A comparative study of radioprotection with Callophyllis japonica extract and amifostine against lethal whole body gamma irradiation in mice

  • The efficacy of the radioprotective effect of Callophyllis japonica ethyl acetate (CJEA) extract was studied by comparing it to that of amifostine, a well-known radioprotective agent, and by evaluating the dose reduction factor, an indicator of radioprotective efficacy. Pretreatment with CJEA extract (100 mg/kg body weight) prior to receiving 12 Gy irradiation significantly improved the survival of jejunal crypts at 3.5 day post-irradiation, but attenuated the level of malondialdehyde compared to vehicle alone (P < 0.01). A similar gastroprotective effect was also obtained in the amifostine-treated irradiated group (P < 0.01). The efficacy of the radioprotective effect was further confirmed by the dose reduction factor, 1.41. Collectively, these results suggest that CJEA extract is a useful radioprotectant whose efficacy is similar to that of amifostine and whose radioprotective mechanism is in part the reduction of lipid peroxidation caused by gamma irradiation.

Reconstruction of Hypopharynx and Cervical Esophagus : Choice of Flap (하인두 및 경부식도 결손의 재건 : 재건술의 선택)

  • Background and Objectives: Various flaps are using for reconstruction of hypopharyngeal and esophageal defect. However, complication and indication of each flap are not fully analyzed. Patient and Methods: Records of 52 hypopharyngeal cancer patients who had surgical treatment and 13 other head and neck cancer patients who underwent hypopharyngeal and/or esophageal reconstruction with flap were retrospectively analyzed. Eighty three percent(54 cases) of patients needed reconstruction other than primary pharyngeal closure. Five split thickness skin graft, 1 pectoralis major myocutaneous flap, 20 forearm free flap, 13 jejunal free flap, 15 gastric pull up were used. Result: Flap failure was noted in 2 cases who had subsequent gastric transposition. Wound dehiscence and fistula were most common problem of forearm free flap. Most fistulas were developed in patients with conduit type reconstruction of forearm flap while there wasn't any fistula in patient with patch type reconstruction. Stenosis of lower anastomosis was the frequent problem of jejunal transfer. Gastric pull-up has frequent com-plication of stomal stenosis. All but three patients had reached oral feeding postoperatively. Conclusion: Based on this study, forearm flap is effective in partial hypopharyngeal defect while jejunum is the choice for circumferential defect. Gastric pull-up is for combined esophageal defect.

Study on Prevention of Adhesion Formation by Use of Sodium Carboxymethylcellulose and Dextran 70 II.   • The effectiveness of sodium carboxymethylcellulose(SCMC) and dextran 70 in the prevention of adhesion formation in abdominal cavity of rabbits following artificial injuries was elucidated and the effects -of these medicines on blood pictures were also examined. After abrasion treatment on jejunum in gonadotropins primed rabbits, 1, 2 and 3% of SCMC solutions, 6 and 10% dextran 70 solutions and a synthetic solution of 1% SCMC and loft dextran 70 in 0.9% saline solution were infused into the abdominal cavity. Four weeks later the abdominal cavities were reopened under general anesthesia. The synthetic solution showed the highest adhesion reduction rate(100%), while 1% SCMC, 6 and loft dextran solutions showed no significantly evident effect of adhesion prevention. The SCMC solutions showed better adhesion reduction effect than dextran 70 solutions. After infusion of these adhesion preventive medicines, the changes of total leucocytes, erythrocytes, lymphocytes, PCV, plasma fibrinogen and protein contents were examined. No remarkable difference in blood pictures was shown between the synthetic solution and the other medicines. Therefore, it can be suggested that the synthetic solution of 1% SCMC and 10% dextran 70 in 0.9% saline solution at dose of 5$m\ell$/kg of body weight is prominently effective in the prevention of postoperative adhesion formation and wolf be safe in animals and human.

Effects of Supplemental Liquid DL-methionine Hydroxy Analog Free Acid in Diet on Growth Performance and Gastrointestinal Functions of Piglets

  • This study was conducted to determine the effect of dietary supplementation of liquid DL-methionine hydroxy analog free acid (DL-MHA) on growth performance and gastrointestinal conditions of piglets. One hundred and eighty crossbred barrow piglets (Large White${\times}$Landrace, body weight: $12.48{\pm}0.33kg$) were divided into three groups with ten replications of six piglets each. Piglets received DL-MHA in diet at a concentration of 0 (control group), 0.15%, or 0.24%. The results indicated that increasing the standardized ileal digestible (SID) of sulfur amino acids (SAA) to lysine (SID SAA:Lys) ratio by supplementation of DL-MHA tended to increase (quadratic; p<0.10) weight gain and ADG, and showed slightly greater (linear; p<0.10) gain:feed ratio. The pH in the diet and cecum linearly decreased (p<0.01), whereas pH in colon had a quadratic response (p<0.01) with increasing supplementation of DL-MHA. By greater supplementation of DL-MHA, the population of Lactobacillus spp. in rectum was likely to increase (quadratic; p<0.10), but Escherichia coli population in the diet was reduced (quadratic; p<0.05). Acetic acid concentration and total short-chain fatty acids in cecum linearly increased (p<0.05), whereas valeric acid in cecum quadratically increased (p<0.05) with increasing DL-MHA levels. Moreover, the villous height of the jejunum quadratically increased (p<0.01) as the supplementation of DL-MHA was increased. It is concluded that the addition of DL-MHA in diet improved the growth performance and the morphology of gastrointestinal tract of piglets.

