Novel Method for Urinary 1-Hydroxypyrene Measurement Using Molecular Imprinting (분자주형을 이용한 요중 1-hydroxypyrene의 측정 방법 개발)

  • This study was performed to determine whether or not urinary 1-hydroxypyrene (1-OHP) levels can be accurately detected by our 1-OHP-detecting $TiO_2$-Bead-HPLC assay that we developed based on the molecular imprinting method. Our method showed a variation coefficient of 4.97% and a between-day variation coefficient of 4.43%, suggesting that this may be a very stable method. In addition, the recovery rate of 1-OHP from a mixture of 1-OHP and similar substances using our $TiO_2$-Bead-HPLC method was estimated to be 105.6%. The correlation coefficient between the conventional enzyme-HPLC method and this new method was 0.74 (p<0.01) when the urine samples were tested. Based on this result, it is conceivable that our method could be a useful technique for measuring urinary 1-OHP levels. Moreover, our method has some advantages of being easier and less expensive than the conventional method. The results of this study suggest that our method can facilitate the development of a urine 1-OHP sensor using $TiO_2$-coating beads and that development of beads by molecular imprinting can be applied to analysis of chemicals other than 1-OHP.

Alterations of Heart Rate Variability upon β3-Adrenergic Receptor Polymorphism and Combined Capsaicin, Sesamin, and L-Carnitine in Humans (복합 캡사이신, 세사인, 그리고 카르니틴과 베타3 유전자 다형에 대한 심박수 변이성의 영향)

  • We investigated whether 1) the combined capsaicin (75 mg), sesamin (30 mg), and L-carnitine (900 mg) (CCSC) ingestion enhances autonomic nervous system (ANS) activities including thermogenic sympathetic activity as energy metabolic modulator, 2) ${\beta}_3-AR$ polymorphism of each subject influences with ANS activity. Seven healthy males $(22.0{\pm}0.5\;yr)$ volunteered for this study. The cardiac autonomic nervous activities evaluated by means of heart rate variability of power spectral analysis were continuously measured during 5 min every 30 min for total 120 min resting condition with CCSC or placebo oral administration chosen at random. The results indicated that, there are not $Arp/Arg^{64}$ variants of the ${\beta}_3-AR$ genotypes in our subjects. There were not also significant differences in heart rate during rest between both trials. The difference of ANS activity did not reach the statistical significance between both trials. However, the significant improvement showed TOTAL power, HF component, and the indices of SNS and PNS activities before and at 30 min after CCSC ingestion (p<0.05, respectively). In conclusions, although each component of combined CCSC is associated with lipolysis and/or fat oxidation, the combined CCSC consumption is not influenced in stimulation of thermogenic sympathetic activity as modulator of energy metabolism. In rather, our results suggested that CCSC ingestion improves the balance of both SNS and PNS activities. Therefore, it will be considered many combined nutrient components for ergogenic and/or lipolysis effects as well as genetic variants affecting ANS activity in further studies.

Physiological and Biochemical Characterization of Bacillus spp. from Polychaete, Perinereis aibuhitensis (갯지렁이(Perinereis aibuhitensis)에서 분리한 Bacillus spp.의 생리생화학적 특성 분석)

  • This study compared the characteristics of five Bacillus strains capable of aerobic and anaerobic growth, CBW3, CBW4, CBW9, CBW14 and EBW10. They were isolated and selected from a polychaete, Perinereis aibuhitensis, which is known as a good degrader of organic compounds in marine wetland. Based on a 16S rRNA sequence, CBW3 and CBW14 were found to share more than 99.8% similarity with B. nanhaiensis, B. arsenicus and B. barbaricus. CBW4, CBW9 and EBW10 shared 92.7%, 99.8%, and 99.8% similarity with B. anthracis, B. algicoa and B. thuringiensis, respectively. The temperature, salinity, and pH ranges of the cell growth of the Bacillus strains were $4-45^{\circ}C$, 0-17%, and pH 5-pH 9, respectively. All Bacillus strains were found to exhibit enzyme activities for the degradation of casein and starch. Notably, strain EBW10 exhibited the enzyme activities for all the tested macromolecules, DNA, casein, starch, cellulose, and four kinds of Tweens, which suggests the possibility that it had protease, amylase, cellulose, and lipase. All five Bacillus strains had alkaline phosphatase activities, and the strains CBW3, CBW4, and EBW10 also had acid phospatase. Strains CBW3 and EBW10 exhibited the enzyme activities both for esterase (C4) and esterase lipase (C8). The analysis of fatty acids revealed that in all strains, major fatty acids were anteiso $C_{15:0}$ and iso $C_{15:0}$.

