Review of Experimental Tracheal Reconstruction (실험적 기관 재건술에 대한 고찰)

  • The purpose of this study was to review the literatures of experimental tracheal reconstruction. Although there have been significant advancements in the surgical treatment of the long circumferential tracheal injuries, there still has been a difficult problem with high morbidity and mortality. The method for tracheal reconstruction after circumferential resection is preferred end-to-end anastomosis for defects up to 6 cm in length, but larger tracheal defects require the use of tracheal allograft, various artificial prosthesis or autogenous organs. The tracheal allotransplantation has been widely used as there was significantly improved the method of surgical technique, preservation and immunosuppression. But it has been limited by a number of factors such as few donor, limited use of immunosuppressant, delayed revascularization and re-epitheliazation. Experimental studies on the tracheal prosthesis have a long history and they tried to use silicone, polytetrafluoroethylene, polypropylene mesh, Dacron, Marlex mesh, external or internal stents. Other experimental studies were reported the use of autogenous tissues that were cartilage. jejunum, aorta, skin, muscle, periostium or esophagus. But a great variety of these protheses have been resulted unsatisfactory in a significant Proportion of cases. Alternatively, the tissue-engineering technique has showed a new approach to reconstruct trachea and much progress in tissue-engineering bas been made recently. In conclusion, although the tracheal allotransplantation and the use of prosthesis and allograft have been reported a lot of limitation to overcome, we could sooner expect good result of ideal tracheal prosthesis.

Recovery, growth and development of Macroorchis spinulosus in albino rats

  • The developmental features, growth and organogenesis of Macroorchis spinuiosus were observed in albino rats. Globular and thick walled metacercariae, possessed a stylet, Y-shaped excretory bladder and extracecal testes. Un albino rats, M. spinulosus showed habitat shifting. The majority of M. spinulosus reside in the jejunum for the first four days post Infection (p.i.) and migrate to the duodenum at the later stage of infection. M. spinulosus grew rapidly during the first four days and reached full maturity at 14 days p.i. and later reduced in size. The ovary was separated from the genital primodium at one day p.i. The seminal vesicle appeared on the third day and divided into two sacs on the fourth day p.i. and intrauterine eggs and sperm mass were produced on the fourth day. Organogenesis and enlargement of reproductive organs governed the growth of M. spinulosus. The similarity of related species of the genus Macroorchis to M. spinulosus was discussed In consideration to developmental features.

Intestinal pathologic findings at early stage infection by Centrocestus ormatus in albino rats (가시입이형흡충에 감염된 흰쥐의 초기 장병변)

  • This study was performed to observe intestinal patholoay ill early Infection by Centrocestus crnlctus. The flukes were in the lowermost part of the intfrvillous space of the duodenum and jejunum from 1 day to 7 days postinfection (PI). Thr stroma of villi around the young fluke was edematous and infiltrated by inflanlrllatory knells Luch as Iymphocytes, plasma cells and eosinophils. The crypt became mildly llyperplastic and villi were moderately atrophied at 4 days Pl. The intestinal lesion produced was confillrd to the areas around the fluke. The pathologic findings were not significarltly different between 1,000 and 5,000 metacercariae infection groups. It is suggested that the lesion should be produced by mechanical destruction of the fluke on the enteroepithelial cells.

Effect of amitraz on intestinal contractility (장평활근의 수축성에 대한 amitraz의 영향)

  • Amitraz frequently causes the side effect of intestinal stasis or bloat in mammals. It is very similar to the side effect of xylazine or clonidine which produce the inhibition of intestinal motility through the stimulation of ${\alpha}_2$ adrenoceptor. Therefore, we examined whether amitraz causes intestinal stasis or bloat through the inhibition of intestinal motility or whether amitraz produces the inhibition of intestinal motility through the stimulation of ${\alpha}_2$ adrenopceptor. Amitraz inhibited the intestinal motility in a dose-dependent manner in isolated rabbit jejunum and isolated pig ileum. These inhibitory effects of amitraz were blocked by yohimbine but not by prazosin. The effect of intestinal contraction of carbachol or high-potassium was not affected by the pretreatment of amitraz. However, the con-traction of histamine was inhibited by the pretreatment of amitraz. It is concluded that amitraz mainly inhibits the intestinal motility through the stimulation of ${\alpha}_2$-adrenoceptor although partially antihistaminic action of amitraz can be involved.