Chemical and Physical Predictors of the Nutritive Value of Wheat in Broiler Diets

  • The aim of this study was to establish relationships between chemical and physical parameters of wheat with performance and digestibilities of feed components in broiler chickens fed on wheat-based diets. Ninety-four wheat samples were selected for inclusion in four bird trials. Birds were housed in individual wire metabolism cages from 7 to 28 d and offered water and feed ad libitum. Dry matter intake (DMI), liveweight gain (LWG) and gain:feed were measured weekly. A balance collection was carried out from 14 to 21 d for determination of apparent metabolizable energy (AME), ME:gain, dry matter retention, oil and neutral detergent fibre (NDF) digestibility. At 28 d the birds were humanely killed, the contents of the jejunum removed for determination of in vivo viscosity and the contents of the ileum removed for determination of ileal dry matter, starch and protein digestibility. When wheat parameters were correlated with bird performance data, it was found that specific weight was not significantly (p>0.05) related to bird performance. Bird DMI, LWG and gain:feed were best correlated (p<0.05) with the rate of starch digestion, although the coefficients of correlation (r) were still low (0.246 to 0.523). A negative relationship (p<0.01) between AME and total (r = -0.432) and soluble (r = -0.304) non starch polysaccharide (NSP) was observed in this study. Thousand grain weight (TG) was positively correlated with DMI (r = 0.299), LWG (r = 0.343) and gain:feed (r = 0.371). When establishing multiple regression relationships, correlation coefficients greater than 0.8 were achieved for DMI, LWG, gain:feed and ileal crude protein digestibility. However, the economics involved in determining the parameters involved in the regressions make the process impractical.

Effects of age on intestinal phosphate transport and biochemical values of broiler chickens

  • Objective: The objective of this experiment was to characterize the mRNA expression profile of type IIb sodium-inorganic phosphate cotransporter (NaPi-IIb) and the biochemical values of serum alkaline phosphatase (AKP), calcium, inorganic phosphorus, tibial ash and minerals of broiler chickens with aging. Methods: A total of 56 one-day-old Arbor Acres male broiler chickens were used. Broiler chickens were weighed and samples were collected weekly from day 1. Results: The result showed that before the growth inflection point, ash, calcium, and phosphorus content in the tibia of broiler chickens increased with growth (before 3 weeks of age), although there were no significant differences in chicks at different ages in the later period of the experiment and weight gain rate was relatively slow at this stage (4 to 6 weeks). NaPi-IIb gene expression in the small intestine in the early growth stage was higher than that in the later growth stage. Expression of calbindin and the vitamin D receptor protein in the intestinal mucosa increased with age in the duodenum and jejunum. Serum AKP activity first increased and subsequently decreased after peaking at 1 week of age, but there was no significant difference after 3 weeks of age. Conclusion: These results show that compared with the early growth stage, the weight-gain rate of broiler chickens in the late growth stage gradually decreased with gradual tibia maturation, along with weaker positive transport of phosphorus in the intestine and reinforced re-absorption of phosphorus in the kidney, which might be the reason that phosphorus requirement in the late growth stage was decreased.

Study on Immunological Response on Salmonella gallinarum in Immunosuppressed Chickens (면역억압계군에서 Salmonella gallinarum 감염에 대한 면역반응)

  • The immune response against Salmonella gallinarum infection was investigated in immunosuppresed chickens. Newly hatched chickens were treated with cyclophosphamide at the first and second day after birth and were challenged intraperitoneally with S gallinarum ($1{\times}10^7CFU/m{\ell}$) on day 6. Group 1, Immunosuppresed and Challenged group, was treated with cyclophaiphamide and challenged with S gallinarum; group 2, Immunosuppressed group, was only treated with cyclophsphamide; group 3, Challenged group, was only challenged with S gallinarum; group 4, Control group. In each group, the localization of lymphocytes of the lymphoid organs and intestine was immunohistochemically compared using a variety of monoclonal antiboies ($CD4^+$, $CD8^+$, and B lymphocyte). Also, S gallinarum were assessed by Maccallum-Goodpasture stain and immunohistochemical analysis in the paraffin-embedded intestinal tissues. In S gallinarum challenged chickens, $CD4^+$ and $CD8^+$ lymphocytes of the intestinal organs such as duodenum, jejunum, ileum and colon were increased. However, in cyclophophamide treated chickens, $CD4^+$ and $CD8^+$ lymphocytes and especially B lymphocytes of the lymphoid organs such as thymus, spleen, and bursa of Fabricius were dramatically decreased. These results suggest that cyclophsophamide is an immunosuppressive agent that especially causes depletion of B lymphocytes, suppress humoral immunity and eventually suppresses avian immune responses. Its protection against S gallinarum infection is mainly dependent on both cell-mediated mechanism and the humoral immune response.

