Effects of Red Light on $\alpha$-Amylase Isozmyes of the Germinated Barley (Hordeum distichum L.) (맥아의 $\alpha$-Amylase Isozyme에 미치는 Red Light의 영향)

  • This study carried out to changes $\alpha$-amylase activity and isozymes in barley during germination in the dark and red light The specific activity of u-amylase was increased during the germination periods in the dark, giving 355.0 and 523.7 units/mg protein at 3 and 5 day, and the activity was increased by the red light up to 48 and 15% at 3 and 5 days of germination, respectively. The ratio of $\alpha$-amylase I and II was approximately 95:5 at both 3 and 5 days of germination in the dark while the different ratio was found by the red light i. e. 60 : 40 and 90 : 10 at 3 and 5 days of germination, respectively.

M Phase-Specific Phosphorylation of DNA Topoisomerase IIα in HeLa Cells

  • Using topoisomerase II (topo II) isozyme-specific antibodies, we investigated the phosphorylation of topo $II{\alpha}$ in mitotic HeLa cells. Topo $II{\alpha}$ was specifically modified in the mitotic cells, resulting in slow migration on SDS-polyacrylamide gel electrophoresis. To characterize the nature of this modification, we treated the nuclear extracts prepared from the mitotic cells with alkaline phosphatase. After the treatment with alkaline phosphatase, the slowly migrated band disappeared and instead a normal (170 kDa) topo $II{\alpha}$ band appeared. These results indicate that human topo $II{\alpha}$ is modified at a specific site(s) in M phase by phosphorylation, supporting the possibility that M phase-specific phosphorylation of topo II is critical for mitotic chromosome condensation and segregation.

Differential Expression of Three Catalase Genes in the Small Radish (Rhaphanus sativus L. var. sativus)

  • Three catalase cDNA clones were isolated from the small radish (Raphanus sativus L.). Their nucleotide and deduced amino acid sequences showed the greatest homology to those of Arabidopsis. Genomic Southern blot analysis, using RsCat1 cDNA as a probe, showed that catalases are encoded by small multigene family in the small radish. Nondenaturing polyacrylamide gels revealed the presence of several catalase isozymes, the levels of which varied among the organs examined. The isozyme activities were assigned the individual catalase genes by Northern analysis using total RNA from different organs. The three catalase genes were differentially expressed in response to treatments such as white light, xenobiotics, osmoticum, and UV. Their expression in seedlings was controlled by the circadian clock under a light/dark cycle and/or in constant light. Interestingly, RsCat1 transcripts peaked in the morning, while those of RsCat2 and RsCat3 peaked in the early evening. Our results suggest that the RsCat enzymes are involved in defense against the oxidative stress induced by environmental changes.

  • Lipopolysaccharide (LPS) endotoxin is an active component in the outer membrane of Gram-negative bacteria. LPS is usually used as an inflammatory animal model. During the inflammation, diarrhea and changes in plasma proteins, in hepatic and/or intestinal microsomal cytochrome P450 (CYP) isozymes, and in the renal and/or biliary excretion of drugs have been reported. Thus, in rats pretreated with lipopolysaccharide endotoxin isolated from Klebsiella pneumoniae (KPLPS rats), the absorption, distribution, metabolism, and excretion of drugs could be expected to be altered. Interestingly time-dependent effects on the hepatic CYP isozymes have been reported in KPLPS rats. Thus, in KPLPS rats, the pharmacokinetics of drugs which are mainly metabolized via CYP isozymes could be expected to be time-dependent. In this review, an attempt to explain changes in pharmacokinetics of drug reported in the literature was made in terms of CYP isozyme changes or urinary and/or biliary excretion changes in KPLPS rats.

