Antioxidant Activity and Phytochemical Study on the Aerial Parts of Oenanthe javanica (미나리 지상부에서 라디칼 소거 활성을 가지는 페놀성 화합물의 분리)

  • Five compounds were isolated from the methanol extract of the aerial parts of Oenanthe javanica(Umbelliferae). Their chemical structures were identified as isorhamnetin(1), uracil(2), adenosine(3), persicarin(4) and uridine(5) by comparison of their spectral data with those of authentic samples and compounds 2, 3 and 5 were isolated from Oenanthe javanica for the first time. Compound 1 showed the strong radical scavenging activity among the compounds isolated from Oenanthe javanica.

  • The antioxidant activity of Persicaria thunberii (Polygonaceae) was determined by measuring the radical scavenging effect on 1,1-diphenyl-2-pocrylhydrazyl (DPPH) radical. The methanol extract of thunbergii showed strong radical scavenging activity at an I $C_{50}$ concentration of 12.2$\mu\textrm{g}$/ml, and thus fractionated by solvent extraction. The EtOAc soluble fraction was stronger than the others, and was further purified by silica gel and Sephadex LH-20 column chromatography, Antioxidant isorhamnetin, quercetin (2), quercitrin (3) and isoquercitrin (4) were isolated from the EtOAc soluble fraction. And isorhamnetin-3-sulfate (5) was isolated from the n-BuOH fraction. Compounds 2, 3 and 4 were found to have strong antioxidative potency.

New Flavonol Glycosides from Leaves of Symplocarpus renifolius

  • A study was carried out to evaluate flavonol glycosides in leaves of Symplocarpus renifolius (Araceae). From the water fraction of the MeOH extract, three new flavonol glycosides were isolated along with three known compounds, Kaempferol-3-O-$\beta$-glucopyranosyl-($1{\rightarrow}2$)-$\beta$-D-glucopyranosyl-7-O-$\beta$-D-glucopyranoside, quercetin-3-O-$\beta$-D-glucopyranosy-1-($1{\rightarrow}2$)-$\beta$-D-glucopyranoside, and caffeic acid. The structures of the new flavonol glycosides were elucidated by chemical and spectral analyses a quercetin-3-O-$\beta$-D-glucopyranosyl-($1{\rightarrow}2$)-$\beta$-D-glucopyranosyl-7-O-$\beta$-D-glucopyranoside, isorhamnetin-3-O-$\beta$-D-glucopyranosyl-(1 2)-$\beta$-D-glucopyranosyl-7-O-$\beta$-D-glucopyranosdie, and quercetin-3-O$\beta$-D-glucopyranosyl-($1{\rightarrow}2$)-$\beta$-D-glycopyranosyl-7-O-($6^{IIII}$-trans-caffeoyl)-$\beta$-D-glucopyranoside.

  • From the dried flower-buds of Syzygium aromaticum Merr. et Perry (Myrtaceae), seven compounds, i.e., eugenol (1), oleanolic acid (2), kaempferol 7-O-metylether (3), 3,3',4-tri-O-methylellagic acid (4), maslinic acid (5), ${\beta}-sitosterol-3-O-glucoside$ (6), and isorhamnetin 3-O-glucoside (7) were isolated. Compound 1 showed cyclooxygenase-2 (COX-2) inhibitory activity.

Effects of the Components and Extracts of Some Edible and Medicinal Plants on the Formation of Lipid Peroxide in Rat Liver Homogenate (식용 및 약용식물 추출물과 화합물이 흰쥐의 간장조직에서 과산화지질 생성에 미치는 영향)

  • We have studied the effect of 31 extracts and 10 components from some edible and medicinal plants on the formation of lipid peroxide in the liver homogenate of rat in vitro. The 70% acetone extracts of Allium tuberosum, Beta vulgaris var. cicla and Brassica juncea var. integrifolia, and methanol extract of Capsicum annuum decreased the formation of lipid peroxide by 33%, 58%, 62% and 56% at the concentration of 1mg/ml, respectively. And these four extracts inhibited the lipid peroxidation at the concentration of $10^{-1}$mg/ml by 17%, 46%, 49% and 45% respectively. Among the component tested, quercetin, quercitrin and isorhamnetin reduced the formation of lipid peroxide by 45%, 15% and 28% respectively at the concentration of $10^{-2}$mg/ml.

Anti-influenza Compounds Isolated from Descurainia sophia Seeds

  • Descurainia sophia seeds methanol extract showed significant anti-influenza activity and we tried to isolate anti-influenza compounds from the D. sophia extract. D. sophia seeds were extracted with 80% methanol and fractionated with n-hexane, ethyl acetate, CHCl3 and n-butanol. The anti-influenza activity of each fraction was assessed using sulforhodamine B (SRB) method in A549 cells, human-derived lung cancer cells. The ethyl acetate and CHCl3 fractions showed the most potent anti-influenza activity. Seven compounds were isolated from CHCl3 fraction and identified 1-decanol (1), 2-(3,4-dihydroxy-2-methylenebutoxy)-6-(hydroxymethyl)tetrahydro-2H-pyran-3,4,5-triol (2), daucosterol (3), isorhamnetin (4), quercetin (5), sinapic acid (6), and helveticoside (7) by spectroscopic data such as UV, IR, 1H-NMR, 13C-NMR and mass spectroscopy. Anti-influenza activities of isolated compounds were evaluated using SRB method in A549 cells. Compounds 3, 4 and 7 had significant anti-influenza activity in a dose-dependent manner.

