Selective Medium for Isolation and Enumeration of Eubacterium sp.from the Feces of the Korean People (한국인의 분변으로부터 Eubacterium을 분리하기 위한 선택 배지 조사)

  • Eubacterium is one of the predominant bacteria in the human large intestine. currently ES (Eubacterium Selective) medium developed by T. Mitsuoka is commonly used as a selective medium. neomycin sulfate which is one of the selective agents of ES medium inhibited about 50% of the growth of Eubacterium isolated, whereas malidixic acid inhibited only 5% while inhibiting other intestinal bacteria. NES medium which replaced neomycin with nalidixic acid in the ES medium was designed and shown to be better for the isolation and enumeration of Eubacterium sp. than ES medium.

A Study on the Salmonella spp. isolation method in the sewage specimens (하수중의 Salmonella spp. 분리방법에 관한 비교연구)

  • This study was carried out in order to compare the methods for Salmonella spp. isolation from sewage and to settle the most appropriate conditions for that isolation. The direct enrichment method was more effective than the pre- enrichment method for Salmonella spp. isolation. The rate of detection was much higher when the specimens were enriched at $41.5^{\circ}C$ than when at $35^{\circ}C$. Usage of XLBG agar medium showed better results for Salmonella isolation than that of SS medium. It can be suggested that the most effective combination for Salmonella spp. isolation was the direct centrifugation(3,000 rpm 100m1)- direct enrichment($41.5^{\circ}$C)-usage of XLBG medium.

  • Seismic isolation technique has been applied mainly in low rise buildings and its seismic performance was satisfactory during Kobe Earthquake. However, in the case of medium and/or high-rise buildings, mid-story isolation could be more technically feasible than base isolation to reduce earthquake forces. In this paper, the seismic effectiveness of mid-story isolation in medium and/or high-rise shear building as well as low rise shear building was evaluated analytically. After verifying the effectiveness of mid-story isolation technique, this method also applied in residential-commercial building. It was found that mid-story isolation, that is isolation between upper residential area and lower commercial area, could reduce inter-story drift and floor shear forces comparing to the conventional fixed base.

  • For the more effective isolation of soil actinomycetes, we have developed HHV (Hair hydrolysate-vitamin) agar medium, containing hair as the sole source of carbon and nitrogen. The HHV agar medium was superior to other media such as colloidal chintin agar, glycerol-arginine agar and starch-casein-nitrate agar, and HV (humic acid-vitamin) agar. The maximum effect of this medium has been shown in hair dry weight 0.4 g/l medium. Of each soil sample, the greatestest number of actinomycetes was isolated from the potato annual planted soil among the tested samp- les. The genus of actinomycetes isolated from the potato annual planted soil sample was identified such 5 group as Stretomyces, Micromonospora, Microbispora, Nocardia and Saccharopolyspora.

Construction of Probability Identification Matrix and Selective Medium for Acidophilic Actinomycetes Using Numerical Classification Data

  • A probability identification matrix of acidophilic Streptomyces was constructed. The phenetic data of the strains were derived from numerical classification described by Seong et al. The minimum number of diagnostic characters was determined using computer programs for calculation of different separation indices. The resulting matrix consisted of 25 clusters versus 53 characters. Theoretical evaluation of this matrix was achieved by estimating the chuster overlap and the identification scores for the Hypothetical Median Organisms (HMO) and for the representatives of each cluster. Cluster overlap was found to be relatively small. Identification scores for the HMO and the randomly selected representatives of each cluster were satisfactory. The matrix was assessed practically by applying the matrix to the identification of unknown isolates. Of the unknown isolates, 71.9% were clearly identified to one of eight clusters. The numerical classification data was also used to design a selective isolation medium for antibiotic-producing organisms. Four chemical substances including 2 antibiotics were determined by the DLACHAR program as diagnostic for the isolation of target organisms which have antimicrobial activity against Micrococcus luteus. It was possible to detect the increased rate of selective isolation on the synthesized medium. Theresults show that the numerical phenetic data can be applied to a variety of purposes, such as construction of identification matrix and selective isolation medium for acidophilic antinomycetes.

Study on Selective Media for Isolation of Entomopathogenic Fungi

  • To determine the optimal isolation conditions of the entomopathogenic fungi from soil, we compared their growth characteristics with non-entomopathogenic fungi on agar media containing various concentrations of cooper (II) chloride ($CuCl_2$) or dodine. The result showed that dodine medium is more selective, and the optimal concentration of dodine is determined with $50{\mu}g$/ml. We could isolate several putative entomopathogenic fungi from soil using this, and identify them using ITS analysis. As a result, 64% fungi were identified as typical entomopathogenic fungi. This selective medium may be useful to the rapid and simple isolation of entomopathogenic fungi from soil.