Purification and Characterization of the Catabolic α-Acetolactate Synthase from Serratia marcescens

  • The catabolic ${\alpha}$-acetolactate synthase was purified to homogeneity from Serratia marcescens ATCC 25419 using ammonium sulfate fractionation, DEAE-Sepharose, Phenyl-Sepharose, and Hydroxylapatite column chromatography. The native molecular weight of the enzyme was approximately 150 kDa and composed of two identical subunits with molecular weights of 64 kDa each. The N-terminal amino acid sequence of the enzyme was determined to be Ala-Gln-Glu-Lys-Thr-Gly-Asn-Asp-Trp-Gln-His-Gly-Ala-Asp-Leu-Val-Val-Lys-Asn-Leu. It was not inhibited by the branched chain amino acids and sulfometuron methyl herbicide. The optimum pH of the enzyme was around pH 5.5 and the pI value was 6.1. The catabolic ${\alpha}$-acetolactate synthase showed weak immunological relationships with recombinant tobacco ALS, barley ALS, and the valine-sensitive ALS isozyme from Serratia marcescens.

High Light-Induced Changes in the Activities of Antioxidant Enzymes and the Accumulation of Astaxanthin in the Green Alga Haematococcus pluvialis

  • We investigated high light-induced alterations in antioxidant enzymes by exposing green vegetative cells of the alga Haematococcus pluvialis to excess irradiance to induce the production of astaxanthin, a carotenoid pigment. Total activity of catalase decreased approximately 70% after high light exposure, whereas glutathione peroxidase (GPX) activity was slightly enhanced. Total activity of superoxide dismutase and ascorbate peroxidase (APX) also slightly decreased. Overall, we did not observe dramatically elevated levels of antioxidant isozymes, although APXn, GPX2, and GPX3 isozyme increased slightly. ${H_2}{O_2}$ content increased about sixfold after high light exposure, demonstrating severe cellular oxidative stress, whereas lipid peroxidation was notably reduced. Concomitantly, astaxanthin accumulation increased about sevenfold. This result suggests that probably massively accumulated astaxanthin may be one of the antioxidant protector against high light stress.

  • Taxonomic relations among three species of some edible fleshy fungi(Lentinus edodes, Pleurotus ostreatus, Flammulina velutipes) in the family Tricholomataceae were examined by using polyacrylamied gel disc electrophoresis. The soluble, crude extract of mycelium grown on potato sucrose broth was subjected to electrophoresis. Similarities in the protein bands for each isolated of one species were compared with those for others. In the banding patterns there was a closer relationship between isolates within one species than among isolates of different species. However, the isozyme patterns obtained from each isolate of Peurotus ostreatus (esterase, peroxidase, tyrosinase) were appeared to represent the degree of geographical variability within one species.

  • 한국산 다슬기류의 종별 유전적 변이를 알아보고 계통분류학적 근거를 제공하기 위하여 다슬기속(Simisulcospira)에 속하는 다슬기(S. libertina),주름다슬기,(S. forticosta), 곳체다슬기(S. gottschei), 좀주름다슬기(S. tegulata)및 참다슬기(S. coreana)등 5종을 전국 각지에서 채집하여 전기영동(starch gelelectrophoresis)를 이용해 분자유전학적 분석을 실시한 결과를 용약하면 아래와 같다. 유전적 다형형의 빈도는 58%에서 83%로 대단히 높았고 이형접합자의 평균 빈도도 50%내외였다. Rogers' D를 근거로 한 각 종간 유연관계는 주름다슬기와 곳체다슬기가 근연관계를 이루고 있었으며 이들 두 군과 다슬기는 유전적으로 거리를 두고 있었다. 이러한 결과는 패각의 형채와 비교할 때 유사한 경향을 보였다.

  • The effect of tannic acid on GAs and cyclic-AMP promoted amylase induction in barley aleurone layers was examined. Of a variety of adenine compounds, only cyclic-AMP and ADP showed significant activity, and these activities were promoted by addition of theophylline to the incubation medium. When aleurone layers of barley endosperm tissues were incubated with GAs in the presence of tannic acid, the amylase activity in the incubation medium was reduced. Cyclic-AMP induced amylase activity was also reduced by additiion of tannic acid. The cyclic-AMP response promoted was more sensitive to tannin inhibition than GAs response. The inhibitory effect of tannic acid shwoed reversibility by addition of higher concentration of GAs or cyclic-AMP. The tannic acid effect on GAs response was also recovered by addition of a higher concentration of cyclic-AMP. Experiment with polyacrylamide disc electrophoresis showed different isozyme patterns according to the additions in the incubation medium. Inhibitory effects of decursinol and coumarin was compared with that of tannic acid. They showed different zymogram patterns.