Abdominal Cystic Lymphangioma (복부 임파관종)

  • Abdominal cystic lymphangiomas arising from the mesentery, omentum and retroperitoneum are rare and occasionally confused with other cause of acute abdomen. Sixteen children who underwent surgery for abdominal cystic lymphangioma between 1984 and 2005 at the Division of Pediatric Surgery, Keimyung University Dongsan Medical Center were evaluated retrospectively. There were 9 boys and 7 girls. Age ranged from 12 days to 13 years (mean age: 4.7years). The cysts were located in the omentum (4 cases),mesentery (9 cases: jejunum: 3, colon: 6). retroperitoneum (2 cases) and pelvic cavity (one case). The cyst content was hemorrhagic (8 cases), serous (5 cases), and inflammatory (2 cases), and chyle (one). Prenatal diagnosis was made in 3. The clinical features were variable, but abdominal pain, mass, and abdominal distention in order. There were 3 emergency operations in patients with complicated cyst, who were suspected of having panperitonitis and volvulus preoperatively. Complete excision was accomplished in all cases. There was one mortalityin a newborn with E.coli sepsis. Intestinal obstruction developed in 2 cases in long-term follow-up. No recurrence was observed. Although intraabdominal lymphangioma arising from mesentery, omentum and retroperitoneum are rare, it should be considered as a possible cause of acute abdomen.

Development of Gut Microbiota in a Mouse Model of Ovalbumin-induced Allergic Diarrhea under Sub-barrier System

  • This study aimed to present a mouse model of ovalbumin (OVA) induced allergic diarrhea under a sub-barrier system and investigate the development of gut microbiota in this model. Male BALB/c mice were systemically sensitized with OVA or sham-sensitized with saline, and followed by oral OVA intubation, leading to OVA-specific acute diarrhea. Compared with sham-sensitized mice, sera OVA-specific IgG1 and total IgE in OVA-sensitized mice were dramatically elevated, and the number of mast cells was greatly increased in the jejunum of the OVA-sensitized mice. Principle component analysis of the DGGE profile showed that samples from group of OVA-sensitized mice and group of sham-sensitized mice were scattered into two different regions. Real-time PCR analysis showed that the number of 16S rRNA gene copies of Lactobacillus in the colon of OVA-sensitized mice decreased significantly, while there was no significant difference in the number of Bifidobacterium and total bacteria. In conclusion, OVA-specific allergic diarrhea was successfully induced under a sub-barrier system, and changes of allergic reactions during induction was coupled with changes in gut microbiota, especially the number of colonic Lactobacillus, but the role of gut microbiota in the development of food allergic reactions needs to be further evaluated.

Studies on the Bioavailability of Berberine Preparations(II) : Antibacterial Activity and Bioavailability of Coprecipitate of Coptidis Rhizoma and Glycyrrhizae Radix (베르베린 제제의 생체이용율에 관한 연구(II): 황련과 감초 공침물의 항균효과 및 생체이용율)

  • Precipitation reaction occured between berberine in Coptidis Rhizoma and glycyrrhizin in Glycyrrhizae Radix when they were boiled together in aqueous solution and the supernatant solution thus obtained did not show any antibacterial activity which was derived from berberine. The content of berberine in BG and CGP by HPLC analysis were 41.1%, 8.3% respectively. BG was occured mostly at pH 5.0. The solubility of berberine was 0.15%, while that of BG and CGP was 0.07%, 0.12%, respectively. CGP shown more increased antibacterial activity to gram positive bacteria, S. dysenteriae and K. pneumoniae than berberine. The absorption rates of CGP in stomach, duodenum and jejunum of rats were compared with those of Coptidis Rhizoma water extracts (CR), which were increased more than CR. The time required for the maximum serum concentration of berberine from CGP in mice was 90 minutes after oral administration. The maximum serum concentration of berberine from CGP was higher than that from CR. The dissolution of CGP was increased more than berberine and BG in both artificial gastric and intestinal fluids. The dissolution of CGP pill made from gelatin was 63.4% in artificial gastric fluids and that made from CMC was 76.0% in artificial intestinal fluids.