Antioxidant Activity and Inhibition of MMP-9 by Isorhamnetin and Quercetin 3-O-$\beta$-D-Glucopyranosides Isolated from Salicornia herbacea in HT1080 Cells

  • Two flavonoids, isorhamnetin 3-O-$\beta$-D-glucopyranoside (1) and quercetin 3-O-$\beta$-D-glucopyranoside (2), from slander glasswort (Salicornia herbacea, Korean name hamcho) were isolated. Antioxidative and matrix metalloproteinase-9(MMP-9) inhibitory effects of these compounds were investigated in HT 1080 cell lines. These compounds suppressed the electron spin resonance (ESR) signal intensity on generation of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical in a free-cellular system. Their scavenging effects on generation of intercellular reactive oxygen species (ROS) also exhibited similar trends with DPPH radical in the free cellular system. Also, a control group combined only with Fe(II)-$H_{2}O_2$ resulted in DNA apoptosis by oxidative stress, whereas treatments with these compounds suppressed radical-mediated DNA damage. Intracellular glutathione (GSH) levels were slightly increased in the presence of compound 1 and 2. Moreover, these compounds led to the reduction of the expression levels of MMP-9 without cytotoxic influence. These results suggest that these compounds have a potential as a valuable natural antioxidant and MMP inhibitor related to oxidative stress. Therefore, these compounds not only can be developed as a candidate for a therapeutic potential but also a source for use as ingredients of health foods or functional foods to prevent metastasis involving MMP-9, closely related to ROS.

Antioxidant Activities of the Ethanol Extract of Hamcho (Salicornia herbacea L.) Cake Prepared by Enzymatic Treatment

  • The antioxidant activities of water ($H_2O$) and ethanol (EtOH) extracts from hamcho (Salicornia herbacea L.) juice and cake prepared by enzymatic treatments were evaluated by in vitro assays against DPPH, superoxide, and hydroxyl radicals. Among the $H_2O$ and EtOH extracts from five different carbohydrases treated, the EtOH extract from viscozyme-treated hamcho cake had higher yield and phenolic content, and exhibited the strongest radical scavenging activity against DPPH ($IC_{50}=186.91\;{\mu}g/mL$), superoxide ($IC_{50}=87.54\;{\mu}g/mL$), and hydroxyl radicals ($IC_{50}=367.07\;{\mu}g/mL$). Antioxidant assay-guided fractionation and purification of the EtOH extract led to isolation and identification of five phenolic compounds, procatechuic, ferulic and caffeic acids, quercetin, and isorhamnetin. Most of these phenolic compounds exhibited considerable DPPH, superoxide, and hydroxyl radical scavenging activities, and in particular, caffeic and ferulic acids had stronger superoxide and hydroxyl radical scavenging activities than the well-known antioxidant radical scavenger, (+)-catechin (p<0.05). Quercetin and isorhamnetin were the primary compounds responsible for the strong antioxidant activity in the EtOH extract of the viscozyme-treated hamcho cake. Meanwhile, these five phenolic compounds were detected in the EtOH extract of the viscozyme-treated hamcho cake at the following levels (dry base of hamcho); procatechuic acid (1.54 mg%), caffeic acid (6.87 mg%), ferulic acid (8.45 mg%), quercetin (12.63 mg%), and isorhamnetin (6.65 mg%). However, three of these phenolic compounds (procatechuic, caffeic acid, and ferulic acids) were detectable in the $H_2O$ extract of viscozyme-treated hamcho juice. These results suggest that the EtOH extract of viscozyme-treated hamcho cake may be a potential source of natural antioxidants.

Isolation and Identification of Flavonoids from Ethanol Extracts of Artemisia vulgaris and Their Antioxidant Activity (쑥의 에탄올 추출물에 함유된 Flavonoid들의 분리 및 동정과 이들의 항산화 효과)

  • Twenty one flavonoids were isolated from ethyl acetate layer of aqueaus EtOH extracts of Artemisia vulgaris and identified as tricin, jaceosidine, eupafolin, diosmetin, chrysoeriol, homoeriodictyol, isorhamnetin, apigenin, eriodictyol, luteolin, luteolin 7-glucoside, kaempferol 3-glucoside, kaempferol 7-glucoside, kaempferol 3-rhamnoside, kaempferol 3-rutinside, quercetin, quercetin 3-glucoside, quercetin 3-galactoside, quercetrin, quercetin 7-glucoside, rutin, and vietexin. The inhibitory activity for all purified flavonoids were examined against lipid peroxidation in rat liver microsome. All examined flavonoids showed considerable antioxidant activity. Among them, $IC_{50}$ value of apigenin, luteolin, isorhamnetin, quercetin, and eriodictyol were showed higher than that of vitamin E used as positive control. And methoxylated flavonoids, tricin, eupafolin, jaceosidine, diosmetin, and isorhamnetin showed considerable antioxidant activity. Each $IC_{50}$ values were shown at 0.9, 1.0, 1.4, 1.0, and $0.7\;{\mu}g/mL$, respectively.

Heptatriacontanol and Phenolic Compounds from Halochris hispida

  • The phytochemical investigation of Halocharis hispida revealed the presence of 1-heptatriacontanol, ${\beta}-sitosterol$, ${\beta}-sitosterol-3-O-glucoside$, kaempferol, vitexin and isorhamnetin-3-O-galactoside in addition to vanillic, ferulic, isoferulic, syringic and caffeic acids. The different isolated compounds were identified by different physical, chemical, chromatographic and/or spectral methods.