Analysis of Physiological Activity and Cytotoxicity of Fermented and Hot Water Extracts Using Residues after Onion Harvest (양파 수확 후 잔재물을 이용한 발효 및 열수 추출물의 생리활성과 세포독성 분석)

  • In order to utilize the residue that is thrown away after an onion harvest, we analyzed the physiological activity and cytotoxicity of fermented and hot water extracts of the residue. The pH of the extracts were all acidic, and organic matter content was 0.75% in the fermented extract and four times more than 0.19% in the hot water extract. The contents of nitrogen, phosphoric acid, calcium, and magnesium components, except for the potassium component among macroelements, were higher in the fermented extract than in the hot water extract. The content of iron and silicon among the micro-elements was also higher in the fermented extract than in the hot water extract. In addition, the content of boron was higher in the hot water extract than in the fermented extract. The total polyphenol contents of the fermented and hot water extracts were $16.2{\pm}3.3mg{\cdot}g^{-1}$ and $14.6{\pm}1.4mg{\cdot}g^{-1}$, respectively, which was $1.6mg{\cdot}g^{-1}$ higher in the fermented extract than in the hot water extract. However, the total flavonoid contents of the fermented and hot water extracts were $0.1{\pm}0.1mg{\cdot}g^{-1}$ and $4.8{\pm}0.7mg{\cdot}g^{-1}$, respectively, which was $4.7mg{\cdot}g^{-1}$ higher in the hot water extract than in the fermented extract. DPPH and ABTS radical scavenging ability for antioxidant activity were higher in the hot water extract than the fermented extract. The cytotoxicity of the extract using MTT assay showed cell viability of 101.6% and 97.9% in the fermented and hot water extracts, respectively. It was confirmed that there was no cytotoxicity in either extract.

Pelvic MRI Application to the Dosimetric Analysis in Brachytherapy of Uterine Cervix Carcinoma (자궁경부암의 강내조사치료에 있어서 흠수선량평가시 골반강 자기공명사진의 응용)

  • Purpose : Before we report the results of curative radiotherapy in cervix cancer patients, we review the significance and safety of our dose specification methods in the brachytherapy system to have the insight of the potential Predictive value of doses at specified points. Matersials and Methods : We analyze the 리5 cases of cervix cancer patients treated with intracavitary brachytherapy in the lateral simulation film we draw the isodose curve and observe the absorbed dose rate of point A, the reference point of bladder(SBD) and rectum(SRD). In the sagittal view of Pelvic MRI film we demarcate the tumor volume(TV) and determine whether the prescription dose curve of point A covers the tumor volume adequately by drawing the isodose curve as correctly as possible. Also we estimate the maximum Point dose of bladder(MBD) and rectum(MRD) and calculate the inclusion area where the absorbed dose rate is higher than that of point A in the bladder(HBV) and rectum(HRV), respectively. Results : Of forty-five cases, the isodose curve of point A seems to cover tumor volume optimally in only 24(53%). The optimal tumor coverage seems to be associated not with the stage of the disease but with the tumor volume. There is no statistically significant association between SBD/SRD and MBD/MRD, respectively. SRD has statistically marginally significant association with HRV, while TV has statistically significant association with HBV and HRV. Conclusion : Our current treatment calculation methods seem to have the defect in the aspects of the nonoptimal coverage of the bulky tumor and the inappropriate estimation of bladder dose. We therefore need to modify the applicator geometry to optimize the dose distribution at the position of lower tandem source. Also it appears that the position of the bladder in relation to the applicators needs to be defined individually to define 'hot spots'.