  • Three different isolation media for Cronobacter sakazakii have been recommended by Korea Food and Drug Administration from 2007. Performance comparison test was carried out between recommended Cronobacter sakazakii isolation medium. Chromogenic Enterobacter sakazakii agar (CESA) and Enterobacter sakazakii agar (ESA) produce more distinctive colonies having characteristic colors and appearance than Violet red bile glucose agar (VRBGA). The sensitivity and specificity of 3 different isolation media was checked. All 3 tested media showed 100% sensitivity when tested with 30 different Cronobacter sakazakii. The CESA and ESA showed 100% specificity when tested with Enterobacteriaceae except Cronobacter sakazakii, However, VRBGA did not show any specificity, showing inadequate selectivity compared to applicable Cronobacter sakazakii isolation medium. Artificially inoculated Cronobacter sakazakii to milk powder was easily recovered with 3 different isolation media and they all showed almost the same recovery activity.

A Semiselective Medium for the Isolation of Xanthomonas campestris pv. oryzae from Rice Seed (벼 종자에서 Xanthomonas campestris pv. oryzae의 분리를 위한 선택배지)

  • A semiselective agar medium (XCO) was developed for the isolation of bacterial blight pathogen, Xanthomonas campestris pv. oryzae, from rice seed. The medium contained yeast extract 1.0 g, peptone 2.0 g, sucrose 5.0 g, sodium glutamate 1.0 g, FeSO4.7H2O 0.05 g, Fe.EDTA 1 mg, cephalexin 20 mg, Evan blue (0.1%) 1.5 ml, bromcresol purple (0.1%) 2.5ml, cycloheximide 100 mg and agar 15.0 g per liter. Colonies of X. c. pv. oryzae were 4~5 mm in diameter, smooth, round, blue (darker center) and convex after 6 days incubation at 28$^{\circ}C$. The recovery of 6 isolates of X. c. pv. oryzae on the XOC medium ranged from 81% to 120% (mean 98.2%) in comparison to Wakimoto's medium. The number of saprophytic bacteria from 10 rice seed lots on XCO medium was reduced to 70.4% of that on Wakimoto's medium. The recovery of X. c. pv. oryzae added to rice seed on XOC medium ranged from 67% to 87% (mean 75.6%) of that on Wakimoto's medium.

Deproteinized Mulberry Leaf Juice - A New Media for Growth of Microorganisms

  • Mulberry being a foliage crop is grown extensively for feeding of silkworms and are also used for cattle feeding. These loaves are highly nutritious, which contain various mineral elements and bio-molecules such as carbohydrates, proteins, lipids and other essential amino acids, etc. In the present study, deproteinized mulberry leaf juice was used for preparation of the medium for cultivation of various types of microbes. Results revealed that deproteinized mulberry leaf juice medium is best for isolation of fungi, bacteria and actinomycetes and this medium can be substituted with synthetic media, which are haying the costly ingredients for isolation and identification of bacteria, fungi and actinomycetes. Further, this deproteinized mulberry juice medium can also be used in mass multiplication of useful/beneficial microbes to enhance soil microflora to improve soil fertility and to avoid root diseases. Perspective enterprises can take up the mass multiplication/large-scale production of useful microbes such as Trichoderma, Rhizobium, Pseudomonas and Bacillus to use in mulberry and in other agricultural crops using deproteinized mulberry leaf juice.

  • The scale and nature of the recent earthquakes in the world and the related earthquake disaster index coerce the concerned community to become anxious about it. Therefore, it is crucial that seismic lateral load effect will be appropriately considered in structural design. Application of seismic isolation system stands as a consistent alternative against this hazard. The objective of the study is to evaluate the structural and economic feasibility of reinforced concrete (RC) buildings with base isolation located in medium risk seismic region. Linear and nonlinear dynamic analyses as well as linear static analysis under site-specific bi-directional seismic excitation have been carried out for both fixed based (FB) and base isolated (BI) buildings in the present study. The superstructure and base of buildings are modeled in a 3D finite element model by consistent mass approach having six degrees of freedom at each node. The floor slabs are simulated as rigid diaphragms. Lead rubber bearing (LRB) and High damping rubber bearing (HDRB) are used as isolation device. Change of structural behaviors and savings in construction costing are evaluated. The study shows that for low to medium rise buildings, isolators can reduce muscular amount of base shears, base moments and floor accelerations for building at soft to medium stiff soil. Allowable higher horizontal displacement induces structural flexibility. Though incorporating isolator increases the outlay, overall structural cost may be reduced. The application of base isolation system confirms a potential to be used as a viable solution in economic building design.