  • As a series of studies to investigate the effect of immunosuppression on Ascaris suum infection in undefinitive hosts, and a delicate relationship between host and parasite, in the present studies, SPF ICR mice were alloted to experiment 1(normal undefinitive host group) and experiment 2(immunosuppressive group treated with prednisolone acetate) and inoculated with a single dose of 1,100 embryonated A suum eggs. In normal group, the infection essentially terminates 4 days after inoculation(DAI) with the attainment of middle third-stage in the liver, although few larvae migrate to the lungs where a few advance to late third stage. In immunosuppressive group, significant numbers developed to late third-stage in liver 8 DAI. In general, increasing of the mast cells and the goblet cells in the jejunum mucosa, of T-cells in the spleen and of activity of peritoneal macrophages followed by expulsion of the worms in the both groups. Considering a series of the results, suitabilities for the host of the worm appeared the highest from rabbit, hamster and mouse in that order. In addition, patent infection of A suum in the mice was also not obviously observed in spite of immunosuppression by prednisolone acetate.

약용식물 추출물의 음수급여가 육계 생산성에 미치는 영향

  • This experiment was conducted to study the effects of medicinal plant extract on performance in broiler chickens. A total of eight hundred forty, 1-d-old broiler chicks(Ross) were randomly allocated to 7 treatments with 4 replicates of 30 birds per replicate. The treatments were negative control (NC, diets without antibiotics), positive control (PC, diets with-containing antibiotics), Schisandrae fructus (T1), Scutellariae radix (T2), Camellia sinensis (T3), Artemisia capillaris (T4) and mixed medicinal plants (T5) extracts added to drinking water. The final body weight, body weight gain and feed conversion rate were significantly improved in all treatments compared to NC (P<0.05). No significant differences were observed in the relative weights of liver, spleen, pancreas, bursa of Fabricius and intestinal tract among the groups. The relative lengths of duodenum, jejunum and ileum were significantly decreased in PC compared with other groups (P<0.05). No significant differences were observed in the cecal microflora. The contents of triacylglycerol (TG), blood urea nitrogen (BUN), glutamic-oxaloacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) in blood serum were not significantly influenced. The concentration of cholesterol in blood serum was significantly decreased in all treated groups as compared to those of the control (P<0.05). There was no difference in the components of leukocytes and erythrocytes among the groups.

Traumatic Diaphragmatic Hernia: A Report of 5 Cases (외상성 횡경막 탈장: 5례 수술 보고)

  • Five cases of traumatic diaphragmatic hernia were repaired in the Department of Thoracic Surgery, Seoul National University Hospital, during the period from 1967 to 1974. The first case, a 14-year aid girl, was diagnosed as diaphragmatic hernia during laparotomy because of jejunal perforation 3 days after traffic accident. Herniated stomach, transverse colon, spleen and left lobe of the liver were repositioned and the diaphragmatic rupture on left posterolateral portion was repaired with two layers of nonabsorbable sutures by transthoracic approach. The second case, a 26-year old man,was diagnosed immediately after traffic accident at a local clinic and transferred to this hospital 24 hours later. Herniated stomach, transverse colon and jejunum were repositioned amd diaphragmatic rupture,about 9 cm in length,from the posterolat.edge to the base of pericardium was sutured in two layers. The third case, a 26-year old man who had stab wound on the left lower lateral chest two years ago,was admitted with sudden abdominal pain and vomiting. Upper gastrointestinal series with barium meal revealed diaphragmatic hernia. The herniated stomach and transverse colon through the defect,about 3.5cm in diameter, at anterolateral portion on the left side,were repositioned and repaired with two layers of nonabsorbable sutures. The forth case, a 26-year old man, sustained blunt trauma to the chest by a roller and was transferred to the emergency room complaining of dyspnea 40 minutes after the accident. The diaphragmatic rupture extended from left midaxillary line to contralateral anterior axillary line,about 20cm long, at anterior portion of diaphragm, which was repaired with two layers, of nonabsorbable sutures. The fifth case, a 4-year old girl, had two separate diaphragmatic ruptures on both sides, which were caused by traffic accident. Immediate upper gastrointestinal series after injury showed herniated stomach, colon and spleen into left Chest cavity. Another small rupture with anterior edge of right lobe of the liver in chest cavity was noted. These were repaired with non-absorbable sutures via thoracotomy.