A Survey on the Perceptions of Consumer Organizations to Promote Risk Communication for Food Additives (식품첨가물의 리스크 커뮤니케이션 촉진을 위한 소비자단체의 인식조사)

  • Consumer organizations usually lead the opinions of the consumers. This study was performed to investigate the perceptions and information needs of consumer organizations on food additives for the promotion of risk communication. A survey was conducted for 4 weeks in March 2012 by using a self-administered questionnaire consisting of 32 questions. Total 111 responses were collected for analysis. 'Safety (91.0%)' was the most important factor influencing the purchase of processed food, and both 'food additives (24.3%)' and 'environmental pollution (23.4%)' were considered as main risk factors. The longer the career of the members had, the higher negative perceptions on food additives (p<0.01). About 70% of the respondents who had known KFDA's researches on evaluating safety of food additives answered that researches were not enough to make sure the safety. Although lectures and other domestic organization were the main channels for getting information, 'TV' was considered as the most suitable way for it. 'Poor risk communication (32.4%)' was pointed out as the serious problem of government's food safety policy. Based on these results, consumer organizations' low level of trust on government should be overcome by continuous supply of information what they need to facilitate risk communication on food additives.

The Additional Effects of Various Materials on Microwave Heating Property of Frozen Dough (품질개량제 첨가가 냉동반죽의 Microwave 가열특성에 미치는 영향)

  • This study was conducted to improve the properties of frozen dough foods (buns and noodles etc.) on the quality deterioration with microwave oven cooking. Microwave is a useful cooking method, but it quickly takes moisture from food surface and makes lowering food quality abruptly. For improvement of these problems, mixing doughs with addition of various additives of 34 types manufactured respectively; starches, modified starches, gums and emulsifiers etc. Each mixing dough produced in sheet type $(30{\times}30{\times}1mm)$ and steamed them, was quickly froze at $-70^{\circ}C$ and packed with polyethylene. Packed samples kept at $-20^{\circ}C$ for 48 hours. After they were steam or microwave treatment packed or non-packed with polyethylene, studied for improvement effects of quality as sensory evaluation and selected 6 type additives; modified starches (TA, ST), gums (AR, GA) and emulsifiers (E, S1) as improvement agent. Because moisture loss from microwave oven cooking leads to quality deterioration of frozen dough foods, additive, such as including starches, modified starch, gums, and emusifiers were added to improve dough properties. Amylogram, scanning electron microscopy, textural analysis, and differential scanning calorimetry revealed addition of additives improved textural properties including surface-hardening of frozen dough foods compared to the control.

Operating Status and Improvement Plans of Ten Wetlands Constructed in Dam Reservoirs in Korea (국내 10개 댐저수지 인공습지의 운영현황 및 개선방안)

  • To propose the improvement and management plans to strengthen the pollutant removal efficiency of dam reservoir's constructed wetlands(CWs), the operation status and configuration of CWs (including water depth, operational flow, water flow distribution, residence time, and pollutant removal efficiency, aspect ratio, open water/vegetation ratio etc.) were analyzed in 10 major wetlands constructed in dam reservoirs. The pollutant concentrations in the inflows of the studied CWs were lower than those of American and European constructed wetlands. Especially, organic matter concentrations in all of inflows were below 3 mg/L(as BOD) due to advanced treatment of sewage disposal plant and an intake of low concentration water during dry and normal seasons. The average removal efficiency of total nitrogen(TN) and total phosphorus(TP) for 10 CWs ranged from 7.6~67.6%(mean 24.9%) and -4.9~74.5%(mean 23.7%), respectively, showing high in wetlands treating municipal wastewater. On the other hand, the removal efficiency of BOD was generally low or negative with ranging from -133.3 to 41.7%. From the analysis of the operation status and configuration of CWs, it is suggested that the low removal efficiency of dam reservoir's CWs were caused by both structural (inappropriate aspect ratio, excessive open water area) and operational (neglecting water-level management, lack of facilities and operation for first flush treatment, lake of monitoring during rainy events) problems. Therefore, to enable to play a role as a reduction facility of non-point source(NPS) pollutants, an appropriate design and operation manuals for dam reservoir's CW is urgently needed. In addition, the monitoring during rainy events, when NPS runoff occur, must be included in operation manual of CW, and then the data obtained from the monitoring is considered in estimation of the pollutant removal efficiency by dam reservoir's CW.