Sea Tangle Supplementation Alters Intestinal Morphology in Streptozotocin-induced Diabetic Rats and Lowers Glucose Absorption

  • This study examined whether dietary supplementation with sea tangle alters the intestinal morphology of streptozotocin-induced diabetic rats and affects the glucose absorption rate. Forty male Sprague-Dawley rats were divided into 2 groups and fed either a control (AIN76-based) diet or a sea tangle-supplemented diet. After 3 weeks, 10 rats in each group received an intramuscular injection of streptozotocin (45 mg/kg BW), and feeding was continued for 3 additional weeks. Dietary supplementation with sea tangle resulted in a lower fasting plasma glucose level compared with the control diet in diabetic rats. Scanning electron micrographs revealed serious damage to the jejunal villi of diabetic rats fed the control diet, whereas supplementation with sea tangle alleviated the damage. In a separate experiment, 20 male Sprague-Dawley rats were divided into 2 groups and fed either a control diet or a sea tangle-supplemented diet for 5 weeks, and fasted rats were subjected to in situ single-pass perfusion. The glucose absorption rate determined in the absence of digesta was decreased by 34% in the jejunum of rats fed a sea tangle diet compared with those fed a control diet. In conclusion, sea tangle supplementation lowered glucose absorption rate, altered intestinal morphology, and appeared to protect villi from damage caused by diabetes mellitus.

Reduction of Postoperative Adhesions by a Synthetic Solution of Sodium Carboxymethylcellulose and Dextran or Interceed Following Intestinal Anastomosis in Dogs (개의 장문합술에 있어서 Sodium Carboxymethylcellulose와 Dextran의 합제 및 Interceed)

  • The effectiveness of a synthetic solution of sodium carboxymethy1cellulose (SCMC) and dextran in the prevention of adhesion formation following intestinal anastomosis of intestine in dogs was elucidated and compared with Interceed. Following induction of adhesion on jejunum of dogs by resection and intestinal anastomosis, the dogs were infused with a synthetic solution of 1% SCMC and 10% dextran in saline in the abdominal cavity at the dose of 5ml/kg of body weight or treated with Interceed membrane. Fourteen days after operation, all the dogs showed adhesion of anastomotic sites to surrounding tissues. The average score of adhesion was 0.50, 0.60 and 0.80 in the dogs infused with the synthetic solution, Interceed membrane and saline solution, respectively. The synthetic solution showed better result of adhesion prevention than Interceed or saline solution. No significant change of body weight, vital signs, hematological values and side effects after operation was found in each group. Therefore, a synthetic solution which is combined with 1% SCMC and 10% dextran in saline can be effectively used for the prevention of adhesion formation comparable to Interceed membrane following intestinal anastomosis in the dogs.

Pathological changes on naturally occuring necrotic enteritis in chicken (닭의 괴사성 장염에 대한 병리학적 연구)

  • From January of 1991 to December of 1992, 42 chickens collected from 21 poultry farms and also diagnosed as necrotic enteritis(NE) was examined clinical signs, gross and histopathological findings. Main clinical signs were characterized by decreased appetite, mild to severe depression, reductance to move, ruffled feathers, greenish to yellow-browinish diarrhea sometimes including blood. As progressed, diseased chickens showed feces mixed with necrotic debris which detached from the intestinal mucosa and mostly resulted in the death. In chronic cases, there were dirty feathers around cloaca due to diarrhea and notably retarded growth. Principle gross lesions were usually confined to the jejunum and ileum, especially toward the lower part of Meckel's diverticulum. The part of small intestine was frequently distended with gas, and also showed mucosal congestion and hemorrhages with varying degrees. Sometimes, the intestinal mucosa was thickened, and also covered with fibronecrotic psuedomembrane. In addition, there were focal necrosis and severely multifocal ulcreation in the mucosa of small intestine. Major histopathological findings included villous necrosis and erosion of the small intestine covering with lots of bacterial colonies, inflammatory cell infiltration in the lamina propria, and dilatation and hyperplasia of crypts. Luminal exudate contained bacterial colonies, fibrin, erythrocytes, and desquamated epithelium. Thirteen(61.9%) out of 21 NE-occurring farms were complicated with intestinal coccidiosis.
