextran (Dx). The data were obtained and analyzed using the Box-Wilson's experimental design protocol and the response surface methodology. To reach this end a multilinear polynomial regres- sion model was developed, which can be utilized for the purpose of optimizing the extractive fermentation. Optimum conditions for batch fermentation with aqueous two phase system were found to be at 4.2~5.4% PEG/3.2~4.2% Dx range. The composition of the center was 4.8% PEG/ 3.6% Dx. Optimum operating conditions for initial sugar concentration and fermentation time were approximately 160 g/l, and 21~22 hr, respectively. Fermentation in the aqueous two phase system composed of 5% PEG/4% Dx showed increase of 23% in ethanol concentration, of 9.5% in ethanol yield, and of 19% in ethanol productivity as compared to the case of fermentation of neat Jerusalem artichoke juice.

Kluyveromyces marxianus var. marxianus IFO 1735에 의한 Inulin Fructotransferase의 생산 및 이용에 관한 연구

  • Kluyveromyces marxianus var. marxianus isolated as an inulin-assimilating microorganism produces inulin fructotransferase (inulaseII) which catalyses the conversion of inulin into di-D-fructofuranose 1, 2' : 2, 3' dianhydrde (DFAIII). The DFA produced by the organism was isolated by using active carbon column, and identified as DFAIII by high performance liguid chromatography. The culture medium giving maximum inulaseII production was found to consist of 1% sucrose and 0.75% yeast nitrogen base (YNB). The inulasell production was induced by inulin or sucrose as a carbon source and increased by addition of YNB as a nitrogen source. Optimal initial pH of the culture medium, culture temperature and medium volume for the enzyme production were pH 4.7, 30$\circ$C and 140 ml, respectively. Under the optimal conditions described above, the enzyme activity in the culture supematant reached 4.2 units/ml after cultivation for 36 h. The DFAIII was accumulated at 13.25 mg/ml after 48 h of culture in the Jerusalem artichoke tuber medium.

추출 발효에 의한 알콜 제조 공정개발 -PEG/Dx 최적 이상계의 선정- (Process Development for Alcohol Production by Extractive Fermentation)

  • The quantitative effects of molecular weight and concentrations of two phase-forming polymers-polyethylene glycol and crude dextran on the two phase extractive ethanol fermentation were investigated using a Box-Wilson central composite protocol. The regression model obtained was used in order to determine optimum compositions of aqueous two phase system. In the aqueous two phase extractive ethanol fermentation of Kluyueromyces fragilis CBS 1555 with Jerusalem artichoke juice, it was found from the regression model that the variables influenlcing on ethanol fermentation were PEG concentration, time, Dx concentration, and PEG molecular weight strongly in order. The interaction of PEG concentration and PEG molecular weight was also found, and the effect of PEG concentration decreased with increase in molecular weight of PEG. The ethanol concentration incresed with increase in molecular weight of PEG, and with decrease in concentration of PEG. In conolusion, maximum concentration of ethanol produced was obtained at the following compositions; PEG MW 20000, Dx concentration ranged from 4% to 5%, and PEG concentration ranged from 3% to 7%.

Flavone Glucosides from the Leaves of Helianthus tuberosus

  • Two flavone glucosides have been isolated from the leaves of Helianthus tuberosus (jerusalem artichoke). Their structures were identified as kaempferol 3-O-glucoside (1) and quercetin 7-O-glucoside (2) by spectroscopic analysis and confirmed by comparison with reported data. These flavonoids were isolated for the first time from this plant part.