Variation of Inflow Density Currents with Different Flood Magnitude in Daecheong Reservoir (홍수 규모별 대청호에 유입하는 하천 밀도류의 특성 변화)

  • Stream inflows induced by flood runoffs have a higher density than the ambient reservoir water because of a lower water temperature and elevated suspended sediment(SS) concentration. As the propagation of density currents that formed by density difference between inflow and ambient water affects reservoir water quality and ecosystem, an understanding of reservoir density current is essential for an optimization of filed monitoring, analysis and forecast of SS and nutrient transport, and their proper management and control. This study was aimed to quantify the characteristics of inflow density current including plunge depth($d_p$) and distance($X_p$), separation depth($d_s$), interflow thickness($h_i$), arrival time to dam($t_a$), reduction ratio(${\beta}$) of SS contained stream inflow for different flood magnitude in Daecheong Reservoir with a validated two-dimensional(2D) numerical model. 10 different flood scenarios corresponding to inflow densimetric Froude number($Fr_i$) range from 0.920 to 9.205 were set up based on the hydrograph obtained from June 13 to July 3, 2004. A fully developed stratification condition was assumed as an initial water temperature profile. Higher $Fr_i$(inertia-to-buoyancy ratio) resulted in a greater $d_p,\;X_p,\;d_s,\;h_i$, and faster propagation of interflow, while the effect of reservoir geometry on these characteristics was significant. The Hebbert equation that estimates $d_p$ assuming steady-state flow condition with triangular cross section substantially over-estimated the $d_p$ because it does not consider the spatial variation of reservoir geometry and water surface changes during flood events. The ${\beta}$ values between inflow and dam sites were decreased as $Fr_i$ increased, but reversed after $Fr_i$>9.0 because of turbulent mixing effect. The results provides a practical and effective prediction measures for reservoir operators to first capture the behavior of turbidity inflow.

Evaluation of near-realtime weekly root-zone Soil Moisture Index (SMI) for the extreme climate monitoring web-service across East Asia (동아시아 이상기후 감시 서비스를 위한 지면모형 기반 준실시간 토양수분지수평가)

  • An extreme climate monitoring is essential to the reduction of socioeconomic damages from extreme events. The objective of this study was to produce the near-realtime weekly root-zone Soil Moisture Index (SMI) on the basis of soil moisture using the Noah 3.3 Land Surface Model (LSM) for potentially monitoring extreme drought events. The Yangtze basin was selected to evaluate the Noah LSM performance for the East Asia region (15-60°N, 70-150°E) and the evapotranspiration (ET) and sensible heat flux (SH) were compared with ET and SH from FluxNet and with ET from FluxCom, Global Land Evaporation Amsterdam Model (GLEAM), ERA-5, and Generalized Complementary Relationship (GCR). For the ET, the coefficients of determination (R2) were higher than 0.96, while the R2 value for the SH was 0.71 with slightly lower than those. A time series of the weekly root-zone SMI revealed that the regions with Extreme drought had been expanded from the northern part of East China to the entire East China between July to October 2019. The trend analysis of the number of extreme drought events showed that extreme drought events in spring had reduced in South Korea over the past 20 years, while those in fall had a tendency to increase. It is concluded that this study can be useful to reduce the socioeconomic damages resulted from climate extremes by comprehensively characterizing extreme drought events.
